Rapid Preparation and Quality Control of $^{99m}Tc$-ECD, $MAG_3$ and MIBI using Microwave Heating and Sep-Pak Cartridges (마이크로웨이브와 Sep-Pak 카트리지를 이용한 $^{99m}Tc$-ECD, $MAG_3$, MIBI의 신속한 제조 및 정도관리)

  • Purpose: We evaluated a rapid preparation procedures for the labeling and quality control of $^{99m}Tc$-ECD, $MAG_3$, and MIBI using microwave heating and Sep-Pak cartridges. Materials and Methods: $^{99m}Tc$ labeling of ECD, $MAG_3$, and MIBI kit preparation was performed according to the package inserts with microwave heating modification. Heating time was 10-15 sec, and heating was performed with 3 mm plastic bottle with screw cap to prevent radiation contamination. Labeling efficiency was obtained with $C_{18}$ or Alumina N Sep-Pak cartridges. Results: The radiochemical purity of $93{\sim}96%$ for $^{99m}Tc$-ECD and $95{\sim}99%$ for $^{99m}Tc$-MIBI was obtained using Alumina N Sep-Pak cartridge. The optimum irradiation time of microwave method for 3 ml $^{99m}Tc$-labeled radiopharmaceutical solution was 10 sec for $^{99m}Tc$-ECD and $^{99m}Tc$-MIBI, and 15 sec for $^{99m}Tc-MAG_3$. The results of quality control data with Sep-Pak cartridges were well correlated with TLC method. The total preparation time of these radiopharmcaeuticals was $5{\sim}6min$ including quality control procedure. Conclusion: This study demonstrates that radiopharmaceuticals preparation by microwave heating and quality control by Sep-Pak cartridges can be efficiently utilized as an alternative to the recommended method by manufacturer's manual.

Protoplast Preparation and Regeneration from Young Hyphae of the Citrinin Producing Fungus Monascus ruber

  • Optimized conditions for protoplast preparation and regeneration from young hyphae of Monascus ruber were established. Heat shock treatment of spores gave rapid and synchronized germination. Spores collected from cultures grown for 7-8 days at $30^{\circ}C$ were germinated until over 70% germ tubes reached to 3-5 spore length. Enzymatic digestion of young hyphae was optimal with 50 mg/mL Glucanex in 0.1 M sodium citrate buffer containing 0.8 M mannitol as an osmotic stabilizer. Regeneration rate was around 10% when 0.8 M sorbitol was used as an osmotic stabilizer in regeneration medium. These conditions will be applied in genetic study of M. ruber that produces citrinin at high level and thus is good model strain for molecular genetic dissection of citrinin biosynthesis.

Preparation of Nano-sized Zirconia Powders by the Impregnation Method (함침법에 의한 지르코니아 나노 분말의 합성)

  • The nano-sized zirconia powders were synthesized in an impregnation method using pulp and $ZrOCl_2{\cdot}8H_2O$ as an initial material. The synthesized powders were characterized by XRD and FE-SEM. The particle size of the powder was controlled by preparation conditions, such as drying temperature and time. As a result of the various drying and calcination conditions, 30~50 nm sized homogeneous zirconia particles were obtained at $800^{\circ}C$ for 1 h. Crystallization and the rapid growth of particles were accelerated with increasing calcination temperature and time. Tetragonal phase generated below $800^{\circ}C$ were transferred to monoclinic phase with increasing calcination temperature and time. Moreover, above $800^{\circ}C$, heat treatment time had very large influence on the particle growth, and the change of drying condition also had large influence on the growth of a crystal.

  • To obtain accurate and repeatable analyses using NIR technology it is important to select an NIR instrument and / or its sample presentation attachments which allow the operator to minimize sampling errors without compromising the benefits of NIR analysis -namely rapid, low cost, minimal sample preparation, minimal structural facilities, minimal hazards. For each sample type and consistency there may be different optimal combinations of instrument, sample presentation attachment, and sample preparation. This paper will consider options available to NIR users in the area of plant and soil analysis and evaluate the potential benefits and disadvantages of crop nutrient diagnoses using laboratory based and airborne imaging techniques.

  • A novel powder processing technique for the preparation of copper activated zinc sulfide (ZnS:Cu,Al) phosphor by combustion process has been proposed. Exothermic reaction between dissolved copper nitrate and carbohydrazide give small-sized particles in presence of alkali metal halides at lower temperature than the traditional method of preparation. This new route takes less than five minutes and requires much less energy. The optical and luminescence characteristics of ZnS:Cu,Al phosphor thus prepared were found to be enhanced significantly. Carbohydrazide acted as fuel at $500^{\circ}C$ with rapid heating and then the phosphors obtained were heated at $900^{\circ}C$ in an inert atmosphere for 3hrs to get better luminescent properties.

  • A simple and rapid method for preparation of standard phosphatidylcholine hydroperoxide (PCOOH) in chemiluminescence-HPLC assay (CL-HPLC) was developed using the rose bengal dependent spectrofluorometric oxidation. The concentration of obtained PCOOH was determined by FOX (ferrous oxidation-xylenol orange) assay and then subjected to a CL-HPLC system.

  • This study presents a rapid and simple ${\alpha}$ source preparation method for a radioactive waste sample. The recovery of $^{239}Pu$, $^{232}U$ and $^{243}Am$ using a micro-coprecipitation method was over 95%. The ${\alpha}$-peak resolution of Pu and Am isotopes through the micro-coprecipitation method is enough to discriminate the Pu and Am isotopes from other Pu and Am isotopes. The determination of the Pu and Am isotopes using the micro-coprecipitation method was applied to the radioactive waste sample, so that the activity concentrations of the Pu and Am isotopes using the micro-coprecipitation method in the radioactive waste sample were similar to those using the electrodeposition method.

Application on Microwave Energy in the Preparation of Fish Samples for Electron Microscopic Observation

  • Chemotherapy can not be applied for the control of fish viral diseases because viruses depend on host machinery for their replication. Although new control strategies including vaccination are under development, avoidance of virus introduction by rapid and correct diagnosis is the best way of fish viral disease control. Although observation of virus particles with an electron microscope is an easy method for virus detection, it take a few days for the sample preparation. In order to shorten the sample preparation time, microwave radiation was applied in the procedure. With this method, 15 seconds was enough for fixation of virus infected fish samples or cultured cells inoculated with infectious hematopoietic necrosis virus, which takes 2-4 hours with routine methods. Also four minutes was enough for polymerization of embedding resin which takes 24-48 hours with routine methods. Samples prepared with microwave were good enough for direct electron microscopic observation and immunogold labeling assay.

  • To establish tile standard condition of uniformed Kimchi product, we introduced the concept of starter and studied the preparation of rapid-fermented Kimchi. Of the strains isolated from Kimchi, Kakdugi and infant's feces, M7 strain grew effectively on aseptic Chinese cabbage juice and on salted Chinese cabbage, the growth of M7 was decreased severely. Inoculated with M7 in salted Chinese cabbage, appropriate range of pH and lactic acid content were reached at 8-13 hrs and 12 hrs after addition of spices, respectively. The result of sensory evaluation was not significant at 5% level.

  • STEP is a neutral model for exchanging CAD modelbetween different CAD systms. The paper presents a laser path contour generation for rapid prototyping originated form STEP format files without using any tools nor libraries for STEP data manipulation. Polygons are generated from every point on the layer of given thickness, which are the intersections of layer plane and edges stored in entity tables. Curves are approximated with polygons whitin segment tolerance limit in order to cover sculptured surfaces. With the advantage of less data loss in direct slicing over STL formats, the system developed in this work shows a good potential to provide data share with various CAD systems including RP data preparation.
