Effects of Browning Reaction Products on DNA Damage (효소적 갈변 생성물의 DNA 손상에 대한 효과)

  • Antimutagenicity profiles of the enzymatic browning reaction products(EBRP) were investigated. The rec-assay with Bacillus subtilis strains $H17(rec^+)$ and $M45(rec^-)$ was carried out using their spores. The biological activities were evaluated for seven different enzymatic browning reaction products, which resulted from the reactions of seven polyphenols with polyphenol oxidase isolated from Ginkgo biloba leaves. In the spore $rec^-$ assay, most of the polyphenolic compounds tested were positive, whereas their enzymatic browning reaction products were tested negative. The mutagenicity of enzymic browning mixtures of the polyphenols and the enzymes obtained from Ginkgo biloba leaves showed negative results in the mutagenicity test using Bacillus subtilis strains $H17(rec^+)$ and $M45(rec^-)$. In the case where polyphenol oxidase inhibitors were added in the enzymatic reaction mixtures with polyphenols, the polyphenols showed mutagenic effect in the spore $rec^-$ assay. This suggests that the activity of polyphenol oxidase is decreased.

  • The antimutagenic effects of enzymatic browning reaction products (MEBRPs) of polyphenol compounds (catechol, homocatechol, hydroxyhydroquinone, pyrogallol) by enzyme extracted from mushroom (Agaricus bisporus) were demonstrated through spore rec-assay using B. subtilis $H17(rec^+)$ and $M45(rec^-)$, Ames test using S. typhimurium TA98 and TA100 and SOS chromotest using E. coli PQ37/plasmid pKM101. In spore rec-assay, the MEBRPs showed antimutagenic effects by decreasing of the inhibition zone induced by MNNG. In Ames test with S-9mix in both TA98 and TA100, all of MEBRPs showed strong antimutagenic effects of about 21 to 99% against mutation by $B({\alpha})P$ and Trp-P-1, as adding $300\;{\mu}l$ of the MEBRPs. In SOS chromotest, MEBRPs showed antimutagenic effects by inhibiting the SOS-inducing function induced by 4NQO and MMC, as increasing in concentration of the MEBRPs. But they did not showed mutagenicity in these bacterial assays.

  • Antimutagenic activity of the enzymatic browning reaction products (EBRPs) was investigated by using the spore rec-assay with Bacillus subtilis strains H17 $(rec^+)\;and\;M45 (rec^-)$. The EBRPs tested were prepared from the reactions of five different kinds of polyphenols with polyphenol oxidase isolated from the leaves Perilla frutescens. In the spore rec-assay, most of the polyphenolic compounds tested showed positive, whereas only their tested compound showed negative respectively. In addition of polyphenol oxidase inhibitors such as cysteine, glutathione and ascorbic acid to the reaction mixtures consisted with the polyphenol oxidase and polyphenols, the mutagenic effects were increased in the spore recassay. These results show that the activity of polyphenol oxidase may play an important role in the reduction of mutagenicity of polyphenols.

  • In order to investigate the antimutagenicity of the sweet potato enzymatic browning reaction products (SPEBRP) were studied the DNA breaking action, spore rec assay and Ames test. In the DNA breaking action of reaction mixture of SPEBRP and polyphenol compounds with an agarose horizonal electrophoresis, catechol (CAT)-SPEBRP and hydroxyhydroquinone (HHQ)SPEBRP inhibited DNA breaking effect in the presence of $Fe^{2+}$. In the spore ree assay using Bacillus subtilis H17(rec+) and M45(rec-), 3,4-dihydroxytoluene (DHT)-SPEBRP showed strong antimutagenic effects on MNNG. In the Ames test using Salmonella tYPhimurium TA 98 and TA 100, pyrogallol(PYR)-, 3,4-dihydroxytoluene (DHT)- and hydroxyhydroquinone (HHQ)-SPEBRPs suppressed about 67%, 71% and 63% in the mutagenesis induced by Benzo($\alpha$)Pyrene(B($\alpha$)p).

  • Gamgung-tang(GGT) that is included in Gamdu-tang(consists of Glycyrrhizae Radix, black beans) and Gunggui-tang(consists of Angelicae Radix and Cnidii Rhizoma), showed therapeutic effects of autoimmume thyroiditis in the previous reports. GGT was tested for the safety using Rec assay and enzymatic methods. In the Rec assay, Bacillus subtilis H-17$(Rec^+)$ and M-45$(Rec^-)$ strains were used to test DNA damage activity. From the results, there was no DNA damage of GGT. Hepatotoxicity of GGT to female ICR mice was also monitored by the measurements of serum(s)-GOT, s-GPT and LDH activities after oral feeding for 15 days. GGT was not shown any significant changes of s-GOT, s-GPT and LDH activities in mice sera.

  • This research was carried out to investigate antimutagent effect of peach enzymatic browning reaction products(PEBRP) obtained by reacting each of polyphenol compounds with oxidase extracted from Korea-cultivated peach. In methods, rec-assay with B. subtilis strains $H17(rec^+)\;and\;M45(rec^-)$, and Ames test with S. typhimurium TA98 and TA100 were used. The spore rec-assay of PEBRP, pyrogallol, hydroxyhydroquinone, homocatechol and caffeic acid were not showed mutagenicity. In the effects of various metal ions$(Al^{3+},\;Cu^{2+},\;Fe^{2+},\;Mn^{2+},\;Ni^{2+},\;Pb^{2+},\;Zn^{2+})$ on the rec-assay, all PEBRP except caffeic acid was increased inhibition zone(5 mm) only with $Zn^{2+}$. In paticular, the Py-PEBRP was decreased the difference of inhibition zone of growth on MMC(mitomycin C). In results of Ames test, all PEBRP were not showed mutagenicity on S. typhimurium TA98 and TA100; however, Ca-PEBRP and Hca-PEBRP were suppressed mutagenic effects on Trp-P-1 and B(a)P in the presence of S-9Mix.

  • This study was carried out to investigate the antimutagenic affect of crude and heated comfrey extract on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), benzo${(\alpha)}$pyrene (B${(\alpha)}$P) and 3-amino-1, 4-dimethyl-5H-pyri-do [4,3-b] indole (Trp-P-1). In spore rec-assay using Bacillus subtilis $H17(rec^+)$ and $M45(rec^-),$ crude comfrey extract showed strong antimutagenic effects on MNNG in the concentration of $40{\mu}l/disc$(p<0.01). In the Ames test using Salmonella typhimurium TA98 and TA100, the crude comfrey extract suppressed about 43% and 52% in the mutagenesis induced by $B{(\alpha)}P.$ However, the heated comfrey extract strongly suppressed about 75% and 76% in the mutagenesis in Salmonella typhimurium TA98 and TA100 induced by Trp-P-1(p<0.01).

  • Antimutagenic effects of boiled water extract and tannin from persimmon leaves were studied by using Ames test, spore rec assay and SOS chromotest. Strong antimutagenic activities toward aflatoxin B1(AFB1), dimethyl-aminobiphenyl(DMAB), N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitroquinoline-1-oxide(4-NQO) were observed when boiled water extract and tannin from the persimmon leaves were added in the Salmonella typhimurium TA 100. In spore rec assay using Bacillus subtilis H17($rec^{+}$) and M45($rec^{-}$), boiled water extract and tannin from the persimmon leaves considerably inhibited the mutagenesis induce by MNNG. In SOS chromotest using Escherichia coli PQ37, these samples also exhibited strong antimutagenic activity toward 4-NQO. The tannin was more effective than boiled water extract of persimmon leaves in the antimutagenicity tests.

  • In order to evaluated the mutagenic potential of Ag-Os produced by receiving Ag ion at the carrier, 2 types of mutagenecity tests were performed. No mutagenic potential was shown in bacterial reverse multation test using Salmonella typhimurim TA 1535, TA 1537, TA 98, TA 100. No DNA-damaging property was shown in Rec-assay using Bacillus subtilis(Rec+) and Bacillus subtilis (Rec-). These results indicate that the Ag-Os does not cause reverse mutation and DNA-damaging property

  • Water extract from Astragali Radix (AR) was tested for the safety using Ames, Bacillus subtilis Rec, and umu gene expression mutagenicity tests. Mutagenic activity in any assays we tested was not found. In Ames test, Salmonella typhimurium TA98 and TA 100 were used to identify mutagenic property, and the number of histidine revertants was measured. In the Recassay, Bacillus subtilis ${H-17(Rec^+)\;and\;M-45(Rec^-)}$ strains were used to test DNA damage activity. In the SOS umu test, Salmonella typhimurium TA1535 containing plasmid pSK1002 was used as a test strain, and we monitored the levels of umu operon expression by measuring the ${\beta}-galactosidase$ activity. From the results, there was no DNA damage and mutagenicity of AR. Hepatotoxicity of AR to female ICR mice was also monitored by the measurements of s-GOT, s-GPT, LDH activities after oral feeding for 15 days. AR was not shown any significant changes of s-GOT, s-GPT and LDH activities in mice sera.
