Effects of Sub-freezing Systems on the Freshness of Pork Loin, Beef Loin and Tuna (냉각방식에 따른 반냉동 돼지고기, 쇠고기와 참치의 신선도 변화)

  • Effects of different types of sub-freezing methods on quality of raw pork loin, beef loin and tuna were studied. Storage tests were undertaken as follows; the three different types of sub-freezing methods, such as regular cold chamber at $-1^{\circ}C$, air-blast sub-freezing at $-5^{\circ}C$ and sharp-plate sub-freezing at $-5^{\circ}C$, were evaluated for extending freshness of samples. Pork loin packed with polyethylene-film showed no significant differences (P<0.05) in physico-chemical properties among the above sub-freezing methods. Air-blast sub-freezing methods revealed faster evaporation of moisture from samples. The same sub-freezing method also showed increased VBN values of pork loin and tuna samples. Regular cold chamber resulted in increased TBA values and K values of beef loin and tuna, respectively. It appeared that sharp-plate freezing system kept the food samples with the least loss of freshness among the above three sub-freezing methods. These results indicated that freshness of raw pork, beef and tuna could be significantly extended by the sharp-plate sub-freezing method.

Study on Processing Quality of Different Parts of Pork and Beef (돈육 및 우육 부위별 가공적성 연구)

  • Purpose: The objective of this study was to investigate the quality characteristics of pork and beef meat according to species (pork: modern genotype pork, Korean native black pork; beef: Holstein, Korean native cattle) and cuts (pork: shoulder, ham, loin; beef: loin, tenderness, round). Methods: The moisture content, protein content, fat content, ash content, fatty acid compositions pH, whater holding capacity, cooking loss, shear force, color, and sensory characteristics were measured in triplicate. Results: The moisture content, pH, cooking loss, and shear force of modern genotype pork were significantly higher than the Korean native black pork; in addition, the moisture content of loin was significantly higher than shoulder and ham. The fatty acid compositions for different parts of pork showed no significant differences. Among the sensory characteristics, the parameters of pork were not significantly different. The moisture content of Holstein was significantly higher than Korean native cattle. The fatty acid composition of beef could not confirm the specific differences. Water holding capacity of Korean native cattle was higher than Holstein, while cooking loss of Korean native cattle was lower than Holstein. Overall acceptability scores of Korean native cattle was higher than Holstein. Conclusion: The study results of several parameters in selected raw meat samples provide useful information for developing new strategies to improve the quality of meat products consumption.

  • Previous paleoparasitological studies of Joseon specimens established that the prevalence of Taenia infection was not much different from that of the early 20th century Korean population. As many of taeniases originally diagnosed as Taenia saginata in South Korea were revealed to be actually Taenia asiatica, which share a common intermediate host with T. solium (the pig), Joseon people must have ingested raw pork frequently. However, the current examination of extant Joseon documents revealed that the population ate significant amounts of beef even if the beef ban was enforced; and pork was not consumed as much as we thought. Considering the meat consumption pattern at that time, Joseon people should have been infected by T. saginata more frequently than T. asiatica. This may suggest a low prevalence of T. saginata metacestodes in cattle compared to that of T. asiatica metacestodes in pigs, possibly due to the traditional way of rearing pigs (using human feces). This letter gives us a chance to reconsider the existing preconception about parasitic infections in Korean history though we are still hard to accurately estimate the historical patterns of taeniases at this stage.

Random amplification of polymorphic DNA typing of Listeria monocytogenes isolates from animal products (축산물유래 Listeria monocytogenes의 RAPD typing)

  • This study investigated the epidemiology of Listeria (L.) monocytogenes, a food-borne pathogen. The epidemiology of food-borne pathogens is of great importance for clarifying bacterial origin and preventing bacterial contamination and infection. This work examined 68 L. monocytogenes strains, including 11 reference strains and 57 isolates from imported US beef, domestic meats (beef, pork, chicken meat), raw milk, and milk plants. The random amplified polymorphic DNA (RAPD) techniques were optimized to develop a standard molecular epidemiological analysis of L. monocytogenes. There was great genetic variability among the isolates, which produced 24 and 34 RAPD patterns with primer HLWL85 and HLWL74, respectively. The discriminatory power of the RAPD methods with HLWL85 and HLWL74 primer were very high (DI = 0.957; S ${\geq}$ 80%, S ${\geq}$ 95%). Some RAPD types were specific to origin. A few RAPD types were specific for L. monocytogenes strains belonging to a particular serotype. Using the HLWL85 primer, the strains isolated from milk plants could be distinguished from the other strains. And using the HLWL74 primer, the strains isolated from imported beef (US) could be distinguished completely from the other strains.

Effects of Marbling on Meat Quality Characteristics and Intramuscular Connective Tissue of Beef Longissimus Muscle

  • This study was designed to explore the effects of marbling on meat quality characteristics and intramuscular connective tissue of beef longissimus muscle. Chemical determinations, histological and mechanical measurements were performed on the raw and cooked meat at d 4 postmortem. The results showed that crude fat, collagen, fiber diameter and maximum transition temperature of intramuscular connective tissue increased (p<0.05) with the increase of marbling score. The cooking losses, collagen solubility, WBSF and perimysial thickness decreased (p<0.05) with the increasing marbling. WBSF correlated (p<0.05) with moisture, crude fat, collagen, cooking losses, sarcomere length and perimysial thickness. The development of marbling results in the decline in cooking losses, the avoidance of sarcomere shortening, and the disorganization of the perimysia, which accounts for the improvement of beef tenderness.

Isolation and PCR detection of Listeria monocytogenes on raw beef and pork carcass (소와 돼지도체에서 Listeria monocytogenes의 분리 및 PCR 검출 방법에 관한 연구)

  • From February 2000 to December 2001, A total of 1,785 samples was taken from beef and pork carcasses in Seoul. Seven(0.69%) Listeria spp. were isolated from the 1,014 of beef carcasses, and five(0.65%) were isolated from the 771 of pork carcasses. The isolates were identified L monocytogenes by API listeria, and VIDAS LMO kit, serological test and PCR assay were preformed. A total 12 strains of L monocytogenes were isolated form samples tested and all of the strains were classified into serotype 1. PCR primers are selected to amplify a 520-base pair(bp) DNA fragment from the listeolysin O gene(hlyA) of Listeria monocytogenes. A 520-bp product was detected in PCR with DNA from L monocytogenes, but not from the other Listeria species tested.

  • To evaluate constitutional preference and clinical symptoms on the foods, we statistically analyzed the 572 patients' data based on the Sasang Constitutional Information Bank (SCIB) in Korea Institute of Oriental Medicine (KIOM). As a result, these are supposed to constitution-related foods ; crucian, watermelon, barley, ginseng, honey, chicken, deer's horn, beef, cold foods. Among these, crucian, barley, cold foods are fit for Soyangin, ginseng, honey, chicken for Soeumin and deer's horn, watermelon, beef for Taeumin. Whereas pork and sliced raw fish are nothing to do with sasang constitution in this study.

Studies on Organochlorine Pesticide Residues in Livestock Products 3. Organochlorine Pesticide Residues in Milk and Meat (축산식품(畜産食品)의 잔류농약(殘留農藥)에 관(關)한 연구(硏究) 제(第) 3 보(報) 우유(牛乳) 및 식육중(食肉中) 유기염소제(有機鹽素劑)의 잔류량조사(殘留量調査))

  • During the period of March, 1976 to December, 1976, 48 raw milk samples were taken from dairy cows at 48 different dairy farms in Korea analyzed by gas liquid chromatography to determine the seasonal variation of the amount of organochlorine pesticide residues. At the same time 80 market milk, 10 beef and 10 pork samples were analyed by the same procedure for checking residual levels. The results were summarized as follows; 1. Milk samples from 17 dairy farms (36 per cent of tatal) were shown to be contaminated with various organochlorine pesticides. The residua lrate of ${\gamma}-BHC$ in tested samples were 44per cent (14 sam ples) that of aldrine was 28 per cent (9 samples) and those of pp'-DDT, dieldrin and heptachlor were 9.3 percent (3 samples) respectively. 2. In raw milk pp'-DDT, ${\gamma}-BHC$, aldrin, dieldrin and heptachlor were detected, and aldrin, dieldrin and heptachlor were detected in the market milk. Any kinds of organochlorine pesticides were not detected in beef samples but dieldrin and heptachlor were detected in pork samples. Average residual values of aldrin, dieldrin and heptachlor in the market milk were 0.0077 ppm (0.0000~1.1100 ppm), 0.0001ppm (0.0000~0.0500 ppm) and 0.0008 ppm (0.0000~2.0520 ppm), respectively, and those of dieldrin and heptachlor in pork samples were 0.0010 ppm (0.0000~0.0100 ppm) and 0.0033 ppm (0.0000~0.0330 ppm). respectively. 3. Residues of organochlorine pesticides in raw milk were extremely variable; in fact pp'-DDT was detected in milk samples from A, B, C and D districts and endrin was not detected from all districts. The ${\gamma}-BHC$ and dieldrin were detected at the district of A, C and D, aldrin at the districts of A and C and heptachlor at the districts of both A and D. 4. Seasonal trends of residual values of organochlorine pesticides were, in general, noticeable. The residual level was much higher in Spring than in other seasons and showed the tendeney of decrease from spring through summer and autumn generally; in the case of pp'-DDT average residual values were 0.0121 ppm in spring, 0.0022 ppm in summer and not detected in autumn. But in winter ${\gamma}-BHC$ and aldrin residues were increased a little. Residual values in raw milk (when cow are fed on hay and silage) were appeared higher in winter than the other seasons. 5. Residues of organochlorine pesticides in raw milk were not related in respect to hygienic conditions of dairy farms pp'-DDT and heptachlor were, in general, detected in all farms and aldrin was more detected in milk from well sanitated farms than poor sanitated ones.

Prevalence and Antimicrobial Resistance of Enterococus faecalis and Enterococcus faecium Isolated from Beef, Pork, Chicken and Sashimi (시판 축산물 및 수산물에서 Enterococcus faecalis와 Enterococcus faecium 분포 및 항생제 감수성에 관한 연구)

  • In this study, a total of 256 samples of retail raw meats (beef, pork and chicken) and sashimi were investigated for the presence of Enterococcus faecalis and Enterococcus faecium. We isolated a total of 117 E. faecalis and E. faecium from the samples, with contamination rates ranging from 18.8% for sashimi samples to 68.8% of chicken samples. E. faecalis was the predominant species recovered from all of the retail raw meats beef (42.2%), pork (42.2%), chicken (65.6%) and sashimi (12.5%). Among 117 isolates, 61 isolates (52.1%) were resistant to tetracycline, 32 isolates (27.4%) were resistant to erythromycin, 23 isolates (19.7%) were resistant to chloramphenicol, 16 isolates (13.7%) were resistant to ripampin, 10 isolates (8.5%) were resistant to gentamycin, 9 isolates (7.7%) were resistant to ciprofloxacin and 1 isolate (0.9%) was resistant to ampicillin and penicillin G. No resistance to amoxicillin + clavulanic acid and vancomycin was observed. Although no strain was resistant to vancomycin, the vanB gene was observed in 9 of 117 of Enterococcus (7.7%) demonstrating potential risk of vancomycin-resistant Enterococcus (VRE). Our results indicate that E. faecalis and E. faecium were highly prevalent in retail raw meats, but most strains were sensitive to tested antibiotics.

Monitoring for β-agonist residues in raw meat in Seoul (식육에서 베타아고니스트계 약물 잔류 연구)

  • We investigated the residues of ${\beta}-agonist$ (zilpaterol, ractopamine and clenbuterol) using LC-MS/MS in raw meat in Seoul. The recoveries ranged between 76.7~89.9% in beef respectively. The limits of detection were $0.01{\sim}0.09{\mu}g/kg$ and the limits of quantification were $0.03{\sim}0.28{\mu}g/kg$ respectively. Residues of ${\beta}-agonist$ drugs which exceeded maximum residue limits (MRL) were not exceed in any of the 267 samples.
