Blood Component Change in Rat by Lipopolysaccharide and Cell Wall Protein-A from Vibrio vulnificus, E. coli, and S. typhimurium

  • Lipopolysaccharide (LPS) and cell wall protein-A (CWP-A) were extracted from the cell wall of Vibrio vulnificus, Escherichia coli and Salmonella typhimurium. LPSs and CWP-As were injected into rat and the changes of the following blood components were examined. The change of the number of white blood cell (WBC), red blood cell (RCB), platelet (PLT), blood urea nitrogen (BUN) and blood glucose in rat blood and interferon (IFN) activity change by LPS and CWP-A were measured. WBC, RETI, PTT, and BUN were increased and RBC and blood glucose were increased slightly, but PLT was decreased.

  • The present study was designed to investigate the effects of green tea extract (GTE) and epigallocatechin gallate (EGCG) on secretion of catecholamines (CA) in the isolated perfused rat adrenal gland. In the presence of GTE (100 ${\mu}$g/$m\ell$) into an adrenal vein for 60 min. CA secretory responses evoked by ACh (5.32 mM), high K+ (56 mM) and Bay-K-8644 (10 ${\mu}$M for 4 min) from the isolated perfused rat adrenal glands were greatly inhibited in a time-dependent fashion.   • The purpose of the present study was to determine the effects of platycodin D and D3 on the contractile force of the isolated rat aorta and blood pressure of the anesthetized rat. and also to establish the mechanism of action. The phenylephrine (10 $\mu$M)-induced contractile responses were greatly inhibited in the presence of platycodin D (4 ∼ 24 $\mu$g/$m\ell$) in a dose-dependent fashion. (omitted)

MRI 데이터를 이용한 쥐의 경추와 인접한 조직의 유한요소 모델화 (Finite Element Modeling of the Rat Cervical Spine and Adjacent Tissues from MRI Data)

  • Traumatic loading during car accidents or sports activities can lead to cervical spinal cord injury. Experiments in spinal cord injury research are mainly carried out on rabbit or rat. Finite element models that include the rat cervical spinal cord and adjacent soft tissues should be developed for efficient studies of mechanisms of spinal cord injury. Images of a rat were obtained from high resolution MRI scanner. Polygonal surfaces were extracted structure by structure from the MRI data using the ITK-SNAP volume segmentation software. These surfaces were converted to Non-uniform Rational B-spline surfaces by the INUS Rapidform rapid prototyping software. Rapidform was also used to generate a thin shell surface model for the dura mater which sheathes the spinal cord. Altair's Hypermesh pre-processor was used to generate finite element meshes for each structure. These processes in this study can be utilized in modeling of other biomedical tissues and can be one of examples for reverse engineering on biomechanics.

Genome wide identification of Staufen2-bound mRNAs in embryonic rat brains

  • Messenger ribonucleoprotein particles (mRNPs) are used to transport mRNAs along neuronal dendrites to their site of translation. Staufen2 is an mRNA-binding protein expressed in the cell bodies and cellular processes of different brain cells. It is notably involved in the transport of dendritic mRNAs along microtubules. Its knockdown expression was shown to change spine morphology and impair synaptic functions. However, the identity of Staufen2-bound mRNAs in brain cells is still completely unknown. As a mean to identify these mRNAs, we immunoprecipitated Staufen2-containing mRNPs from embryonic rat brains and used a genome wide approach to identify Staufen2-associated mRNAs. The genome wide approach identified 1780 mRNAs in Staufen2-containing mRNPs that code for proteins involved in cellular processes such as post-translational protein modifications, RNA metabolism, intracellular transport and translation. These results represent an additional and important step in the characterization of Staufen2- mediated neuronal functions in rat brains.

피하조직에 투여된 수은과 카드뮴의 효소활성과 과산화지질에 미치는 영향 (The Effects of Mercury and Cadmium Administered in Subcutaneous Tissue on Enzymatic Activity and lipidperoxidation)

  • Heavy metals like Mercury and Cadmium cause various kinds of toxicities in the organs of Liver and Kidney. To observe the results of toxicity in the liver, kidney, and serum when the rats were injected subcutaneously with HgCl$_2$ and CdC1$_2$ and sacrificed after 24 hours and 72 hours from the last injection, we measured variation of lipidperoxide values in rat liver homogenate, variation of aspartate aminotransferase and alanine aminotransferase in rat serum. Variation of lipidperoxide values in rat kidney homogenate and variation of BUN in rat serum. It was found that Mercury and Cadmium administered subcutaneously to the skin in the air could cause the damages of liver and kidney.

Is Nitric Oxide Involved in Relaxation of Urinary Bladder\ulcorner

  • We investigated whether nitric oxide (NO) may serve a role in bladder function by immunohistochemical analysis of the distribution of intrinsic NADPH-diaphorase and functional study of isometric tension recordings via a photo-induced adequate nitric oxide (PIANO) generating system using rat bladder. Results suggest that a small number of NADPH-diaphorase-positive perikarya are present within the bladder wall and within adjacent small ganglia. Furthermore, NADPH-diaphorase-positive nerve fibers were observed in the adventitial and muscular layers, subjacent to the urothelium and perivascular fibers. Rat bladder strips precontracted with 3$\mu$M carbachol were reversibly relaxed upon NO generation by UV irradiation. PIANO-mediated relaxation was sensitive to oxygen free radicals. In addition, tissue cGMP levels were increased by the PIANO generating system and elevated cGMP levels were decreased by pretreatment of guanylate cyclase inhibitor, methylene blue. These results indicate that NO may serve a role in modulating bladder tone in the rat.

랫드의 두개골세포 활성 및 mRNA 발현에 미치는 홍화자의 영향 (Effects of Carthami Tinctorius on Rat Calvarial Cell Activity and MG63 Cell mRNA Synthesis)

  • This study was performed to evaluate the effect of Carthami tinctorius(HH) on osteoblast function and gene expression. The osteoblasts separated from the rat calvariae were cultivated to evaluate the cell function, and MG-63 cell was also cultivated for the test of mRNA synthesis. In this experiments, cell proliferation of rat calvarial cells was increased by HH. PKC activity, intracellular free calcium level and collgen synthesis from calvarial cells were increased by HH, but not PKA activity. And the mRNA of $PLA_2$, COX-2, and $PGE_2$ synthase from MG-63 were decreased by HH, but the mRNA of prostacyclin synthase was increased. It is concluded that HH might increase the proliferation of calvarial cell resulted from augumentation of osteoblast activity and its mRNA synthesis.

  • Spherical multicellular aggregates of adult rat hepatocytes (spheroid) which have tissue like structure, were formed and immobilized in the pores of polyurethane foam (PUF) which was used as a culture substratum. These hepatocyte/spheroids, about 100 $\mu\textrm{m}$ in diameter, have maintained higher differentiated functions than those of hepatocyte/monolayer for about 3 weeks in serum-free medium. Then, we designed a prototype module of an artificial liver support system using a PUF/spheroid packed-bed, in which hepatocyte/spheroids were immobilized at high density. The urea synthesis activity of the artificial liver was maintained at least 10 days in 100% rat blood plasma. We start examining the performance of hybrid artificial liver in an ex vivo extracorporeal experiment with an acute hepatic failure rat.

Effects of Prenatal Cnidium officinale Makino Treatment on Spatial Memory and Neurogenesis in the Hippocampus of Rat Pups Born from Maternal Rats Exposed to Noise Stress during Pregnancy

  • During the prenatal period, the development of the individual is influenced by a host of environmental factors. Exposure to noise stress during pregnancy was determined to result in the retardation of growth, a reduction in neurogenesis, and an impairment of spatial learning ability in the rat pups. In the present study, we have attempted to characterize the effects of prenatal treatment with Cnidium officinale Makino on spatial memory and neurogenesis in the hippocampus of rat pups born from maternal rats exposed to noise stress during pregnancy. Prenatal treatment with Cnidium officinale Makino was shown to increase neurogenesis and enhanced spatial learning ability in rat pups born from maternal rats exposed to noise stress. In this study, we have determined that prenatal treatment with Cnidium officinale Makino can stimulate spatial development and neurogenesis in the brain of the fetuses exposed to prenatal stresses.
