Antioxidative Effects of Scutellariae Radix Aaquaacupuncture Solution on Lipid Peroxidation Induced by Free Radicals (자유기에 의한 지질과산화 반응에 대한 황금 약침액의 항산화 효능)

  • Scutellariae radix, has been used as a natural drug for fever, inflammation, cataract, and liver disease in traditional medicine. This study was performed in order to investigate the antioxidative effects of Scutellariae radix aqua-acupuncture solution (SRAS) on lipid peroxidation by free radicals. Lipid peroxidation levels were determined by TBA method during the autoxidation of linoleic acid. In this linoleic acid autoxidation system, SRAS markedly exhibited antioxidant activity, which inhibited 89% of linoleic acid peroxidation. SRAS showed scavenging effects on ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$(DPPH) radical, inhibited superoxide generation in xanthine-xathine oxidase system, and also inhibited lipid peroxidation of rat liver tissue by hydroxyl radical derived from $H_2O_2-FE^{+2}$ system. These effects were similar to those of $dl-{\alpha}-tocopherol$, BHA and BHT. In addition, SRAS protected the cell death induced by ter-butyl hydroperoxide (t-BHP) and significantly increased cell viability in the normal rat liver cell (Ac2F). On the basis of these results, it is suggested that SRAS might play a protective role in lipid peroxidation by free radicals.

  • Objective : This study on Sipimikwanjung-tang was undertaken to evaluate its antioxidant capacities and antiperoxidation activities in rat liver tissues. Sipimikwanjung-tang which has been one of the prescriptions in sasang constitutional medicine is usually applied for the therapy of various liver diseases. It is elucidated that Sipimikwanjung-tang has antioxidants on liver tissue of rat and the cytotoxic effects on human hepatoblastoma Hep G2 cells. Methods: Sipimikwanjung-tang extract in antioxidant effects of Hep G2 cells is evaluated by MTT assay, DAPI staining, DNA fragmentation assays and FACS can analysis. Results: Sipimikwanjung-tang induced apoptosis in Hep G2 cells, and induced G1 and G2M arrest of the cell cycle as well as a significant increase in PARP and caspase-3 activity. It induced an increase in $H_2O_2$ generation and the subsequent $NF-{\kappa}B$ activation and also induced cell apoptosis through the caspase-3-dependent pathways in the low concentration of Sipimikwanjung-tang extracts. However, the high dose of Sipimikwanjung-tang extract in Hep G2 cells inhibited $TGF-{\beta}l-induced$ apoptosis via increase in cellular $H_2O_2$, formation and $NF-{\kappa}B$ activation in human hepatoblastoma Hep G2 cells. Conclusion: From this study, the possibility that Sipimikwanjung-tang extracts apply to antioxidant and apoptotic treatment of disease is revealed.

  • A study on the antitoxic effects of Herba Taraxaci extracts against Cadmium Chloride Toxicity in Rats. This study was performed to find out the effect of taraxacum platy carpum against Cadmium toxicity. The experimental rats were divided into 5 groups such as control group, Cadmium alone treatment group, three simultaneous treatment groups of taraxacum platy carpum and cadmium. Rat were given pellets administration with three dosage of taraxacum platy carpum such as 4 mg/kg body weight for four weeks. The results were summarized as follows on: 1. The simultaneously administration of taraxacum platy carpum and cadmium significantly more decreased cadmium concentration in liver and kidney tissues compared to the administration of cadmium alone. (p<0.05) 2. The simultaneously administration of taraxacum platy carpum and cadmium significantly more increased metallothionein concentration in liver and kidney tissues compared to administration alone. (p<0.05) 3. When liver and kidney tissues were observed with optical microscope obvious changes were visible in those tissue.

  • This paper presents the effect of the supramolecular complex of GA (Glycyrrhizic acid) and Mt(menthol) and GA: Mt (4: 1) obtained on their basis can restore functional dysfunction of the liver mitochondria in alloxan diabetes, ie, inhibit lipid peroxidation. The hypoglycemic activity and mitochondrial membrane stabilizing properties of the supramolecular compound GA: Mt (4: 1) in alloxan diabetes were more pronounced than those of menthol, GA and its GK: Mt (2: 1) and GA: Mt (9: 1) compounds. According to the results obtained, the concentration of GA did not affect the peroxidation of lipid membranes of the liver mitochondria. However, a concentration of 15 μM of GA was found to reduce LPO (lipid peroxidation) formed by the effect of Fe2+ / ascorbate on the mitochondrial membrane by 58.0 ± 5.0% relative to control. Effects of Scutellaria baicalensis Extract on Gout-Induced Rats

  • Background: This study aimed to investigate hyperuricemia, renal inflammation, and xanthine oxidase (XO) activity improvement in a rat model treated with Scutellaria baicalensis extract (SBE). Methods: The rats were divided into 4 groups (n = 5 each), including sham, potassium oxonate (PO) injected hyperuricemia (control group), PO + 10 mg/kg allopurinol administrated (allopurinol group), and a PO + 50 mg/kg SBE administrated (SBE group), to investigate the effectiveness and molecular mechanisms of SBE. The effects of SBE on PO-induced hyperuricemia rats, renal inflammation, and XO activity were measured. Body weight and organ index of the kidney and liver were measured in PO-induced hyperuricemia rats, and serum uric acid level was extracted from whole blood and was measured. Renal inflammation was observed under a microscope after sections. XO activity was measured by liver tissue and serum XO levels. Results: Organ indexes of the kidney and liver in rats were significantly decreased in the allopurinol group than in the control group and with no significant difference in the SBE group. A PO injection for 5 days significantly increased serum uric acid levels in the control group compared to the sham group. Meanwhile, the SBE and allopurinol groups have significantly decreased serum uric acid levels compared to the control group. The SBE group revealed effectively improved renal histopathological changes compared to the control group. The XO inhibitor, allopurinol, significantly decreased XO activity. Additionally, SBE significantly lowered XO activity in rats. Conclusion: SBE can be used as an effective treatment for gout in the future.

Measuring the Light Dosimetry Within Biological Tissue Using Monte Carlo Simulation (Monte Csrlo 시뮬레이션을 이용한 생체조직내의 광선량 측정)

  • As the correct measuring of the light dosimetry in biological tissues give the important affection to the effect of PDT treatment we used Monte Carlo simulation to measure the light dosimetry on this study. The parameters using in experiments are the optical properties of the real biological tissue, and we used Henyey-Greenstein phase function among the phase functions. As we results, we displayed the result the change of Fluence rate and the difference against the previous theory was at least 0.35%. Biological tissues using in experiment were Human tissue, pig tissue, rat liver tissue and rabbit muscle tissue. The most of biological tissue have big scattering coefficient in visible wavelength which influences penetration depth. The penetration depth of human tissue in visible region is 1.5~2cm. We showed that it is possible to measure fluence rate and penetration depth within the biological tissues by Monte Carlo simulation very well.

Effects of vitamin C on the formation of aflatoxin B1-DNA adduct in rat livers treated with radiation and aflatoxin B1 (Vitamin C가 방사선과 Aflatoxin B1을 처리한 흰쥐의 간세포에서 Aflatoxin B1-DNA Adduct 형성에 미치는 영향)

  • The objective of this study was to examine the effects of vitamin C on the formation of aflatoxin $B_1$ ($AFB_1$)-DNA adduct and $AFB_1$-induing cellular oxidative damage in rat livers treated with radiation and $AFB_1$. Six-week-old male Sprague-Dawley rats were randomly divided into five groups: the control group, the $AFB_1$-treated group, the group treated with $AFB_1$ and vitamin C, the group treated with X-ray and $AFB_1$, and the group treated with X-ray and $AFB_1$ with vitamin C. On the first day of the experiment, only one dose of X-rays was exposed to the entire liver at 1,500 cGy. Next, vitamin C was injected at 10 mg/kg body weight via intraperitoneal injection, followed an hour later by the administration of 0.4 mg/kg of $AFB_1$ via intraperitoneal injection. These treatments were administered every three days for 15 days. On the 16th day, the animals were sacrificed. The $AFB_1$ contents of the rat sera were determined via indirect competitive ELISA. In the quantitative analysis of $AFB_1$ in the rat sera via ELISA, $5.17{\pm}0.34ng/mL$ of $AFB_1$ was detected in the $AFB_1$-treated groups, but the amount decreased more significantly to $3.23{\pm}0.76ng/mL$ in the groups treated with $AFB_1$ and vitamin C (p<0.01) than in the $AFB_1$-treated groups. The effect of vitamin C on $AFB_1$-DNA adduct formation was determined via ELISA. The values of $AFB_1$-DNA adduct formation were $9.38{\pm}0.41ng/mL$ in the $AFB_1$-treated groups, but the amount decreased more significantly to $5.28{\pm}0.32ng/mL$ in the groups treated with $AFB_1$ and vitamin C (p<0.01) than in the $AFB_1$-treated groups. Immunohistochemistry revealed that the accumulation of the $AFB_1$ was not observed in the normal liver tissue (G1). The $AFB_1$-positive materials were observed in the central vein and the portal vein of the liver tissue from the $AFB_1$(G2) treatment or the X-ray and $AFB_1$(G4) co-treatment, but the $AFB_1$-positive materials were observed weakly in the group treated with vitamin C (G3 and G5). These results indicate that vitamin C had ameliorating effects on the $AFB_1$ accumulation of liver tissue.

  • The therapeutic effect of tissue cultured root of mountain ginseng (Panax ginseng) (tcMG) on 2,3,7,8-tetrachlorodaibenzo-p-dioxin (TCDD) induced toxicity in rat was investigated. The rats were assigned into three groups (10 rats/group), control, TCDD exposed group and tcMG treated group after TCDD exposed. $50\;{\mu}g/kg$ of TCDD was injected by i.p. for TCDD exposed group and 30 mg/kg of tcMG saponin was administered for 4 weeks by oral gavage for tcMG treated group. The weights of body, spleen, kidney, thymus, testes and epididymides were decreased in the single TCDD treatment. However these organs was significantly recovered by tcMG saponin except thymus (p<0.05). tcMG decreased the level of hepatic demage maker enzymes, AST and ALP. It also lowered total cholesterol and triglyceride. The level of serum triglyceride was significantly decreased in tcMG saponin treated group compared with the control. Histopathological examination revealed morphological change in the liver spleen, thymus and testes of TCDD treated rats. However they were relatively well preserved in the tcMG treatment group. In conclusion, TCDD induced toxicity was some repaired by tcMG. tcMG may be useful for prevention and treatment of TCDD induced toxicity.

Effects of Hwanggimacmundong-tang on Body Weight and Gene Expression in Obese Rats with Estrogen Deficiency (황기맥문동탕이 에스트로겐 결핍 Rat의 체중 변화와 비만 관련 유전자 발현에 미치는 영향)

  • Objectives: This study was conducted to assess the effects of Hwanggimacmundong-tang extract (HM) on obese rats with estrogen deficiency. Methods: The experiments were performed with ovariectomized rats as estrogen-deficient obesity model. Rats were grouped NC (Normal Control Group; sham operation group), OC (Obesity Control Group; estrogen-deficient group), HML (20 mg/kg/day), HMH (100 mg/kg/day). HM was administered orally for six weeks. Body weights and serum lipid level were measured, and real-time PCR was performed to estimate the effect of HM on gene expression in liver. Results: HM decreased the total cholesterol and triglyceride in serum. And HM increased leptin, CPT1 gene expression in liver tissue of obese rats with estrogen deficiency. However HM decreased $PPAR{\gamma}$, $PGC-1{\alpha}$, HMGCR, CYP8B1, LPL, ACAT1, ACAT2, ApoB, ACOX gene expression in liver tissue of obese rats with estrogen deficiency. Conclusions: It is concluded that HM reduced total cholesterol and triglyceride in serum, and regulate gene expression related to lipid metabolism. These results indicate Hwanggimacmundong-tang that might have potential for treatment of obesity and complications during the menopause caused by estrogen-deficiency.

Intragastrically Applicated CCl4-Thiopental Sodium Enhanced Lipid Peroxidation and Liver Fibrosis (Cirrhosis) in Rat: Malonedialdehyde as a Parameter of Lipid Peroxidation Correlated with Hydroxyproline as a Parameter of Collagen Synthesis (Deposition)

  • We investigated the pathogenesis of liver tissue damage during the lipid peroxidation and fibrogenesis with the observation of correlations between the parameters of collagen synthesis (and deposition) and lipid peroxidation in liver fibrosis (cirrhosis) rats. Rats were randomly divided into two groups, normal and $CCl_4$-thiopental sod. intoxicated group. And the one group was treated intragastrically with the mixture of $CCl_4$-thiopental sod. 3 times per week for 3 weeks. The liver tissue and sera were used for the measurement of hydroxyproline (HYP), malonedialdehyde (MDA) and superoxide dismutase (SOD). Biochemical parameters such as aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), total-bilirubin and blood urea nitrogen (BUN) were measured. Additionally, the expression of collagen ${\alpha}1$(III) and $\beta$-actin mRNA was observed by RTPCR. The histological change in liver tissue was also observed by Masson's trichrome and H&E staining. Correlation analysis was carried by Spearman's rho method. All biochemical parameters except total-bilirubin were significantly higher in the $CCl_4$-thiopental sod. treated group than that of the normal group (p < 0.01). In the $CCl_4$-thiopental sod. treated group, Hyp as a parameter of collagen synthesis (deposition) and MDA as a metabolite of lipid peroxidation, were significantly elevated by 1.98 and 2.11 times higher than that of the normal group (p < 0.001) respectively. The activity of SOD in the $CCl_4$-thiopental sod. treated group is decreased significantly by 44.8% (p < 0.001). And collagen ${\alpha}1$(III) mRNA was more expressed in the $CCl_4$-thiopental sod. treated group than that of the normal group. However, the expression of $\beta$-actin mRNA is showed similar in both of groups. A good correlation was observed between the content of hyp and MDA concentration (r = 0.70, n = 40) in the two groups. And the correlation between the levels of hyp and SOD (r = -0.71, n = 25) is also reliable. However, no correlation were observed between MDA concentration and SOD (r = -0.40, n = 25) in the two groups. Elevated levels of MDA in $CCl_4$-thiopental sod. treated rats indicated enhancement of lipid peroxidation, which is accompanied by a decrease in SOD activity. Moreover, we could confirm that the parameters of collagen synthesis (and deposition) is in good correlation with the metabolite of lipid peroxidation (MDA) and the lipid peroxidation antagonizing enzyme (SOD). Hence, we propose that (1) lipid peroxidation and collagen synthesis (and deposition) could be enhanced by intragastrically application of $CCl_4$-thiopental sod. during a short terms. And (2) the intoxication of $CCl_4$-thiopental sod. could be used for monitoring of lipid peroxidation and collagen synthesis (and deposition) for test of antioxidant and antifibrotic agent.
