EXPERIMENTAL STUDY ON THE EFFECTS OF THE ANTICANCER AGENT(ENDOXAN) TO THE RAT'S TEETH (항암제가 치아조직에 미치는 영향에 관한 실험적 연구)

  • The author studied the effects of anticancer Endoxan to the rat's teeth. Two hundred pregnant rats were used. The experimental rats were given an intramuscular injection of Endoxan as nitrogen mustard derivatives 10mg. per 40gm of body weight. In order to investigate effects of Endoxan to the teeth of rats the control and experimental rats group wee sacrificed on 1,3,5,7 and 14days, and made preparation for histopathologic findings. The rusults were as follows: 1. In several days (about 5days) after injection of Endoxan, tooth germs of cap stage, bell stage, and dental lamina were irregular arranged form as compared with in the control group teeth. 2. Irregular arranged cell were recovered from 14days after injection of Endoxan.

Studies on the Air-Liquid Interface Culture as an Experimental Model for Physiology and Pharmacology of Tracheal Epithelial Cells (기관(氣管) 상피세포 생리 및 약리 실험모델로서의 공기-액체 접면 일차배양법 연구)

  • In this study, we intended to get a preliminary data for establishing rat tracheal surface epithelial(RTSE) cell culture system as an experimental model for physiology and pharmacology of tracheal epithelial cells. Primary culture on the membrane support and application of the air-liquid interface system at the level of cell layer were performed. The cell growth rate and mucin production rate were measured according to the days in culture. The results were as follows: this culture system was found to manifest mucocilliary differentiation of rat tracheal epithelial cells, the cells were confluent and the quantity of produced and released mucin was highest on culture day 9, the mucin was mainly released to the apical side and tbe free $^3{H}$-glucosamine which was not incorporated to process of synthesis of mucin was left on the basolateral side. Taken together, we suggest that air-liquid interface culture system can be used as a substitute for immersion culture system and as an experimental model for in vivo mucus-hypersecretory diseases.

The Effects of Sophorae radix Extracts on CGRP Immunoreactive Neurons of Spinal Cord and Ganglia in Experimental Arthritic Rat Model (실험적 관절염 흰쥐 모델에서 고삼추출액이 척수와 척수신경절의 CGRP 면역반응 신경원에 미치는 영향)

  • To investigate the antiinflammatory and analgesic effects of Sophorae radix extracts administered to the arthritic rat model, immunohistochemical stains for CGRP in the L4, L5 and L6 spinal cord and ganglia were done, and paw swelling thickness were measured. Complete Freund,s Adjuvant(CFA) were injected to subcutaneous tissue of left foot paw of rats to induce arthritis. Sophorae radix extracts was administered immediately after CFA injection for 10 days. The spinal cord and ganglia were frozen sectioned(30㎛). These sections were stained by CGRP immunohistochemical staining method, and observed with light microscope. The results were as follows : 1. The change of paw swelling thickness of experimental group decreased from 4 day to 10day after CFA injection compared to control group. 2. The change of differential leukocytes counts of experimental group increased the ratio of lymphocytes. and decreased the ratio of neutrophils compared to control group. 3. The change of CGRP immunoreactive nerve fiber of dorsal horn of experimental group was dense stained compared to control group. 4. The number of CGRP immunoreactive neurons of L4 and L5 spinal cord of experimental group was less than in those control group. These results suggested that Sophorae radix extracts reduces the number of CGRP immunoreactive neurons and nerve fibers of spinal cord and ganglia, and decrease paw swelling thickness in arthritic rat model, which may be closely related to analgesic and antiinflammatory effects of Sophorae radix.

Effects of ${\omega}-3$ and ${\omega}-6$ Fatty Acids from Prenatal to Growing Period on the Brain Growth and Behavioral Development of the Rats ($\omega3$$\omega6$계 지방산이 흰 쥐의 뇌 성장과 지능발달에 미치는 영향)

  • This study was designed to find out the effects of $\omega$-3 and $\omega$-6 polyunsaturated and saturated fatty acid from prenatal to growing period on the brain growth and behavioral development of rats. Rats(Sprague-Dawley strain) were fed experimental diets-fish oil, corn oil or beef tallow-with different contents of $\omega$-3 and $\omega$-6 fatty acids throughout the prenatal and lactational period and up to 10 weeks of age. DNA and RNA concentration of rat brain were determined at 0, 3, 6 weeks of age and choline and acetylcholine concentrations were analyzed at 10 weeks of age. When the rats were 7 weeks of age, position reversional test in a Y-shaped water maze for 4 weeks was measured. The experimental results obtained are summarized as follows. Food intakes were significantly lower in fish oil group and body weight gain was low in the group fed beef tallow and the groups fed fish oil and corn oil were somewhat good. Food efficiency ratio was not significantly different among the groups. Brain weight was not affected by the fatty acid composition of experimental diets and DNA and RNA concentration of the rat brain were consistently maintained at the same level. It was not different significantly among the dietary groups in the DNA and RNA concentrations of the rat brain during the experimental period. The acetylcholine concentration in the fish oil group was somewhat higher than the other groups. The position reversional test in a Y-shaped water maze showed a significant difference the score of test among the experimental groups. The score of the rats fed the fish oil diet was significantly higher than the other groups and the concentration of acetylcholine in brain were too. Therefore the correlatin between the Y-shaped water maze test score and the acetylcholine concentratin in the brain was found. Above finding support the content that dietary fatty acid composition does not affect to the brain cell number and cell size but the behavior development is influenced. Therefore, the improvement of behavior development is required the effective usage of finny tribe.

Modulation of Branched-Chain Amino Acid Metaolism by Exercise in Rats

  • A variety of important roles for branched-chain amino acids in metabolic regulation has been suggested. Branched-chain $\alpha$-keto acid dehydrogenase(BCKAD) complex is a rate limiting enzyme in branched-chain amino acid metabolism. The purpose of this study was to examine the effects of exercise on the activity and activity state of branched-chain $\alpha$-keto acid dehydrogenase in rat hert and liver thssues. Forty-eight Sprague-Dawley rats were assigned into three experimental groups : sedentary control, exercised, or exercised-rested. Submaximal exercise(running) for two hours significantly increased basal activity without a change in total activity in both tissues, with a concomitiant increase in activity state of the enzyme complex. At 10 min post-exercise, heart enzyme activity significantly decreased, though not to the control level, while liver enzyme activity remained unchanged. These data suggested that the exercise-induced increase in branched-chain $\alpha$-keto acid decarboxylation in rat tissues may not be the result of enzyme synthesis, but rather is due to increased activity of the BCKAD.

  • Currently numerous methods are in use for myocardial hypothermia as a myocardial preservation modality for cardiac operation. During cardiac ischemia after crystalloid cardioplegia[4C GIK solution], topical cold saline[Group I, a=9], topical ice slush[Group II, n=9] and topical ice chip[Group III, a=10] have been compared for myocardial surface cooling in the isolated rat heart model of cardiopulmonary bypass. During postischemic period, hemodynamic functions[aortic flow, coronary flow, peak aortic pressure and heart rate], biochemical enzymatic activities and cellular injuries with electron microscope were evaluated in this isolated rat heart perfusion model. Postischemic aortic flow, cardiac output and peak aortic pressure in Group I and Group II recovered better than Group III.[p< 0.05] Postischemic creatine kinase and lactate dehydrogenase leakages in Group II and Group III increased more than Group l and postischemic mitochondrial swelling in Group III was more severe than Group I, and Group II.[p< 0.05] These results suggest that topical cold saline was the better method than topical ice slush or topical ice chip as a myocardial preservation modality in the isolated rat heart model of cardiopulmonary bypass.

  • It is reasonably well known that there is a relationship between myelin formation and methylation of myelin basic protein in nerve cells. One of the suggestions is that arginine methylation of myelin basic protein could be of aid in the conjugation of myelin protein with the nonpolar lipid to form myelin. Abnormality in methylation of myclin basic protein might induce the neurological diseases in experimental animals as well as in human being. In the biological system, the methylation reaction is catalyzed by protein methaylse I using S-adenosyl-L-methionine as methyl donor. In this study, we examined the changes of S-adenosyl-L-methionine concentration and protein methylase I activity in developing rat brain tissues. The results are sumraerized as followings: (1) In brain tissues of fetus rat, the concentration of S-adenosyl-L-methionine was gradually decreased until to birth. However, the concentration in brain tissues of infant rat was suddenly increased at 7th day(just before myelination occur) birth. (2) Protein methylase I activity was decreased until to birth in brain of fetus rat and increased temporally just after birth, However, the enzyme activity showed no changes around 7th day after birth.

Effects of Garlic on the Blood Lipids and Other Serum Components in Rats (흰쥐에 마늘 투여로 혈액의 지질량과 그외 혈액성분 변화 고찰)

  • In present study possible hypolipidemic effects of garlic were investigated in an experimental model which consisted of 4 groups of Wistar rats(three weeks old): CO group received a basal diet(as a standard diet) contained 6.3% of corn oil. LO group was taken the experimental diet in which only 6.3% corn oil in basal diet was substituted by 12.6% of lard oil. So the experimental diet was somewhat high saturated fat diet. LG1 group was treated orally with garlic juice as 1% of raw garlic in the diet together with the same diet as the diet for LO group. LG2 group was taken 2% of garlic under the same dietary condition as that of LG1 group. The rat body weights prior and posterior to the experimental period were measured and the amount of the experimental dietary intake was determined at every 3 day interval. After the experimental dietary period of 30 days the blood obtained from all the sacrificed rats were analyzed for the biochemical parameters. Over the one month period of experiment there was no abnormality or apparent change in appearance and activity or diet consumption in all experimental rat groups. LO group fed a diet rich in lard showed significantly increased body weight gain rate by 30% and also elevated levels of triglyceride, total cholesterol and LDL cholesterol by 29.9%, 27.4% and 62.3% respectively and no significant difference in the levels of HDL cholesterol, GOT, GPT, blood glucose and blood urea nitrogen as compared to those of control group. So it seemed that the high calorie diet in LO group mainly contributed to the significant increase in body weight gain rate and other biochemical parameters. In spite of 1% garlic treatment LG1 group had significantly increased body weight gain rate by 25.2%, levels of triglyceride, total choesterol and LDL cholesterol by 25.2%, 24.6% and 50.5% respectively as compared to control and LO group, and no increase in the levels of HDL cholesterol, blood glucose and blood urea nitrogen. At garlic 1% diet there was no hypolipidemic efficacy in rat serum. Under the treatment with garlic 2% in diet LG2 group showed significantly increased body weight gain rate by 19.7% as compared to control and elevated levels of GOT, GPT and BUN by 85.8~96.4%, 127~148% and 88% respectively as compared to control, but LG2 group had significantly reduced levels of triglyceride, total cholesterol and LDL cholesterol as compared to LOand LG1 group. In present study there were no significant change in HDL cholesterol and blood glucose value. In LG2 group GOT, GPT and blood urea nitrogen values were slightly increased presumably due to the inhibiting effects of garlic on the hepatic or renal function of rats. Nevertheless in this study garlic may have some demonstrable hypolipidemic effects in rat.

A Study of the Effects of Bone Morphogenetic Protein on the Characteristics of Rat Periodontal Ligament and Calvaria Cells (골형태형성단백질이 백서치주인대세포와 두개관세포에 미치는 영향)

  • Bone morphogenetic protein-2/4 (BMP-2/4) are members of Transforming Growth $Factor-{\beta}\;(TGF-{\beta})$ superfamily and they may differentiate the osteoprogenitor cell and induce formation of cartilage and bone in vivo. This study was performed to investigate the effects of bone morphogenetic protein-2/4 on the characteristics of rat periodontal ligament cells(RPDL) and rat calvaria cells(RCV). In the control group, the cells were cultured alone with Dulbeco's Modified Eagle's Medium contained with 20% fetal bovine serum, $100{\mu}/ml$ penicillin, $100{\mu}/ml$ streptomycin. In the experimental groups, recombinant human bone morphogenetic protein-2/4 (25ng, 100ng, 250ng/ml) were added into the above culture condition. And then each group was characterized by examing the cell proliferation at 1, 2, 3, 5, 7th day, the amount of total protein synthesis and alkaline phosphatase activity at 2, 5, 7th day. And also, the calcified nodule was examed. The results were as follows ; 1 . Both RCV and RPDL cells in both control and experimental groups proliferated during the entire experimental period, but there is no stastically significant difference according to the BMP-2/4 concentration. 2 . Amount of total protein synthesis of both cells in both groups was steadily increased until 5th day, but all experimental groups were significantly different from the control group at 7th day. 3. Alkaline phosphatase activity of both cells in both groups was increased during the entire experiment period. In RCV cells, the experimental group treated with 100ng/ml and 250ng/ml BMP-2/4 were significantly different from the control group at 7th day. In RPDL cells, the experimental group treated with 100ng/ml and 250ng/ml BMP-2/4 were significantly different from the control group at 5th day, and all experimental groups were significantly different from the control group at 7th day. 4. In the both of the cultured Rat Periodontal ligament and calvaria cell treated with BMP-2/4 to compared with control group, it revealed more rapid cell polarization, cell aggregation and hyperchromatic stained on HE agent, and even though only 1 day treated with BMP-2/4 both RPDL and RCV showed more rapid cell reaction than control group. More sensivitve cell reaction of RCV were observed than RPDL in this experiment. From the above results, we could conclude that BMP-2/4 influenced the induction, proliferation and differentiation of bone forming cells

Effects of continuous force application for extrusive tipping movement on periapical root resorption in the rat mandibular first molar

  • Objective: The purpose of this study was to clarify the effects of continuous force application for extrusive tipping movement and occlusal interference on periapical root resorption in the rat mandibular first molar. Methods: We constructed an appliance comprising a titanium screw implant with a cobalt-chromium post as the anchorage unit and a nickel-titanium closed coil spring (50 cN) as the active unit. Force was applied on the mandibular left first molar of rats for 8 (n = 10) and 15 days (n = 10; experimental groups), with the tooth in occlusion. Five rats were included as a non-treated control group to examine the body effect of the appliance. Active root resorption lacunae, identified using tartrate-resistant acid phosphatase, were evaluated in terms of the length, depth, and area. Results: The rat mandibular first molars were mesially tipped and extruded in the occlusal direction. This mesio-occlusal tipping movement and occlusion resulted in the formation of a compression zone and active root resorption lacunae in the distoapical third of the distal roots. However, there was no significant difference in the amount of root resorption between the two experimental groups. The control group did not exhibit any active root resorption lacunae. Conclusions: Periapical root resorption was induced by continuous extrusive tipping force and occlusal interference in rat mandibular molars. These data suggest that we orthodontists had better take care not to induce occlusal interference during our orthodontic treatment.
