Effect of Amniotic Membrane to Reduce Postlaminectomy Epidural Adhesion on a Rat Model

  • Objective : Epidural fibrosis and adhesion are the main reasons for post-laminectomy sustained pain and functional disability. In this study, the authors investigate the effect of irradiated freeze-dried human amniotic membrane on reducing epidural adhesion after laminectomy on a rat model. Methods : A total of 20 rats were divided into two groups. The group A did not receive human amniotic membrane implantation after laminectomy and group B underwent human amniotic membrane implantation after laminectomy. Gross and microscopic findings were evaluated and compared at postoperative 1, 3 and 8 weeks. Results : The amount of scar tissue and tenacity were reduced grossly in group of rats with human amniotic membrane implantation (group B). On a microscopic evaluation, there were less inflammatory cell infiltration and fibroblast proliferation in group B. Conclusion : This experimental study shows that implantation of irradiated freeze-dried human amniotic membrane reduce epidural fibrosis and adhesion after spinal laminectomy in a rat model.

ET-1 RIA and Immunocytochemistry on EAE-induced lewis rat

  • Endothelin-1, which is a peptide originally isolated from cultured porcine aortic cell, has been found to play a crucial role in potentiating the vasoconstriction mitogenesis, and chemotaxis. In the present study, we examined the level of endothelin-1 in the brain, spinal cord and blood from rat with experimental allergic encephalomyelitis(EAE). At the peak stage of EAE(grade 3), endothelin-1 level in the spinal cord of rat with EAE increased two folds as compared with that of sham-treated rats, and subsequently decreased to the level of control at the recovery stage. In the endothelin-1 immunocytochemistry, endothelin-1 immunopositive cells and ED-1, macrophage marker immunopositive cells observed in the spinal cord of peak stage(grade 3). These Findings suggest that endothelin-1 play the important role in progression of EAE.

대장내 항염증 작용이 있는 천연 소재의 개발 (Studies on Protective Effect of Herbal medicines against Experimental Inflammtory Bowel Disease Model)

  • The Efficacy of PSB-2061, was investigated in comparision with predinisolone in acetic acid and Picrylsulfonic acid solution (TNBS)-induced rat inflammatory bowel disease (IBD) for 5 days. 5 % TNBS solution were administered with polyethylene (P.E) tube inserted to rats intracolon, which causing colitis to the rats. The TNBS control group (the saline treated colitic rat) exhibited ulceration and inflammation of the distal colon with formation of granuloma and pathologic connections. We checked the inflammatory parameters like rat's weight, food intake quantity change during administration. After 5 days, we sacrificed the rats and checked the colon's length, ulcer and pathologic condition. Oral treatment with PSB-2061 resulted in significant recovery of macroscopic parameters like weight and diet intake change. Especially, PSB-2061 extract had a more potent effect than $mesalazine^{(R)}$ on macroscopic colonic damage score. We can suggest that PSB extract could be a promising drug in the treatment of IBD.

Antioxidant Effects of Berchemia berchemiaefolia in Nerve Pain Models

  • Berchemia berchemiaefolia (BB) are climbing plants or small to medium-sized trees that live in Africa, Asia and America. We performed the present study to investigate whether oral administration of Berchemia berchemiaefolia extract (BBE) protects SD rats from pain. The SD rat experimental groups were divided into four groups. Two of the animal model groups were fed on BBE (200 mg/kg or 100 mg/kg). We performed oral acute toxicity test to determine the optimal oral dose of BBE. To explore if BBE alleviated pain in the SD rat, we undertook the tail flick latency test and formalin test. Additionally, we conducted the anti-oxidative test. The findings of the present study suggest that Berchemia berchemiaefolia extract exhibits strong antioxidant and analgesic activities.

  • The present study was designed to determine whether cyclosporine A inhibits Major Histocompatibility Complex Class II antigen[MHC II expression in the allograft rat heart myocardium. In this rat allograft study we also tried to elucidate whether CSA inhibits MHC II in a dose dependent way. Hearts were isolated from LBN rats weighing 200-250 grams and heterotopically transplantated into the abdomen of weight-matched ACI rats. The ACI rats were randomly assigned to one of the three experimental groups according to cyclosporine dosage: {1}control [no CSA , n=6 {2}CSA 5 mg/day , n=5 {3}CSA 10 mg/day, n=5. The transplanted hearts were harvested 5 days postoperatively and analyzed. MHC II expression was detected by indirect immunoperoxidase staining and quantified via computer image analysis system. The % positive area reading was obtained in each slide [50 areas per group and compared to other groups. Significant differences were noted between three groups [p<0.05 . We conclude that CSA inhibits MHC II in heterotopically transplanted allograft rat heart in a dose dependent way.

에탄올 장기 투여에 의한 쥐 심근조직의 산화적 스트레스와 생체내 항산화 효소활성의 변화 (Effect of Chronic Ethanol Administration on Oxidative Stress and Cellular Defence System in Rat Myocardium)

  • The level of oxidative tissue damage caused by free radicals generated from ethanol oxidation was determined in the myocardium of chronic ethanol fed-rats and the protective action of various radical scavenging enzymes was monitored, also. Adult male Sprague-Dawley rats were given ethanol in an amount of 36% of total calories via Lieber-DeCarli liquid diet for 6 weeks. Control group was pair-fed with the diet containing isocaloric amount of dextrin-maltose instead of ethanol. Chronic ethanol administration resulted in the increased amount of myocardial thiobarbituric acid reactive substance(TBARS), th parameter of lipid peroxidation, under our experimental condition. Chronic ethanol ingestion did not cause any change in activities of either glutathione peroxidase or glutathione reductase and glucose-6-phosphate dehydrogenase were decreased after ethanol treatment. Therefore, chronic ethanol administration seemed to cause considerble changes in cellular defense function against oxidative tissue damage in rat myocardium through glutathione utilizing system and radical generation system. However the ultimate net result of chronic ethanol inestion on the myocardium of rat was the oxidative tissue damage revealed by increased TBARS content.

  • In this study, the effects of Boshimdan, which is used in oriental medicine for the treatment of palpitation, chest discomfort, impatience, insomnia, forgetfulness ete., were tested as a part of series of experimental verification of prescriptions treatments for preventing and curing ischemic heart. The results were as follows: 1. Boshimdan did not effectively work on heart rate of the perfused rat heart. 2. Boshimdan did not effectively work on heart pressure of the perfused rat heart. 3. Boshimdan did not effectively work on flow rate of the perfused rat heart. 4. Boshimdan was not effective in activation of GOT. 5. Boshimdan was not effective in activation of GPT. 6. Boshimdan was not effective in activation of LDH. 7. Boshimdan was not effective in activation of CPK.

T-2 toxin이 흰쥐 혈철 중 지질농도에 미치는 영향 (Effects of T-2 Toxin on Lipid Concentration in Rat Serum)

  • This study was designed to observe the effects of T-2 toxin on total cholesterol and lipid concentration in rat serum. T-2 toxin is a secondary metabolite produced by Fusarium sp. which is often found on agricultural products including cereals, and it is a causal material of liver injuries in cattle and humans. When we fed rats with standard diet treated with T-2 toxin, the body weight and feed consumption of rats treated T-2 toxin were decreased. As the results of lipid analysis, the concentrations of total cholesterol and free cholesterol in serum of treated rats were increased compared to non-fed control group, On the other hand, the levels of triglyceride and phospholipid in the serum of T-2 toxin treated experimental groups were declined. In conclusion, T-2 toxin largely influenced on the total cholesterol and lipid levels in rat serum.

Somatic Cell Nuclear Transfer in Rodents, the Little Big Animals

  • Transgenic rats and mice are useful experimental animal models for medical research including human disease model studies. Somatic cell nuclear transfer (SCNT) technology is successfully applied in most mammalian species including cattle, sheep, pig and mouse. SCNT is also considered to increase the efficacy of transgenic/knockout mouse and rat production. However, in the area of reproductive biotechnology, the rodent model is inadequate because of technical obstacles in manipulating the oocytes including intracytoplasmic sperm injection and SCNT. In particular, success of rat SCNT is very limited so far. In this review, the history of rodent cloning is described.

  • The effects of Ginseng saponin on respiration and $Na^+$, $K^+$ content of rat cerebral cortex slices were investigated to determine the action of Ginseng saponin on brain cortex at cellular level. There are many reports for the study of Ginseng on central stimulatory action in experimental animals. The electrical stimulation of slices of cortex causes a loss of potassium. And the respiration is needed to maintain a supply of energy for active cation transport. The reduction in $Qo_2$ is a consequence of primary cessation of active cation transport. Ginseng saponin stimulated respiration which was depressed by Morphine. But there was no significant change of electrolyte. It is suggested that the Ginseng saponin act rather on metabolic process than neural excitatory mechanism in vitro.
