전북지역 한우의 red cell fragility와 glutathione peroxidase활성에 관한 연구 (Studies on red cell fragility and glutathione peroxidase activities in Korean native cattle of Chonbuk region)

  • The tests related to red cell fragility were performed. Samples of blood anticoagulated with heparin were obtained from Korean native cattle in Chonbuk region abattoir, and classified by the district(Kun) with reference to breeding location. Hemolysis test for red cell fragility was performed with whole blood and glutathione peroxidase activity was measured spectrophotometrically. Blood concentration of selenium, inorganic component of glutathione peroxidase, was also determined fluorophotometrically. The results obtained were summerized as follows; 1. Percent hemolysis of erythrocytes ranged from 13.53 to 20.74%, and its mean Palue was low as $17.11{\pm}9.91%$. Means in all were not district(Kun) in Chonbuk region significantly different. 2. Glutathione peroxidase activity ranged from 2,881 to 4,000mU/ml, and high mean values, $3,352{\pm}1,872mU/ml$, reflected low percent hemolysis. 3. There was a highly negative correlation between the red cell fragility(Y) and blood glutathione peroxidase activity(X). The linear regression equation for these data was: Y=29.86-3.75X with a correlation coefficient of r=-.6886 (p<0.01) 4. Blood selenium concentration ranged from 0.16 to $0.24{\mu}g/ml$, and mean values was normal level as $0.2{\pm}0.11{\mu}g/ml$. 5. There was a highly positive correlation between blood selenium concentration(X), and blood glutathione peroxidase activity(Y). The linear regression for these data was: Y=230+15,790X, with a correlation coefficient officient of r=0. 8635.

건강인(健康人) 및 직업적(職業的) 매혈(賣血)로 인(因)한 실혈성(失血性) 빈혈자(貧血者)의 적혈구(赤血球) 취약성(脆弱性)에 관(關)하여 (On the Red Cell Fragility in the Normal and Anemic Professional Blood Donors)

  • The osmotic and mechanical red cell fragility of the professional blood donors, who were found to be anemic as the result of frequent and repeated blood loss the past 5-6 years, were compared with that of the normal person while incubating the blood at $4-6^{\circ}C$ for 28 days. The fragility was expressed as % hemolysis occured during the incubation, and the following results were obtained: 1. The osmotic fragility in the normal persons (i.e, ; control group) progressively increased as the incubation became longer, and % hemolysis in 0.42% NaCl solution at 0, 10, 15, 21 and 28 incubation day was 31.90, 50.20, 41.68, 43.50 and 55.40 respectively. The mechanical fragility. in the normal red cells ranged between the minimum of 0.00% to the maximum of 5.80% both in 0.90 and 0.66% of NaCl solutions. 2. The hemolysis curve obtained in the red cell osmotic fragility from three cases of the anemic persons (i.e,; experimental group) showed a significant left side shift comparing with the normal in general which indicates that the fragility was more increased in the experimental group. The mechanical fragility in the experimental group ranged between the minimum 0.00% to the maximum 19.00% both in 0.90 and 0.66% of NaCl solutions. 3. The red cells of the chronic anemic person due to the frequent blood loss as the professional blood donor exhibit significantly marked increase both in osmotic and mechanical fragility comparing with the normal, and the tendency was more prominent as the incubation period became longer.

Histamine에 의한 적혈구 취약성의 변화에 관한 연구 (A Study on Osmotic Fragility of the Red Blood Cell in Histamine-treated Rabbit)

  • Adult rabbits were anesthetized with nembutal, 30 mg/kg. Carotid artery and jugular vein were exposed surgically and cannulated with polyethylene tubing. Arterial blood pressure was recorded via pressure transducer on the physiograph and $100{\mu}g/ml$ of histamine solution was infused through the jugular vein by using the constant infusion pump with a rate of 0.92 ml/min or 1.40 ml/min. Mean arterial blood pressure was maintained at $40{\sim}70 mmHg$ and hypotension was kept for 2 hours. After the termination of this period, blood was taken and osmotic fragility was mea sured immediately. Also, every sample of normal blood and shocked blood was incubated for 1 hour or 2 hours at $37^{\circ}C$ in order to see whether or not there was some influence of incubation. Furthermore to clarify which component was responsible for the change on the fragility, the mixtures of normal blood cells with shocked plasma and shocked blood cells with normal plasma were also incubated at $37^{\circ}C$ for one or two hours and fragility in such cases was measured. The data obtained were analysed by probit-plot method and the concentration of saline solution at which the hemolysis started to occur, 50% of blood cells were hemolysed and that at which the red blood cells hemolysed completely were determined. The values for the blood of hypotension stage were compared with those of the control blood. The results obtained were as fellows: 1. Osmotic fragility of red blood cell was increased in hypotensive state induced by histamine. 2. The differences of osmotic fragility after two hours of incubation were negligible both in normal blood and in that of hypotensive state. 3. Osmotic resistance of normal red blood cell incubated in shock plasma was less than that of shock red blood cell incubated in normal plasma. It was suggested that plasma in hypotensive state caused by histamine might be primarily responsible for the alteration of red blood cell fragility.

  • The effects of Gypsophila saponin, sodium dodecylsulfate (SDS) and Triton X-100 on volume changes and fragility of red blood cells were compared to ginseng saponin to elucidate whether there are any difference in their action on membrane lipid. Cell volume was decreased to about 38% in 1M NaCl and increased to about 20% in 1/10M NaCl. Hematocrit value was decreased by Gypsophila saponin, SDS, and Triton X-100 which caused hemolysis in isotonic NaCl solution. These detergents also inhibited increase of cell volume and accelerated hemolysis in hypotonic solution. However, ginseng saponin did not influence to osmotic volume changes and hemolysis of red blood cells. These results suggest that the disruptive effect of plant saponin on membrane barriers induced by removing membrane lipid is different from their source and ginseng saponin has very low affinity to membrane lipid.

The Effect of the Aging of Red Blood Cells on Rheological Properties and Hemolysis

  • It is well known that red blood cells (RBCs) are suffered from chronic stresses in systemic circulation. The objective of this study is to clarify the effect of the aging of RBCs on rheological properties and hemolysis. Initially, RBCs age fractionation was performed by using a high-speed centrifugation (15[min] at 1500[G]), then young and aged RBCs were suspended in plasma to adjust the hematocrit level of 40[%]. After this pretreatment, the viscosity was measured by using a capillary type and a cone-plate type viscometers, respectively, and the hemolysis test was carried out by a seesaw type shaker. Results from these experiments showed that the viscosity of the aged RBCs measured by the capillary viscometer was increased by 10[%] as compared with that of the young RBCs. Under the condition of all shear zones, the viscosity of the aged RBCs was increased in case of using the cone-plate type viscometer. And the hemolytic level was increased twice as the aging. The data obtained in this study indicated that the ability of aggregation of RBCs was increased and the deformability of RBCs membrane got lower with the aging. Furthermore, it was exhibited that the fragility of RBCs ’ membrane was increased with the aging.

  • The Present study was conducted to investigate the effects of Ginseng extract on the tension-area curve for stearic acid monolayer. At the same time, the effects of Ginseng extract on osmotic and mechanical fragility of human red cells and histamine release from rabbit leukocytes were studied, The results are summarized as follows. 1. The Ginseng alcohol extract was found to expand liquid expanded phase of stearic acid monolayer, thus it is speculated that this agent may be acting as a surface active substance. 2. Osmotic hemolysis was inhibited by the Ginseng alcohol extract and the same effect was also observed in the presence of Ginseng saponin. However, the Ginseng alcohol extract was found to decrease hematocrit ratio of the RBC suspension, therefore, the inhibition of the osmotic hemolysis by this agent may be secondary effect to the reduced cell volume. 3. The mechanical hemolysis was also inhibited by the Ginseng alcohol extract but the inhibition was independent of changes in hematocrit ratio. 4. Histamine release from rabbit leukocytes was significantly increased in vitro in the presence of the Ginseng alcohol extract.(p<0.05)

혈색소 농도에 따른 대학생의 골밀도 분석 (Analysis of Bone Mineral Density according to Hemoglobin in University Students)

  • 본 연구는 대학생을 대상으로 혈색소와 골밀도를 평가하고, 혈색소와 골밀도 정량 분석 결과를 보기 위하여 시작되었다. 본 연구는 골밀도에 미치는 혈색소의 영향을 목적으로 하였다. 연구대상자는 20~30세의 건강한 대학생 52명을 직접 실험을 진행하였다. 골다공증은 전통적 노화 여성의 질병으로 간주되었지만, 점차 남성의 건강 문제로 되고 있다. 골다공증의 진단은 골밀도의 정량적 평가로 계산된다. 검사실에서 실시하는 혈액과 소변 검사는 주로 뼈의 신진 대사의 장애의 원인을 파악하기 위해 낮은 BMD 또는 취약성 골절에 사용된다. 본 연구는 성별에 따른 골밀도의 차이는 없었다. 평균 적혈구수, 혈색소 및 적혈구 용적은 남성에게서 상당히 높은 값을 나타내었다(p<0.01). 요추 척추, 골격근양, 기초대사량 사이의 상관 관계는 통계적으로 유의 한 수준에서 정의 관계로 나타났다(p<0.01). 다중회귀분석결과 혈색소는 골밀도에 51.7% 통계적 영향을 나타냈다. 이러한 연구 결과는 골밀도와 혈색소에서 유의한 관계를 이해하는데 유용하며, 혈색소 수준은 골밀도 수준을 예측하는데 강력한 인자이다. 결론적으로, 본 연구에서 낮은 Hb 값이 낮은 뼈 질량을 가진 피험자에서 유의한 결과를 보여, 낮은 Hb 값이 골밀도를 감소시키는 뼈 회전율 변화의 위험 요소로서의 규칙을 가짐을 의미한다고 사료된다.

소아에서 유전성 구상 적혈구증의 비장 적출술 (Splenectomy in Hereditary Spherocytosis in Childhood)

  • 저자들은 1987년 12월부터 1993년 8월까지 영남대학교 의과대학 부속병원에 입원하여 유전성 구상 적혈구증으로 진단받고 비장적출술을 시행한 9례를 대상으로 임상분석하여 다음과 같은 결론을 얻었다. 1. 총 9명중 남아 5명, 여아 4명으로 남녀비가 비슷하였으며, 6세에서 10세 사이가 9례중 5례로 전체의 반이상을 차지하였다. 2. 가족력을 나타낸 경우는 9례중 4례(44.4%)이었다. 3. 주요임상 증상으로는 빈혈, 황달 및 비장종대가 주증상이었으며, 담석증 및 총담관결석을 동반하였던 2례에서는 우상복부 동통을 나타내었다. 4. 검사실 소견으로는 말초혈액 도말검사상 구상 적혈구를 발견할 수 있었으며, 혈색소 $8.1{\pm}2.4gm/dl$, 헤마토크리트 $23.4{\pm}10.2%$ 망상적혈구 $15.9{\pm}11.7%$이었으며, 삼투압 취약성은 모두에서 증가되어 있었다. 혈청 전 빌리루빈 $8.6{\pm}10.9mg/dl$, LDH $370.1{\pm}169.1IU/L$으로 증가되었다. Coombs 검사상 9례 모두 음성반응을 보였다. 5. 9례 모두 비장적출술을 시행하였으며 이들중 담석증을 동반한 1례에서는 담낭절제술, 총담관 결석을 동반한 1례에서는 담낭절제술, 총담관 결석제거술 및 T-tube 담관조루술을 동시에 시행하였다. 수술 소견상 비장의 무게는 350(150-600)gm이었고, 9례중 3례에서 부비장(accessory spleen)이 관찰되었다. 6. 비장 적출후 유전성 구상 적혈구증 9례에서 수술전 혈색소 $8.1{\pm}2.4gm/dl$, 헤마토크리트 $23.4{\pm}10.2%$, 수술후 혈색소 $12.3{\pm}1.2gm/dl$, 헤마토크리트 $37.0{\pm}5.3%$로 증가되어 술후 빈혈이 교정되었음을 관찰할 수 있었다. 수술후에 수혈의 필요성은 없었으며, 술후 특별한 합병증없이 9명 모두 현재까지 건강하게 잘 자라고 있다.
