Studies on the Effects of Co-culture of Cumulus Cell, Oviduct Epithelial Cell and Hormones and Freezing on !fl Vitro Developmental Rates of Bovine Embryos (소 수정란의 난구세포, 난관 상피세포, 호르몬과의 공배양 및 동결이 체외발생에 미치는 영향에 관한 연구)

  • The studies were carried out to investigate the effects of co-culture with cumulus cells and oviduct epithelial cells on the in vitro fertilization and cleavage rate of bovine follicular cocytes and to determine the optimum thawing temperature and equilibration time on in vitro developmental rate of frozen bovine embryos. The ovaries were obtained from slaughtered Korean native cows. The follicular oocytes were cultured in TGM-199 medium containing 10 IU /ml의 PM SG, 10 IU /ml의 hCG, ip g/ml의 $\beta$-estradiol and 10% FCS for 24~48 hrs in incubator with 5% $CO_2$ in air at 38.5$^{\circ}C$. The bovine embryos following dehydration by cryoprotective agents and a various concentration of sucrose were directly plunged into liquld nitrogen and thawed in 3$0^{\circ}C$ water. Survival rate was defined as developmental rate on in vitro culture or FDA-test. The results are sunanarized as followes :1. The in vitro fertilization and in vitro developmental rates of bovine oocytes co-cultured with cumulus cells in TCM499 medium were 75.0~76.8% and 17.3~27.6%, respect-ively. And in-vitro fertilization rates of cumulus-enclosed oocytes(55.4%)were significantly(p<0.05) higher than cumulus-denuded oocytes (23.1%). 2. The in vitro fertilization and in vitro developmental rates of bovine oocytes co-cultured with l$\times$ l04cells /ml, 1 x l06cells /ml, lx l08cells /ml and 1 x l015cells /ml oviduct epithelial cells in TCM-199 medium were 74.5~77.8% and 15.7~21.20 respectively.3. The in-vitro fertilization and in vitro developmental rates of bovine oocytes cocultured in '1CM-199 media containing PMSG, hCG, PMSG+hCG. PMSG+$\beta$-estradiol, hCG+$\beta$-estradiol 0 to 40 hrs after insemination were 74.0~77.4% and l8.9~23.l%, re-spectiv ely.4.The survival rates of bovine embryos thawed after rapid freezing in the freezing medium containing a various concentration of sucrose added 1.5M and 2.OM glycerol,DMSO and propanediol were 23.5~31.4% and 20.6~34.l%, respectively. 5. The temperature thawed at 3$0^{\circ}C$ after rapid freezing of bovine embryos resulted in a significantly higher embryos survival rate than did at 2$0^{\circ}C$ and 35$^{\circ}C$.6. The equilibration time on the survival rates of bovine embryos was attained after short period of time(2.5~5 min.) in the freezing medium higher than long period of time (10~20min.).   • This study was conducted to investigate the effect of freezing methods, slow freezing at $-20^{\circ}C$ and rapid freezing at $-70^{\circ}C$, on physical properties of Chinese cabbage in frozen Kimchi during storage at $-20^{\circ}C$. Elasticity of midrib of Chinese cabbage in frozen Kimchi was decreased until 15 days and did not changed thereafter during storage at $-20^{\circ}C$. Hardness of that was not changed during storage. Those results of elasticity and hardness of slow frozen sample are similar to rapid frozen sample. By the morphological observation through transmission electron microscope, more of cellular structure of Chinese cabbage in slow frozen was destructed than that of rapid frozen sample. Drip loss was more in slow frozen sample than that in rapid one.

Quality Characteristics of Wheat Flour Breads with the Doughs Frozen at the Different Freezing and Storage Conditions (반죽의 냉동과 저장 조건에 따른 빵의 품질 특성)

  • The dough was frozen either before or after fermentation at the five different freezing and storage conditions. Although fermentation before freezing was effective for rapid freezing, it reduced bread volume of the dough frozen at both air freezer and liquid immersion freezer. Freezing at the air freezer set to $-70^{\circ}C$ took more time for freezing and resulted in lower bread volume than freezing at the immersion freezer set to $-20^{\circ}C$. Therefore, the freezing in the liquid immersion freezer was more effective to reduce the freezing time and increase the bread volume. At the liquid immersion freezer, the higher temperature was more effective than lower temperature. The doughs frozen in a liquid immersion freezer set to $-10^{\circ}C$ and fermented after de-frosting produced higher bread volume than control unfrozen dough. And also there was no significant difference in bread volume between the control unfrozen dough and the dough frozen in a liquid immersion freezer set to $-10^{\circ}C$, fermented before freezing and re-fermented after defrosting. The longer proof time and greater loaf volume obtained for the dough frozen and stored at the air freezer set to $-70^{\circ}C$. Therefore the optimum process for freezing the dough was freezing immediately after mixing, storing at $-10^{\circ}C$ in a liquid immersion freezer and fermented after defrosting.

Studies on Rapid Freezing and Thawing of Porcine Embryos III. Factors affecting the survival rate of porcine embryos cryopreserved and diluted by one-step straw method (돼지 수정란의 급속 동결 융해법에 관한 연구 - 돼지 동결 수정란에 대한 1단계 Straw법이 배의 생존성에 미치는 영향)

  • This study were carried out to investigate the effective concentration of cryoprotective agents and sucrose by one-step straw method, and to determine the optimum thawing temperature and equilibration time of frozen porcine embryos. The porcine embryos foflowing dehydration by cryoprotective agents and a various concentration of sucrose were directly plunged into liquid nitrogen and thawed in 3$0^{\circ}C$ water. Survival rate was defined by FDA test. The results are sunnnarized as follows : 1. The survival rates of porcine embryos after ultrarapid frozen4hawing in the freezing medium with a various concentration of glycerol, DMSO and propanediol added 0.25M sucrose were higher survival rate than those of sucrose concentration of 0.50M. 2. The survival rates of porcine embryos after ultrarapid ftozen4hawing in the freezing medium added 0.25M and 0.SOM sucrose were higher survival rate than those of sucrose concentration of 0.75M and 1.00M. 3. The temperature thawed at 2$0^{\circ}C$ and 3$0^{\circ}C$ resulted in a significantly higher embryos survival rate after 72 hrs in culture than did at 35$^{\circ}C$. 4. The equilibration time on the survival rate of porcine embryos was attained after short period of time(2.5~5 min.) in the freezing medium higher than long period of time(10~20 min.).

Developmental Capacity of Bovine Follicular Oocytes after Ultra-Rapid Freezing by electron Microscope Grid II.Cryopreservation of In Vitro Matured Bovine Oocytes (Electron Microscopic Grid를 이용한 초급속 동결이 소 난포란의 발달능에 미치는 영향. II. 체외 성숙된 소 미수정란의 동결에 관한 연구)

  • This study was carried out to confirm whether the developmental capacity of bovine mature oocytes frozen ultra-rapidly using electron microscopic(EM) grids and EFS30 can be obtained, and whether the cryoprotectants and the freezing method used in this study effect detrimentally to the bovine oocytes by indirect immunocytochemistry. As freezing solution, we used EFS30 which consisted of 30% ethylene glycol, 0.5 M sucrose, 18% ficoll and 10% FBS added in D-PBS. The results obtained in this experiment were summarized as follows: When the effects of cryoprotectant and freezing procedure on the microtuble, micrfilament and chromatin morphology of oocytes were evaluated using indirect immunocytochemistry, the results of freezing as well as exposure group were not different with that of the control oocytes. When the fertilization abnormality after ultrarapid freezing of bovine mature oocytes was examined by Hoechst staining, the rates of total penetration(96.7, 9.0%), normal two pronuclei formation(74.6, 68.9%) and mean number of sperm / oocyte(1.50, 1.44) were not different between control and freezing group. In addition, when the developmental capacity of frozen-thawed of oocytes(85.5%) was survived, 74.5% of them were cleaved and 31.4% of cleaved embryos were developed to blastocyst. These data were similar to those of the control(76.0%, 34.6%) and exposure(74.5%, 33.0%) except survival rates. Also, when the total cell number of blastocysts produced from the each treatment at day after IVF was examined by hoechst staining, there were not different among groups. There results demonstrate that developmental capacity of frozen-thawed bovine mature oocytes can be successfully obtained by ultra-rapid freezing method using EM grid and EFS30 solution.

  • This study was conducted to assess the durability of Chloride-inhibiting Low-Heat Cement while being subjected to freezing-thawing during winter seasons. Although durability varies slightly depending on the conditions of the jobsite, frost damage to concrete resulting from repeated freezing and thawing over the course of seasonal changes is the leading cause behind lowered concrete durability. in addition, concrete that has been subjected to freezing and thawing during the winter season develops a significant amount of expansive force at the core and begins to exhibit signs of damage, such as cracking, peeling, and detachment from the aggregate. Therefore, this study fabricated test specimens using a Chloride-inhibiting Low-Heat Cement(CLC) and the widely used blast furnace slag cement(BFS) and Ordinary Portland Cement(OPC) with water-to-cement ratios of 35%, 40% and 45%, respectively, to assess the durability index of the CLC as per resistance to freezing-thawing. The specimens were then tested using the KS F 2456 method (Testing method for resistance of concrete to rapid freezing and thawing) to measure the dynamic modulus of elasticity. The dynamic modulus of elasticity measurements were then used to derive the durability indices. By comparing the durability indices, it was confirmed that CLC, BFS, and OPC all had superior durability.

Evaluation of Cold Tolerance of Blueberry (Vaccinium corymbosum L.) and Diagnosis of Freezing Injury Using Timber Moisture Meter (블루베리의 내한성 평가 및 목재수분계측기를 이용한 동해피해 진단)

  • This study was conducted to evaluate on the freezing tolerance of introduced blueberry cultivars in Korea and to investigate availability of portable timber moisture meter for simple and rapid diagnosis of blueberry-shoot damage by freezing during wintering. Frost tolerance of blueberry cultivars showed big difference that rates of blueberry-shoot death were widely distributed from about 0% to 100% after wintering. Optical density in TTC reduction of blueberry twig treated low temperature was low in order of $-40^{\circ}C$ < $-21^{\circ}C$ < $4^{\circ}C$. Hardiness evaluation of visible injury in the cross-sectional surface color did not agree with that of rates of blueberry-shoot death during wintering. Lowest water content of blueberry stem measured by timber moisture tester during wintering was about 15%. During wintering, water contents of blueberry stems were higher at lower part of tree, but were low at end part of stems, and then when the blueberry grew again for spring, the water content gradually increased to 20~40%. Water content of blueberry stem with freezing injury during wintering decreased to under 5% by desiccation. Therefore it is assummed that the moisture content of blueberry stem injured by freezing during wintering was about under 14%, and it is expected that portable timber moisture meter could be available for rapid diagnosis of blueberry freezing injury in field.

Changes of Internal Pressure during Freezing, Frozen Storage and Thawing of Meats (식육의 동결, 냉동냉장 및 해동시 내부압력 변화)

  • The destruction of tissues by volume increase at food freezing is accepted as one of the factor responsible for quality damage. For this reason, the internal pressure developed in meats were investigated with a pressure transducer during freezing, frozen storage and thawing. Increasement of 6.33% for volume and $942.17\;kg/cm^2$ for density at $-20^{\circ}C$ for beef were shown. In quick and slow freezing of beef, internal pressure reached to highest point after reached to the lowest point at initial of the zone of ice crystal formation. The internal pressure was approximately $8{\sim}10\;psig$ and pressure difference was about 1 psig, which was bigger in immersion freezing than that of still-air freezing. During frozen storage of pork, internal pressure of $1.84{\sim}2.32\;psig$ occurred repeatedly as a function of sample weight at material temperature difference of ${\pm}1^{\circ}C$. The internal pressure during thawing of pork was decreased slowly after rapid increase to the maximum for less than 5min at the beginning of thawing. Internal pressure value at thawing was higher than that at freezing in most cases. Internal pressure of beef with thermal equalized freezing was about $1{\sim}4\;psig$, which was lower than that of non-thermal equalized freezing. Also, freezing time was shortened to $10{\sim}20%$.

Effect of Freezing, Thawing and Blanching on the Pigment of Purple Sweet Potato (동결, 해동 및 Blanching이 자색고구마 색소에 미치는 영향)

  • The effect of freezing, thawing and blanching on the change of extractable pigment content of purple sweet potato (PSP) was investigated. Freezing at $-5^{\circ}C$ was more effective than freezing at $-20^{\circ}C\;or\;-40^{\circ}C$, and rapid thawing methods such as microwave heating or hat air blast heating were effective than slow thawing methods such as thawing at $4^{\circ}C\;or\;20^{\circ}C$. Inactivation of enzymes, which cause pigment destruction during thawing, by blanching before freezing was necessary to obtain the highest possible amount of extractable pigment from PSP. Microwave blanching for $3{\sim}4$ min or hot air blanching for $10{\sim}15$ min were effective in extracting pigment from PSP.

Experimental Study on Performance of MgO-based Patching Materials for Rapid Repair of Concrete Pavement (콘크리트 포장의 급속 보수를 위한 산화마그네슘계열 단면복구재의 성능에 대한 실험적 연구)

  • PURPOSES : This study aims to develop a repair material that can enhance pavement performance, inducing rapid traffic opening through early strength development and fast setting time by utilizing MgO-based patching materials for repairing road pavements. METHODS : To consider the applicability of MgO-based patching materials for repairing domestic road pavements, first, strength development and setting time of the materials were evaluated, based on MgO to $KH_2PO_4$ ratio, water to binder ratio, and addition ratio of retarder (Borax), by which the optimal mixture ratio of the developed material was obtained. To validate the performance of the developed material as a repair material, the strength(compressive strength and bonding strength) and durability (freezing, thawing, and chloride ion penetration resistance) was checked through testing, and its applicability was evaluated. RESULTS : The results showed that when an MgO-based patching material was used, the condensation time was reduced by 80%, and the compressive strength was enhanced by approximately 300%, as compared to existing cement-based repair materials. In addition, it was observed that the strength (compressive strength and bonding strength) and durability (freezing and thawing, and chloride ion penetration resistance) showed an excellent performance that satisfied the regulations. CONCLUSIONS : The results imply that an emergent repair/restoration could be covered by a rapid-hardening cement to meet the traffic limitation (i.e. the traffic restriction is only several hours for repair treatment). Furthermore, MgO-based patching materials can improve bonding strength and durability compared to existing repair materials.
