백서 두개골 결손부에서 입자 크기에 따른 Calcium aluminate cement의 효과 (The effects of calcium aluminate cement according to particle sizes on calvarial bone defects in rats)

  • This present study was carried out to find the effects of calcium aluminate cement($CaO\;{\cdot}\;Al_2O_3$, CAC), which has been developed with bio-compatibility and mechanical properties, in biological environments. Two different particle sizes of CAC - 3.5${\mu}m$ vs. 212${\sim}$250${\mu}m$ which is recommended in periodontal bone grafting procedures-were filled in 8mm calvarial defect in Sprague-Dawley rat. The specimens were examined histologically, especially the bone-cement interface and the response of surrounding tissues. The results are as follows; 1. In the control group, inflammatory cells were observed at 2 weeks. At 8 weeks, periosteum and dura mater were continuously joined together in the defect areas. But in the center of defect area were filled up with the loose connective tissues. 2. In the experimental group l($212{\mu}m{\sim}250{\mu}m$ particle), immature bone was formed and outermost layer was surrounded by osteoid layer at 2 weeks. Osteoblasts were arranged between immature bone and osteoid layer. And, osteoid layer was remained until 8 weeks after surgery. 3. In the experimental group 2, periosteum and dura mater lost its continuity at 2 weeks. Scattering of CAC particles and infiltration of inflammatory cells were observed, which this findings deepened at 8 weeks. The result of this study shows that when calvarial defects in white rats are filled with calcium aluminate cement of 212${\sim}$250${\mu}m$, the materials are to be bio-compatible in growth and healing on surrounding tissues. When further researches are fulfilled, such as direct bone adhesion and bone regeneration ability, it's possible that CAC could be applied to various periodontology fields in the future.

간섬유화(경화)를 유도한 실험동물에서 생약 추출물(LH)의 항산화와 항섬유화 효과 검색 (The Screening of Antioxidant and Antifibrotic Effect from Water Extracts of Herbs(LH) in Biliary Liver Fibrosis (Cirrhosis) Induced Rsts)

  • The pathogenesis of cholestatic liver injury as well as the modulation of hepatic fibrogenesis is causally associated with involvement of reactive oxygen species (ROS) and free radical reactions. In this study, we investigated whether dried extracts of oriental medicine (LH) have antioxidant and antifibrotic effect under the biliary liver fibrosis (cirrhosis) c ondition. The female Sprague-Dawley rats were divided in six groups (Normal, N-LH, op-2, op-4, opLH-2, opLH-4) and were observed in 2 weeks or 4 weeks. For this purpose the rats were operated by bile duct ligation/scission (BDL/S), which induced to liver fibrosis and cirrhosis. After surgery, the prepared LH was administered p.o. 2 mι/day/rat in 2 weeks or 4 weeks for opLH groups. During the observation period, jaundices appeared in eyes, ears and tail of all BDL/S operated rats. And at the time of sacrifice, cholestasis was observed in proximal bile duct, especially the color of bile juice and urine in opLH-4 group showed more clear than op-2, op-4 and opLH-2 group. The value of clinical parameters and product of lipid peroxidation (MDA) in sera and the hydroxyproline (hyp) content in liver tissue were significantly increased in all liver fibrosis (cirrhosis) developed rats (p<0.001~0.05). Among the clinical parameters of sera, value of BUN, ALP in opLH-4 group showed significantly lower than in op-4 group (p<0.05, p<0.001). The content of hyp in opLH-2, opLH-4 group (478.0 $\pm$ 134.3 $\mu\textrm{g}$/g 897.5 $\pm$ 118.2 $\mu\textrm{g}$/g) showed lower than in op-2, op-4 group (528.9 $\pm$ 220.7 $\mu\textrm{g}$/g, 1023.8 $\pm$ 277.1 $\mu\textrm{g}$/g) and then the value of MDA in opLH-4 was also significantly reduced to 59.4% of that in op-4 group (p<0.001). The histological change (bile duct proliferation, fibrosis, collagen bundle) was similarly observed in op-2 group and in opLH-2 group, but the weak fibrosis and bile duct proliferation were observed in opLH-4 group compared with in op-4 group. Our data indicate that the 4 weeks treatment with LH extract suppressed lipid peroxidation and inhibited fibrotic (cirrhosis) process, and experimental cholestatic liver disease is associated with increased lipid peroxidation in BDL/S operated rats. Hence we concluded that the measurement of MDA and hyp can be useful monitor for the screening of antioxidant and antifibrotic effect in experimental liver fibrosis (cirrhosis), and LH has been shown to have hepatoprotective effect, antifibrotic effect and antioxidant effect.

퇴행성 골관절염에 대한 HPL-04의 효과 (Effects of HPL-04 on Degenerative Osteoarthritis)

  • 본 연구에서는 HPL-04가 골관절염의 예방 및 치료 약물로서의 가능성을 탐색하여 다음과 같은 결론을 얻었다. HPL-04는 연골세포 생존율과 연골형성과 관련된 collagen type II, SOX 9 그리고 aggrecan의 유전자 발현을 유의성 있게 증가시킬 뿐만 아니라 염증성 인자와 관련 있는 MMP-2, 9도 유의성 있게 감소시켰다. HPL-04는 MIA에 의해 유도된 퇴행성 골관절염에서 관절연골의 파괴와 골 침식 등 연골의 변성을 억제했으며 proteoglycan의 소실을 유의성 있게 감소시켰다. 이에 본 연구는 HPL-04가 부작용이 적고 약리효과가 뛰어나 골관절염 예방 및 치료제 개발에 활용될 수 있을 것으로 본다.

β-Carotene의 섭취가 당뇨 유도 흰쥐의 간조직 항산화효소 활성과 Glutathione 함량에 미치는 영향 (Effect of Dietary Supplementation of β-Carotene on Hepatic Antioxidant Enzyme Activities and Glutathione Concentration in Diabetic Rats)

  • 본 연구에서는 당뇨병의 치료에 중요한 저해요인으로 제기되고 있는 당뇨합병증을 예방하고자 녹황색 채소 등을 통해 식사 시 한국인들이 쉽게 섭취할 수 있는 ${\beta}$-carotene의 혈관계 합병증 예방효과를 분석하고자 하였다. 간 미토콘드리아에서의 catalase 활성은 ${\beta}$-carotene을 급여한 비당뇨군에서 유의적으로 높은 활성을 나타내었으나 당뇨군에서는 ${\beta}$-carotene 급여에 의한 영향을 나타내지 않았다. 간 사이토졸 내의 superoxide dismutase 활성은 ${\beta}$-carotene을 급여하지 않은 당뇨군에서 그 활성이 가장 저하되었으나 ${\beta}$-carotene 공급에 의해 유의적으로 증가되었다. 간 사이토졸 내의 glutathione-S-transferase 활성은 ${\beta}$-carotene의 급여에 의한 차이가 없었다. 간 마이크로솜에서의 glucose-6-phosphatase 활성은 대조군에 비하여 ${\beta}$-carotene을 급여하지 않은 당뇨군에서 유의적으로 높게 나타났으나 ${\beta}$-carotene을 급여한 당뇨군에서는 활성이 유의적으로 저하되었다. 간조직의 환원형 glutathione 함량은 당뇨군에서 유의적으로 감소되었으며, 당뇨군에서 ${\beta}$-carotene의 급여로 증가하였다. 이상의 결과에서 볼 때 ${\beta}$-carotene의 섭취는 당뇨 유도에 의한 산화스트레스가 증가된 생리적 상태에서 glutathione의 소모를 감소시키고, 항산화 효소계 중 특히 superoxide dismutase 활성을 유도하여 과산화 라디칼을 환원시킴으로써 활성산소에 의해 유발되는 산화적 손상의 일차적 방어에 관여한 것으로 여겨진다. 따라서 인체시험 등을 통한 후속연구와 연계하여 ${\beta}$-carotene 수준을 적절하게 보충 섭취한다면 당뇨로 인한 항산화계 활성의 변화를 조절함으로써 산화스트레스에 대한 보호효과를 나타내어 당뇨병 합병증의 예방에 기여할 수 있을 것으로 여겨진다.

계면활성제 유발된 피부장애의 아로마 에션셜 오일의 유효성 연구 - Palmarosa, Neroli essential oil을 중심으로 - (Efficiency of Essential oil about the Skintroubles induced Surfactants - Palmarosa, Neroli essential oil -)

  • 주방세제로 유도한 건성피부에 대한 Palmarosa, Neroli essential oil의 유효성 연구결과는 다음과 같다. 단백질 분석결과 Neroli essential oil 처리 군이 대조군과 가장 유사하게 나타났고 그 다음으로 Palmarosa + Neroli essential oil 처리 군이 대조군과 유사하게 복구되어 가고 있는 것으로 나타났고 그 다음으로 Palmarosa의 순이다. 표피층의 형태적 관찰결과로는 Palmarosa essential oil에 의한 각질층의 이탈과 Neroli essential oil에 의한 각질층의 복구에 의한 표피층의 비후 현상을 볼 수 있었으나 각각의 오일 처리군에서 계면활성제 처리 후 피부 염증에 대한 재생효과가 있음을 알 수 있었다. 염증 반응을 알아보기 위한 Mast Cell 실험에서 보면 대조군에 비해 계면활성제 연속처리군에서는 mast cell의 크기와 수가 증가되었음을 확인 할 수 있다. Neroli essential oil이 처리된 각각의 군에서는 mast cell의 수가 확연히 감소된 것을 볼 수 있다.

새롭게 개발된 비정질의 Calcium Phosphate가 백서두개골의 골재생에 미치는 영향 (The effects of novel biodegradable amorphous Calcium Phosphate on bone regeneration in rat calvarial defects)

  • Purpose: The purpose of this study was to evaluate the bone regeneration of novel biodegradable amorphous calcium phosphate. Materials and Method: An 8-mm, calvarial, critical-size osteotomy defect was created in each of 20 male Sprague-Dawley rats(weight $250{\sim}300g$). The animals were divided into two groups of 10 animals each and allowed to heal for 2 weeks(10 rats). The first group was the control group and the other group was the experimental group which received the novel biodegradable calcium phosphate. Results: The healing of the calvarium in the control group was uneventful. The histologic results showed little bone formation in the control group. The experimental group which received the novel biodegradable calcium phosphate showed a normal wound healing. There were a lot of new bone formation around the biomaterial in 2 weeks. The bone formation increased in 8 weeks when compared to 2 weeks and there was a significant bone increase as well(P<0.01). The nobel biodegradable calcium phosphate showed statistical significance when compared to the control group (P<0.05). The novel biodegradable calcium phosphate in 8 weeks showed a significant increase in bone formation when compared to 2 weeks $(40.4{\pm}1.6)$(%). The biodegradable calcium phosphate which is made from mixing calcium phosphate glass(CPG), NaCO and NaOH solution, is biocompatible, osteoconductive and has a high potency of bone formation. Conclusion: We can conclude that the novel biodegradable calcium phosphate can be used as an efficient bone graft material for its biodegradability and osteoconductivity.

Effects of Green Tea Infusion on the Preneoplastic Lesions and Peroxidation in Rat Hepatocarcinogenesis

  • The effect of green tea drinking on the hepatocellular chemical cacinogenesis have been studied. Placental glutathione S-transferase(GST-P) positive foci area in a liver tissue, contents of thiobarbituric acid reactive substances(TBARS), total cytochrome P450 and glucose 6-phospphatase(G6P) activity in hepatic microsomes were investigated. Weaning Sprague-Dawley male rats were fed AIN-76A diet with deionized water or green tea infusion, Rats of CTR and CTR+ groups were provided deionized water while GTI and GTI+ groups were provided green tea instead of deionized water for the entire experimental period of 13weeks. Rats of GTP and GTP + groups had deionized water for the first 6 weeks and switched to green tea for the last 7weeks of the experimental period. CTR+, GTI +, and GTP + groups were carcinogen treated groups, Diethylnitrosamine(DEN) was injected as a single dose of 200mg/kg body weight intraperitoneally after 4 weeks of feeding. 2-Acetyla-minofluorene(AAF) was used as a carcinogen proliferater and suppled in the diets of carcinogen treated rats as 0.02% content for the last 6weeks starting from 2weeks after DEN injection. Rats were sacrificed after 13week weeks of feeding. The area and number of GST-P positive foci detected in carcinogen treated rats were decreased by green tea ingestion but when timing and duration of green tea ingestion was delayed after promotion period as in GTP + group, GST-P positive foci were not decreased as much as in GTI+ group. TBARS contents of carcinogen treated rats decreased by 13weeks of green tea ingestion but GTP groups did not show statiscally significant differences. G6P activities tended to decrease by carcinogen treatment but changes were not statiscally significant by green tea ingestion. Total cytochrome P450 contents were increased by carcinogen treatment. Thirteen weeks of green tea ingestion (GTI) also increased to total cytochrome P450 contents while 7weeks of green tea ingestion(GTP) did show any effects. These results suggest that green tea has suppressive effects on hepatocellular chemical carcinogenesis probably through the activities of antioxidant compounds. Chronic Alcohol Consumption Results in Greater Damage to the Pancreas Than to the Liver in the Rats

  • Alcohol consumption increases the risk of type 2 diabetes. However, its effects on prediabetes or early diabetes have not been studied. We investigated endoplasmic reticulum (ER) stress in the pancreas and liver resulting from chronic alcohol consumption in the prediabetes and early stages of diabetes. We separated Otsuka Long-Evans Tokushima Fatty (OLETF) rats, a type-2 diabetic animal model, into two groups based on diabetic stage: prediabetes and early diabetes were defined as occurrence between the ages of 11 to 16 weeks and 17 to 22 weeks, respectively. The experimental group received an ethanol-containing liquid diet for 6 weeks. An intraperitoneal glucose tolerance test was conducted after 16 and 22 weeks for the prediabetic and early diabetes groups, respectively. There were no significant differences in body weight between the control and ethanol groups. Fasting and 120-min glucose levels were lower and higher, respectively, in the ethanol group than in the control group. In prediabetes rats, alcohol induced significant expression of ER stress markers in the pancreas; however, alcohol did not affect the liver. In early diabetes rats, alcohol significantly increased most ER stress-marker levels in both the pancreas and liver. These results indicate that chronic alcohol consumption increased the risk of diabetes in prediabetic and early diabetic OLETF rats; the pancreas was more susceptible to damage than was the liver in the early diabetic stages, and the adaptive and proapoptotic pathway of ER stress may play key roles in the development and progression of diabetes affected by chronic alcohol ingestion.

흰쥐대구치 치수노출후 치수조직내 CGRP함유 신경섬유의 분포에 관한 면역조직화학적 연구 (AN IMMUNOHISTOCHEMICAL STUDY ON THE DISTRIBUTION OF CGRP CONTAINING NERVE FIBERS AFTER PULP EXPOSURE IN RAT MOLAR)

  • The purpose of this study was to investigate the distribution of calcitonin gene-related peptide(CGRP) containing nerve fivers after pulp exposure in rats. The Spague-Dawley rats weighing about 250 - 300g were used. The animals were devided into normal control group and experimental groups. Experimental animals were sacrified on 2, 4, 7, 10 days after pulp exposure. The maxillary teeth and alveolar bone were removed and immersed in the 4% paraformaldehyde plus 0.1M phosphate buffer (pH 7.4). Serial frozen $50{\mu}m$ thick sections were cut with a cryostat. In the immunohistochemical staining procedure, the rabbit CGRP antibody was used as a primary antibody. The sections were incubated for 48 hours at $4^{\circ}C$, and placed into biotinylated anti-rabbit IgG as a secondary antibody and incubated in ABC (avidin-biotin complex), The sections were visualized by 0.05% 3.3 diaminobenzidine tetrahydrochloride. The results of this study were as follows: 1. In control group, CGRP containing nerve fibers ran parallel to the long axis of root and reached the coronal pulp. They were distributed on Raschkow plexus under the odontoblastic layer. 2. In 2 day group after pulp exposure, tissue necrosis and acute inflammation occurred and CGRP containing nerve fibers increased. In 4 day group, the necrotic tissue extended to the pulp and CGRP containing nerve fibers were distributed around the inflammation zone. 3. In 7 day group after pulp exposure, pulp necrosis occurred, and in 10 day group, the abscess under the necrotic pulp extended to the root apex area and CGRP containing nerve fibers were not observed in root canals. 4.The sprouting of CGRP nerve fibers was most remarkable at the pulp chamber under injury in 4 day group, and it was found at inflammation zone under the necrotic tissue in 7 day group and the remaining root pulp tissue in 10 day group. As mentioned above, CGRP nerve fibers had a tendency to increase around the inflammatory zone, especially around the acute inflammation tissue, when compared with control group. It is suggested that CGRP nerve fibers maybe related to the control of inflammatory response of pulp tissue.

트레드밀 운동이 척수손상 백서의 운동기능 회복에 미치는 효과 (The Effects of Treadmill Exercise on the Recovery of Functional Capacity in Spinal Cord Injured Rats)

  • Purpose: This study was designed to investigate the effects of treadmill exercise on functional recovery after rat with experimental SCI. Methods: SCI was induced by the NYU-spinal cord impactor(NYU, USA) dropped a weight of 10 gm after laminectomy. Experimental groups were divided into the Group I (normal), Group II (control) and Group III(treadmill exercise). After 2 days of the operation, 24 rats(group II, III) were trained to walk on treadmill for 21 days twice/day, 15 min/session. After operation, rats were tested at modified Tarlov scale at 1, 2, 3, 4 days with divided into 2 groups, and Motor behavior test(BBB locomotor rating scale, Grid walking test, Narrow beam crossing test, Modified inclined plane test) was examined at 1, 3, 7, 14 and 21 days. Histopathological study were performed at 1. 3, 7, 14 and 21 days by H&E, Luxol Fast Blue staining were same times. Results: After SCI an improvement of motor behavior was shown group II, III. The motor behavior test of group Ill showed considerable improvement until 14 days. Conclusion: These results suggest that treadmill exercise treatment can playa role in facilitating recovery of locomotion following spinal cord injury.
