Transcription Profiles of Human Cells in Response to Sodium Arsenite Exposure

  • Arsenic exposure is associated with several human diseases, including cancers, atherosclerosis, hypertension, and cerebrovascular diseases. In cultured cells, arsenite, an inorganic arsenic com-pound, was demonstrated to interfere with many physiological functions, such as enhancement of oxidative stress, delay of cell cycle progression, and induction of structural and numerical changes of chromosomes. The objective of this study is to investigate the effects of arsenic exposure on gene expression profiles by colorimetric cDNA microarray technique. HFW (normal human diploid skin fibroblasts), CL3 (human lung adenocarcinoma cell line), and HaCaT (immortalized human keratinocyte cell line) were treated with 5 $\mu\textrm{M}$ or 10 $\mu\textrm{M}$ sodium arsenite for 6 or 16 h, respectively. By a dual-color detection system, the expression profile of arsenite-treated cultures was compared to that of control cultures. Several genes expressed differentially were identified on the microarray membranes. For example, MDM2, SWI/SNF, ubiquitin specific protease 4, MAP3K11, RecQ protein-like 5, and Ribosomal protein Ll0a were consistently induced in all three cell types by arsenite, whereas prohibitin, cyclin D1, nucleolar protein 1, PCNA, Nm23, and immediate early protein (ETR101) were apparently inhibited. The present results suggest that arsenite insults altered the expression of several genes participating in cellular responses to DNA damage, stress, transcription, and cell cycle arrest.

  • STI CMP process are substituting gradually for LOCOS(Local Oxidation of Silicon) process to be available below sub-0.5um technology and to get planarized. The other hand, STI CMP process(especially STI CMP with RIE etch back process) has some kinds of defect like Nitride residue, Torn Oxide defect, etc. In this paper, we studied how to reduce Torn Oxide defects after STI CMP with RIE etch back process. Although Torn Oxide defects which occur on Oxide on Trench area is not deep and not sever, Torn oxide defects on Moat area is sometimes very deep and makes the yield loss. We did test on pattern wafers witch go through Trench process, APCVD process, and RIE etch back process by using an REC 472 polisher, IC1000/SUV A4 PAD and KOH base slurry to reduce the number of torn defects and to study what is the root causes of torn oxide defects.

  • Due to the international environmental regulations and changes in policies, the demand for generators using the renewable energy is increasing. However, renewable energy generators with intermittent output characteristics such as solar and wind power generators, need the buffer facilities such as ESS during system operations. However, because of low price competitiveness in energy storage system, it is difficult to operate the renewable energy generator with ESS. Therefore, the government has recently proposed a policy to compensate the REC for renewable energy system with ESS. For all this, since the initial cost of the ESS is high, it is the most important to calculate and operate the optimal capacity of the ESS through an economic analysis. In this paper, we proposed the method of calculation the optimal capacity of ESS and analyzed economic feasibility of renewable energy system using the ESS according to depreciation in ESS price.

Characterization of Lactobacilli with Tannase Activity Isolated from Kimchi

  • Tannase catalyzes the hydrolysis of gallic acid esters and hydrolysable tannins. Twenty-two Lactobacillus strains with tannase activity were isolated from 7 types of kimchi. A polymerase chain reaction-based assay targeting the recA gene assigned all isolates to either Lactobacillus plantarum or Lactobacillus pentosus. The tannase activities of isolates measured in whole cells and cell-free extracts varied even within each species. The activities of the isolates varied with the assay method, but both methods indicated that isolate LT7 (identified as L. pentosus) showed the highest activity. The results of thin layer chromatography and high performance liquid chromatography, respectively, showed that tannic acid and gallic acid degraded to pyrogallol in resting L. pentosus LT7 cells. Therefore, the putative biochemical pathway for the degradation of tannic acid by L. pentosus implies that tannic acid is hydrolyzed to gallic acid and glucose, with the formed gallic acid being decarboxylated to pyrogallol. This study revealed the possible production of pyrogallol from tannic acid by the resting cell reaction with L. pentosus LT7.

  • In order to deploy the large-scale energy storage (ES) service in the various industry, it is very important to develop a business model with high technological and economic feasibility through detailed valuation of cost and expected benefits. In relation to this, this paper established an optimal scheduling plan for electric vehicle charging stations connected with photovoltaic (PV) and ES technologies in Korea using the distributed energy resource valuation tool and analyzed the feasibility of the project. In addition, the impact of incentives such as REC (Renewable Energy Certificate) to be given to electric vehicle charging stations in accordance with the relevant laws to be revised in the future was analyzed. As a results, the methodology presented in this paper are expected to be used in various ways to analyze the feasibility of various business models linked to renewable energy and ES technologies as well as the electric vehicle market.

우리별 인공위성의 지상 촬영 장치에 쓰여진 렌즈의 MTF 측정 (MTM MEASUREMENT OF THE LENS ON THE KITSAT-1 EARTH IMAGING SYSTEM)

  • 한국과학기술원 인공위성 연구센터에서는 1992년 8월과 1993년 9월에 마이크로 위성인 우리별 1호와 2호를 발사, 운용중이다. 두 위성 모두에는 지표면을 촬영하기 위한 CCD 카메라가 실려있다. 이들 카메라에 쓰여진 렌즈(촛점거리: 50mm)를 평가하기 위해 MTF(Modulation Transfer Function) 측정을 하였다. 실험을 위해 표준과학연구소의 MTF 측정장치를 사용하였으며, f-number와 field angle을 변화시켜 가면서 측정하였다. 측정 결과로부터 f-number가 4.0 보다 작을때, 점광원으로부터 나온 빛이 CCD 칩의 한 화소(pixel) 안으로 들어 간다는 사실을 알 수 있었으며, field angle을 변화시켜가면서 tangential MTF와 sagital MTF를 측정한 결과, field angle이 증가함에 따라 MTF 값이 떨어짐을 알 수 있었다.

식물에서의 상동재조합을 이용한 효율적인 진타겟팅 시스템 (An efficient gene targeting system using homologous recombination in plants)

  • The plant breeding technology was developed with genetic engineering. Many researchers and breeders have turned from traditional breeding to molecular breeding. Genetically modified organisms (GMO) were developed via molecular breeding technology. Currently, molecular breeding technologies facilitate efficient plant breeding without introducing foreign genes, in virtue by of gene editing technology. Gene targeting (GT) via homologous recombination (HR) is one of the best gene editing methods available to modify specific DNA sequences in genomes. GT utilizes DNA repair pathways. Thus, DNA repair systems are controlled to enhance HR processing. Engineered sequence specific endonucleases were applied to improve GT efficiency. Engineered sequence specific endonucleases like the zinc finger nuclease (ZFN), TAL effector nuclease (TALEN), and CRISPR-Cas9 create DNA double-strand breaks (DSB) that can stimulate HR at a target site. RecQl4, Exo1 and Rad51 are effectors that enhance DSB repair via the HR pathway. This review focuses on recent developments in engineered sequence specific endonucleases and ways to improve the efficiency of GT via HR effectors in plants.

명태(Theragra chalcogramma) 종묘에서 분리된 Vibrio anguillarum의 생화학적, 분자생물학적 특성 (Biochemical and Molecular Characteristics of Vibrio anguillarum Isolated from Alaska Pollock Theragra chalcogramma Seedlings)

  • The health of Alaska pollock Theragra chalcogramma seedlings was monitored during February and April 2015. After microscopic examination for parasites, 20 samples sets were made by pooling 50 individuals for each sample set. Then, they were homogenized and examined for viral and bacterial pathogens. No parasites or viruses were detected using either microscopy or PCR. Colonies suspected of belonging to the genus Vibrio were isolated from Tryptic Soya Agar and Thiosulfate Citrate Bile Salts Sucrose Agar plate incubations, and identified as Vibrio anguillarum based on biochemical and physiological examinations and PCR amplification of the 16S rDNA, recA, and pyrH genes. Although there was no mortality during the sampling period, 65.0% (13/20) of the pooled samples were PCR-positive for V. anguillarum. To prevent possible outbreaks, the pathogenic potential of V. anguillarum should be investigated in the future.

Karman 경계층 유동의 안정성에 관한 연구 (Stability of the K rm n Boundary Layer Flow)

  • The Karman boundary-layer, has been numerically investigated for the disturbance wave number, wave velocity, azimuth angle and radius (Reynolds number, Re). The disturbed flow over rotating disk can lead to transition at a much lower Re than that of the well-known Type 1 mode of instability. This early transition is due to the excitation of the Type II mode. Presented are the neutral stability results concerning these modes by solving new formulated vorticity equations with consideration of whole convective terms. When the present numerical results are compared with the previously known results, the value of critical Re corresponding to Type I is moved from Rec,! =285.3 to 270.2 and the value corresponding to Type II is from $Re_{c,2}$=69.4 to 36.9, respectively. Also, the corresponding wave number is moved from $k_1$ =0.378 to $k_1$ =0.389 for Type I; from $k_2$ =0.279 to $k_2$=0.385 for Type II. For Type II, the upper limit of wave number and azimuth angle is $k_U$=0.5872,$varepsilon_U=-18^{\circ}$ , while its lower limit is$k_L$ =0.05, $varepsilon_L=-27^{\circ}$ This implies that the disturbances will be relatively fast amplified at small Re and within narrow bands of wave number compared with the previous results.

First Report of Pectobacterium aroidearum Causing Soft Rot on Ficus carica in Korea

  • In July 2021, symptoms of soft rot were observed on the stems of Ficus carica in Yeongam, Jeollanamdo, Korea. To accurately diagnose the cause, infected stem was collected and bacterial strain was isolated. Among these, the pathogenic strain KNUB-08-21 was identified as Pectobacterium aroidearum through 16S rRNA gene sequencing and phylogenetic analysis based on the concatenated sequences of the dnaX, leuS, and recA genes. The affiliation of the isolate with this bacterial species was also confirmed by its biochemical characteristics obtained using API ID 32 GN system. Artificial inoculation confirmed the strain's pathogenicity in figs, causing significant damage to both stems and fruits. To our knowledge, this is the first report of P. aroidearum causing soft rot disease in F. carica in Korea.
