Micropropagation of Echinosophora koreensis Nakai, a Korean Endemic Species in Danger, Using Axillary Buds

  • To establish in vitro axillary bud culture conditions of Echinosophora koreensis Nakai, one of Korean endemic endangered species famous for beautiful flowers, we tested the influence of plant growth regulators (PGRs) in shooting and rooting stage from in vitro plants. In shoot multiplication, addition of 6-benzylaminopurine (BA) to the media induced 2.5 to 3 shoots per bud during 4 weeks of culture. And media including 0.5 mg L-1 thidiazuron (TDZ) produced 3 to 4 shoots per bud. However, zeatin and isopentenyl adenine (2-ip) were not successful to increase shoot number, and the combination treatments of BA with other PGRs were also not effective. Shoots were smaller than 2 cm in length, in most of the treatments. In rooting, naphthalene acetic acid (NAA) treatments in the range of 0.5 to 4.0 mg L-1 appeared to increase rooting rate by 10% to 60% approximately when compared with the control but roots developed with callus clusters. Indole butyric acid (IBA) addition had little effect on rooting (below 10%), while some roots were longer than in NAA treatments and some shoots were longer on high IBA concentrations (4.0 to 8.0 mg L-1). It is suggested that micropropagation is a highly applicable and promising to multiplication and conservation of rare and endangered endemic species.

보길도(전남) 적자봉 일대의 관속식물상 유형별 특성 (Characteristics Type of Vascular Plants in Jeokjabong, Bogil Island(Jeonnam))

  • The vascular plants in Jeokjabong, Bogil Island were listed 350 taxa composed of 89 families, 233 genera, 309 species, 1 subspecies, 36 varieties and 4 forms. Divided into woody plants were 142 taxa (40.6%) and herbaceous plants were 208 taxa (59.4%). Based on the list of rare and endangered species by Korea Forest Service and Korea National Arboretum, 3 taxa were recorded; Celtis choseniana, Asarum maculatum, Rubus sorbifolius, and endemic plants, 10 taxa were recorded; Asarum maculatum, Cephalotaxus koreana, Carpinus coreana, Celtis choseniana, Lespedeza maritima, Indigofera koreana, Dendropanax morbifera, Ligustrum quihoui var. latifolium, Weigela subsessilis, Carex okamotoi, and plant species subject to permission for taking abroad, 4 taxa were recorded; Celtis choseniana, Asarum maculatum, Vaccinium oldhami, Carex okamotoi. Based on the list of floristic regional indicator plants by Korean Ministry of Environment were total 64 taxa (18.3% of all 350 taxa of vascular plants); Ligustrum quihoui var. latifolium in class IV, 19 taxa (Stauntonia hexaphylla, Cinnamomum japonicum, Zanthoxylum ailanthoides, Daphniphyllum macropodum, Centella asiatica, Verbena officinalis, Mitchella undulata, etc.) in class III, 5 taxa (Potentilla dickinsii, Ostericum melanotilingia, Sanicula rubriflora, Caryopteris incana, Teucrium veronicoides) in class II and 39 taxa (Gleichenia dichotoma, Cyrtomium fortunei, Aphananthe aspera, Rubus hirsutus, Meliosma myriantha, Hedera rhombea, Ainsliaea apiculata, etc.) in class I. Based on the list of naturalized plants, 5 families, 17 genera, 20 taxa (Phytolacca americana, Lepidium virginicum, Robinia pseudo-acacia, Verbesina alternifolia, Lolium perenne, etc.) and naturalization rate was 5.7% of all 350 taxa of vascular plants. Hereafter natural ecosystem into disturbance on naturalized plants and legal protection species and rare and endangered species distribute in the Bogil Island, precision investigation after management counterplan of the monitoring a point of view be required.

천마산(경기도)의 식물상 (Flora of Mt. Cheonma (Gyeonggi-do Prov.))

  • 본 연구는 경기도 남양주시에 위치한 천마산의 관속식물상의 조사를 위해 수행되었으며, 이를 대상으로 한반도고유종, 희귀 및 멸종위기식물, 적색식물, 구계학적특정식물종, 기후변화 적응대상 식물, 귀화식물을 조사하였고 또한 이미 조사가 이루어진 주변지역의 산과 종 조성을 비교분석하였다. 2012년부터 2014년까지 총 22회에 걸쳐 현지조사를 실시한 결과, 94과 266속 378종 2아종 46변종 7품종의 총 433분류군의 관속식물이 확인되었다. 이 중에서 산림청(국립수목원)이 지정한 특산식물은 13분류군이, 환경부지정 고유종은 10분류군이 확인되었다. 희귀 및 멸종위기식물은 IUCN 평가기준에 따라 국립수목원이 제시한 기준에 따라 약관심종(LC) 9분류군, 취약종(VU) 2분류군, 위기종(EN) 1분류군 등 총 12분류군이 확인되었으며, 환경부에서 제시한 적색자료집에 따르면 관심대상(LC) 7분류군, 미평가(NE) 1분류군 등 총 8분류군이 확인되었다. 환경부지정 멸종위기 야생식물 I, II 등급종은 확인되지 않았다. 환경부지정 구계학적 특정식물종은 49분류군이 확인 되었으며, 이 중 정밀생태 조사종인 IV등급에 해당하는 종은 도깨비부채, 만주바람꽃, 미역고사리, 점현호색 4분류군이 확인되었다. 국립수목원지정 기후변화 적응대상식물 300종 중에서 14분류군이 확인되었다. 귀화식물은 19분류군이 확인되어 귀화율 4.3%, 도시화지수 5.9%를 보였다. 광주산맥내 위치하는 산인 축령산, 화악산, 명지산과 비교하여 190분류군이 공통적으로 분포하였으며, 51분류군은 천마산에서만 확인되었다.

금정산의 관속식물상 (Floristic Study on Mt. Kumjung)

  • The floristic characteristics and ecological conservation measures for the plant resources of the Mt. Kumjung(801m), Pusan, Korea were discussed. The field works was done for about twenty times in nine major areas from March, 1995 to Octover, 1996. The total number of vascular plants In this area were summarized as 103 families, 312 genera, 493 species and 564 kinds. The fourteen kinds of artificially planted, ten kinds of Korean endemics and twenty-nine kinds of Korean rare and endangered species in the area were revised and compiled in the list. The flora of the area were categorized into the South-coast put of the Korean Region, where the species such as Platycarya strobilacea, Quercug serrata, Eutya japonica, Camellia japonica, Trachelosperm urn asia talcum vu . in termedium, Quercus acu tissima were roles as major floristic components in the Korean Region and endemized in the south-cosat part of Korea. The natural communities such as Pinus thunbergii Community, Quercus mongolica community, Cupinus laxiflora Community, Wistria floribunda Community and swamp vegetation in Bukmun area as well as upland grassland at the summit of Kodangpong were represented for their floristic values in the area. The thirty kinds of medichinal plant, fifteen kinds of ornamental plants and thirteen kinds of edible plants are faced potentially in endangered from the outrageous collections by the local peoples. The action plan for the conservation management of the area which enables sustainable use by the local peoples must have established in the near feature.

주왕산국립공원 내 주산지 유역의 관속식물상 및 관리방안 (Management Plan and Vascular Plants of the Jusan Reservoir Watershed in Juwangsan National Park)

  • This study is carried out to survey and analyse the vascular plants distributed around the Jusan Reservoir, Juwangsan national park. The results are as follows. The numbers of vascular plants were 377 taxa; 85 families, 253 genera, 332 species, 3 subspecies, 37 varieties and 5 forma. The endangered plants designated by Ministry of Environment were 2 taxa; Berchemia berchemiaefolia (Makino) Koidz. and Iris odaesanensis Y.N.Lee. The rare plants designated by Korea Forest Service were 6 taxa; Eranthis stellata Maxim., Aristolochia contorta Bunge, Aristolochia manshuriensis Kom., Berchemia berchemiaefolia (Makino) Koidz., Scopolia japonica Maxim. and Iris odaesanensis Y.N.Lee. The endemic plants were 4 taxa; Carpinus laxiflora (Siebold & Zucc.) Blume, Pseudostellaria coreana (Nakai) Ohwi, Philadelphus schrenkii Rupr. and Weigela subsessilis (Nakai) L.H.Bailey. The specific plants by floristic region were 38 taxa; Juglans mandshurica Maxim., Salix chaenomeloides Kimura, Hylomecon vernalis Maxim., Lonicera praeflorens Batalin, Erythronium japonicum (Baker) Decne., Potentilla dickinsii Franch. & Sav., Celtis aurantiaca Nakai, Anemone reflexa Steph. & Willd., Cimicifuga heracleifolia Kom. and so forth. The naturalized plants were 20 taxa; Fallopia dumetorum (L.) Holub, Chenopoidum album L., Lepidium apetalum Willd., Veronica persica Poir., Conyza canadensis (L.) Cronquist, Taraxacum officinale Weber and so forth.

멸종위기 식물 덩굴용담의 기내번식에 미치는 생장조절제 효과 (Effect of plant growth regulators on micropropagation of a rare and endangered species, Tsuru-rindo (Tripterospermum japonicum))

  • 멸종위기 식물 덩굴용담의 기내증식법 개발을 위하여 기내증식에 미치는 생장조절제의 효과를 시험하였다. 6가지의 기본배지 시험에서 MS 배지가 줄기증식에 가장 적합한 것으로 나타났다. 싸이토키닌 가운데 BA 처리는 3.0 mg/L농도까지 증식에 유의적인 증식효과를 보였고, 그 이상 농도에서는 증식이 억제되었다. Kinetin과 TDZ는 BA 만큼 증식에 효과적이지 못했다. 발근에 있어 TDZ는 저농도에서도 발근을 현저히 억제하였으나, BA와 kinetin은 저농도 처리 조건에서 자발적인 발근이 이루어 졌다. 한편 지베렐릭산 ($GA_3$)의 처리는 줄기신장의 유의적인 효과가 있었다. 증식중인 줄기는 인공상토에 기외삽목하여 90% 이상 발근이 가능하고 정상적인 생장이 이루어졌다. 대부분의 식물체는 토양이식 다음해에 개화되고 과실을 맺었다.

Effects of Culture Mechanism of Cinnamomum kanehirae and C. camphora on the Expression of Genes Related to Terpene Biosynthesis in Antrodia cinnamomea

  • The rare edible and medicinal fungus Antrodia cinnamomea has a substantial potential for development. In this study, Illumina HiSeq 2000 was used to sequence its transcriptome. The results were assembled de novo, and 66,589 unigenes with an N50 of 4413 bp were obtained. Compared with public databases, 6,061, 3,257, and 2,807 unigenes were annotated to the Non-Redundant, Gene Ontology, and Kyoto Encyclopedia of Genes and Genomes databases, respectively. The genes related to terpene biosynthesis in the mycelia of A. cinnamomea were analyzed, and acetyl CoA synthase (ACS2 and ACS4), hydroxymethylglutaryl CoA reductase (HMGR), farnesyl transferase (FTase), and squalene synthase (SQS) were found to be upregulated in XZJ (twig of C. camphora) and NZJ (twig of C. kanehirae). Moreover, ACS5 and 2,3-oxidized squalene cyclase (OCS) were highly expressed in NZJ, while heme IX farnesyl transferase (IX-FIT) and ACS3 were significantly expressed in XZJ. The differential expression of ACS1, ACS2, HMGR, IX-FIT, SQS, and OCS was confirmed by real-time quantitative reverse transcription PCR. This study provides a new concept for the additional exploration of the molecular regulatory mechanism of terpenoid biosynthesis and data for the biotechnology of terpenoid production.

마이산도립공원의 관속식물 분포 및 관리방안 (Distribution and Management plan of Vascular Plants in Maisan Provincial Park)

  • This study aims to examine and analyze the diverse vascular plants in Maisan Provincial Park, with the objective of establishing fundamental data for the systematic preservation and efficient management of plant resources. A total of 23 field surveys were conducted from February 2022 to March 2023, and a total of 733 taxa were identified, including 120 families, 410 genera, 644 species, 17 subspecies, 52 varieties, 13 formas, 2 cultivars, and 5 hybrids. Among the surveyed taxa, 20 taxa were identified as rare plants designated by the Korea Forest Service. Additionally, one taxa of Cyrtosia septentrionalis, a grade II endangered species designated by the Ministry of Environment, was recognized. There were 26 taxa categorized as Korean endemic plants, and a total of 101 taxa represented floristic regional indicator plants. Moreover, a total of 53 taxa were identified as naturalized plants and 5 taxa were classified as ecosystem-disturbing plants. The urbanization index and naturalized plant index were calculated to be 16.5% and 7.2%, respectively.

덕적도(옹진군) 및 인근 도서지역의 관속식물상 (Flora of Vascular Plants in Deokjeokdo (Ongjin-gun) and Its Adjacent Regions, Korea)

  • This study was carried out to inventory the vascular plant flora in Deokjeokdo and its adjacent regions (Mungapdo, Soyado), Ongjin-gun, South Korea, from April to October 2014. Based on the voucher specimens, vascular plants in this area consisted of 108 families, 362 genera, 578 species, 5 subspecies, 66 varieties and 4 forms, totally 653 taxa. In the flora of this area, 5 taxa of Korean endemic plants were found distributed in the sites, including Hepatica insularis Nakai and Asarum glabrata (C.S.Yook & J.G.Kim) B.U.Oh. Korean rare and endangered plants found in this area were 1 taxa of Critical Endangered Species (CR) and 5 taxa of Vulnerable Species (VU). The floristic regional indicator plants found in this area were 67 taxa comprising 3 taxa of grade V, 2 taxa of grade IV, 12 taxa of grade III, 3 taxa of grade II and 47 taxa of grade I. Naturalized plants consisted of 67 taxa, such as Chenopodium album L. and Erigeron annuus (L.) Pers. were recognized widely distributed in the area.

가야산국립공원의 식물상과 보전방안 (Flora and Conservation Plan of Gayasan National Park)

  • This study is to offer the raw data for conservation and restoration of national park by surveying and analysing the vascular plants in the Gayasan national park. The flora were summarized as 102 families, 328 genera, 469 species, 4 subspecies, 59 varieties and 9 forms. The endangered plants designated by Ministry of Environment were 2 taxa; Pedicularis hallaisanensis Hurus. and Gymnadenia conopsea for. albiflora Y.N.Lee. The rare plants designated by Korea Forest Service were 13 taxa; Crypsinus hastatus (Thunb.) Copel., Ligusticum tachiroei (Franch. & Sav.) M. Hiroe & Constance, Primula modesta var. fauriae (Franch.) Takeda, Lilium distichum Nakai ex Kamibay and so forth. The korean endemic plants were 15 taxa; Betula ermanii Cham., Carpinus laxiflora (Siebold & Zucc.) Blume, Stewartia pseudocamellia Maxim., Galium koreanum (Nakai) Nakai, Heloniopsis koreana Fuse, N.S. Lee & M.N. Tamura and so forth. The specific plants by floristic region were 66 taxa; Betula chinensis Maxim., Spiraea fritschiana Schneid., Vaccinium hirtum var. koreanum (Nakai) Kitam., Cimicifuga heracleifolia Kom., Adoxa moschatellina L. and so forth. The plants with approval for delivering oversea were 22 taxa; Salix hallaisanensis H.L$\acute{e}$v., Sanguisorba argutidens Nakai, Viola albida Palib., Weigela subsessilis (Nakai) L.H.Bailey and so forth. The naturalized plants were 30 taxa; Chenopodium album L., Lepidium apetalum Willd., Trifolium pratense L., Bidens frondosa L., Helianthus tuberosus L. and so forth.
