Effects of Dietary Supplementation of a New Probiotic CS61 Culture on Performance in Broiler Chickens (새로운 생균제 CS61 배양액의 사료 내 급여가 육계의 생산성에 미치는 영향)

  • Bacterial resistance to antibiotics and residues of antibiotics in poultry products have encouraged the use of probiotics, prebiotic substrates, and synbiotic combinations of prebiotics and probiotics as alternative approaches to the use of antibiotics in poultry. The present study was carried out to evaluate the effect of a new probiotic CS61 culture on growth performance, feed conversion efficiency, and safety in broiler chickens, and to evaluate its value as an alternative for antibiotics used as a feed additive. Two dosages of CS61 culture (0.1% and 1%) were fed to chickens for 28 days. The results showed that terminal body weight and daily weight gain in the treatment groups increased in a dose-dependent manner when compared with the control group. Dietary supplementation with CS61 culture also improved feed conversion rate compared to the control group. There were no treatment-related toxic effects in terms of clinical findings, mortality, necropsy findings, hematology, or serum biochemistry parameters in any group tested. The nitric oxide assay showed that CS61 peptide has a dose-dependent inhibitory effect on lipopolysaccharide-induced nitric oxide production in RAW 264.7 cells. The results of this experiment indicated that dietary supplementation of CS61 culture may improve growth performance and feed conversion efficiency in chickens through its anti-inflammatory effect.

Tyrosinase-inhibitory and Radical Scavenging Activities from the Seeds of Coix lachryma-jobi L. var. ma-yuen [Roman] Stapf (율무의 tyrosinase 저해활성과 radical 소거효과)

  • The seeds of Coix lachryma-jobi L. var. ma-yuen(yullmoo) were investigated for the free radical scavenging and mushroom tyrosinase inhibitory activities. Brown yullmoo, which contains bran in the seed, has also been evaluated for comparison. After partitioning of the total methanol extracts of yullmoo and brown yullmoo, each organic layer exhibited mild or potent dose-dependent inhibition(about 20-80%) on 1,1-diphenyl picryl hydrazyl(DPPH) free radical with the concentration of $300\;{\mu}g/ml$ to $800\;{\mu}g/ml$. n-Butanol fraction of yullmoo showed the most potent scavenging effect on DPPH free radical. $IC_{50}$ of n-butanol fraction was $423\;{\mu}g/ml$, about six to seven fold higher concentration than standard BHT at the same inhibition rate. As for the tyrosinase inhibitory activity, all of the fractions including the methanol extract exhibited the similar activities even after partitioning. The chloroform fraction of brown yullmoo was found to show the most potent inhibitory activity with an $IC_{50}$ of $321.5\;{\mu}g/ml$.

Skin Moisturizing Properties and Anti-Inflammatory effects of extracts from Coptis chinensis in HaCaT cells (HaCaT cell에서 황련 추출물(Coptis chinensis)의 피부보습과 항염증 효과)

  • Coptis chinensis has been used in the treatment of various diseases such as soothing, anti-inflammation, antimicrobial and antipyretic in oriental traditional medicine. In this study, we investigated the effect of hot water extract of Coptis chinensis(CCW) on skin barrier and inflammation-related factors in UVB and TNF-α/IFN-γ-induced HaCaT cells and evaluated its potential as a moisturizing and anti-inflammatory material. Based on result, the amount of HA (Hyaluronic acid) production and protein and mRNA expression of filaggrin were measured. In TNF-α/IFN-γ-induced HaCaT cells, CCW increased the amount of HA production in a concentration-dependent manner. In the measurement of protein and mRNA expression of filaggrin, the expression rate increased as the concentration of CCW increased. In UVB-induced HaCaT cells, CCW decreased the production of ROS and showed significant results with EGCG ((-)-epigallocatechin-3-gallate), a positive control. In addition, CCW inhibited the expression of inflammatory cytokines TNF-α, IL-1β, IL-6, and IL-8 in TNF-α/IFN-γ-induced HaCaT cells. It was confirmed that the protein and mRNA expression of COX-2, a major factor in skin inflammation, was decreased in a concentration-dependent manner. These results suggest that hot water extract from Coptis chinensis can be used as a cosmetic material having a moisturizing and anti-inflammatory effect.

  • Depression has a negative impact on social functioning due to its high prevalence and increased suicide rate, and is a disease with a high economic burden. Depression is related to diverse brain-related phenomena, such as neuroinflammation, synaptic dysfunction, and cognitive deficit. As antidepressant drugs used in clinical trials have shown poor therapeutic effects, antidepressant drugs that show rapid efficacy urgently need to be developed. Although studies on various genes, proteins, and signaling pathways related to depression have been conducted, the pathogenesis of depression has not been clearly elucidated. Sirtuin 1 is a nicotinamide-adenine dinucleotide- (NAD+-) dependent histone deacetylase and is involved in cell differentiation, apoptosis, autophagy, and cancer metabolism. Recent genetic studies found that sirtuin 1 is a potential target gene for depression. In addition, preclinical studies reported that sirtuin 1 signaling affects depression-like behavior. In this review, we attempt to present up-to-date knowledge of depression and sirtuin 1. We describe the various roles of sirtuin 1 in the regulation of glial activation, circadian rhythm, neurogenesis, and cognitive function and the effects of its expression on depression. Further, we discuss the effect of sirtuin 1 on the impairment of neural plasticity, one of the key mechanisms of depression, and the associated mechanisms of sirtuin 1.

Anticancer Effect of Novel Peptide from Abalone (Haliotis discus hannai) based on Next Generation Sequencing Data (차세대염기서열분석 데이터 기반으로 선별한 전복(Haliotis discus hannai) 유래 신규 펩타이드의 항암 효과)

  • Glioblastoma is one of the highly aggressive central nervous system tumors and it is difficult to treat owing its anatomical location. Peptides are novel class of drugs which has the potential to cross the blood brain barrier and exerts its anti-tumor activity. Here, we discovered a novel peptide from abalone (Haliotis discus hannai) next generation sequencing (NGS) data and tested its anticancer effect on glioblastoma cell line SNU-489. The anticancer activity was measured using a cytotoxicity assay in a time and dose-dependent manner. A concentration and time dependent increase in the cytotoxicity was seen in cells treated with the novel peptide. The highest cytotoxicity rate of about 67% was observed in SNU-489 cells treated with 200 µM peptide for 48 hrs. However, the cytotoxic effect was not or less observed in a normal skin cell line HaCaT at similar concentration, thus, evident of peptide's cell specific anticancer activity. In addition, the gene expression level of necroptosis-related genes was analyzed by qRT-PCR to elucidate the anticancer mechanism of the novel peptide. RIPK3 expression was significantly increased by 9.6-fold in 200 µM of novel peptide treatment group, and MLKL expression level was significantly elevated by 2-fold in 100 µM treated group compared to the control group. Therefore, this study confirmed that the novel abalone-derived peptide has anticancer potency, and it causes cancer cell death through the necroptosis mechanism. Collectively, these results suggest that the novel peptide could be candidate anticancer agent for the treatment of glioblastoma in the future.

Effect of Stewartia koreana Nakai Branch Extracts on the Biofilm Formation of Porphyromonas gingivalis (노각나무 가지 추출물이 Porphyromonas gingivalis의 바이오필름 형성에 미치는 영향)

  • This study was investigated to evaluate the potential of Stewartia koreana Nakai as an oral healthcare material. The inhibitory effects of extracts on the biofilm formation and fimbriae genes expression of Porphyromonas gingivalis were determined by minimal inhibitory concentrations (MIC), biofilm biomass staining, SEM, and qRT-PCR analysis. The S. koreana Nakai branch was extracted into 70% ethanol, and bacteriostatic MIC of extracts against P. gingivalis were 0.6 mg/ml. In P. gingivalis cultures treated with 0.2-2.0 mg/ml of extract, biofilm production rate was significantly decreased in a concentration-dependent manner. The morphology of treated and untreated samples was observed by SEM, and cell aggregation and biofilm were only observed in those treated with extract. Subsequently, qRT-PCR analysis showed that the mRNA expression on fimbriae genes fimA and fimB was suppressed in a concentration-dependent manner. Based on these results, it can be suggested that S. koreana branch extract has the potential to be used as naturally derived oral healthcare material because of its bacteriostatic action and inhibition of P. gingivalis biofilm formation.

  • This paper analyzes the problem of the economic order quantity (lot size) of a retailer in a two-stage supply chain consisting of a supplier, a retailer(distributor), and a customer. In this two-stage supply chain, the supplier permits the retailer to defer payment for a certain fixed period of time for the purchase cost to be paid by the retailer as a price differentiation strategy with his competitor. In addition, in the case of customer goods such as food and grain, it is common to see that end-customer demand is generally depend on the level of inventory displayed by the retailer. From this perspective, this paper analyzes the inventory problem of retailers under the assumption that the supplier may allow a certain period to suspend payments for the purchase of goods and the end customer demand is a function of the retailer's inventory level increasing with size. In this regard, we need to analyze how much the length of the grace period for product purchase costs affect the retailer's lot-sizing policy. Therefore, we formulate the retailer's annual net profit and analyze the effect of the length of credit period on the retailer's inventory policy numerically.

A Data-driven Classifier for Motion Detection of Soldiers on the Battlefield using Recurrent Architectures and Hyperparameter Optimization (순환 아키텍쳐 및 하이퍼파라미터 최적화를 이용한 데이터 기반 군사 동작 판별 알고리즘)

  • The technology that recognizes a soldier's motion and movement status has recently attracted large attention as a combination of wearable technology and artificial intelligence, which is expected to upend the paradigm of troop management. The accuracy of state determination should be maintained at a high-end level to make sure of the expected vital functions both in a training situation; an evaluation and solution provision for each individual's motion, and in a combat situation; overall enhancement in managing troops. However, when input data is given as a timer series or sequence, existing feedforward networks would show overt limitations in maximizing classification performance. Since human behavior data (3-axis accelerations and 3-axis angular velocities) handled for military motion recognition requires the process of analyzing its time-dependent characteristics, this study proposes a high-performance data-driven classifier which utilizes the long-short term memory to identify the order dependence of acquired data, learning to classify eight representative military operations (Sitting, Standing, Walking, Running, Ascending, Descending, Low Crawl, and High Crawl). Since the accuracy is highly dependent on a network's learning conditions and variables, manual adjustment may neither be cost-effective nor guarantee optimal results during learning. Therefore, in this study, we optimized hyperparameters using Bayesian optimization for maximized generalization performance. As a result, the final architecture could reduce the error rate by 62.56% compared to the existing network with a similar number of learnable parameters, with the final accuracy of 98.39% for various military operations.

Population dynamics of the red-eared slider (Trachemys scripta elegans) with changes in the population dependent carrying capacity in Republic of Korea

  • Background: In this study, we proposed that the population dynamics of non-native red-eared sliders (Trachemys scripta elegans) depends on the species' habitat extension and survivorship. We used a logistic equation with time-dependent habitat carrying capacity. In detail, the present carrying capacity depends on the red-eared slider population of the previous year. Anthropogenic activities such as the abandonment of previously captive red-eared sliders or the release due to religion customs would supply new habitats to the species. Therefore we assumed that anthropogenic spread increases the habitat carrying capacity. Based on the urbanization increase rate of 3% in Korea from 1980 to 2000, we assumed an annual spread of 3% to simulate the population dynamics of the red-eared slider. In addition, the effect on the population of an increase of natural habitats due to migration was simulated. Results: The close relationship between the distributions of non-native red-eared sliders and of urbanized areas demonstrates that urbanization plays an important role in providing new habitats for released individuals. Depending on the survivorship, the population of the red-eared slider in Korea increased 1.826 to 3.577 times between 1980 and 2000. To control population growth, it is necessary to reduce carrying capacity by reducing habitat expansion through prohibition of release into the wild ecosystem and careful managements of the wetland or artificial ponds. Changes in the habitat carrying capacity showed that the population fluctuated every other year. However, after several years, it converged to a consistent value which depended on the survivorship. Further, our results showed that if red-eared sliders expand their habitat by natural migration, their population can increase to a greater number than when they have a 99% survivorship in a fixed habitat. Conclusions: Further introductions of red-eared sliders into wetlands or artificial ponds should be prohibited and managed to prevent future spread of the species. Moreover, it is important to reduce the species' survivorship by restoring disturbed ecosystems and maintaining healthy ecosystems.

Skin barrier and anti-inflammatory effect of petasites japonicus (머위(Petasites japonicus)의 피부장벽과 항염증 효과)

  • In this study, to investigate the possibility of using ethanol extract of Petasites japonicus (PJE) as a functional material, we investigated the activity of improving skin barrier and inflammation through UVB-induced human keratinocyte (HaCaT cell). As a result of confirming the antioxidant effect through DPPH radical scavenging activity, ABTS+ radical scavenging activity, and hydrogen peroxide scavenging activity, it was confirmed that it had an antioxidant effect similar to that of ascorbic acid, a control, at a concentration of 1 mg/ml. As a result of confirming the mRNA expression of the production ability of filaggrin and aquaporin-3 in HaCaT cells induced by UVB, it was confirmed that the reduced expression level by UVB stimulation increased in a concentration-dependent manner when the PJE was treated. It was confirmed that the mRNA expression of TNF-𝛼 and IL-1𝛽 were increased by UVB stimulation and decreased when the PJE was treated. As a result of the migration assay, it was confirmed that the proliferation of skin keratinocytes and the recovery rate of wounds were increased in a concentration-dependent manner. Based on the experimental results, it suggests that Petasites japonicus can be used as a functional cosmetic product that can improve skin moisturizing and skin barrier function.
