Studies on the Optimal Culture Condition for Production of Red Pigments by Monascus rubber on Liquid Culture (액체배양에 의한 Monascus rubber의 적색 색소 생산을 위한 최적 배양조건에 관한 연구)

  • The optimum cultural conditions for production of red pigment from Monascus rubber KCTC 6122 is liquid culture were studied. Monascus robber KCTC 6122 was shown to give the maximum production of red pigment in the medium containing 4% rice powder, 0.2% NaNO$_3$, 0.3% Na$_2$HP0$_4$ and 0.15% MgSO$_4$. The optimum culture conditions, temperature, initial pH and shaking speed were 30$^{\circ}C$, 6.5 and 150 rpm, respectively. The red pigment production reached a maximum level at 8days of cultivation.

  • Harmful pet insects, if not controlled, can negatively affect people, plants and their surrounding environment. In Vietnam, all crops are regularly impacted by pest insects. In serious cases, crops can be totally destroyed by insect pests. Harmful insects that damage crops often grow fast and increase rapidly. Therefore, research on insects is crucial for managing pests, protecting crops, and forecasting pest situation in the following years. This study aimed to collect data regarding changes of pests on rice and corn as two main crops in four provinces in Red River Delta of Vietnam, including Thai Binh, Nam Dinh, Ha Nam, and Hung Yen, from 2018 to 2022. Primary data were collected from reports of government agencies and official statistics. Based on these data, this study evaluated changes of pest insects in five years, discussed reasons for such changes and response methods, and forecasted pest's behavior in the following years. Significant findings of this study include the fact that Vietnam has to face many difficulties to develop its agricultural sector. For insect management, an essential action is to do ground surveys to gather all related data including weather data, pesticide data, crop yield, and product quality. This information is meaningful for finding out causes of changes, understanding relationships between insects and surrounding factors, and predicting the situation in the following years.

Silk Dyeing Method in Natural Pigments - In Case of Korean Colored Rice Bran - (한국산 유색미 속겨의 안토시아닌 색소에 의한 견직물염색)

  • This research was conducted to establish the efficient use of the colored rice bran fur dyeing textiles. To investigate the fabric dyeability of the colored rice bran extract, the anthocyanin pigments were extracted with water of different temperature ranges of 40 ~ 8$0^{\circ}C$ and were dyed on silk fabrics with different dyeing temperature $25^{\circ}C$~6$0^{\circ}C$, at acidic pH and neutral pH, respectively. Aluminum chloride was preheated with mordant K/S value and dyeing fastness of dyed silk fabrics were examined. The anthocyanins of the colored rice bran were stable and red color at acidic pH, red purple or purple blue at neutral pH, but unstable, blue color at alkaline pH. If extracting temperature and dyeing temperature of dyeing solution were higher, the dyeability was high, but the color of dyed fabric showed red tone. When extracting temperature was 8$0^{\circ}C$ and dyeing temperature of dyeing solution was 6$0^{\circ}C$, the dyeability was best. Without mordant, the dyeability of silk fabrics was higher in acidic pH than in neutral pH solution. With mordant, the dyeability was higher than without mordant, and also higher in acidic pH. Pretreatment of aluminum chloride resulted in the increase of color intensity and stability. The laundering fastness of dyed fabrics was good from grade 5 to grade 3-4. Because of the anthocyanins sensitivity on light radiation, the light fastness of dyed fabrics was poor from grade 3 to grade 1-2.

Effects of Soil Types on Methane Gas Emission in Paddy During Rice Cultivation (논토양 종류가 메탄배출에 미치는 영향)

  • Anaerobic decomposition of organic materials in flooded rice fields produces methane ($CH_4$) gas, which escapes to the atmosphere primarily by transport through organs of the rice plants such as arenchyma etc., Although the annual amount of methane emitted from a given area is influenced by cultivation periods of rice and organic/inorganic amendments etc., soil type also affects methane emission from paddy soil during a rice cultivation. A field experiment was conducted to evaluate effects of soil type on $CH_4$ emission in two paddy soils. One is a red-yellow soil classified as a Hwadong series (fine, mixed, mesic family of Aquic Hapludalfs), and the other is a gley soil classified as a Shinheung series (fine loamy, mixed, nonacid, mesic family of Aeric Fluvaquentic Endoaquepts). During a flooded periods, redox potentials of red-yellow soil were significantly higher than gley soil. $CH_4$ emission in red-yellow soil ($0.21kg\;ha^{-1}\;day^{-1}$) was lower than that in gley soil ($5.25kg\;ha^{-1}\;day^{-1}$). In the condition of different soil types, $CH_4$ emissions were mainly influenced by the content of total free metal oxides in paddy soil. The results strongly imply that iron- or manganese-oxides of well ordered crystalline forms in soil such as goethite and hematite influenced on a $CH_4$ emission, which is crucial role as a $CH_4$ oxidizers in paddy soil during a rice cultivation.

Mycotoxin Production and Animal Toxicity of Molds Isolated from Discolored Sun-Dried Red Pepper(Capsicum annuum L.) (변질된 건조고추에서 분리한 곰팡이의 독소 생성 및 독성작용)

  • Mycotoxins produced by molds isolated from discolored sun-dried red pepper fruits were determined and the toxicity to animals was also tested by feeding mold-grown unpolished rice to rats. Among the mold species tested, only Alternaria alternata was toxic to experimental animals, while other mold species belonging to the genera of Colletotrichum, Diaporthe, Diaporthopsis, Botryosphereia, Aspergillus and Fusarium were not. Rats fed Alternaria-grown rice lost weight and died within two weeks of feeding period. Succumbed rats during the process of feeding study showed extreme cases of enlargements of stomach, small intestine and liver. Among the 17 Alternaria isolates, 8 species produced considerable amount of tenuazonic acid along with small amounts of other toxins including alternariol and monomethyl ether derivative of alternariol in both red pepper homogenate and unpolished rice. It is therefore advised that red pepper fruits infested by molds during the sun-drying process be discarded to avoid unnoticeable health hazards.

Screening and Identification of Monascus Strain Producing Monacolin K (Monacolin K 대량생산 Monascus 균주의 탐색 및 동정)

  • We had screened the Monascus strain capable of producing monacolin K dominantly among 29 Monascus strains. Red yeast rice was prepared by culturing each Monascus sp. with 200 g of steamed rice (12$0^{\circ}C$, 20 min) at 3$0^{\circ}C$ for 10 days and drying at 8$0^{\circ}C$ for 20 min. As a result, red yeast rice cultured by M. purpureus ATCC16457, M. purpureu IFO 32316, M. purpureus IFO 32228, M. kaoliang ATCC 46595 and M. kaoliang ATCC 46596 produced lots of red pigment and monacolin K. An unidentified Monascus sp. showed the highest productivityof red pigment and monacolin K among 29 Monascus strains. Its production of red pigment and monacolin K was 1.3∼39 times and 2.4∼8 times higher than other strains, respectively. Although the morphological characteristics of unidentified Monascus strain were a little different from the typical M. purpureus, it was identified as M. purpureus CBS 281.34 from the result of sequencing of ITS (Internal transcribed spacer) and 28S ribosomal RNA (partial).

An Experimental Study for Dryer (건조기 고안 제작에 관한 연구)

  • A newly devised dryer with heated air for the farm products, especially suited for high water content materials such as red pepper, Beer ground, each Vegetables, and Low water content materials such as Rough rice was tested for its thermal efficiency and drying mechanism, and the optimum conditions for each sample were established. In order to improve the present rural situation of drying farm products which entirely dependent upon natural solar radiation, a study upon an economic multi-parpose dryer was conducted. A series of drying tests were run first with red pepper which is one of the important cash crop in Korean farm. And successive series of tests were also run with such proaucts as garlic, sweet potatoes, green onion, radish, Beer ground and Rough rice. The results from the above experiment in drying system with heat dryer can be summarized as follows. 1. Drying duration could be shortened by the tempering effect in high water content crop such as red pepper and beer ground. 2. The color changes occured in around 20% water content in red pepper. The degree of color change was heavily affected by high temperature and short drying duration. 3. The drying condition of red pepper was most favourable at the temperature of 85$^{\circ}C$ in early stage and 80$^{\circ}C$ in middle stage and 75$^{\circ}C$ at the final stage, and with the air rate of 0.81㎥/sec and with sample amount of 200kg. 4. The drying condition of Rough rice(I.R.667) was most favourable at the templature of 40$^{\circ}C$ in early stage and 35$^{\circ}C$ in middle stage and final stage and with the air rate of 0.2㎥/sec and with sample amount of 75kg. 5. In order to prevent the color change of red pepper and to assure high efficiency in drying mechanism, it was necessary to lower the temperature as the time passes in drying process. 6. For vege tables, the drying rate were short in early stage and there was also tempering effect. However, for garlics, Constant drying rates through the early and final stages were observed and there were no tempering effects. 7.   • A total of 40 Korean and Japanese rice varieties were evaluated for their main chemical components, physical properties, cooking quality, pasting properties, and instrumental measurements. Based on their quality evaluations, it was concluded that Korean and Japanese rice varieties were not significantly different in the basic components of NIR (Near Infra Red) data and the chemical analysis from the uncooked brown and milled rices. Korean rice had a little bit higher protein and amylose contents but much lower fat acidity than those of Japanese rice from the chemical analysis. From all the data of three different kinds of NIR methods, Korean and Japanese milled rice were very similar except the taste score. Japanese rice showed a slightly higher taste score, a little bit higher lightness and whiteness, but lower yellowness and redness than Korean one. From all those data of NIR and the chemical analysis, Korean and Japanese rices had very similar components except the fat content.

Sensory Profiling of Rice Wines Made with Nuruks Using Different Ingredients (누룩원료를 달리하여 제조한 쌀약주의 관능적 특성)

  • The quantitative sensory profiles of rice wines made with nuruks using eight different cereal ingredients were developed using sensory descriptive analysis. Two appearances, eight aromas, eight flavors and tastes, and two mouthfeel related sensory attributes were evaluated by a panel of 10 judges. The sample made of black rice nuruk had the highest intensity in red color, while the other samples had similar ranges in yellow color. The mean sensory intensities of the samples prepared with black rice and glutinous rice nuruks were high in 'sweet', 'fruit taste', 'pungent', and 'sour', while those samples prepared using non-glutinous rice, buckwheat, hull-less barley, unpolished rice nuruks had overall high intensities in 'grain', 'fermented aroma', 'bitter', and 'astringent' attributes. Based on the principal component analysis of the descriptive data, samples were primarily separated along the first principal component, which accounted for 53% of the total variance between the rice wines with high intensities of 'red color', 'sweet', and 'fruit taste' versus 'bitter', 'astringent', and 'yellow color'.

  • This experiment was carried out to evaluate the germination characteristics of Korean red rice seeds. Ten lines of the long grain and ten lines of the short grain Korean red rices were tested for dormancy, shoot emergence rate at different water and soil depth. Both the long and short grain type seeds of Korean red rice germinated over than 99% immediately after harvest, so that no dormancy was observed in the Korean red rices. Shoot emergence rate of the long grain red rices at 7cm of water depth and that of the short grain red rices at 11cm were 83.3% and 83.5% respectively, while those of the indica check Samgangbyeo and japonica check Seomjinbyeo were 0%. Shoot emergence rates of the long and short grain red rices at 7cm of soil depth were 71.1% and 73.9% respectively, while those of the check varieties Samgangbyeo and Seomjinbyeo were 40.0% and 28.0% respectively. Thus, the Korean red rices were expected to be useful germplasm source for direct seeding.
