  • To improve the quality of environmental measurements and evaluation of the workplace air in the pigment manufacturing industries, we analyzed metal(chromium, cadmium, lead, iron, cobalt, manganese, antimony, titanium, arsenic, and selenium) concentrations by ICP-AES in sixty seven samples of inorganic bulk pigments which are produced and/or used in Korea. We also collected MSDS which has to be supplied by manufacturer and/or supplier and posted in the workplace according to the Hazard Communication Standards, and compared the number of metals listed in each MSDS with the number of metals determined by ICP-AES. Results were as followed; 1. Among seventeen yellowish-colored samples, chromium(2~19%) and lead(0.1~61%) were the two major metals. In thirteen reddish-colored samples, iron was the major component with 37~81%. Cobalt and manganese were detected in blue-colored samples with less than 1%, while antimony and titanium were the major two metals in white-colored pigments with 178~300 ppm and with 36~65%, respectively. 2. In area samples collected in workplace air(one pigments producing factory and five retailer stores), iron and manganese were detected but the concentrations not exceeded the TLVs(1 and $5mg/m^3$, respectively). In three of fifteen samples, the concentrations of lead exceeded the TLV ($0.05mg/m^3$). 3. Two out of seven companies provided MSDS, and the average provision rate was 22.4%. And the coincidence rate of the number of metals referenced in MSDS and determined by ICP-AES mostly accorded, but in one sample, different metal was detected from MSDS. In summary, metals have to be concerned in evaluation of the workplace air dealing with compounds of inorganic pigments dust are cobalt, chromium, iron, manganese, lead and antimony, and these are simultaneously determined by ICP-AES. Taking this opportunity, it is needed to reinforce that the personnel is to be concerned about prevention of workers' ill health regarding to provision of MSDS.

  • Pigments were extracted by aqueous acetone from the boiled canned oyster colored during storage and then the components were separated by thin layer chromatography. Totally eleven pigments could be isolated including one yellow, one red, five orange or reddish orange and four green components, and their UV-visible spectra were measured. It can be envisioned from the electronic spectral study and color reaction on the indivisual pigments isolated from the brown acetone extracts that the green pigments as well as most of the yellow orange ones may be porphyrin derivatives originated probably from the chlorophyll and some of the orange pigments contains ketocarotenoids. In particular, the pigment of band 8 which is expected to be pheophytin a or its derivatives and the carotenoid band 7 seem to be the major pigment. The close resemblance of the chromatogram of the colored muscle extract to that of the viscera suggests that the brown coloring material is probably originated from the viscera pigments.

  • To investigate Phomopsis species causing fruit decays of Japanese apricot, apple and kiwifruit, we collected diseased fruits from the fruit markets in 1995 and 1996 respectively. Phomopsis mali Roberts was identified based on cultural characteristics, morphological aspects and pathogenicity. There were no remarkable differences with respect to $\alpha$ and $\beta$ conidia, growth rates and colony characters among the isolates from Japanese apricot, apple and kiwifruit. The pathogens grew more than 70 mm on potato dextrose agar in 5 days at $25^{\circ}C$. The agar was slightly discolored by the production of a reddish purple pigment under the light at $25^{\circ}C$ and 3$0^{\circ}C$ respectively. Only $\alpha$ spores of the different isolates of P. mali were formed at 15$^{\circ}C$ and $\beta$ spores were mainly produced at 3$0^{\circ}C$, but and $\alpha$ and $\beta$ spores were produced in approximately equal numbers at 2$0^{\circ}C$ and $25^{\circ}C$. Pycnidia were a few under the dark condition but were abundant at wide range of 15~3$0^{\circ}C$ under near ultra violet illumination. Conidia were two types : $\alpha$ spores were unicellar, fusoid, hyaline and biguttulate, whereas $\beta$ sores were unicellar, acicular to filiform, straight or hooked and hyaline. An ascigerous stage was not formed in cultures or in nature. Isolates of Phomopsis mali from japanese apricot, apple and kiwifruit could infect fruits of apple, pear, apricot, Japanese apricot and kiwifruit. There were some differences in pathogenicity depending on stocks of fruit crops tested.

  • Studies were carried out to examine the effects of simulated acid rain on the various crops of rice, peper, tomato, pumpkin, egg plant, potato, soybean, radish, cucumber, common pea, sweet potato, corn, lettuce, chinse cabbage, spinach, under the condition of the rains of pH 3.0. The rain was adjusted to pH 4.0, 3.0, 2.0 with sulfuric acid, and soybeans were exposed with the various pH levels, eighteen times every other days. The symptoms of damage, ratios of destroyed leaf and sulphur content by leaves were investigated. The results obtained were as follows. 1. The exposure to the rain resulted in the reddish brown or white pigment spots in the leaves surface and the colour was developed red and darker along the edge of spots. 2. Most crops were affected with acid rain of pH 3.0 at which the amount of chlorophyl was decreased. 3. The spinach, chinese cabbage, lettuce were sensitive to the acid rain, however, the peper, tomato pumpkin were resistance to it. 4. The content of chlorophyl was decreased with increasing leave injury. 5. The sulphur content in leave was increased with increasing acidity of the rain, however the yield was decreased.

  • 1,225 healthy day-old avian broiler chicks were used to investigate the effects of activated charcoal (AC, made from willow tree), hydrated sodium calcium aluminosilicates (HSCAS) and esterified glucomannan (EGM) supplementation on broiler performance, blood profiles and meat characteristics when less moldy or moldy corn was included to formulate seven isocaloric and isonitrogenous diets: Positive Control (less moldy corn diet, PC), PC+2% AC, Negative Control (moldy corn, NC), NC+0.05% EGM, NC+0.1% EGM, NC+0.5% HSCAS and NC+1% AC. PC+2% AC resulted in lower growth rate, poorer feed conversion ratio (FCR), more leg problems and higher mortality of birds than those fed PC diet (p<0.05). Inclusion of 0.05% EGM, 0.1% EGM, 0.5% HSCAS and 1% AC in NC diet did not improve average daily weight gain (ADG) or affect feed intake of birds during the first or the second three-week periods. However, 0.05% EGM tended to (p>0.05) and 0.1% EGM significantly (p<0.05) improved FCR during the first three-week period. Breast meat of NC birds had higher Minolta $L^*$ values (white) but lower $a^*$ (reddish) and $b^*$ (yellowish) values (p<0.01) than the PC birds. Addition of 0.05% EGM and 0.1% EGM in NC diet reduced the $L^*$ values (p<0.05), improved $a^*$ and $b^*$ values (p<0.05) of breast meat of birds fed NC diet, but had no effect on meat color when 0.5% HSCAS or 1% AC was included (p>0.05). Relative weight of liver to body was reduced by feeding NC diet (p<0.05) and could not be normalized by different mycotoxin adsorbents (p>0.05) to the ratio of the PC birds. Relative weight of cholecyst of NC birds was increased compared with PC birds and could only be normalized by addition of 0.05% EGM and 1% AC (p<0.05) in NC diet. NC birds had lower serum albumin level than the PC birds (p<0.05) and addition of 0.05% EGM or 1% AC in NC diet did normalize serum albumin level. Addition of 0.5% HSCAS in NC diet further reduced serum albumin, globulin, total protein and uric acid levels (p<0.05). It was concluded that lower FCR during the first three-week period of growth and deterioration of meat quality observed in bird fed moldy corn with moderate T2 and fuminisin contamination and damaged nutrients and pigment availability, might be improved by dietary supplementation of 0.05% to 0.1% EGM, but not by 1% AC or 0.5% HSCAS supplementation.

  • A bacterial disease of small red bean (Phaseolus angularis) was observed on field-grown plants in Suwon in year 2003. Leaf symptoms initially appeared as water-soaked spots that gradually enlarged, became flaccid and necrotic and were often bordered by a small zone of lemon yellow tissue. In the case of severe infection, dead leaves were defoliated. Pod symptoms consisted of the lesions that were generally circular, slightly sunken and dark reddish brown. Isolation made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow-pigmented bacterium typical of a xanthomonad. Three bacterial strains were purified and used for further tests. Pathogenicity of strains was confirmed on 3-week-old small red bean plants sprayed with bacterial suspensions containing $10^8 cfu/ml$ of phosphate buffered saline. The representative Xanthomonas strains isolated from small red bean were compared with X. axonopodis pv. phaseoli and X. axonopodis pv. phaseoli var. fuscans type strains for fatty acid profiles, biochemical tests and metabolic fingerprints using Biolog GN2 microplate, showing that all outcomes were indistinguishable between our isolates and reference strains. Two of three strains produced a melanin-like brown pigment extracellularly on King's medium B agar. These results suggest that this new small red bean disease observed in Suwon is bacterial fuscous blight caused by X. axonopodis pv. phaseoli and X. axonopodis pv. phaseoli var. fuscans.

  • Sarracenia purpurea as a carnivorous plant in the family Sarraceniaceae is known to require strong light for its growth and to absorb nutrients from the decomposed molecules of insects that are attracted by color, sweet juice, and the like. S. purpurea grew greenish in whole body under weak light conditions, while the whole of the insectivorous sac including leaves, is changed to dark red under strong light conditions. The phenomenon of reddish S. purpurea is thought to be related to the flavonoid pigment anthocyanin. Interestingly, the color change was not observed when S. purpurea was grown in a growth condition with abundant nitrogen fertilizer. The expression levels of anthocyanin contents and biosynthesis-related genes were strongly correlated with light intensity and nitrogen fertilizer. The anthocyanin content in the strong light condition ($240{\mu}mol\;m^{-2}s^{-1}$) was 6.15 times higher than that in the weak light ($40{\mu}mol\;m^{-2}s^{-1}$). In contrast, the anthocyanin contents were not significantly changed when 0.8% urea solution was supplied as nitrogen fertilizer. Consistently, CHALCONE SYNTHASE (CHS) gene was up-regulated by strong light and down-regulated by nitrogen fertilizer. These results suggest that the environmental changes of light and nitrogen in soil regulate the anthocyanin content in S. purpurea.

  • The seasonal variations of environmental sea water, arid carotenoid and lipid composition of reddish muscle tissue of arkshell, Anadara broughtonii in Chungmu and $Y{\check{o}}su-area$ were investigated. In the sea water, pH value, salinity, electrical conductivity of Chungmu and $Y{\check{o}}su-area$ were similar tendency, but water temperature in annual average at Chungmu-area was about $2^{\circ}C$ higher than that of $Y{\check{o}}su-area$. The concentration of nitrite-nitrogen, ammonia-nitrogen, phosphate-phosphours at Chungmu-area were slightly higher than those at $Y{\check{o}}su-area$. Meanwhile the concentration of silicate-silicious at $Y{\check{o}}su-area$ was about 2 times higher than that of Chungmu-area. The bacterial density of the sea water was ranged from 3.6 to 93/100ml for coliform and 3.0 to 15/100ml for fecal colifrom at Chungmu-area, but the coliform was ranged 7.3 to 150/100ml and the fecal coliform was ranged 3.6 to 20/100ml at $Y{\check{o}}su-area$. Among the coliform, 8.9% Escherichia coli, 33.9% Citrobacter freundii, 41.1% Enterobacter aerogenes groups were classified and 16.1% was not identified. In the hemoglobin content of reddish muscle tissue of arkshell, one from Chungmu-area was ranged from 1.1 to 2.5 g /dl and one from $Y{\check{o}}su-area$ was ranged from 1.7 to 4.4 g /dl. In total carotenoid content, 0.80 to 1.28mg/100 g muscle was in Chungmu-area and 0.45 to 0.99mg/100 g muscle was in $Y{\check{o}}su-area$. Among the carotenoid compositions, pectenolone content was 63.0% in annual average from Chungmu area and 59.6% from $Y{\check{o}}su-area$, 6.5% from Chungmu area and 18.9% from $Y{\check{o}}su-area$ in ${\beta}-carotene$, 9.9% from Chungmu-area and 9.1% from Yosu-area in pectenoxanthin, 11.2% from Chungmu-area and 5.2% from $Y{\check{o}}su-area$ in diatoxanthin monoester were observed. In the seasonal variation for the major carotenoids, the content of pectenolone was continuously incresedbefore spawning period and showed the maximum in July, but decresed after spawning period. Diatoxanthin monoester was gradually incresed during all growing period from March to next January. And ${\beta}-carotene$ was decresed before spawning period and showed the maximum in July but incresed after spawning period. The major fatty acids of total lipid and fractionated lipid classes from total lipid namely neutral-, glyco- and phospholipid were $C_{16:0},\;C_{18:1},\;C_{22:6},\;C_{20:2},\;C_{18:2},\;C_{20:5}\;and\;C_{18:3}$ acid in the muscle tissue of arkshell. The content of neutral lipid showed about 2 times higher than polar lipid in the muscle tissue of arkshell from both Chungmu and, $Y{\check{o}}su-area$. The content of total and neutral lipid was decresed during spawning period. In the seasonal variation for fatty acid compositions, the content of polyenenoic acid in total and neutral lipid was decresed and saturated acid was incresed. On contrary, polyenoic acid in glyco and phospholipid was incresed and saturated acid was decresed during spawning period from July to September. The sterol composition in total lipid were mainly consisted of cholesterol, campesterol, brassicasterol and ${\beta}-sitosterol$. Especially, the rate of content in cholesterol showed decrese during spawning period butcampesterol showed increse from March to November.
