Inhibition of Cronobacter sakazakii by Lactobacillus acidophilus n.v. Er2 317/402

  • Lactobacillus acidophilus n.v. Er2 317/402 strain Narine is known as a health beneficial functional probiotic culture and supplementary source of nutrition for newborns. In this study, in vitro antimicrobial activities of Narine-lyophilized (Narine-L), Narine-heat treated (Narine-HT), and Narine crude cell-free extract (Narine-CCFE) were evaluated against pathogen Cronobacter sakazakii (C. sakazakii) in agar as well as in a reconstituted powdered infant formula (RPIF) model. Inhibition zones of 30 mg Narine-L and Narine-HT were both 150 U, whereas inhibition zone of 30 mg Narine-CCFE was 200 U. Narine-L (1 g) and Narine-HT (1 g) were added to 10 mL of artificially contaminated RPIF, respectively, containing 100 μL of C. sakazakii (1.62×108 colony forming unit (CFU)/mL). After treatment with Narine-L and Narine-HT for 3 h and 6 h at 37℃, less than ≤107 CFU/mL of C. sakazakii was detected in RPIF. Without Narine-L and Narine-HT treatment, the population of C. sakazakii increased up to 5.36×109 CFU/mL after 6 h. Examination by transmission electron microscopy confirmed C. sakazakii cells were damaged by Narine-CCFE. Thus, employing Narine culture as a natural and safe bio-preservative may protect infants from C. sakazakii.

Evaluation of the Influence of the Method of Sample Preparation on the Shearing Behavior of Sands using Elastic Waves (탄성파를 통한 시료성형방법에 따른 모래 전단거동특성 평가)

  • For economic and technical reasons, it is difficult to obtain high quality undisturbed cohesionless samples, hence most researchers rely on preparing remolded and reconstituted representative samples of sandy soils. In this study, moist tamping, air pluviation, and dry deposition methods were applied to make remolded samples at similar relative densities. A series of isotropically consolidated drained tests were conducted with accompanied by measured elastic wave velocities in order to evaluate a difference between sample preparation methods and relative densities. For the elastic wave velocity measurements, piezoelectric elements were installed on the top and bottom cap of the triaxial device. The results showed that soil behavior relies on sample preparation methods, and that the trend of shear wave velocity was the same with volumetric strain behavior.

  • A potent fungus producing milk clotting enzyme with fairly weak proteolytic activity was isolated from various soil and sewage, which the selected strain, SA-101, was identified as Mucor sp. with microbiological characteristics. Its milk clotting enzyme production was maximized when grown on 10g of wheat bran media added to 8ml of tap water containing 0.1M HCl for 60hrs at $30^{\circ}C$. This enzyme production was stimulated by addition of 6% lactose, 0.05% NaCl and reached a maximal level of 9810 unit/g wheat bran. The crude enzyme product could be produced effectively by salting out with ammonium sulfate fractionation and lyophilization. The ratio of milk clotting activity to proteolytic activity of crude enzyme product was lower than Hansen rennet, but resembled to Meito rennet. The optimal temperature of milk clotting activity of crude enzyme product was abound $60^{\circ}C$ on a substrate of 10% reconstituted skim milk containing 1/100M $CaCl_2$.

Fermentation and Quality Characteristics of Soy Yogurt incorporating Insoluble Components of Domestic Soybeans (국내산 대두의 불용성 물질을 포함한 두유 요구르트의 발효 및 품질 특성)

  • The objective of this research was to determine the fermentation and quality properties of soy yogurt incorporating insoluble components fermented with Streptococcus thermophilus (S. thermophilus) and Lactobacillus bulgaricus (L. bulgaricus) for 24h. Boiled soybeans were ground and reconstituted into equivalent total solids (18%). After soymilk was homogenized with sugar, it was pasteurized in a water bath at $90^{\circ}C$ for 30 min. Two kinds of lactic acid bacteria were inoculated into two types of soymilk after cooling down $40{\sim}45^{\circ}C$ and fermentation at $37^{\circ}C$ for 24h. Titrable acidity, pH, viscosity, sugar content, and number of viable cells were determined in triplicate. Soy yogurt fermented with L. bulgaricus did not reach pH 4.5, where as S. thermophilus was considered good for achieving pH 4.08 and titratable acidity of 2.27% for 24 h. Soy yogurt fermented with S. thermophilus had a higher viscosity and lower sugar contents than that fermented with L. bulgaricusas incubation time increased. Total viable count was $1.80{\times}10^{10}CFU/mL$ on S. thermophilus and $2.16{\times}10^8CFU/mL$ on L. bulgaricus after 24 h at $37^{\circ}C$. However, there was no significant difference in sensory intensities and preference between the two samples. As a result, S. thermophilus was identified as a better culture than L. bulgaricus for the manufacture of soy yogurt incorporating insoluble components.

Inhibitory actions of the antidepressant/antipanic drug phenelzine on brain GABA transaminase

  • Brain GABA transaminase is inactivated by preincubation with antidepressant/antipanic drug pheneizine (${\beta}$ethylphenylhydrazine) (mixing molar ratio 10:1) at pH 7.4. The reaction of enzyme with phenelzine was monitored by absorption and fluorescence spectroscopic methods. The inactive enzyme was fully reconstituted by addition of cofactor pyridoxal-5-phosphate. This result implies that the blocking of 1 mol of pyridoxal-5-phosphate per enzyme dimer is needed for inactivation of the enzyme. The time course of the reaction is significantly affected by the substrate .alpha.-ketoglutarate, which afforded complete protection against the loss of catalytic activity. The kinetic studies shows that phenelzine reacts with the cofactor of enzyme with a second-order rate constant of $2.1{\times}10^3M^{-1}s^{-1}$. It is postulated that the antidepressant/antipanic drug phenelzine is able to elevate the neurotransmitter GABA levels in central nervous system by inhibitory action on GABA degradative enzyme GABA transaminase.

Stability of Cytarabine Admixtures (Cytarabine 혼합조제액의 안정성 연구)

  • The stability of cytarabine (manufactured by two pharmaceutical companies) in intravenous admixture and in plastic syringe was investigated. The admixures containing cytarabine 0.35, 8.6, and 17.1 mg/ml in $0.9\%\;NaCl\;or\;5\%$ D5W were placed in PVC bags or glass bottles, and the reconstituted cytarabine (50 mg/ml) was placed in plastic syringe. One ml aliquots were withdrawn immediately after each preparation and stored at $4^{\circ}C\;or\;24^{\circ}C$ for 1, 2, 4, 6, 8, 10, and 14 days unprotected from light. Each sample was tested for pH and visually inspected for precipitation and change in color. Cytarabine concentrations were measured using high-performance liquid chromatography. Neither precipitation nor change in color was noted, and there were no change in pH during 14 days of testing. The changes in cytarabine concentrations were less than $10\%$. In conclusion, cytarabine in IV admixture or plastic syringe was stable for at least 14 days at $4^{\circ}C\;and\;24^{\circ}C.$(Kor. J. Clin. Pharm. 1998; 8(1): 59-67)

Improved Viability and Proteome Analysis of Lactobacillus fermentum KLB12 upon Heat Stress (Lactobacillus fermentum KLB12의 열 전처리에 따른 열 스트레스 내성 증진 및 프로테옴 변화)

  • In the previous study, we have isolated several vaginal lactobacilli from Korean woman and selected one of them (KLB12) for further study, which was indentified as Lactobacillus fermentum by sequence analysis of 16S rRNA gene. Formulated L. fermentum KLB12 can be used for ecological treatment of bacterial vaginosis. For pharmaceutical formulation, the spray-drying process is required where stress such as high temperature is routinely applied. In this study, we found that heat stress at 60$^{\circ}C$ for 20∼30min reduced the viable cell population of KLB12 by 10$\sub$6/~10$\sub$9/. However, adaptation of KLB12 cells at 52$^{\circ}C$ made them more thermotolerant upon exposure to 60$^{\circ}C$. The level of thermal protection in RSM (reconstituted skim milk) by adaptation in acid (pH 5), cold (4$^{\circ}C$), ethanol (3%), NaCI (0.3M) was also examined. Although not as efficient as the homologous stress, adaptations in both cold and NaCI gave considerable cross protection against heat stress. When chloramphenicol was added during heat adaptation, adaptation effect was abolished. This suggests that de novo protein synthesis is necessary during the adaptation process. Important changes in proteome during heat adaptation was examined with two-dimensional gel electrophoresis.

Studies on Wood-based Composite Panel with Waste Tire - Properties of Composite Boards in Relation to Hot Pressing Conditions - (폐타이어를 이용한 목질계 복합판넬의 연구 - 열압조건에 의한 재질특성 -)

  • The effect and control of wood property of reconstituted composite panels for improved board properties by wood-waste materials and development of combination method for heterogeneous materials have been of interest to the wood science researchers. The purpose of this study is to consider the properties in relation to hot pressing conditions and to develope the optimum hot pressing condition with waste wood and waste tire for the manufacturing of composite boards. The study of composite boards for recycling of wood and waste tire is nothing up to the present. Physical and mechanical properties such as specific gravity, moisture content, swelling coefficient, modulus of rupture and modulus of elasticity in bending test were studied. The condition of 3-stage press time for the lowest moisture content of composite board was $4{\rightarrow}3{\rightarrow}3$ minutes. Specific gravity of composite panels was affected mainly by the amount of rubber chip. Because of the low rigidity and high elasticity in rubber chip, it is considered the composite panel was adequate material in the place of compression load, but not bending load. Therefore, it was concluded that a use of rubber-based wood composite panel is proper to the interior materials such as floor a room than exterior materials. From the test results, the most optimum hot pressing conditions were $4{\rightarrow}3{\rightarrow}3$ minutes for 3-stage press time and $45{\rightarrow}20{\rightarrow}5kg/cm^2$ for 3-stage press pressure. The rubber-based wood composite panel was very excellent in elasticity by combination of rubber chip in comparison with existing other wood-based materials. Therefore, it was considered that rubber-based wood composites can be applicable to every interior materials such as floor a room and will be expected to effective reuse and recycle of waste tires and wood-waste materials, and will be contribute to protection of environment pollution in earth.

  • ATP synthase produces ATP, a major energy source for metabolic processes in organisms, from ADP and inorganic phosphate in cellular membranes. ATP synthase is known as a rotary motor, in which the c-subunit ring functions as a rotor. In this work, we have tried to develop a more general preparation procedure of thermophilic $F_oc$-ring ($TF_oc$-ring) for NMR measurements. The expression of $TF_oF_1$ is easily affected by various experimental conditions such as temperature, shape and size of a flask, a volume of medium, and shaking rate of an incubator. Accordingly, we have tried to optimize the expression conditions of $TF_oF_1$. $TF_oc$-rings were purified from $TF_oF_1$ according to a reported method. We modified purification procedures to improve purity and yield of $TF_oc$. On top of them, we found a new combination of detergents for the purification at anion-exchange column chromatography. To examine the effect of lipid environments on the structure, the $TF_oc$-rings were reconstituted into two kinds of lipid bilayers, namely, saturated and unsaturated lipid ones. Then, we have compared characteristics of the $TF_oc$-ring structures in these membranes with solid-state NMR.

Liquefaction Characteristic of Pohang Sand Based on Cyclic Triaxial Test (진동삼축시험을 통한 포항 지역 사질토의 액상화 저항 특성 연구)

  • In this study, series of cyclic triaxial tests and shear velocity measurement were conducted using Pohang sand, which was taken from liquefaction observed area, to verify the liquefaction characteristics of Pohang. The cyclic resistance ratio(CRR) was derived based on the test results. A specimen was reconstituted into 40% and 80% relative density conditions and then a series of cyclic triaxial tests and shear-wave velocity measurement were conducted. As a result, the effect of particle distribution and relative density to liquefaction resistance was verified. The liquefaction resistance of Pohang sand was evaluated by comparing with other liquefaction resistance of sands from previous research. In addition, the liquefaction resistance curve from field observation data was used to verify the reliability of results from this study by measured shear-wave velocity.
