Expression of Sex-Related Genes in the Fetus of Mouse: 2-Bromopropane and Sex Differentiation (생쥐 태자의 성 관련 유전자 발현: 2-Bromopropane과 성 분화)

  • The recent reports that endocrine disruptors(EDs) bring about abnormalities in reproductive organs and functions of invertebrates suggest that mammals be affected by the EDs. The present study examined the influence of 2-bromopropane(2-BP) by looking at the sexes of litters in mouse. The expression of sex-related genes during sex differentiation was also investigated in the fetus of mouse. The male and female mice were infused with 2-BP for 3 weeks before mating. The litters were sexed at the weaning time from the 4 different groups. The sex-related genes were identified by RT-PCR from the fetuses at gestation 10 days. The sequences of the genes were analysed by comparing to those of other animals. The mean numbers of litters survived by the weaning time were slightly reduced in the only group of both female and male mice treated with 2-BP. The female litters were greater than male litters in the only group of female treated with 2-BP. The other groups showed male litters greater than female litters. The sex-related genes, SRY, DAX1, SF1 , and AMH genes were identified and sequenced, showing 416, 466, 326, 389 base pairs, respectively. All of the genes had the homology of 89~90% with rat and 81~92% with human within the range of bases identified. They were expressed at the time of sex determination. Therefore, it appears that 2-BP somewhat affects the reproductive activity of adult mouse. Influence of 2-BP on the reproductive function is expected to be studied through the expression of the sex-related genes.

  • Molds are eucaryotic, multicellular, multinucleate, filamentous organisms that reproduce by forming asexual and sexual spores. The spores are readily spread through the air and because they are very light-weight and tend to behave like dust particles, they are easily disseminated on air currents. Molds therefore are ubiquitous organisms that are found everywhere, throughout the environment. The natural habitat of most molds is the soil where they grow on and break down decaying vegetable matter. Thus, where there is decaying organic matter in an area, there are often high numbers of mold spores in the atmosphere of the environment. Molds are common contaminants of plant materials, including grains and seeds, and therefore readily contaminate human foods and animal feeds. Molds can tolerate relatively harsh environments and adapt to more severe stresses than most microorganisms. They require less available moisture for growth than bacteria and yeasts and can grow on substrates containing concentrations of sugar or salt that bacteria can not tolerate. Most molds are highly aerobic, requiring oxygen for growth. Molds grow over a wide temperature range, but few can grow at extremely high temperatures. Molds have simple nutritional requirements, requiring primarily a source of carbon and simple organic nitrogen. Because of this, molds can grow on many foods and feed materials and cause spoilage and deterioration. Some molds ran produce toxic substances known as mycotoxins, which are toxic to humans and animals. Mold growth in foods can be controlled by manipulating factors such as atmosphere, moisture content, water activity, relative humidity and temperature. The presence of other microorganisms tends to restrict mold growth, especially if conditions are favorable for growth of bacteria or yeasts. Certain chemicals in the substrate may also inhibit mold growth. These may be naturally occurring or added for the purpose of preservation. Only a relatively few of the approximately 100,000 different species of fungi are involved in the deterioration of food and agricultural commodities and production of mycotoxins. Deteriorative and toxic mold species are found primarily in the genera Aspergillus, Penicillium, Fusarium, Alternaria, Trichothecium, Trichoderma, Rhizopus, Mucor and Cladosporium. While many molds can be observed as surface growth on foods, they also often occur as internal contaminants of nuts, seeds and grains. Mold deterioration of foods and agricultural commodities is a serious problem world-wide. However, molds also pose hazards to human and animal health in the form of mycotoxins, as infectious agents and as respiratory irritants and allergens. Thus, molds are involved in a number of human and animal diseases with serious implication for health.

Experimental Study of Friction Factors for Laminar, Transition, and Turbulent Flow Regimes in Helical Coil Tubes (헬리컬 코일 튜브에서의 층류, 천이, 난류 영역의 마찰계수에 대한 실험적 연구)

  • The friction factors according to the flow regimes in helical coil tubes depend on the coil diameter, the tube diameter, and the coil pitch. In previous studies, correlations for the laminar flow regime in helical coil tubes have been proposed. However, studies on the transition flow regime and the turbulent flow regime are insufficient and further researches are necessary. In this study, characteristics of the friction factors for the laminar, transition and turbulent flow regimes in helical coil tubes were experimentally investigated. The helical coil tubes used in the experiments were made of copper. The curvature ratios of the helical coil tubes, which means the ratio of helical coil diameter to inner diameter of the helical coil tube are 24.5 and 90.9. Experiments were carried out in the range of $529{\leq}Re{\leq}39,406$ to observe the flows from the laminar to the turbulent regime. The friction factors were obtained by measuring the differential pressures according to the flow rates in the helical coil tubes while varying the curvature ratios of the helical coil tubes. Experimental data show that the friction factors for the helical coil tube with 24.5 in the curvature ratio of the helical coil tube were larger than those in the straight tube in all flow regimes. As the curvature ratio of the helical coil tube increases, the friction factor in turbulent flow regime tends to be equal to that of the straight tube. In addition, it was confirmed that the transition flow regimes in the helical coil tubes were much wider than those in the straight tube, also the critical Reynolds numbers were larger than those in the straight tube. The results obtained in this experimental study can be used as basic data for studies on the water hammer phenomenon in helical coil tubes.

Changes of the Physico-Chemical and Microbiological Properties during Composting for Composting of Sewage Sludge (하수슬러지의 퇴비화과정 중 이화학성 및 미생물상 변동)

  • To study the possibility of agricultural utilization of sewage sludge, the changes of the substances physico-chemical and microbiological properties composting periods such as inorganic matter, the form of organic matter and nitrogen and the kinds and the population number of microorganisms were investigated. The results were summarized as follows ; Temperature and $CO_2$ generation, they were the highest in the second day of composting periods and then were gradually fallen. pH value was not changed in first day during composting periods, but in second day was rapidly increased and then it was constant of the range of 8.4∼8.6. The contents of $P_2O,\;K_2O$, CaO and Fe were a little increased during composting periods, while that of ${SO_4}^{2-}$and Mn were big increased with 253${\sim}$331% and 191${\sim}$208% in late composting periods in comparing with early composting periods, respectively. The contents of ether extracted materials, water soluble polysaccharides, hemicellulose and cellulose were decreased but that of resins and lignin were not changed during composting periods. The contents of total and organic nitrogen during composting periods were decreased with 15${\sim}$20% and 22${\sim}$35%, respectively, while that of inorganic nitrogen was decreased with 75${\sim}$116%. The population numbers of microorganism during composting periods was much too changed according to the kinds of microorganism and composting periods.

A Study on Wear Characteristics of Cutting Tools in a Titanium Roughing Cut Machining (티타늄 황삭가공에 있어서 절삭공구의 마모 특성에 관한 연구)

  • The application of titanium has been gradually rising because the utilizing ranges for low weight and high strength are rapidly increased by the need for improving the fuel economy in production industries such as the aviation and automotive in recent. The purpose in this study is to investigate the appropriate cutting conditions on the life of flat and round end mills by measuring the maximum cutting temperature relative to the machining time, and calculating the wear rates of cutting tool with the spindle speed and feed rate of vertical machining center as a parameter in the titanium roughing cut machining which is widely used in critical parts of aircraft, cars, etc. When the wetted roughing cut machining of titanium with a soluble cutting oil is conducted by the flat and round end mills, the maximum cutting temperatures for a variety of spindle speed and feed rate are measured at ten-minute intervals during 60 minutes by an infrared thermometer, and the wear rates of cutting tool are calculated by the weight ratios based on tool wear before and after the experiment. It is found that the maximum cutting temperature and the wear rates of cutting tool are raised as the cutting amount per tool edge is increased with the rise of feed rate, in this experimental range, and as the frictional area due to the rise of contacting friction numbers between tool and specimen is increased with the rises of cutting time and spindle speed. In addition, the increasing rate of maximum cutting temperature in the flat and round end mills are the highest for the cutting time from 50 to 60 minutes, and the wear rate of cutting tool in the flat end mill is 1.14 to 1.55 times higher than that in the round end mill for all experimental conditions.

Automatic Traffic Data Collection Using Simulated Satellite Imagery (인공위성영상을 이용한 교통량측량 자동화)

  • The fact that the demands on traffic data collection are imposed by economic and safety considerations raisese the question of the potential for complementing existing traffic data collection programs with satellite data. Evaluating and monitoring traffic characteristics is becoming increasingly important as worsening congestion, declining economic situations, and increasing environmental sensitivies are forcing the government and municipalities to make better use of existing roadway capacities. The present system of using automatic counters at selected points on highways works well from a temporal point of view (i.e., during a specific period of time at one location). However, the present system does not cover the spatial aspects of the entire road system (i.e., for every location during specific periods of time); the counters are employed only at points and only on selected highways. This lack of spatial coverage is due, in part, to the cost of the automatic counters systems (fixed procurement and maintenance costs) and of the personal required to deploy them. The current procedure is believed to work fairly well in the aggregate mode, at the macro level. However, at micro level, the numbers are more suspect. In addition, the statistics only work when assuming a certain homogenity among characteristics of highways in the same class, an assumption that is impossible to test whn little or no data is gathered on many of the highways for a given class. In this paper, a remote sensing system as complement of the existing system is considered and implemented. Since satellite imagery with high resolution is not available, digitized panchromatic imagery acquired from an aircraft platform is utilized for initial test of the feasibility and performance capability of remote sensing data. Different levels of imagery resolutions are evaluated in an attempt to determine what vehicle types could be classified and counted against a background of pavement types, which might be expected in panchromatic satellite imagery. The results of a systematic study with three different levels of resolutions (1m, 2m and 4m) show that the panchromat ic reflectances of vehicles and pavements would be distributed so similarly that it would be difficult to classify systematically and analytically remotely sensing vehicles on pavement within panchromatic range. Anaysis of the aerial photographs show that the shadows of the vehicles could be a cue for vehicle detection.

Optimal Condition of Operation Parameter for Livestock Carcass Leachate using Fenton Oxidation Process (가축 사체 매몰지 침출수 처리를 위한 Fenton 산화공정의 최적조건)

  • Outbreak of animal infectious diseases such as foot-and-mouth disease, avian influenza are becoming prevalent worldwide. For prevent the further infection, tremendous numbers of the infected or culled stocks are buried around farm. This burial method can generate a wide range of detrimental components such as leachate, nutrient, salt, and pathogenic bacteria, consequently. In this study, for the stabilization of livestock carcasses leachate, advanced oxidation processes utilizing the Fenton reaction was investigated in lab-scale experiments for the treatment for $COD_{Cr}$ of livestock carcass leachate. $COD_{Cr}$ reduction by the Fenton oxidation was investigated response surface methodology using the Box-Begnken methods were applied to the experimental results. A central composite design was used to investigate the effects of the independent variables of pH ($x_1$), dosage of $FeCl_2{\cdot}4H_2O$ ($x_2$) and dosage of $H_2O_2$ ($x_3$) on the dependent variables $COD_{Cr}$ concentration ($y_1$). A 1 M NaOH and $H_2SO_4$ was using for pH control, $FeCl_2{\cdot}4H_2O$ was used as iron catalyst and NaOH was used for Fenton reaction. The optimal conditions for Fenton oxidation process were determined: pH, dosage of $FeCl_2{\cdot}4H_2O$ and dosage of $H_2O_2$ were 3, 0.6 g (0.0151 M) and 7 mL(0.259 M), respectively. Statistical results showed the order of significance of the independent variables to be pH > initial concentration of ferrous ion > initial concentration of hydrogen peroxide.

Fermentation and Quality Characteristics of Kwamaegi added Kimchi (과메기 첨가 김치의 숙성과 품질특성)

  • The effects of kwamaegi (0, 6, 12 and 18%, all w/w) on the quality characteristics of Kimchi during fermentation at $10^{\circ}C$ were investigated. The mixtures are denoted KK. The late of decrease in pH during KK fermentation was lower than that of control. The number of total microbes (T) in Kimchi controls fermented for 14 days did not differ, and were in the range of $8.64{\sim}8.68\;log\;CFU/g$, whereas the numbers of lactic acid bacteria (L) in KK were higher than in controls. The L/T(%) was 78.41 in control, 85.76 in 6% KK and $93.68{\sim}94.25%$ in the 16% or 18% KK samples. The hardness of tissue ($2.98{\sim}3.66{\times}10^7\;dyne/cm^2$) in KK fermented for 14 days was higher than that of control ($2.67{\times}10^7\;dyne/cm^2$). The content of alcohol-insoluble substances in KK juice (1.281.08 g/100 g) was lower than that of control (1.45 g/100 g). In sensory evaluation, the appearance and texture of KK were better than those of control. Although some fishy flavor was noted in KK, this did not affect acceptability. Sourness was less, whereas ripened taste and overall acceptability were higher in KK (especially 12% KK) compared to control.

The Necessity of Resetting the Filter Criteria for the Minimization of Dose Creep in Digital Imaging Systems (디지털 영상 시스템에서 선량 크리프 최소화를 위한 부가 필터 두께 권고 기준의 재설정에 대한 연구)

  • Recently, Following the recent development of flat panel detector with wide dynamic ranges, increasing numbers of healthcare providers have begun to use digital radiography. As a result, filter thickness standards should be reestablished, as current clinical practice requires the use of thicknesses recommended by the National Council on Radiation Protection and Measurements, which are based on information, acquired using conventional analog systems. Here we investigated the possibility of minimizing dose creep and optimizing patient dose using Al filters in digital radiography. The use of thicker Al filters resulted in a maximum 19.3% reduction in the entrance skin exposure dose when medical images with similar sharpness values were compared. However, resolution, which is a critical factor in imaging, had a significant change of 1.01 lp/mm. This change in resolution is thought to be due to the increased amount of scattered rays generated from the object due to the X-ray beam hardening effect. The increase in the number of scattered rays was verified using the scattering degradation factor. However, the FPD, which has recently been developed and is widely used in various areas, has greater response to radiation than analog devices and has a wide dynamic range. Therefore, the FPD is expected to maintain an appropriate level of resolution corresponding to the increase in the scattered-ray content ratio, which depends on filter thickness. Use of the FPD is also expected to minimize dose creep by reducing the exposure dose.

Establishment of National Quality Control System for Analytical Laboratory of Pesticide Products by Proficiency Testing (농약 이화학시험 분석기관의 숙련도시험을 통한 정도관리체계 확립 연구)

  • Performance of proficiency testing and the validation of analytical method was included a scheme of quality assurance in analytical chemistry laboratory to monitor a laboratory's performance abilities and produce consistently reliable data. This study was assessed the applicability of proficiency testing scheme proposed for analytical laboratories of pesticide product in domestic. The validation of analytical methods, stability and homogeneity for formulated pesticide products (emulsifiable concentrate) of emamectin benzoate and lufenuron was confirmed for the proficiency testing. The z-score of 33 participation laboratories for emamectin benzoate were that the numbers of outlier were 2 laboratories (6.0%), z-score outside the range from -3 to 3 designated "unaccptable" were 2 laboratories and z-score in the ranges -2 to -3 and 2 to 3 designated "questionable" were 3 laboratories (9.0%). Three laboratories (9.0%) showed the z-score designated "questionable" for lufenuron. The additional proficiency testing for various product types will be needed to establish the scheme of quality control.
