Phylogenetic relationships and distribution of Gelidium crinale and G. pusillum (Gelidiales, Rhodophyta) using cox1 and rbcL sequences

  • The taxonomic distinctiveness and cosmopolitan distributions of the red algae $Gelidium$ $crinale$ and $G.$ $pusillum$ remain unclear. Both species were first described in Devon in southwestern England; namely in Ilfracome for $G.$ $crinale$ and Sidmouth for $G.$ $pusillum$. We analyzed mitochondrial $cox$1 and plastid $rbc$L sequences from specimens collected in East Asia, Australia, Europe and North America. In all phylogenetic analyses of $cox$1 and $rbc$L sequences, $G.$ $crinale$ was distinct from congeners of the genus. The analyses also revealed a sister relationship with the $G.$ $coulteri$ and $G.$ $capense$ clade. Nineteen $cox$1 haplotypes were identified for $G.$ $crinale$, and they were likely geographically structured. Despite the distinctiveness in both $cox$1 and $rbc$L datasets, the sister relationship of $G.$ $pusillum$ in the genus was not resolved. Our $cox$1 and $rbc$L datasets indicate that $G.$ $crinale$ is a cosmopolitan species, found in East Asia, Australia, Europe and North America, while the distribution of $G.$ $pusillum$ is restricted to Europe and Atlantic North America. Our results suggest that infraspecific classification of $G.$ $pusillum$ may be abandoned.

The Occurrence of Griffithsia okiensis (Ceramiaceae, Rhodophyta) from Korea on the Basis of Morphology and Molecular Data

  • Despite continued studies on red algal flora in Korea, the taxonomy of the tiny ceramiaceous algae has received little attention. We report for the first time Griffithsia okiensis from Korea on the basis of morphology and molecular data. The species is small in thalli height (0.3-1.5 cm), and in diameter of vegetative cells (50-500 μm), and the ratio of cell length/breadth is 2-3 times. It has two carpogonial branches from the supporting cell of procarp. We generated psbA and rbcL sequences from ten specimens of G. okiensis isolated from Korea and Japan and from one G. japonica species isolated Japan. Eight specimens of G. okiensis from Korea were almost identical in both psbA and rbcL regions, nevertheless they differed from Japanese specimens by 4 ucleotides in psbA and 7 in rbcL. In all analyses of psbA, rbcL, and psbA + rbcL data sets, G. okiensis was determined to be a different species from G. japonica isolated from Japan, although both species showed a sister relationship. For all that extensive collection trips, we found no evidence for the occurrence of G. japonica in Korea.

Subcloning and Sequencing of Maize rbcL Promoter Region

  • pRLYS1 containing intact rbcL gene of maize (Zea mays L. cv Golden X Bantam T-51; Zm-A) was digested with several restriction enzymes to construct subcones carrying promoter region of rbcL. The DNA fragments of 0.20, 0.19, 0.92 and 1.55 kb among the EcoRI digests, the EcoRI-DdeI digests, the AvaI digests and the EcoRI-BamHI digests of pRLYS1 were subcloned into pBluscriptSK+and named pRLPS2, pRLPS3, pRLPS14 and pRLPS35, respectively. Four subclones contain the 1.92 kb portion from 136 nucleotide downstream to 1780 nucleotide upstream from the ATG initiation codon of rbcL gene. pRLPS2 (-29 to -229) and pRLPS3 (-239 to -420 from the ATG) were sequenced. When nucleotide sequence of Zm-A was compared with sequence of rbcL promoter region of a different cultivar of maize (Zea mays L. cv WFG TMS X BS7; Zm-B), the difference rate between two cultivars was 4.3%. The mean of sequence divergence between Zm-A and three grass species in the same tribe, Andropogoneae, in the upstream region from 29 to 420 of ATG was 1.8%, whereas between Zm-B and above-mentioned three species was 5.4%. Therefore, Zm-A seems to evolutionarily closer to three other species in Andropogoneae tribe than Zm-B is.

PCR-aided RFLP기술을 이용한 인삼의 DNA분석 (DNA Analysis of Ginseng Using PCR-aided RFLP Technology)

  • 본 연구는 DNA수준에서 인삼의 종내 및 종간 개체간의 유전변이를 확인할 수 있는 새로운 방법인 PCR-aided RFLP를 사용하여 품종육성의 기초자료로 삼고자 수행하였다. 인삼의 엽록체 DNA중 psbA gene과 rbcL gene을 제한효소처리하여 그 band 양상을 조사하고자 하였다. Chloroplast DNA 중 psbA gene과 rbcL gene을 분리하기 위하여 각각 psbA-N, psbA-C primer 및 rbcL-N, PX-1 primer를 사용한 결과 적정 분자량인 psbA gene은 1,008bp에서, rbcL gene은 1,336 bp에서 band가 나타났다. 또한 atpB gene, rpoB gene, trn gene을 분리하기 위한 primer를 사용한 결과 역시 예상 대로 1,366bp, 900bp, 1,500bp, 1,008bp에서 band가 나타났다. PCR에 의하여 분리한 psbA gene과 rbcL gene을 sau3A, Taq1, Alu1, HaeIII 등의 제한효소로 절단하여 RFLP양상을 조사한 결과 모든 인삼에서 TaqI 제한효소 처리구에서 KG Line 과 종 및 변종간 모두 절단이 되었으며 800bp에서 band가 위치하고 있다. AluI의 제한효소 처리구에서도 KG Line과 유전자원에서 800bp인 동일한 band를 보였다. 제한효소 HaeIII에서는 KG Line의 경우 500bp의 위치에서 희미하게 band를 동일하게 보였다. 그러나 유전자원에 있어 HaeIII 제한효소처리구에서는 band가 관찰되지 않아 KG Line과 차이를 보였다. 모든 chloroplast gene은 PCR 증폭에 의하여 밴드를 형성하였으나 제한효소 처리후 각 인삼 종내 또는 종간 식별이 용이하지 않아서 좀 더 많은 제한효소를 사용하거나 증폭된 DNA를 염기서열을 분석하여 비교하는 방법이 고려 되어야 할 것으로 사료된다.

Hybrid-RBC와 BAF의 조합공정을 이용한 고농도 도축폐수의 처리 특성 (High Strength Slaughter Wastewater Treatment in a Novel Combined System of Hybrid-Rotating Biological Contactor and Biological Aerated Filter)

  • 본 연구에서는 부착/부유 미생물 혼재형 RBC (Rotating Biological Contactor)와 BAF (Biological Aerated Filter) 공정이 가지는 각각의 장점을 조합한 호기성 생물막 처리 시스템을 고안하여 고농도 도축폐수의 처리 특성을 평가하였다. 본 공정은 상당량의 부유 미생물이 함께 있는 RBC와 침전지 그리고 BAF 공정 순으로 구성되었다. 첫 번째 공정인 RBC와 침전지에서는 유기물의 산화, 질소의 질산화/탈질 및 부유물질의 제거가 이루어지며, 후속 공정인 BAF에서는 일부 제거되지 않은 유기물과 질소의 산화 및 부유물질의 여과가 이루어진다. 돈까스 소스(시판용)와 돼지피을 이용하여 모사한 고농도 도축폐수(TCOD $5.2{\sim}40.4g/m^2{\cdot}d$, TN $0.44{\sim}4.17g/m^2{\cdot}d$)의 처리 특성을 평가한 결과, 침전지를 포함한 RBC 공정에서 Soluble COD와 $NH_3-N$의 평균 제거율은 각각 90%와 82% 이상으로 양호하였으나 TCOD와 TN은 Suspended Solid (SS)의 대량 유출과 돼지피에서 기인한 콜로이드 물질 생성 때문에 각각 60%와 69%의 다소 낮은 제거율을 보였다. 후속 공정인 BAF가 잔존 TCOD와 TN을 제거하는 생물 반응조의 역할과 SS를 제거하는 여과기의 역할을 충분히 수행해 약 100 mg/L의 TCOD와 약 140 mg/L의 SS 추가 제거가 가능하였으나, 처리수질은 TCOD 300 mg/L, SS 180 mg/L 그리고 TN 53 mg/L로 상당히 높았다. RBC 유출수에 Polyaluminium Chloride를 투입한 결과, 침전성이 크게 향상되어 RBC+침전지 공정 유출수의 TCOD와 TN은 각각 93.8%, 25.6%의 제거율을 보였으며, BAF 유출수 수질은 TCOD 16.5 mg/L, SS 0 mg/L, NH3-N 12.0 mg/L, TP 1.3 mg/L로 우수하였다. 따라서 별도의 추가 처리공정 없이 본 연구에서 고안한 RBC+BAF 조합공정에 의한 처리만으로 고농도 도축폐수를 성공적으로 처리할 수 있었다.

Phylogenetic Relationships of Soranthera ulvoidea (Chordariaceae, Phaeophyceae) on the Basis of Morphology and Molecular Data

  • The brown algal family Chordariaceae sensu lato is a focus of taxonomy because recent studies suggest a broad concept of the family, including genera formerly classified in the Dictyosiphonales. Using morphology, plastid rbcL and nrDNA ITS sequences, we evaluated relationships of the monotyic genus Soranthera (S. ulvoidea), which has been classified in the Punctariaceae. The species occurs in Bering Sea and Aleutian Islands, Alaska to Baja California. Thalli are globose to lobed, hollow, 3-5 cm in diameter, and covered with evenly distributed sori. However, two forms within the species are recognized: f. ulvoidea for globose forms and f. difformis for lobed forms. Plastid rbcL and nuclear ITS region sequences were newly determined in samples of S. ulvoidea from the Pacific coast of the North America. We found little variations in the ITS sequences among samples of S. ulvoidea from five different locations and in the rbcL region from two different locations. These results do not support previous classification of f. ulvoidea and f. difformis within the species. All analyses of our rbcL sequence dataset show that Soranthera was placed in the Chordariaceae s.l., but more related to Botrytella than Punctaria and Asperococcus.

Waxy와 atpB-rbcL 염기서열 분석에 의한 Coryloideae의 계통 유연관계 (Phylogenetic relationships of Coryloideae based on waxy and atpB-rbcL sequences)

  • Coryloideae 35집단에 대한 계통 유연관계를 알아보기 위하여 핵 DNA의 waxy 유전자와 엽록체 DNA의 atpB-rbcL intergenic spacer 지역에 대한 염기서열을 분석하였다. Waxy 유전자 분석에서 Coryloideae의 4개 속은 단계통군을 형성하였으며, 개암나무속은 단계통군을 형성하면서 군내군의 가장 기부에 분계조(clade)를 형성하였다. Ostryopsis속은 서어나무속과 새우나무속을 위한 자매군을 형성하였으며, 새우나무속 역시 단계통군을 형성하였다(BS=86, PP=99). AtpB-rbcL 분석에서는 Ostryopsis속이 아과 내에서 가장 기부에 분계조를 형성하였다. 개암나무속은 서어나무속-새우나무속과 Ostryopsis속의 중간에 위치하였고(BS=98, PP=100), 서어나무속은 새우나무속 분류군들과 함께 높은 지지도(BS=100, PP=100)를 가지고 하나의 clade를 형성하였다. Waxy 유전자 분석에서 서어나무속의 Carpinus절은 단계통을 형성하였지만 Distegocarpus절은 병계원군(paraphyletic group)으로 나타났다. 개암나무속의 경우는 2개의 subclade를 형성하였지만 속내 절 또는 아절 등의 분류계급과는 일치하지 않았다. AtpB-rbcL 분석에서는 대부분의 분류군들이 각각의 clade내에서 polytomy를 형성하여 차이를 보였다. 이상의 결과에서 두 유전자의 계통분석 결과가 뚜렷하게 불일치하였고, 특히 Ostryopsis속의 위치와 새우나무속의 단계통 여부, 그리고 서어나무속과 새우나무속의 유연관계가 가장 큰 차이를 보였다. 따라서 이들에 대한 추가적인 연구가 필요한 것으로 판단된다.

범용성 DNA 바코드(matK, rbcL) 분석을 통한 독활(獨活) 유전자 감별용 Marker Nucleotide 발굴 (Identification of Marker Nucleotides for the Molecular Authentication of Araliae Continentalis Radix Based on the Analysis of Universal DNA Barcode, matK and rbcL, Sequences)

  • Objectives : Araliae Continentalis Radix and Angelicae Pubescentis Radix have been used as the same medicinal name Korean and Chinese traditional medicines, respectively. The authentic Araliae Continentalis Radix is described only the root of Aralia continentalis in the Korean Pharmarcopoeia. However, the dried root of Angelica biserrata, Levisticum officinale, or Heracleum moellendorffii also has been distributed adulterants of Araliae Continentalis Radix. To develop a reliable method for identifying Araliae Continentalis Radix from adulterants, we carried out the analyses of universal DNA barcode sequences.Methods : Four plants species were collected from different habitate and nucleotide sequences of matK and rbcL were analyzed. The species-specific sequences and phylogenetic relationship were estimated using entire sequences of two DNA barcodes, respectively.Results : In comparative analysis of matK sequences, we were identified 104 positions of marker nucleotide for Ar. continentalis, 3 for An. biserrata, 4 for L. officinale and 8 for H. moellendorffii enough to distinguish individual species, respectively. Furthermore, we obtained marker nucleotides in rbcL at 42 positions for Ar. continentalis, 5 for An. biserrata and 2 for H. moellendorffii, but not for L. officinale. The phylogenetic tree of matK and rbcL were showed that all samples were clustered into four groups constituting homogeneous clades within the species.Conclusions : We confirmed that species-specific marker nucleotides of matK sequence provides distinct genetic information enough to identify four species. Therefore, we suggest that matK gene is useful DNA barcode for discriminating authentic Araliae Continentalis Radix from inauthentic adulterants.

바베시아 원충감염 예방과 치료에 대한 비타민 E의 응용 (Effects of vitamin E on prevention and treatment of canine babesiosis)

  • This study was carried out to investigate the effects of vitamin E on the prevention and treatment of Babesia gibsoni. Fifteen mongrel dogs, uninfected with Babesia spp, were assigned to three groups according to vitamin E(${\alpha}-tocopherol$) concentrations in the RBC. The concentrations in each of the three groups were, respectively : ${\alpha}-tocopherol$ in RBC less than $30{\mu}g/{\mu}l$(Group I), $30{\mu}g/{\mu}l{\sim}60{\mu}g/{\mu}l$(Group II), more than $60{\mu}g/{\mu}l$(Group III). Artificial infection was accomplished by injecting $2{\times}10^7{\sim}2{\times}10^8$ erythrocyte of Babesia gibsoni-infected dog into the cephalic vein. We investigated the clinical signs, vitamin E concentrations in RBC and serum, Vitamin A concentrations in serum, hematological values, white blood cell(WBC) viability and RBC membrane osmotic fragility after infection of Babesia gibsoni for a period of 20 days at 5 day intervals. The results obtained are summarized as follows : 1. After infection by Babesia gibsoni, clinical examination revealed depression, anorexia, pale mucous membranes, dark brown urine and diarrhea in proportion as time went on. After 10 days of infection, one dog each of Groups I, II and III revealed depression and anorexia. Two dogs in Group I and one dog each of Groups II and III showed dark brown urine after 15 days. Diarrhea was observed in one dog in each of the 3 groups after 20 days of infection. 2. After 5 days of infection, two dogs in each of Groups I, II and III showed Babesia gibsoni in RBC of blood smear stained with Giemsa. At the 15th day after infection with Babesia gibsoni, they were observed in all experimental animals. After both 5 days and 10 days of infection, the rate of Babesia gibsoni parasitized RBC(permillage, ‰) was 1‰, and increased as time went on. 3. After 5 days of infection by Babesia gibsoni, Group I, which had the lowest vitamin E concentration, showed significantly decreased RBC and PCV levels(p < 0.01). Group II and group III also showed significantly decreased RBC and PCV levels after 15 days of infection(p < 0.05). Particularly after 10 days of infection, Group I showed lower values in RBC and PCV levels compared to Groups II and III. WBC, RBC, fibrinogen and total protein levels between the groups did not differ during experimental periods. 4. According to the WBC differential counts, the ratios of neutrophil to lymphocyte showed a tendency to be slightly higher in Group III (more than $60{\mu}g/{\mu}l$) than in Groups I and II. 5. WBC viability did not differ between the groups. 6. RBC membrane osmotic fragility did not differ between the groups.

Reinstatement of Gracilariopsis chorda (Gracilariaceae, Rhodophyta) Based on Plastid rbcL nad Mitochondrial cox1 Sequences

  • Two different molecular markers, the plastid rbcL and mitochondrial cox1 genes, were used to define the taxonomic position of the northwest Pacific Ocean species currently named Gracilaria chorda. We analyzed both genes (1,222 bp for rbcL and 1,245 bp for cox1) from 18 specimens collected in Korea, Japan, and China. Phylogenetic reconstruction revealed that this organism should be classified in the genus Gracilariopsis, rather than in the Gracilaria. Thus, Gracilariopsis chorda (Holmes) Ohmi is the legitimate name for Gracilaria chorda Holmes. Within the species, the sequences differed by 8 bp (0.7%) in rbcL and 5 bp (0.4%) in cox1. Six haplotypes of cox1 tended to be geographically organized. Gp. chorda is characterized by coarse, elongate terete axes, short filiform branchlets usually at irregular intervals, an abrupt transition in cell size from medulla to cortex, cystocarps without tubular nutritive cells connecting the gonimoblast to the upper pericarp, and relatively large gonimoblast cells of the cystocarp in the specimens collected from Wando in southern Korea.
