Protective Effects of Folium Artemisiae Argyi Herbal Acupuncture on Transient Forebrain Ischemic Injury in Rats (흰쥐의 일과성 전뇌 허혈 손상에 대한 애엽 약침의 신경보호 작용)

  • Objectives : Recently, the new therapeutic tool, that is herbal acupuncture, has been developed and applied to various diseases including the cerebrovascular accident. The main characteristics of herbal acupuncture are a combination of acupuncture and herbal medicine. It was not well known the therapeutic effect and the mechanism of herbal acupuncture on transient forebrain ischemic injury, although it has been used frequently in clinics. The objective was to determine the effect of folium artemisiae argyi (艾葉) herbal acupuncture on the trasient forebrain ischemic injured rat. Methods : In this study, the effects of folium artemisiae argyi (艾葉) herbal acupuncture on the $LR_3$ named Taechung, on neuroprotection after the transient forebrain ischemia were investigated in Sprague-Dawely rats. Expressions of cFos, FosB and BDNF protein in the hippocampus and cortex were observed at 2 hrs and 48 hrs after transient forebrain ischemia by immunohistochemistry and ELISA technique. Results : Expression of cFos protein was increased slightly in the hippocampus and cortex at 2 hrs after transient forebrain ischemia, but FosB protein was increased highly comparing to cFos protein. However, pretreatment with folium artemisiae argyi' herbal acupuncture on $LR_3$ significantly increased expression of cFos protein and significantly decreased expression of FosB protein compared to control group. These features were observed in the retrosplenial granular cortex as well as the hippocampus. Also, pretreatment with folium artemisiae argyi' herbal acupuncture on $LR_3$ significantly increased the expression of BDNF protein in the hippocampus ($263.26{\pm}44.56{\;}pg/ml$ extracted by water, $275.35{\pm}51.47{\;}pg/ml$ extracted by moxa tar)and the cortex ($102.33{\pm}13.65{\;}pg/ml$ extracted by water, $109.54{\pm}9.37{\;}pg/ml$ extracted by moxa tar) compared to the hippocampus ($134.07{\pm}2.96{\;}pg/ml$) and the cortex ($61.16{\pm}4.11{\;}pg/ml$) in control group at 48 hrs after transient forebrain ischemia. Conclusions : These results suggest that pretreatment with folium artemisiae argyi' herbal acupuncture on $LR_3$ has neuroprotective effect on transient forebrain ischemia and the herbal acupunture on $LR_3$ may be related to antioxidative function of folium artemisiae argyi.

Study of Apoptotic Effect on Hydrogen Peroxide-induced Rat PC-12 Cells by Aster tataricus Water Extract (자완 수추출물(水抽出物)이 $H_2O_2$에 의해 유도(誘導)된 PC-12 세포주(細胞株)의 세포사(細胞死)에 미치는 영향(影響))

  • Objective : Alzheimer's disease (AD) is a geriatric dementia that is widespread in old age. With an aging populace, AD is a looming problem in public health service. Alzheimer's disease is characterized by specific neuronal degeneration in certain areas of the brain. Mutations and abnormal expression of several genes are associated with ${\beta}-amyloid$ deposits and Alzheimer's disease; among them APP, PS1, and PS2, SOD, free radical, ROS. Methods:We studied herbal medicines that have a relationship to brain degeneration. From pre-modern times, although a variety of oriental prescriptions of Aster tataricus have been traditionally utilized for the treatment of AD, their pharmacological effects and action mechanisms have not yet been fully elucidated. Result : Based on morphological observations by phase-contrast microscope, TUNEL assay and MTT in the culture media, $H_20_2-induced$ cell death was significantly inhibited by Aticus. We examined by ROS formation, catalase activity and GSH activity. We studied the protective effect and inhibitory effects of neurotoxicity in $H_20_2-induced$ PC-12 cells by Aticus. Findings from our experiments show that Aticus inhibits apoptosis, which has neurotoxicities and cell damage in PC-12 cells. In addition, treatment with Aticus ($>25{\mu}g/ml$ for 6hrs) partially prevented $H_20_2-induced$ cytotoxicity in PC-12 cells, and induced a protective effect. Conclusion : As the result of this study, in the Aticus group, the apoptosis in the nervous system was inhibited, protected against the degeneration of PC-12 cells by $H_20_2$. Taken together, Aticus exhibited inhibition of $H_20_2-induced$ apoptotic cell death. Aticus was found to induce protective effect on GSH and catalase in PC-12 cells. Based on these findings, Aticus may be beneficial for the treatment of AD.

Effect of Gamiondam-tang (GMODT), a Polyherbal Formula on the Pharmacokinetics Profiles of Tamoxifen in Male SD Rats

  • Objectives: The effects of Gamiondam-tang (GMODT) co-administration within 5min on the pharmacokinetics (PK) of tamoxifen were observed as a process of the comprehensive and integrative medicine, combination therapy of tamoxifen with GMODT to achieve synergic pharmacodynamics and reduce toxicity on the breast cancer. Methods: After 50mg/kg of tamoxifen treatment, GMODT 100mg/kg was administered within 5min. The plasma were collected at 30 min before administration, 30 min, 1, 2, 3, 4, 6, 8 and 24 hrs after end of GMODT treatment, and plasma concentrations of tamoxifen were analyzed using LC-MS/MS methods. PK parameters of tamoxifen (Tmax, Cmax, AUC, $t_{1/2}$ and $MRT_{inf}$) were analysis as compared with tamoxifen single administered rats using noncompartmental pharmacokinetics data analyzer programs. Results: Co-administration with GMODT induced increased trends of plasma tamoxifen concentrations to 1hr after end of administration, and then showed decreased trends of plasma tamoxifen concentrations, and especially significant (p<0.05) increases of plasma tamoxifen concentrations were demonstrated at 0.5hr after end of co-administration with GMODT and also related significant (p<0.05) decreases of $AUC_{0-inf}$ and $MRT_{inf}$ as compared with tamoxifen single formula treated rats, at dosage levels of tamoxifen 10 mg/kg and GMODT 100 mg/kg within 5 min, in this experiment. Conclusion: Based on the results of the present study, it is considered that single co-administration GMODT within 5min significantly inhibited the oral bioavailability of tamoxifen through variable influences on the absorption and excretion of tamoxifen, can be influenced on the toxicity or pharmacodynamic of tamoxifen.

Protective effects of Jijang-kimchi extracts on diabetes mellitus and alcoholic liver injury in laboratory rats (랫드에서 김치가 당뇨병 및 알코올성 간 손상 예방에 미치는 효과)

  • The current study was carried out to investigate the effect of oral administration for 30 days of the Jijang kimchi extracts on prevention of diabetes, alcoholic liver injury and reduction of blood lipids in laboratory rats with alcoholic liver injury and diabetes induced by streptozotocin. In a diabetic model animals, the blood lipid profile, ALT, and AST levels were lower in kimchi extract groups compared to DC (diabetes control) group, and blood glucose level of DCJK (DC+oral administration with Jijang kimchi extracts) group was lower than that of DCCK (DC+oral administration with commercial kimchi extracts) group. Insulin levels were increased in order of NC (normal control), DCJK > DCCK > DC groups. In alcoholic liver injury model animals, ALT, AST and bilirubin were lowed in order of AC (alcohol group received 1 bottle of soju) > ACCK (1 bottle of soju plus oral administration with commercial kimchi extracts) ACJK (AC plus oral administration with Jijang kimchi extracts) > NC groups. In the clinical pathologic findings of liver tissue, AC group was severely injured, and tended to be improved in groups eating a 1 bottle of soju plus oral administration with kimchi extracts, especially Jijang kimchi extract group. The results suggest that eating Jijang kimchi can improve insulin secretion ability while lowering blood lipid profile, blood sugar and ALT, AST, and bilirubin levles in diabetic and alcoholic liver injury model animals.

The Effects of Gobonyangjeonbang Administration on Antioxidative of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) induced Rats. (고본양정방 투여가 TCDD 유도 랫드의 항산화 변화에 미치는 영향)

  • The purpose of this study was to evaluate the effects of gobonyangjeonbang (GYB) on the endocrine function and the antioxidant efficacy of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-induced oxidative stress in rats. In 2017, to evaluate the efficacy of GYB on oxidative stress, 35 male SD rats were divided into five groups and tested. The normal control group was administered saline as a vehicle, while the TCDD-alone group was administered TCDD (2 ㎍/kg per week) intraperitoneally and with physiological saline, and the test group was administered GYB orally by dividing it into three concentrations (75, 150, and 300 mg/kg) for six weeks. Bodyweight decreased significantly after six weeks of TCDD exposure, when compared to rats in the NC group (p<0.001). However, weight loss from TCDD was significantly protected by administration of GYB at 300 mg/kg (p<0.01). The rat liver induced by TCDD showed cytoplasmic vacuole degeneration, and the hepatic sinusoid and weight increased. As a result of measuring MDA and SOD, both items tended to decrease under TCDD administration. On the other hand, there was no change due to GYB administration, and significance was observed in the GYB 300 mg/kg group compared to the NC group in the SOD result (p<0.05). These findings demonstrated that GYB may have a protective effect against TCDD-induced liver toxicity in rats.

Effect of Dietary n-3 Fatty Acids and Fat Unsaturation on Plasma Lipids and Lipoproteins in Rats (식이의 n-3 지방산과 지방의 불포화도가 혈장지질 조성에 미치는 영향)

  • This study was to compare the effects of dietary n-6 and n-3 fatty acids and fat unsaturation on plasma lipids and chemical composition of VLDL and LDL fraction and lipogenic enzymes activity in rat liver under the conditions providing 1) a similar amount of n-6, n-3 fatty acids(LA, ALA, EPA+DHA) in diets and 2) the various degree of fat unsaturation. Male Sprague-Dawley rats weighing 420g were treated for 6-n with six experimental diets providing 25% of energy as fat and which were different only in fatty acid composition. The fats used for a source of each fatty acid were beet tallow for saturated fatty acid corn oil for n-6 linoleic acid(LA) perilla oil for n-3 $\alpha$-linolenic acid(ALA) and fish oil n-3 eicosapentaenoic acid (EPA) and n-3 docosahexaenoic acid(DHA). Plasma cholesterol level was increased by corn oil to compare with beef tallow but was decreased by perilla oil or fish oil. Plasma TG level was significantly decreased by perilla oil or fish oil. Fish oil significantly reduced the level of HDL-Chol and the proportion of Chol in LDL fraction and that of TG in vVLDL fraction. Overall there was a singificant negative correlation between the level of each plasma lipid(Chol TG, VLDL-TG, LDL-C) and the degree of fat unsaturation. However this rerlationship is not always true when compared the hypolipidemic effect of each fatty acid at a similar level of fat unsaturation. There was a trend such taht glucose 6-P dehydrogenase 6-phosphogluconate dehydrogenase and malic enzyme activites were reduced by n-3 fatty acids. Perilla oil significantly increased the incorporation of c20:5 and c22:5 into liver tissue and fish oil suignificantly increased the incorporation of c20:5, c22:6 into liver tissue and the effect of long chain n-3 fatty acid incorporation was greater by fish oil. therefore the hypotriglyceridemic effect of n-3 fatty acid could be resulted from the interference of hepatic lipogenesis by long-chain n-3 fatty acids and the reduced proportion of TG in VLDL fraction and its effect was greater by n-3 EPA+DHA than n-3 ALA even though plasma Chol and TG levels were also influenced by the degree of dietary fat unsaturation.

Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats (감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level.   • The authors has observed the histopathologically and histochemically on the effect of the garlic and garlic oil to the liver and kidney of rats. In order to confirm the histochemical changes of the metabolism of polysaccarides, the periodic acid Schiff reaction was applied. The 30 albino adult male rats weighing about 150 grames from the National Institute of Health were housed individualy and devide into 3 experimental groups: Group C: stock diet group Group B: stock diet-garlic group Group A: stock diet-garlic oil group Group C was fed with stock diet only through out this experimental period, Group B was fed with stock diet supplemented with garlic homogenator to be 1%, and Group A was fed with stock diet supplemented with the garlic oil to 0.05%. The garlic oil used in this experiments was extracted by author. And all rats was fed during 10 weeks. The histopathological and histochemical results were shown in each figure. According to the all results, the following concIusions were drawn. 1) In the garlic oil administrated groups, congestion of the sinusoid was subsided and the liberation of the Kupffer's cells were observed. 2) In garlic administrated groups, fatty metamorphosis in hepatic cells, and slight liberation of Kupffer's cells in sinusoidal walls were observed. Connective tissue proliferation and collagen bundle were observed. 3) The connective tissue and blood vessel wall in portal area Were reacted intensely with PAS stain. The hepatic cells Were reacted intensely with PAS stain in control group and moderately or slightly in garlic and garlic oil administrated group. 4) There were no significant differences in collecting and Henle's loops in each groups, but narrowing of lumen of the distal tubules were observed in garlic oil administrated group. 5) The basement membrane of the tubules and the connective tissues of the vessel wall in Kideny were reacted intensely with PAS stain in each groups. In control and garlic administrated groups. the brush border of the proximal tubules were reacted intensely with PAS stain, but epithelium of the Heine's loop, proximal, distal and collecting tubules were reacted moderately. In garlic oil administrated-group, there were tendency of decreasing of PAS stain in each tubules.

Effect of Long-Term Pyridoxine Depletion on Asparte Aminotransferase and Pyridoxal 5'-Phosphate of Rat Liver Mitochondrial and Cytosolic Fractions (장기간의 Pyridoxine 부족이 쥐 간의 Mitochondria 및 Cytosolic Fraction 에 있는 Aspartate Aminotransferase 및 Pyridoxal 5'- Phoshate에 미치는 영향)

  • Weanling female Sprague Dawley rats were fed d diets containing 22mg pyridoxine. BCI/kg diet (control diet) and l.2mg pyridoxine. BCI/kg diet (deficient diet). One control group and one defi­c dent group were fed their diet throughout growth, g gestation and lactation. After the pups were born and weaned, the deficient group was divided into two groups. One switched to control diet(supple­I mented group) and the other continued the same d deficient diet( deficient group) until 10 week -old. The liver mitochondrial and cytosolic asparate a aminotransferase activity and pyridoxal phosphate content were determined in offspring rats. The aspartate aminotransferase activities in both liver mito$\phi$ondrial and cytosolic fractions of den­d cient group were significantly lower than those of controls, but there were no significant differences between two groups after addition of 1O^{-4}M pyri­d do뼈I phosphate to the medium. By pyridoxine s supplementation after weaning, the reduced aspar­a tate aminotnmsferase activities were only partialy I restored to control levels. The pyridoxal phospha­t te content of deficient group in Iiver mitochondr­ial and cytosoIic fractions were alo significantly different from those of controls, but readily restored by dietary supplementation. These results suggest that there is a quantitative and a qualitative changes of aspartate amino trans­f ferase and pyridoxal phosphate in liver mitochon­d drial and cytosolic fraction by long-term pyrido­x xine deficiency and these reductions can partially recovered by dietary pyridoxine supplementation after weaning.

Effect of Coix on Plasma Cholesterol and Lipid Metabolism in Rats (율무쌀이 쥐의 혈장콜레스테롤 및 지질대사에 미치는 영향)

  • This study was done to determine the effects of coix on the content of cholesterol. lipids, phosphoilpids, total bile acids in liver, plasma, and feces of rats fed containing 1% cholesterol. Four groups of experimental diets, such as soybean oil-starch diet, soybean oil-coix diet, lard-starch diet, lard-coix diet were fed ad libitum to the 6 weeks old white male rat of Spargue-Dawley strain for 27 days. The results obtained are summarized as follows: 1) Plasma cholesterol level of the lard-starch group was significantly higher than the other groups (p<0.05), and that of the lard-coix group was the lowest. The content of plasma triglyceride, total bile acids was not different among the lard-starch group was significantly higher than the other groups(p<0.05). 2) The contents of the liver lipids, cholesterol, and triglycerides of the lard-coix group were significantly lower than the other groups(p<0.05). The contents of phospholipid of the liver of the lard-starch group was significantly lower than the other groups(p<0.05), and those of the soybean oil-coix group and the lard-coix group were high. 3) The contents of intestinal lipids, triglyceride, and phospholipid of the lard-coix group were significantly lower than the other groups(p<0.05). The contents of the intestinal tissue cholesterol of the lard-coix group was significantly lower than the other groups(p<0.05). 4) Cholesterol content of the feces was not different among the experimental groups. Triglyceride content of the feces of the soybean oil-starch group was significantly higher than the other groups, but total bile acid content of the feces was not different among the experimental groups. 5) Dry matter digestibility was not different among the experimental groups, but cholesterol retention was rather low when soybean oil-diet was fed. The results of this study reveales that coix has an effect to lower the plasma cholesterol level. Coix were decreased the content of liver cholesterol, increased the contend of liver phospholipid, and induced triglyceride excretion of the feces in the lard diet which lead to hypercholesterolemia. It could be suggested that the coix has influesced liped metabolsim in body.
