Thiol-dependent Redox Mechanisms in the Modification of ATP-Sensitive Potassium Channels in Rabbit Ventricular Myocytes

  • Cellular redox state is known to be perturbed during ischemia and that $Ca^{2+}$ and $K^2$ channels have been shown to have functional thiol groups. In this study, the properties of thiol redox modulation of the ATP-sensitive $K^2$ ($K_{ATP}$) channel were examined in rabbit ventricular myocytes. Rabbit ventricular myocytes were isolated using a Langendorff column for coronary perfusion and collagenase. Single-channel currents were measured in excised membrane patch configuration of patch-clamp technique. The thiol oxidizing agent 5,5'-dithio-bis-(2-nitro-benzoic acid) (DTNB) inhibited the channel activity, and the inhibitory effect of DTNB was reversed by dithiothreitol (disulfide reducing agent; DTT). DTT itself did not have any effect on the channel activity. However, in the patches excised from the metabolically compromised cells, DTT increased the channel activity. DTT had no effect on the inhibitory action by ATP, showing that thiol oxidation was not involved in the blocking mechanism of ATP. There were no statistical difference in the single channel conductance for the oxidized and reduced states of the channel. Analysis of the open and closed time distributions showed that DTNB had no effect on open and closed time distributions shorter than 4 ms. On the other hand, DTNB decreased the life time of bursts and increased the interburst interval. N-ethylmaleimide (NEM), a substance that reacts with thiol groups of cystein residues in proteins, induced irreversible closure of the channel. The thiol oxidizing agents (DTNB, NEM) inhibited of the $K_{ATP}$ channel only, when added to the cytoplasmic side. The results suggested that metabolism-induced changes in the thiol redox can also modulate $K_{ATP}$ channel activity and that a modulatory site of thiol redox may be located on the cytoplasmic side of the $K_{ATP}$ channel in rabbit ventricular myocytes.

Unpolished Thai Rice Prevents ACF Formation and Dysplastic Progression in AOM-Induced Rats and Induces Apoptosis Through Redox Alteration in CaCo-2 Cells

  • Oxidative stress is associated with colon carcinogenesis including aberrant crypt foci (ACF) formation and it plays an important role in pathophysiological changes in cancer cells. The aims of this study were to investigate the effects of dietary unpolished Thai rice (UTR) on ACF formation and dysplastic progression in azoxymethane (AOM)-treated rats. Anti-cancer efficacy of UTR regarding apoptotic induction and oxidative redox status in human colon cancer (CaCo-2) cells was also investigated. Rats given 20% and 70% of UTR in the diet showed significantly and dose-dependently decreased total number of ACF. UTR treatment also was strongly associated with the low percentage of dysplastic progression and mucin depletion. In addition, we found that UTR significantly induced cancer cell apoptosis, increased cellular oxidants, and decreased the level of GSH/GSSG ratio in CaCo-2 cells. Our study suggests that UTR supplementation may be a useful strategy for CRC prevention with the inhibition of precancerous progression, with induction of cancer cell apoptosis through redox alteration.

Facile Electrodeposition Technique for the Fabrication of MoP Cathode for Supercapacitor Application

  • The continued environmental pollution caused by fossil fuel consumption has prompted researchers around the world to develop environmentally friendly energy technologies. Electrochemical energy storage is the significant area of research in this development process, and the research significance of supercapacitors in this field is increasing. Herein, a simple electrodeposition synthetic route was explored to develop the MoP layered cathode material. The layered structure provided a highly ion-accessible surface for smooth and faster ion adsorption/desorption. After Fe was doped into MoP, the morphology of MoP changes and the electrochemical performance was significantly improved. Specific capacitance value of the binder-free FeMoP electrode was found to be 269 F g-1 at 2 A g-1 current density in 6 M aqueous KOH electrolyte. After adding Fe to MoP, an additional redox contribution was observed in the redox conversion from Fe3+ to Fe2+ redox pair, and the charge transfer kinetics of MoP was effectively improved. This research can provide guidance for the development of supercapacitor electrode materials through simple electrodeposition technology.

Development of a Redox Dye-Based Rapid Colorimetric Assay for the Quantitation of Viability/Mortality of Pine Wilt Nematode

  • Control of pine wilt disease, which is caused by pine wilt nematode Bursaphelenchus xylophilus, is heavily dependent on the use of chemicals such as abamectin. Although such chemicals are highly effective, demands for alternatives that are derived preferentially from natural sources, are increasing out of environmental concerns. One of the challenges to discovery of alternative control agents is lack of fast and efficient screening method that can be used in a high-throughput manner. Here we described the development of colorimetric assay for the rapid and accurate screening of candidate nematicidal compounds/biologics targeting B. xylophilus. Contrary to the conventional method, which relies on laborious visual inspection and counting of nematode population under microscope, our method utilizes a redox dye that changes its color in response to metabolic activity of nematode population in a given sample. In this work, we optimized parameters of our colorimetric assay including number of nematodes and amount of redox dye, and tested applicability of our assay for screening of chemicals and biologics. We demonstrated that our colorimetric assay can be applied to rapid and accurate quantification of nematode viability/mortality in a nematode population treated with candidate chemicals/biologics. Application of our method would facilitate high-throughput endeavors aiming at finding environment-friendly control agents for deadly disease of pine trees.

Chemical and Electrochemical Synthesis of Highly Conductive and Processable PolyProDOP-alkyl Derivatives

  • New monomers, possessing various alkyl substituents on propylene dioxypyrrole, were synthesized. The monomers could be easily polymerized to produce highly conductive and soluble polymers. The corresponding polymers showed excellent solubility, retaining electrochemical and optical properties of their parent polymer [poly(propylene dioxypyrrole)]. The conductivities of chemically prepared polymers were quite high in a range of 20 and $60\;Scm^{-1}$. Solubility of the polymer in a common organic solvent was as high as no polymer is deposited on an electrode. The redox potentials of the electrochemically prepared polymers revealed quite stable electro-activity during repeated redox switching up to 500 times. The optoelectrochemistry studies also showed distinct color changes of the polymers upon changing the doping state, indicating strong absorption peaks at 400~600 nm in reduced states and complete bleaching in fully oxidized states.

Analysis of the Changes in Metabolic Diversity of Microbial Community in pH-gradient Microcosm

  • The Biolog redox technology was carried out for evaluation of acidification effect on microbial communities at each stage of pH gradient microcosm. While the number of heterotrophic bacterial population and activities of extracellular enzyme decreased as the pH decreased, the number of total bacteria in the microcosm was not affected. The average color development of sample at each pH-gradient showed a sigmoidal curve, and at higher pH, more overall color development appeared in Biolog plates. Average color development value in Biolog plates was stabilized at 50 hours as an optimum incubation time. The color production in the Biolog plates was caused by cell density at above pH 5.0, but by cell activity below pH 4.0. Principal component analysis of color responses revealed distinctive patterns among the pH-gradient microcosm samples.

Characteristics of Groundwater Quality in a Riverbank Filtration Area (강변여과수 부지 내 충적층 지하수의 수질특성과 변화)

  • Characteristics and changes of groundwater qualify were investigated in a riverbank filtration area at Daesan-myeon, Changwon City, Korea. The total dissolved solids (TDS) in groundwater samples collected in October were much less than that in March, indicating the mixing with recharged water from precipitation, as well as the changes of dissolved oxygen profiles at monitoring wells from March to October. Redox processes at depths appeared to trigger Fe and Mn contamination of groundwater in riverbank deposits. Amorphous oxyhydroxides md carbonate minerals such as $MnCO_3$ were probably the reactive phases for dissolved Fe and Mn, respectively. Groundwater contamination by nitrate-nitrogen $(NO_3-N)$ was controlled by the redox processes and subsequent denitrification at the sampled depths. Distribution of $NO_3-N$ concentrations at monitoring wells suggested that the nitrate contaminants were originated from agricultural facilities on the riverbank deposits. Some of monitoring wells, DS-2, D-2, DS-3, SJ-1, and SJ-3, were only partially penetrated into the sand/gravel aquifer, and subsequently, could not fully function to detect the water quality changes for the pumping wells. Proper measures, with regulating agricultural activities in the riverbank deposits, should be carried out to prevent groundwater contamination of the riverbank filtration area.

A Study on the Electrochemical Performance of Fe-V Chloric/Sulfuric Mixed Acid Redox Flow Battery Depending on Electrode Activation Temperature (Fe-V Chloric/Sulfuric Mixed Acid 레독스흐름전지 전극의 활성화 온도에 따른 전기화학적 성능 고찰)

  • Among the components of redox flow battery (RFB), the electrode serves as a diffusion layer of an electrolyte and a path for electrons and also is a major component that directly affects the RFB performance. In this paper, chloric/sulfuric mixed acidwas used as a supporting electrolyte in RFB system with Fe2+/Fe3+ and V2+/V3+ as redox couple. The optimum electrode and activation temperature were suggested by comparing the capacity, coulombic efficiency and energy efficiency according to the electrode type and activation temperature. In the RFB single cell evaluation using 5 types of carbon electrodes used in the experiments, all of them showed close to the theoretical capacity to retain the reliability of the evaluation results. GFD4EA showed relatively excellent energy efficiency and charge/discharge capacity. In order to investigate the electrochemical performance according to the activation temperature, GFD4EA electrode was activated by heat treatment at different temperatures of 400, 450, 500, 600 and 700 ℃ under an air atmosphere. Changes in physical properties before and after the activation were observed using electrode mass retention, scanning electron microscope (SEM), XPS analysis, and electrochemical performance was compared by conducting RFB single evaluation using electrodes activated at each temperature given above.

Methylglyoxal-Scavenging Enzyme Activities Trigger Erythroascorbate Peroxidase and Cytochrome c Peroxidase in Glutathione-Depleted Candida albicans

  • γ-Glutamylcysteine synthetase (Gcs1) and glutathione reductase (Glr1) activity maintains minimal levels of cellular methylglyoxal in Candida albicans. In glutathione-depleted Δgcs1, we previously saw that NAD(H)-linked methylglyoxal oxidoreductase (Mgd1) and alcohol dehydrogenase (Adh1) are the most active methylglyoxal scavengers. With methylglyoxal accumulation, disruptants lacking MGD1 or ADH1 exhibit a poor redox state. However, there is little convincing evidence for a reciprocal relationship between methylglyoxal scavenger genes-disrupted mutants and changes in glutathione-(in)dependent redox regulation. Herein, we attempt to demonstrate a functional role for methylglyoxal scavengers, modeled on a triple disruptant (Δmgd1/Δadh1/Δgcs1), to link between antioxidative enzyme activities and their metabolites in glutathione-depleted conditions. Despite seeing elevated methylglyoxal in all of the disruptants, the result saw a decrease in pyruvate content in Δmgd1/Δadh1/Δgcs1 which was not observed in double gene-disrupted strains such as Δmgd1/Δgcs1 and Δadh1/Δgcs1. Interestingly, Δmgd1/Δadh1/Δgcs1 exhibited a significantly decrease in H2O2 and superoxide which was also unobserved in Δmgd1/Δgcs1 and Δadh1/Δgcs1. The activities of the antioxidative enzymes erythroascorbate peroxidase and cytochrome c peroxidase were noticeably higher in Δmgd1/Δadh1/Δgcs1 than in the other disruptants. Meanwhile, Glr1 activity severely diminished in Δmgd1/Δadh1/Δgcs1. Monitoring complementary gene transcripts between double gene-disrupted Δmgd1/Δgcs1 and Δadh1/Δgcs1 supported the concept of an unbalanced redox state independent of the Glr1 activity for Δmgd1/Δadh1/Δgcs1. Our data demonstrate the reciprocal use of Eapx1 and Ccp1 in the absence of both methylglyoxal scavengers; that being pivotal for viability in non-filamentous budding yeast.

Solute Transport Model with Cation Exchange under Redox Environment and its Application for Designing the Slow Infiltration Set-up

  • The present trend of disposing treated sewage water by allowing it to infiltrate the soil brings a new dimension to environmental problems. It is therefore necessary to identify the chemicals likely to be present in treated sewage water. A soil column experiment was conducted to determine the behavior of chemical species in soil columns applied with secondary treated sewage water. To predict the behavior of chemical species, a multicomponent solute transport model that includes the biochemical redox process and cation exchange process was developed. The model computes changes in concentration over time caused by the processes of advection, dispersion, biochemical reactions and cation exchange reactions. The solute transport model was able to predict the behavior of the different chemical species. The model reproduced the sequential reduction reaction. To design the safe depth of plow layer where $NO_3^-$ is totally reduced, a numerical study of $NO_3^-$ leach was done and it was found out that the pore velocity and concentration of $CH_2O$ at the inject water was found to affect $NO_3^-$ reduction in the mobile pore water phase. It is revealed that the multicomponent solute transport model is useful to design the land treatment system for $NO_3^-$ removal from wastewater.
