Signal Analysis of Optical Biosensor to Detect Peroxide Using Electrically Controlled Release System (전기적 방출 조절 시스템을 이용한 광 페록사이드센서의 개발)

  • The optical biosensor using the electrically controlled release of reactive reagent is developed for the detection of peroxide. Rapid degradation of polymer complex of PEOx and PMAA occurs as the applied current increases and thus released amount of HPA increases. The degradation velocity of polymer and the amount of HPA released are linearly proportional to the applied current. Peroxide is reacted with the released reagent by peroxidase and then the product, a fluorescent dimer DBDA, is formed. The monochromic light from light source (150W Xe arc ramp) excites the DBDA and the excited light is transmitted through an optical fiber to be detected by a photodiode array. The change of fluorescence intensity is related to the change of peroxide concentration. The peroxidase is entrapped in Ca-alginate get on the inner surface. The biosensor has the linear signal range of 0.025mM-10.mM peroxide. By applying the step function of peroxide, reproducibility of biosensor has been investigated. The mathematical model is constructed by the combination of enzyme kinetics with reactor flow model. Good agreement is obtained between the experimental result and model prediction in the sensor signal.

Pretreatment of Hyperbaric Oxygenation Increases the Activities of Myocardial Antioxidant Enzymes and Protects the Ischemia-Reperfusion Injury of the Heart (고압산소 전처치의 심근 항산화효소 활성 증가 및 허혈-재관류손상 보호 효과)

  • Myocardial ischemia-reperfusion injury is known to be mediated by reactive oxygen species. The myocardial cell is equipped with endogenous antioxidant defensive system which can be adaptively stimulated by various oxidative stress. It is postulated that an increased oxygen partial pressure induced by hyperbaric oxygenation impose an oxidative stress on the cells, resulting alterations in the endogenous antioxidant system. In this study we investigated the effect of hyperbaric oxygenation on the activities of myocardial antioxidant enzymes and observed whether the hyperbaric oxygenation could protect the ischemia-reperfusion injury of heart. Rats or rabbits were pretreated with hyperbaric $oxygenation(2{\sim}3\;atm\;O_2/1{\sim}3\;hrs/1{\sim}10\;days)$. The changes in activities of major antioxidant enzymes(superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, glucose-6-phasphate dehydrogenase), functional recovery and infarct size were observed in the experimentally induced ischemia-reperfused hearts. In the hearts isolated from rats pretreated with $2\;atm\;O_2/1{\sim}2\;hrs$ for 5 days, the functional recovery after reperfusion(20 min) following global ischemia(25 min) was significantly increased without any observable oxygen toxicity. Lactate dehydrogenase release was also significantly reduced in this hyperbaric oxygenated rat hearts. In in vivo regional ischemia(30 min) model of rabbit hearts, pretreatrment with $2\;atm\;O_2/1\;hr$ for 5 days significantly limited the infarct size. Among the myocardial antioxidant enzymes of rat hearts pretreated with the hyperbaric oxygenation, the activities of catalase, superoxide dismutase and glucose-6-phosphatase dehydrogenase were increased, while those of glutathione peroxidase and reductase were not changed. There were lethal cases in the groups of rats exposed to 3 atm $3\;atm\;O_2/2{\sim}3\;hrs$ for 5 days. A lipid-peroxidation product, rnnlondialdehyde was increased in brains and livers of the rats exposed to$2\;atm\;O_2/2{\sim}3\;hrs/5\;days\;and\;3\;atm\;O_2/1\;hr/5days$. The present results suggest that the pretreatment of hyperbaric oxygenation can protect the post-ischemic rererfused hearts in association with a stimulation of the activities of myocardial antioxidant defensive enzymes, and that the hyperbaric oxygenation of $2\;atm\;O_2/1\;hr$for 5 days would be a safe condition which does not produce any oxygen toxicity.

  • Antioxidant enzymes, scavengers of the reactive oxygen intermediate (ROI), are involved in numerous defense systems in cells. In the present study, we investigated the effects of the hot-water extracts of two medicinally potent mushrooms (Ganoderma lucidum and Phellinus linteus) on the activity and expression of antioxidant enzymes in vitro and in vivo. The mushroom extracts stimulated the catalase activity in a dose-dependent manner in vitro, whereas the other antioxidant enzymes (such as superoxide dismutase (SOD), glutathione peroxidase (GPx)) were unaffected by the extracts. The catalytic activity of catalase in the liver and brain was significantly increased after the oral treatment of the mushroom extracts (2.5 g/kg) to ICR mice for 2 months. Western blot analysis of the liver and brain tissues revealed that the expression level of catalase in the mice, treated with both mushroom extracts, was significantly increased compared to that of the control mice. However, the level of the SOD expression in the mice treated with the natural product extracts was unchanged under the same experimental conditions. Although the mechanisms for the stimulatory effect of the catalase expression by these extracts remains unclear, these results suggest that the ingredients of the Ganoderma lucidum and Phellinus linteus extracts act as an activator of catalase, and regulate the expression of catalase at the translational or transcriptional level.

Antioxidant Enzyme Activity and Cell Membrane Stability of Korean Bermudagrass Genotypes Different in Ploidy at Dormant Stage (배수성이 다른 자생 버뮤다그래스의 휴면 전후 항산화 효소활성 및 세포막 안정성 변화)

  • Korean bennudagrass collections showed diverse genetic variations in their morphology, growth habit, and cytological aspects. Chromosome number and nuclear DNA content of the bennudagrasses indicated a ploidy level ranging from triploid (2n=3x) to hexaploid (2n=6x). In this study, we investigated the different responses of antioxidant enzymes (superoxide dismutase, catalase, peroxidase, ascorbate peroxidase) and cell membrane stability of those bennudagrass cytotypes to lower temperature and shorter day length, which meets a dormant induction in Korea. All the antioxidant enzymes were found to be higher during dormant stage, while the heme-containing catalase which converts hydrogen peroxide ($H_2O_2$) to water and oxygen molecules was activated before dormant initiation in the three cytotypes except for hexaploid bennudagrass. The triploid and tetraploid which exhibited relatively finer leaves and a rapid establishment speed were found to show increased activities of superoxide dismutase and peroxidase enzyme. The malondialdehyde(MDA) which is a product of lipid peroxidation in the cell membrane damaged by the hydroxyl radical was increased in all cytotypes as temperature declined, and tri- and tetraploids which had more protective antioxidant enzymes demonstrated a significantly lower MDA production. Similarly electrolyte leakage was higher in penta- and hexaploidy, seemingly more damage to cell membrane when low temperature was implemented. Results indicated that antioxidant responses of different cytotypes were genetically specific, which needs to be investigated the relevance with the low temperature tolerance in the bermudagrass further at the molecular level.

Numerical simulation of gasification of coal-water slurry for production of synthesis gas in a two stage entrained gasifier (2단 분류층 가스화기에서 합성가스 생성을 위한 석탄 슬러리 가스화에 대한 수치 해석적 연구)

  • Oxy-gasification or oxygen-blown gasification, enables a clean and efficient use of coal and opens a promising way to CO2 capture. The coal gasification process of a slurry feed type, entrained-flow coal gasifier was numerically predicted in this paper. The purposes of this study are to develop an evaluation technique for design and performance optimization of coal gasifiers using a numerical simulation technique, and to confirm the validity of the model. By dividing the complicated coal gasification process into several simplified stages such as slurry evaporation, coal devolatilization, mixture fraction model and two-phase reactions coupled with turbulent flow and two-phase heat transfer, a comprehensive numerical model was constructed to simulate the coal gasification process. The influence of turbulence on the gas properties was taken into account by the PDF (Probability Density Function) model. A numerical simulation with the coal gasification model is performed on the Conoco-Philips type gasifier for IGCC plant. Gas temperature distribution and product gas composition are also presented. Numerical computations were performed to assess the effect of variation in oxygen to coal ratio and steam to coal ratio on reactive flow field. The concentration of major products, CO and H2 were calculated with varying oxygen to coal ratio (0.2-1.5) and steam to coal ratio(0.3-0.7). To verify the validity of predictions, predicted values of CO and H2 concentrations at the exit of the gasifier were compared with previous work of the same geometry and operating points. Predictions showed that the CO and H2 concentration increased gradually to its maximum value with increasing oxygen-coal and hydrogen-coal ratio and decreased. When the oxygen-coal ratio was between 0.8 and 1.2, and the steam-coal ratio was between 0.4 and 0.5, high values of CO and H2 were obtained. This study also deals with the comparison of CFD (Computational Flow Dynamics) and STATNJAN results which consider the objective gasifier as chemical equilibrium to know the effect of flow on objective gasifier compared to equilibrium. This study makes objective gasifier divided into a few ranges to study the evolution of the gasification locally. By this method, we can find that there are characteristics in the each scope divided.

Protective Effect of Flavonoids on Lymphocyte DNA Damage Using Comet Assay (Comet Assay를 이용한 Flavonoids와 항산화 비타민의 인체임파구 세포 DNA 손상 보호 효과)

  • The present study was attempted to investigate and compare the antioxidant potency of several well-know flavonoids, antioxidant vitamin and commercially available popular beverages. The antioxidant potency was assessed by the effect on reducing oxidative DNA damage of human lymphocytes. Cellular oxidative DNA damage was measured by SCGE (single-cell gel electrophoresis), also known as comet assay. Lymphocytes were pre-treated for 30 minutes with wide ranges of doses of apigenin, kaempferol, luteolin, myricetin, rutin, quercetin, $\alpha$-tocopherol (10,25,50,100,200,500,1000 $\mu$M) ,green tea extract or grape juice (10,50,100,250,500,1000 $\mu$g/mL) followed by a $H_2O$$_2$(100 $\mu$M) treatment for 5 min as an oxidative stimulus. The physiological function of each antioxidant substance on oxidative DNA damage was analyzed as tail moment (tail length $\times$ percentage migrated DNA in tail) and expressed as relative DNA damage score after adjusting by the level of control treatment. Cells treated with $H_2O$$_2$alone (positive control) had an extensive DNA damage compared with cells treated with phosphate buffered saline (PBS, negative control) or pre-treated with all the tested samples. Of all the six flavonoids, quercetin was the most potent antioxidant showing the lowest $ED_{50}$/ of 8.5 $\mu$g/mL (concentration to produce 50% protection of relative DNA damage). The antoxidant potency of individual flavonoids were ranked as follows in a decreasing order; luteolin (18.4 $\mu$g/mL), myricetin (19.0 $\mu$g/mL) , rutin (22.2 $\mu$g/mL) , apigenin (24,3 $\mu$g/mL) , kaempferol (25.5 $\mu$g/mL). The protective effect of $\alpha$-tocopherol was substantially lower (highest $ED_{50}$value of 55.0 $\mu$g/mL) than all the other flavonoids, while the protective effect was highest in green tea and grape juice with low ED5O value of 7.6 and 5.3, respectively. These results suggest that flavonoids, especially quercetin, and natural compounds from food product, green tea and grape juice, produced powerful anti-oxidative activities, even stronger than $\alpha$-tocopherol. Taken together, supplementation of antioxidants to lymphocytes followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species.

Effect of Inorganic Admixture for Magnesia Cement Using MgCO3 and Serpentine (MgCO3와 사문석을 사용한 마그네시아 시멘트의 무기 첨가제 영향)

  • The carbon dioxide($CO_2$) released while producing building materials is substantial and has been targeted as a leading contributor to global climate change. One of the most typical method to reducing $CO_2$ for building materials is the addition of slag and fly ash, like pozzolan material, while another method is reducing $CO_2$ production by carbon negative cement development. The MgO-based cement was from the low-temperature calcination of magnesite required less energy and emitted less $CO_2$ than the manufacturing of Portland cements. It is also believed that adding reactive MgO to Portland-pozzolan cements could improve their performance and also increase their capacity to absorb atmospheric $CO_2$. In this study, the basic research for magnesia cement using $MgCO_3$ and magnesium silicate ore (serpentine) as main starting materials, as well as silica fume, fly ash and blast furnace slag for the mineral admixture, were carried out for industrial waste material recycling. In order to increase the hydration activity, $MgCl_2$ was also added. To improve hydration activity, $MgCO_3$ and serpentinite were fired at $700^{\circ}C$ and autoclave treatment was conducted. In the case of $MgCO_3$ as starting material, hydration activity was the highest at firing temperature of $700^{\circ}C$. This $MgCO_3$ was completely transferred to MgO after firing. This occurred after the hydration reaction with water MgO was transferred completely to $Mg(OH)_2$ as a hydration product. In the case of using only $MgCO_3$, the compressive strength was 3.5MPa at 28 days. The addition of silica fume enhanced compressive strength to 5.5 MPa. In the composition of $MgCO_3$-serpentine, the addition of pozzolanic materials such as silica fume increased the compression strength. In particular, the addition of $MgCl_2$ compressive strength was increased to 80 MPa.

Evaluation for Applicability as the Inorganic Binder with Rapid Setting Property for Construction Material of LFS Produced from Various Manufacturing Process (다양한 철강제조공정에서 부산되는 전기로 환원슬래그의 급경성 무기결합재로의 적용성 검토)

  • The Ladle Furnace Slag, about 20% of the electric arc furnace slag, has high content of free CaO and free MgO, which generates the expansion collapse by hydration reaction. Although many researchers have been endeavoring to recycle the EAF reducing slag in construction fields, there is not found the effective recycling method up to now. However, the LFS(Ladle Furnace Slag) contains mineral composition of the system of calcium aluminate with high-reactivity. Therefore, it is possible to developed the quick setting property and the high strength at the early age by the rapid cooling. This study aimed to check the reactive minerals and predict the reactivity with water on the LFS discharged from different steel product plants. The test results show that many types of LFS has hydration reactivity and can use in construction field as a inorganic binder with the rapid setting property.

Effects on Quality Characteristics of Pork Loin Fed with Wild Grape(Vitis amurensis Ruprecht) Wine By-product (머루주 박의 급여가 돈육 등심의 품질에 미치는 영향)

  • This study was carried out to investigate the feeding effect of wild grape wine by-products on pork qualities. The chemical composition, volatile basic nitrogen, thiobarbituric acid reactive substances and pH were not significantly different between control and wild grape pork, but the cholesterol and roast loss of wild grape polk. were lower than those of the control, and 1l1e salt soluble protein extractability of the control was lower than that of the wild grape pork. The Hunter's $a^{\ast}$ and $b^{\ast}$ value of wild grape polk. meat were higher than the control, the $a^{\ast}$ value of wild grape polk. meat was higher than the control, but the $L^{\ast}$ value of meat and fat were not significantly different between control and wild grape pork. The cohesiveness, gumminess and chewiness of control(respectively $66.2\%$ 428kg and 189g) were higher than wild grape polk. (respectively $61.4\%$ 357 kg and 154 g). The total amino acid composition of polk. were not significantly different between control and wild grape wine, The glutamic acid, leucine, arginine and aspartic acid were major amino acids in control and wild grape pork. The arachidonic acid $(C_{20:4})$, EPA $(C_{20:5})$ and DHA $(C_{22:6})$ of wild grape polk. were higher than those of the control. The taste (p<0.001), aroma (p<0.05), flavor (p<0.001), juiciness (p<0.01) and palatability (p<0.01) of wild grape polk. were higher than those of the control, but the texture between control and wild grape polk. were not significantly different.

Effects of Dietary Bio Ethanol By-product and Complex Enzyme on Meat Quality of Pork Loin (바이오 에탄올 부산물 DDGS와 복합 효소제 첨가급여가 돈육의 품질 특성에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary complex enzyme (${\beta}$-mannanase 800 IU/kg and xylanase 700 IU/kg) in a diet containing corn distiller's dried grain with soluble (DDGS) on meat quality and pork fatty acid composition. Ninety-six pigs ([(Landrace${\times}$Yorkshire)${\times}$Duroc], with an average body weight of 68.77 kg were used in the 8 wk growth assay. Dietary treatments included 1) corn-soybean meal diet, 2) corn-soybean meal diet + 0.05% enzyme complex, 3) cornsoybean meal diet with DDGS and 4) corn-soybean meal diet with DDGS + 0.05% enzyme complex. The pigs were allotted randomly into pens (n=4 per pen) with six replicate pens per treatment by a completely randomized design. Pigs were slaughtered at the end of the experiment and the loin muscle was obtained for meat quality. Meat pH (p<0.01), firmness (p<0.01) and redness (p<0.05) were higher in DDGS-supplemented diet than in the corn-soy bean meal diet. However, color, marbling, lightness, yellowness, thiobarbituric acid reactive substances, water holing capacity, driploss, cooking loss and loin muscle area were not significantly different among the diets. The pigs fed the DGGS-supplemented diet had higher total unsaturated fatty acids (UFA) and total UFA/saturated fatty acid (SFA) ratio of loin and backfat. The results indicate that a diet containing DDGS can influence pH, firmness, redness and total UFA concentration and total UFA/SFA ratio of meat and backfat, but that enzyme addition has no affect on meat quality.
