Diisocyanate Exposure Assessment for Polyurethane Foam Manufacturing Workers (우레탄 폼 제조방식에 따른 작업자의 디이소시아네이트 노출수준 평가)

  • Objectives: Polyurethanes are usually formed by the reactions of liquid diisocyanate components with liquid polyol resin components. Although polyurethanes have advantageous properties, such as their versatility, the manufacturing process generates diisocyanates, which can cause asthma and respiratory irritation in exposed workers. This study compared the differences in diisocyante concentrations between two different (molded foam and slabstock foam) polyurethane foam manufacturing methods. Materials and Methods: Active samples and direct reading samples of diisocyanates (MDI, TDI) were collected in five polyurethane foam manufacturing companies. Results: Workers' exposure concentrations of diisocyanate (GM: 4.078 ppb, range: 1.190~23.770 ppb) in a slabstock foam manufacturing company were much higher than those (GM: 0.011 ppb, range: 0.001~0.055 ppb) in molded foam manufacturing companies. The results of the direct reading samples of diisocyanate indicated that the rapid reaction zone of the slabstock foam processes emitted large amounts of diisocyanates. Conclusions: The exposure levels of diisocyanates for all molded foam workers were much lower than the occupational exposure standard (5 ppb); however, exposure levels for many slabstock foam workers exceeded the standard.

  • Because the electronic book field tends to be marked by rapid changes, keeping current with the events and the literature can be difficult. The electronic book means for reading on PCs or hand-held devices were also limited by the quality of the screen. The electronic book will survive and reading devices become much cheaper and much better in quality. This paper, based on recent electronic book technologies, addresses some of the issues that are central to the acceptance and integration of electronic book into academic library. It also addresses the current electronic book environment, notes the increasing issues, details the management factors in academic library, and examines potential future electronic book directions.

Efficacy of intensive treatment of dysarthria for people with multiple system atrophy (다계통위축증 환자를 대상으로 한 마비말장애 집중 치료의 효과)

  • A mixed dysarthria with combinations of hypokinetic, ataxic, and spastic components is a common clinical feature of multiple system atrophy (MSA). Due to the rapid progress of dysarthria after diagnosis, people with MSA experience difficulty with verbal communication, which eventually affects their quality of life negatively. In this study, SPEAK $OUT!^{(R)}$, an intensive 1:1 treatment of dysarthria for improving functional communicative ability, was provided to twelve people with MSA. To evaluate the efficacy of SPEAK $OUT!^{(R)}$ in people with MSA, aerodynamic, acoustic, and perceptual analyses were conducted. Pre-and post-therapy data included maximum phonation time, vocal intensity, and fundamental frequency during /a/ sustained phonation and passage reading; frequency range between high /a/ and low /a/ phonation; jitter, shimmer, and HNR for vocal quality; speech rate during passage reading; and perceptual evaluation scores for articulation precision and intonation. The participants achieved statistically significant improvement in vocal intensity, pitch range, vocal quality, speech rate, and speech intelligibility. In conclusion, SPEAK $OUT!^{(R)}$ is a feasible treatment for people with MSA to efficaciously improve their speech ability.

An Analysis of Online Readers' Advisory Services Offered by Public Libraries (공공도서관의 온라인 독자상담 서비스에 관한 연구)

  • Due to the rapid advancement of information technology and changes in the reading environment, the readers' advisory services provided by public libraries are facing challenges and opportunities. Recognizing the need for public libraries to respond and propel their services forward, this study analyzed the current status of online readers' advisory services offered by public libraries in Korea and discusses areas that require improvement. To achieve this, the study analyzed the online readers' advisory services in 50 public libraries located in Seoul and six metropolitan cities (Incheon, Daejeon, Daegu, Gwangju, Busan, and Ulsan) across seven categories. These categories encompass various aspects, such as announcing reading programs and new books, identifying and addressing reader needs, developing content by librarians, generating content by readers, providing guidance on external reading information, connecting with professional book review databases and online websites, and utilizing social media services. Through this analysis, the study identified areas for improvement to enhance online readers' advisory services in public libraries and ultimately improve reader satisfaction. It suggested enhancing activities such as librarians' book recommendations and reviews for individual readers, reader participation, and interaction.

Isolation and Characterization of a Novel Broad-host-range Bacteriophage Infecting Salmonella enterica subsp. enterica for Biocontrol and Rapid Detection

  • Bacteriophages have gained substantial attention as biocontrol and biorecognition agents, substituting antibodies. In this study, a Salmonella Enteritidis-specific bacteriophage, KFS-SE1, was isolated, identified, and characterized. This Siphoviridae phage infects S. Enteritidis with high specificity. This phage is highly stable under various pH (5-11), temperature ($4-60^{\circ}C$), and organic solvent conditions. The KFS-SE1 genome consisted of 59,715 bp with 73 predicted open reading frames and 57.14% GC content; it had a complete set of genes required for phage reconstruction. Comparative phylogenetic analysis of KFS-SE1 revealed that it was very similar to the other Salmonella phages in the Siphoviridae family. These characteristics suggest that KFS-SE1 with its high specificity and host lysis activity toward S. Enteritidis may have various potential applications.

A Gene Encoding $\beta$-amylase from Saprolegnia parasitica and Its Expression in Saccharomyces cerevisiae

  • The ${\beta}$-Amylase cDNA fragment from the oomcete Saprolegnia parasitica was cloned by reverse transcription-polymerase chain reaction (RT-PCR) using degenerate oligonucleotide primers derived from conserved ${\beta}$-amylase sequences. The 5'and 3'regions of the $\beta$-amylase gene were amplified using the rapid amplification of cDNA ends (rACE) system. It consisted of an open reading frame of 1,350 bp for a protein of 450 amino acids. Comparison between the genomic and cDNA sequences revealed that the intron was not present in the coding region. The deduced amino acid sequence of the ${\beta}$-amylase gene had a 97% similarity to the ${\beta}$-amylase of Saprolegnia ferax, followed by 41% similarity to those of Arabidopsis thaliana, Hordeum vulgare, and Zea mays. The ${\beta}$-amylase gene was also expressed in Saccharomyces cerevisiae by placing it under the control of the alcohol dehydrogenase gene (ADC1) promoter.

A Novel Rapid Fungal Promoter Analysis System Using the Phosphopantetheinyl Transferase Gene, npgA, in Aspergillus nidulans

  • To develop a convenient promoter analysis system for fungi, a null-pigment mutant (NPG) of Aspergillus nidulans was used with the 4'-phosphopantetheinyl transferase (PPTase) gene, npgA, which restores the normal pigmentation in A. nidulans, as a new reporter gene. The functional organization of serially deleted promoter regions of the A. nidulans trpC gene and the Cryphonectria parasitica crp gene in filamentous fungi was representatively investigated to establish a novel fungal promoter assay system that depends on color complementation of the NPG mutant with the PPTase npgA gene. Several promoter regions of the trpC and crp genes were fused to the npgA gene containing the 1,034-bp open reading frame and the 966-bp 3' downstream region from the TAA, and the constructed fusions were introduced into the NPG mutant in A. nidulans to evaluate color recovery due to the transcriptional activity of the sequence elements. Serial deletion of the trpC and crp promoter regions in this PPTase reporter assay system reaffirmed results in previous reports by using the fungal transformation step without a laborious verification process. This approach suggests a more rapid and convenient system than conventional analyses for fungal gene expression studies.

  • Chemical accidents are the cause of the accident site during the initial responders to quickly and easily see materials and concentration method for the U.S. Environmental Protection Agency(EPA) is widely used in the initial response team direct reading detection equipment used. Ministry of the tubular gas detection equipment to detect direct reading detection equipment used in the event of an accident scene, and shell-and-tube gas detector for rapid detection and identification and precise analysis of causative pollutants before about strategically can identify the quantitative and qualitative useful equipment. However, those who initially respond to the scene of a direct reading detection equipment and a simple lack of understanding of how to use the numbers only because of the way you want to check the accuracy of detection results have been raising questions about the increase. The scene of the accident in order to obtain an accurate detection results used in this paper, the Ministry of Environment of gas detectors detect tubular Kitagawa and Draeger detector tube to check the reactivity of the material on-site detection of early response of those who were to raise the accuracy of the results.

Cloning and Expression of a Farnesyl Diphosphate Synthase in Centella asiatica (L.) Urban

  • A cDNA encoding farnesyl diphosphate synthase (FPS; EC2.5.1.1/EC2.5.1.10) was isolated from Centella asiacita (L.) Urban, using degenerate primers based on two highly conserved domains. A full-length cDNA clone was subsequently isolated by rapid amplification of cDNA ends (RACE) PCR. The sequence of the CaFPS (C. asiatica farnesyl diphosphate synthase) cDNA contains an open reading frame of 1029 nucleotides encoding 343 amino acids with a molecular mass of 39.6 kDa. The deduced CaFPS amino acid sequence exhibits 84, 79, and 72%, identity to the FPSs of Artemisia annua, Arabidopsis thaliana, and Oryza sativa, respectively. Southern blot analysis suggested that the C. asiatica genome contains only one FPS gene. An artificially expressed soluble form of the CaFPS was identified by SDS-PAGE. It had high specific activity and produced farnesyl diphosphate as the major isoprenoid.

Simple and Rapid Detection of Potato leafroll virus by Reverse Transcription Loop-mediated Isothermal Amplification

  • A new reverse transcription loop-mediated isothermal amplification (RT-LAMP) method for the Potato leafroll virus (PLRV) was developed and compared with conventional reverse transcription polymerase chain reaction (RT-PCR) to address its advantages over RTPCR. RT-LAMP primers were designed from the open reading frame 3 (ORF3) sequence of PLRV. The RT-LAMP reactions were conducted without or with a set of loop primers. By real-time monitoring using Turbimeter, the RT-LAMP (with loop primers) detects PLRV in less than 30 min, compared to 120 min of RT-PCR. By adding fluorescent reagent during the reaction, final products of the RT-LAMP were fluorescently visualized under UV light or could be differentiated by naked-eye inspection under normal light. The RT-LAMP was extremely sensitive, about 2000-fold more sensitive than RT-PCR. This study presents great potential of the RT-LAMP for diagnosis and PLRV epidemiology because RT-LAMP method is speedy, sensitive, inexpensive, and convenient.
