Effect of Kimchi Extracts to Reactive Oxygen species in Skin Cell Cytotoxicity (김치 추출물의 활성산소에 대한 피부세포 독성 완화효과)

  • Kimchi is composed of many ingredients such as Chinese cabbage, garlic, ginger, and red pepper and fermented fish extract. Some of them were known to have antioxidative activities due to their scavenging effect against reactive oxygen species(ROS). To study the health effects of kimchi on human skin cells, keratinocyte(A431, epidermoid carcinoma, human) and fibroblast(CCD-986SK, normal control, human) were cultured in oxidative stress condition provoked by paraquat, a superoxide anion generator, and hydrogen peroxide in the absence and presence of kimchi extract. The survival rate of keratinocyte was greatly reduced when exposed over 1mM concentration of hydrogen peroxide($H_{2}O_{2}$), but cytotoxicity of $H_{2}O_{2}$ was significantly reduced by kimchi extracts on cells. Especially 2 week-fermented kimchi decreased remarkably the cytotoxicity by $H_{2}O_{2}$ to keratinocyte cells. Over 1mM of paraquat concentration showed strong cell toxicity on keratinocyte, but the extracts from kimchi fermented for 1, 2 and 3 weeks showed protective effects in order. Fibroblast cells were significantly affected by $H_{2}O_{2}$ as were keratinocyte cells. Although almost all extacts of kimchi of different fermentation periods showed protective effect against cell killing at 0.5mM concentration of $H_{2}O_{2}$ week-fermented kimchi extract showed the strongest protective effect on fibroblast cells treated with 1mM $H_{2}O_{2}$ for either 1 day or 4 days. However most of kimchi extracts showed weak preventive effect or no effect on oxidative stress produced by paraquat. In conclusion, 2 week-fermented kimchi extract seems to have the best potential in preventing skin cells against oxidative damage which might be related to their scavenging effects of kimchi components produced during their fermentation process.

Educational attainment and differences in fruit and vegetable consumption among middle-aged adults in the Korean National Health and Nutrition Examination Survey IV

  • We investigated whether socioeconomic differences affect fruit and vegetable (FV) consumption with respect to total intake and intake of various FV subgroups. Our study included 6667 adults aged 40-64 years who completed a dietary survey in the fourth Korean NHANES (2007-2009). FV intake was estimated from 24-hour recalls and food frequency questionnaires. Differences in FV consumption related to educational attainment were analyzed according to different nutritional categories of FV. Both men and women in the low-education group had the lowest intake of total FV and total fruits, and women also had the lowest intake of total vegetables. Also lowest in this group was consumption of mushrooms and vegetables (excluding kimchi) among men, and cruciferous and allium vegetables (excluding Chinese cabbage and radish) among women, while kimchi consumption was the highest in this group. Additionally, an association between educational level and intake of citrus fruits was evident among men. Adults in the low-education group consumed less carotene-rich FV, red fruit and/or vegetables, and dark-green leafy vegetables, fewer total vegetable dishes, and fewer types of fruit than in other groups. Men in this group had the lowest intake of yellow/orange fruit and/or vegetables, and women consumed the least folate-rich FV. There is a clear association between educational attainment and FV intake with regard to total intake, and to specific nutrients, bioactive compounds, colors, and variety.

  • Three species of Pythium previously not recorded in Korea were found during 1975. Pythium spinosum Sawada was isolated from diseased cabbage seedlings, P. myriotylum Dreschl. was isolated from kidney bean and P. butleri Subramanium from cucumber, spinach, red bean and radish. Pathogenicity of isolates of P. butleri and P. myriotylum was confirmed in pathogenicity tests but P. spinosum appeared to be non-pathogenic. Several isolates failed to sporulate satisfactorily on cornmeal agar and some produced degenerate oogonia after sub-culturing on this medium. Sunflower seed agar was found to be a suitable alternative medium. The validity of previous records of P. debaryanum Hess in Korea is discussed.

Physiological Characteristics and Manufacturing of the Processing Products of Sprout Vegetables (새싹채소의 생리활성 특성 및 그 가공품의 제조)

  • Sprout vegetable and general vegetable were obtained from radish, red cabbage, vegetable green, buckwheat and broccoli seeds. The vitamin contents had a large deviation, and the vitamin B1 contents decreased and/or was not detected according to the sprouting. This tendency occurred because of the use for growth of sprouts. The mineral contents in sprout vegetables was the highest and was 2~10 times greater than general vegetables. In the growing pathway, potassium was a large consumption nutrient and consumption of sodium was low. $\alpha$-Amylase and total enzyme activities were higher in sprout vegetable than general vegetable, which was also the case of the corresponding freeze dried powders. Sprouting was shown to increase the enzyme activities. Thus, sprout vegetables had several advantages including a rich level of nutrients and safety from agricultural pesticides.

  • The assortment of endophytic lactic acid bacteria (LAB) in kimchi derives from its raw vegetables, which include Chinese cabbage, radish, welsh onion, onion, garlic, red pepper, and ginger. These vegetables were examined during mulkimchi fermentation using gene-specific multiplex polymerase chain reaction and 16S ribosomal RNA sequence analysis. Sixteen species from five LAB genera (Leuconostoc, Lactobacillus, Lactococcus, Pediococcus, and Weissella) appeared in the raw kimchi materials. Interestingly, nine LAB species were identified in mulkimchi on fermentation day 0 as follows: Leuconostoc carnosum, Leuconostoc citreum, Leuconostoc gelidum, Leuconostoc inhae, Leuconostoc mesenteroides, Lactobacillus plantarum, Lactobacillus sakei, Lactococcus lactis, and Weissella confusa. Seven additional LAB species were present in mulkimchi at fermentation day 9 as follows: Leuconostoc gasicomitatum, Leuconostoc kimchii, Lactobacillus brevis, Lactobacillus curvatus, Lactobacillus pentosus, Pediococcus pentosaceus, and Weissella koreensis. These species corresponded completely with the LAB in kimchi vegetables. Wei. confusa was the predominant LAB during early fermentation (pH 6.20 to 4.98 and acidity 0.20 to 0.64%), while Lac. sakei, Lac. plantarum, and Wei. koreensis became dominant later in fermentation (pH 4.98 to 3.88 and acidity 0.64 to 1.26%). These results collectively demonstrate that the LAB involved in mulkimchi fermentation originates from the raw vegetables examined.

The Effects on Antimicrobial and Cytotoxicity of Brassica oleracea L. Fractions. (적채 분획물의 항균, 암세포 증식 억제효과)

  • This study was carried out to investigate the affects on antimicrobial and cytotoxicity of red cabbage (Brassica oleracea L.,BO). In the paper disc test, antimicrobial activity of BO fractions was increased in proportion to its concentration. Among the various solvent fractions of methanol extract (BOM) of BO, the ethylacetate partition layer (BOMEA) showed the strongest antimicrobial activity We also determined the cytotoxicity and Quinone Reductase induced effect of BO extract and fractions on human cancer cells. The cytotoxicity of BO fractions on HepG2, HeLa and MCF-7 cells was evaluated by MTT assay. The BOMEE and BOMEA showed strong cytotoxic effects on all cancer cell lines we used. The quinone reductase induced effect of BO fractions on HepG2 cells, the hexane partition layer (BOMH) at a dose of 200 $\mu{g}$/ml was 2.88 times more effective compared to the control values of 1.0.

  • This study was investigated traditional food utilization actual conditions of Gwangju and Chollanamdo. Determination of sulforaphane in cruciferous vegetables by SIM (선택이온 측정법에 의한 십자화과 채소중의 Sulforahane 함량)

  • Quantitative determination of sulforaphane, S-methylsulfinylbutyl isothiocyanate in 20 cruciferous vegetables of Korean origin was performed. Homogenate of vegetable was extracted with dichloromethane, and the extract, after drying, was subjected to GC/MS analysis, which was based on single ion monitoring (SIM) at m/z 72, 160, 55, 114 and 177. The content of sulforaphane was found to be the highest in the extract of broccoli $(80.2{\sim}617.7\;ppm)$ followed by turnip $(15.4{\sim}23.1\;ppm)$, red cabbage $(9.9{\sim}32.1\;ppm),\;radish\;(5.5{\sim}8.8\;ppm)$ and kale (8 ppm). Among various cultivars of broccoli, broccoli '1243' and broccoli 'Pilgrim' showed higher content of sulforaphane than others. Especially, in the stem of broccoli '1243' and the floret of broccoli 'Pilgrim' the amount of sulforaphane was the highest (>700 ppm). Thus, the content of sulforaphane differed according to the cultivars and the portion of the vegetables.

Projection of Consumptive Use and Irrigation Water for Major Upland Crops using Soil Moisture Model under Climate Change (토양수분모형을 이용한 미래 주요 밭작물 소비수량 및 관개용수량 전망)

  • The impacts of climate change on upland crops is great significance for water resource planning, estimating crop water demand and irrigation scheduling. The objective of this study is to predict upland crop evapotranspiration, effective rainfall and net irrigation requirement for upland under climate change, and changes in the temporal trends in South Korea. The changes in consumptive use and net irrigation requirement in the six upland crops, such as Soybeans, Maize, Potatoes, Red Peppers, Chinese Cabbage (spring and fall) were determined based on the soil moisture model using historical meteorological data and climate change data from the representative concentration pathway (RCP) scenarios. The results of this study showed that the average annual upland crop evapotranspiration and net irrigation requirement during the growing period for upland crops would increase persistently in the future, and were projected to increase more in RCP 8.5 than those in RCP 4.5 scenario, while effective rainfall decreased. This study is significant, as it provides baseline information on future plan of water resources management for upland crops related to climate variability and change.

The Systematic Study on Reduction of Food Waste Products(I) -Survey on Waste Rates of Frequently Consumed Korean Foods and Effect of Education on Kitchen Wastes of Institutional Foodservices - (음식물 쓰레기를 줄이기 위한 체계화 연구(I) -상용식품의 폐기율 조사와 급식소의 유형별 음식물 쓰레기량과 교육에 의한 개선효과-)

  • This research was conducted to estimate waste rates of 69 food items after pre-preparation and amounts of food wastes disposed and it's reduction effect by education in some institutional foodservices for reduction of the food wastes. The result are as follows: 1. The waste rates were 55.40% for sweet corn, 5.73% for potatoes, 19.14% for sweet potatoes, and 33.47% for chestnuts. 2. The waste rates of fruits were 0.21-49.36%. Food items with over 40% waste rate were banana, watermelon, and pineapple. 3. The waste rates of vegetables were 1.14-52.90%. The number of foods with under 10% waste rate were 14 items(red pepper, green pepper, chard, and so on), 10-20% was 9 items(perilla leaf, chinese cabbage, ginger root, and so on), 20-30% was 5 items(root of chinese bellflower, garlic, radish-leaves, burdock, welsh onion-large type), 30-40% was 3 items(shepherd's purse, head lettuce, kale) and over 40% was 3 items(water dropwort, crown daisy, mallow). 4. The waste rates were 24.30% in chicken, 9.53-13.79% in eggs, and 9.30-55.32% in fishes. The waste rates of vegetables and fishes were higher than those of other food groups. 5. There were significantly different in amount of food wastes disposed (g/person/day) to institutional foodservices (hospital>industrial institution> Korean restaurant>elementary school). The amount of food wastes disposed, especially amount in pre-preparation phase, after education for reduction of food wastes was significantly reduced. Since these study results show significant deviations in food waste rates and education effect, there should be more studies for standard waste rates of each food and systematic education method for reduction of food wastes.
