The semicontinuous production of red pigment by immobilized cells of Bacillus sp. B H-99 was investigated in comparison with free cells. The red pigment produced highest productivity under the conditions of aeration of 0.2 mL/min and 2 mm diameter of gel beads by using 3.0% sodium alginate. Semicontinuous production by immobilized cells showed the highest productivity with replacement of fresh production medium in every 72 h for fourth fermentation cycle following the conditions of red pigment productivity.
Mycelial growth, color difference and productivity of red pigment of beni-koji by Monascus anka KCCM 11832 were examined with respect to it's pigment in submerged culture with various medium and culture conditions. Shaking incubation was more promoted mycelial growth and the production of pigments than that for non-shaking incubation, and red color became ten times deeper. The production of red pigment was the highest when incubated at 25$\circ$C for 7 days in pH 6.0, but mycelial growth was showed the highest at 32.5$\circ$C. The levels of carbon and nitrogen source for maximum red pigment production were 2% rice powder and 0.05% peptone, respectively and the level of peptone for maximum pigment production was lower than that for maximum mycelial growth. Among pigmentation promoting agents tested, MgSO$_{4}$, was found to be suitable for the production of red pigment, and the optimum level was 0.1%.

In general, pigment production can be influenced by the medium composition, pH and physical factors such as aeration, agitation, and visible light. The influence of gaseous environments on the pigment production by Monascus purpureus ATCC 16365 was investigated by controlling the DO (dissolved oxygen) concentration through aeration and agitation. When the DO concentration was controlled below 20%, the production of red pigment significantly increased whereas the biomass production decreased. Therefore, the dissolved oxygen concentration could significantly affect the biosynthesis of red pigment as a secondary metabolite by a wild-type filamentous fungus under the anaerobic condition. The results indicate a high potential of enhancing the productivity of the red pigment as a secondary metabolite through controlling the DO concentration.
To maximize and sustain the productivity of Monascus pigments, various environmental and nutritional parameters, such as the initial moisture content, pH, inoculum size, sample size, and nutrient supplement, that influence pigment production were evaluated in solid-state cultures as follows: initial moisture content, $50\%;$ pH, 6.0; inoculum size $1\;\times\;10^4$ spore cells $(grams\;of\;dry\;solid\;substrate)^{-1};$ sample size, 300 g. All supplementary nutrients (carbon, nitrogen, and mineral sources) added has inhibitory effects on the cell growth and red pigment production. In open tray culture the maximum biomass yield and specific productivity of red pigments were 223 mg DCW $(grams\;of\;initial\;dry\;substrate)^{-1}$ and, $47.6\;OD_{500}\;(DCW\;grams)^{-1}h^h{-1}$ respectively.
The characteristcs of monascus fermentation using a hyperpigment-producing mutant, Monascus sp. J101, were analyzed, and the extractive fermentations employing permeabilizing agents and resin were carried out to increase the productivity of red pigment. And the kinetic analysis was also carried out in case of the monascus fermentation using Amberlite XAD-7. The extracellular content of the red pigment produced by Monascus sp. J101 was about 17% of the total, and the production of pigment was regulated by its own product. The cell growth reached a stationary phase at 48 hours ofter inoculation, whereas the pigment production continued up to 100 hours, which showed the pattern of a mixed growth-associated type. During the fermentation, various permeabilizing agents were added to the culture medium and their effects on pigment production were examined. By adding 0.05% Triton X-100 at 48 hours of cultivation, about an 18% increase in pigment production was accomplished as compared to the control, 12% ethyle acetate and 15% for 0.05% deoxycholate, respectively. When a nonionic adsorbent, Amberlite XAD-7 was added to the culture medium at a concentration of 12.0% at 48 hours of cultivation, the pigment production was enhanced by about 48.9% as compared to the control.
Isolation of a pigment overproducing mutant, P-57, by ultraviolet irradiation of Monascus purpureus KCCM 60016 and investigation of the optimal conditions for pigment production of the mutant were carried out. P-57 mutant produced pigment on solid state culture. Unpolished rice was the best cereal source for pigment production among eight kinds of tested cereal sources for the solid culture of the mutant. The optimal culture condition for pigment production were obtained from the cultivated at $30^{\circ}C,\;90\%$ humidity for 30 days. The P-57 mutant strain showed the best pigment productivity of 160.0 unit at red pigment, 193.6 unit at orange pigment, and 141.6 unit at yellow pigment on solid state culture under optimal condition.
Optimal conditions of producing red pigment by Monascus anka, Nakazawa, et Sato, IFO 4478 and 6540 were found to be at pH 6.0 and $30^{\circ}C$ for 10 days. When 3.0% steamed rice and 1.0% defatted sesame exfracts were used as substrates, the highest production of red pigment was yielded. Furthermore, mutants such as Monascus anka 4475-27 and 6540-185 induced by N -methyl-N-nitro-N-nitrosoguanidine (MNNG) treatment were found to produce pigment much more than parent strains.
Cultural conditions for the production of water-soluble pigment from mycelial culture of Cordyceps scarabaeicola KEFC-C252 and antimutagenic activity of the pigment were investigated. To obtain the maximum productivity of the pigment from mycelial culture of C. scarabaeicola KEFC-C252, the optimized medium was made with 1.5% sucrose, 2.5% yeast extract and initial pH 5.5. C. scarabaeicola KEFC-C252 was cultivated to reach the maximum concentration of the pigment at $26^{\circ}C$ for 108 hrs. C. scarabaeicola KEFC-C252 produced about 1.2 g/liter pigment under the optimized condition. The pigment was isolated from the culture filtrate by ethylacetate extraction, acidic precipitation and crystallization. The isolated pigment was scarlet hexagonal column crystal, and the color of the pigment was changed according to pH of the solution. The pigment showed violet in the alkaline water but showed red color in the acidic water. The pigment showed inhibitory activity against mutagenic activity induced by 4-nitroquinoline N-oxide. Furthermore, the pigment showed inhibitory activity against spontaneous mutation on Salmonella typhimurium TA98 and TAlOO.
For the purpose of mass producing Monascus red pigments optimum medium composition and environmental conditions were investigated in submerged flask cultures. The optimum carbon and nitrogen sources were determined to be 30g/L of glucose and 1.5 g/L of monosodium glutamate (MSG). Of the three metals examined, Fe$\^$2+/ showed the strongest stimulatory effect on pigment production and some stimulatory effect was also found in Mn$\^$2+/. Optimum pH and agitation speed were determined to be 6.5 and 700 rpm, respectively. Under the optimum culture conditions batch fermentation showed that the maximum biomass yield and specific productivity of red pigments were 0.20 g DCW/g glucose and, 32.5 OD$\sub$500/g DCW$\^$-1/h$\^$-1/, respectively.
Three marine bacteria producing pigments were isolated from seawater of Jeju-Do and local solar saltern in Korea. Based on phenotypic characteristics and 16S rRNA sequence analysis, the strains were identified as Pseudoalteromonas spp., which produced red (Ju11-1), yellow (Ju14), and orange (TA20) pigments. The pigments showed UV absorption maxima at 537, 378 and 387 nm, respectively. The strains were growing well on Marine broth 2216 culture medium. The productivity of pigments reached the maximum value after 28 hours (Ju11-1, Ju14) and 24 hours (TA20) at $30^{\circ}C$, 2% NaCl and pH 6-7. The best pigment production of strains were supported by 1% of lactose (Ju11-1) and maltose (Ju14, TA20) as a carbon source and 1% of beef extract as a nitrogen source.
