Studies on Pollutant Concentrations in Ground Water and Their Behavior in Soils , Cheju Island. I. Changes in Nitrate-nitrogen Concentration in Ground-water near Hotel complexes and Business district During the Period form 1987 to 1992 (제주도 지하수중 오염물질의 농도와 토양중 그의 행동에 관한 연구 1. 호텔 및 상가주변에서의 지하수중 $NO_3-N$의 농도 변화 (1987-1992))

  • This study was conducted to investigate the degree of ground water pollution used for drinking water in Cheju Island. Samples were collected monthly from 31 wells of 10 divided areas and were analyzed for $NO_3-N$, pH, $SO_4$, Cl and hardness from 1987 to 1992. $NO_3-N$ concentrations in the samples, with the exception of sample No.23, did not exceed the standard concentration of drinking-water(10mg/l). $NO_3-N$ concentrations at area 10, unpolluted area, ranged from <1.0 mg/l to just over 1.2 mg/l. However, samples from the business district of the area 8 showed the concentrations ranging from 5 mg/l in 1987 to 8.l mg/l in 1992 with a mean of 6.8 mg/l, about 5 times higher than those from the area 10. $NO_3-N$ concentration in sample No. 23 increased from 4 mg/l in 1987 to 12.6 mg/l in 1991. Average rate of increase in $NO_3-N$ in samples of No. 1, 2, 4, and 8 at area 8 was about 1.2mg/l per year in the study period. The ground water at area 1, 2, 3, 4, 5, 6, and 7 showed in the range of pH 7 to 8.1, being similar to area 10. However, pH at area 8 was in the range of 6.6 to 7.3, being lower than that in the other areas. Hardness at area 1, 2, 3, 4, 5, and 6 were in the range of 30 to 80 mg/l, being higher than that at area 10. Those at area 8 were the highest among all the other areas tested. The results of this study suggest that $NO_3-N$ contamination in ground water could be a problem at hotel complexes and business district in Cheju Island.

Risk Factor Analysis for Preventing Foodborne Illness in Restaurants and the Development of Food Safety Training Materials (레스토랑 식중독 예방을 위한 위해 요소 규명 및 위생교육 매체 개발)

  • Recently, with the rapid expansion of the franchise restaurants, ensuring food safety has become essential for restaurant growth. Consequently, the need for food safety training and related material is in increasing demand. In this study, we identified potentially hazardous risk factors for ensuring food safety in restaurants through a food safety monitoring tool, and developed training materials for restaurant employees based on the results. The surveyed restaurants, consisting of 6 Korean restaurants and 1 Japanese restaurant were located in Seoul. Their average check was 15,500 won, ranging from 9,000 to 23,000 won. The range of their total space was 297.5 to $1322.4m^2$, and the amount of kitchen space per total area ranged from 4.4 to 30 percent. The mean score for food safety management performance was 57 out of 100 points, with a range of 51 to 73 points. For risk factor analysis, the most frequently cited sanitary violations involved the handwashing methods/handwashing facilities supplies (7.5%), receiving activities (7.5%), checking and recording of frozen/refrigerated foods temperature (0%), holding foods off the floor (0%), washing of fruits and vegetables (42%), planning and supervising facility cleaning and maintaining programs of facilities (50%), pest control (13%), and toilet equipped/cleaned (13%). Base on these results, the main points that were addressed in the hygiene training of restaurant employees included 4 principles and 8 concepts. The four principles consisted of personal hygiene, prevention of food contamination, time/temperature control, and refrigerator storage. The eight concepts included: (1) personal hygiene and cleanliness with proper handwashing, (2) approved food source and receiving management (3) refrigerator and freezer control, (4) storage management, (5) labeling, (6) prevention of food contamination, (7) cooking and reheating control, and (8) cleaning, sanitation, and plumbing control. Finally, a hygiene training manual and poster leaflets were developed as a food safety training materials for restaurants employees.

  • The quality of meat is highly variable in many properties. This variability originates from both animal production and meat processing. At the pre-slaughter stage, animal factors such as breed, sex, age contribute to this variability. Environmental factors include feeding, rearing, transport and conditions just before slaughter (Hildrum et al., 1995). Meat can be presented in a variety of forms, each offering different opportunities for adulteration and contamination. This has imposed great pressure on the food manufacturing industry to guarantee the safety of meat. Tissue and muscle speciation of flesh foods, as well as speciation of animal derived by-products fed to all classes of domestic animals, are now perhaps the most important uncertainty which the food industry must resolve to allay consumer concern. Recently, there is a demand for rapid and low cost methods of direct quality measurements in both food and food ingredients (including high performance liquid chromatography (HPLC), thin layer chromatography (TLC), enzymatic and inmunological tests (e.g. ELISA test) and physical tests) to establish their authenticity and hence guarantee the quality of products manufactured for consumers (Holland et al., 1998). The use of Near Infrared Reflectance Spectroscopy (NIRS) for the rapid, precise and non-destructive analysis of a wide range of organic materials has been comprehensively documented (Osborne et at., 1993). Most of the established methods have involved the development of NIRS calibrations for the quantitative prediction of composition in meat (Ben-Gera and Norris, 1968; Lanza, 1983; Clark and Short, 1994). This was a rational strategy to pursue during the initial stages of its application, given the type of equipment available, the state of development of the emerging discipline of chemometrics and the overwhelming commercial interest in solving such problems (Downey, 1994). One of the advantages of NIRS technology is not only to assess chemical structures through the analysis of the molecular bonds in the near infrared spectrum, but also to build an optical model characteristic of the sample which behaves like the “finger print” of the sample. This opens the possibility of using spectra to determine complex attributes of organic structures, which are related to molecular chromophores, organoleptic scores and sensory characteristics (Hildrum et al., 1994, 1995; Park et al., 1998). In addition, the application of statistical packages like principal component or discriminant analysis provides the possibility to understand the optical properties of the sample and make a classification without the chemical information. The objectives of this present work were: (1) to examine two methods of sample presentation to the instrument (intact and minced) and (2) to explore the use of principal component analysis (PCA) and Soft Independent Modelling of class Analogy (SIMCA) to classify muscles by quality attributes. Seventy-eight (n: 78) beef muscles (m. longissimus dorsi) from Hereford breed of cattle were used. The samples were scanned in a NIRS monochromator instrument (NIR Systems 6500, Silver Spring, MD, USA) in reflectance mode (log 1/R). Both intact and minced presentation to the instrument were explored. Qualitative analysis of optical information through PCA and SIMCA analysis showed differences in muscles resulting from two different feeding systems.

Characterization of Bacillus amyloliquefaciens DA12 Showing Potent Antifungal Activity against Mycotoxigenic Fusarium Species

  • In an attempt to develop a biological control agent against mycotoxigenic Fusarium species, we isolated Bacillus amyloliquefaciens strain DA12 from soil and explored its antimicrobial activities. DA12 was active against the growth of mycotoxigenic F. asiaticum, F. graminearum, F. proliferatum, and F. verticillioides both in vitro and in planta (maize). Further screening using dual culture extended the activity range of strain DA12 against other fungal pathogens including Botrytis cinerea, Colletotrichum coccodes, Endothia parasitica, Fusarium oxysporum, Raffaelea quercus-mongolicae, and Rhizoctonia solani. The butanol extract of the culture filtrate of B. amyloliquefaciens DA12 highly inhibited the germination of F. graminearum macroconidia with inhibition rate 83% at a concentration of $31.3{\mu}g/ml$ and 100% at a concentration of $250{\mu}g/ml$. The antifungal metabolite from the butanol extract was identified as iturin A by thin layer chromatography-bioautography. In addition, volatile organic compounds produced by DA12 were able to inhibit mycelial growth of various phytopathogenic fungi. The volatile compounds were identified as 2-heptanone, 5-methyl heptanone and 6-methyl heptanone by gas chromatography-mass spectrometry (GC-MS) analysis. These results indicate that the antagonistic activity of Bacillus amyloliquefaciens DA12 was attributable to iturin A and volatile heptanones, and the strain could be used as a biocontrol agent to reduce the development of Fusarium diseases and mycotoxin contamination of crops.

Changes of multi-purpose solutions for soft contact lens depending on using period or keeping temperature (사용기간 및 온도에 따른 소프트콘택트렌즈 다목적용액의 변화)

  • To investigate changes of multi-purpose solutions for soft contact lens(MPS) depending on using period or keeping temperature, we evaluate four brands of MPS. No significant difference was seen in protein deposit removing efficacy after samples had used for 24 weeks and kept at $4^{\circ}C$, $20^{\circ}C$ or $30^{\circ}C$. The pH values of the samples of 4 brands measured weekly over the 24 week testing period. The initial average pH value of samples were 7.0, 7.5, 7.6 or 8.2. One brand of MPS was in the range of the threshold for ocular awareness, which is outside the zone of 6.6 ~ 7.8. During the testing period, the pH value were decreased in using period-dependent manner. At the 24th week, the average pH values of samples turned to 6.6, 7.2, 7.2 or 7.7. However, the difference of keeping temperature was not associated with decreased levels of pH values. After 24 weeks, one of total 36 samples was contaminated by bacteria. Furthermore, the change of components was shown after 24 weeks in the analysis using thin layer chromatography and the analysis of UV absorption pattern. The results of our study provides that the keeping temperature of MPS is not the important factor of changes of MPS, but the using period of MPS can cause contact lens wearers discomfort.

The Effect of Physico - Chemical Environmental Parameters on Early Composting of Paper - Mill Sludge Cakes (이화학적 환경변수가 제지슬러지케이크의 조기부숙에 미치는 영향)

  • The paper manufactories in Sangpyeong Industrial Estates, Chinju, produce more than 80 tons of paper-mill sludge cakes every day, which costs about 840 million won for dump per year. Since the paper-mill sludge is biodegradable, the sludge can he utilized as an organic fertilizer if it is properly decomposed. This may lead to not only dramatic cut of the treatment cost but prevention from secondary environmental contamination due to dump. The primary objective of the study was to explore the quantitative range of major enviromental parameters influencing early composting of paper-mill cakes, naturally pretreated in warn and cold weather. The automatically-controlled reactor was designed, manufactured, and operated for nine days to decompose about 2201 of raw sludge cakes. Four tests(Test 1-Test 4) were implemented for the study of Phase I. Treatments of two levels of initial temperature (40˚C and 28˚C) and two levels of water content + C/N ratio (35% + 40 and 63% + 80) were made to test the significance of their parameters for decomposition of raw sludge cakes pretreated in warm weather. Another four tests (Test 5- Test 8) were implemented for the study of Phase II. Treatments of initial temperature and water content(W/C) + C/N ratio of raw sludge pretreated in cold weather were made to 16˚C and 13% +58 for Test 5, 6˚C and 53% +55 for Test 6, 7˚C and 36% +81 for Test 7, 31˚C and 30% +81 for Test 8. Natural weater condition(pretreatment condition) revealed the importance m composting of the paper-mill sludge cakes. Combination of water content adjustment to about 30% with C/N ratio amendment of about 20 and initial temperature of 30~40˚C was concluded to be the best for early composting of paper-mill sludge cakes with aeration rate and pH fixed. Temperature and C/N ratio were adapted as judging variables for composting degree. In addition, tests for microbial activity were performed to validate the experimental results. Since the temperature and C/N ratio did not coincide in some tests as judging variables for the maturity of the composting sludges, taking one of these parameters could mislead the concept of the maturity (composting conceptually new criterion to provide more reliable information for early composting of paper-mill sludge cakes.

Monitoring of Endocrine Disruptor-suspected Pesticide Residues in Greenhouse Soils and Evaluation of Their Leachability to Groundwater (시설재배 토양 중 내분비계장애 추정농약의 잔류 모니터링 및 지하수 용탈 가능성)

  • This study was carried out to survey the residual characteristics of endocrine disruptor (ED)-suspected pesticides in greenhouse soils and assess their leachabilites to groundwater. Greenhouse soils were collected from 40 sites of greenhouse in 2008 in Korea. Sixteen ED-suspected pesticides which had been using in Korea, such as alachlor, benomyl, carbaryl, cypermethrin, 2,4-D, dicofol, endosulfan, fenvalerate, malathion, mancozeb, metribuzin, metiram, methomyl, parathion, trifluralin, and vinclozolin, in the soils, were analyzed by chromatographic methods using GLC-ECD and HPLC-DAD/FLD. Limits of detection (LODs) of the test pesticides ranged from 0.0004 to 0.005 mg/kg. Recoveries of the target pesticides from soil ranged from 72.69 to 115.28%. Four pesticides including cypermethrin were detected in the range of from 0.001 to 2.019 mg/kg, representing that their detection rate from greenhouse soils was 37.5%. The highest detection rate was observed from endosulfan which was detected from 16 site soils of the total samples, indicating that endosulfan is persistent in soil because of its very low mobility and high adsorption characteristics in soil. Based on the groundwater ubiquity scores (GUSs) of the pesticides detected from greenhouse soils, most of them have little possibilities of groundwater contamination except the fungicide vinclozolin with some leaching potential because of high water solubility and very low soil adsorption property.

Heavy Metal Concentrations in the Mollusc Gastropod, Cipangopaludina chinensis malleata from Upo Wetland Reflect the Level of Heavy Metals in the Sediments

  • Upo wetland is the largest inland wetland in Korea as Ramsar Convention Area. The purposes of the study were to investigate the levels of heavy metals (Cd, Cr, Cu, Ni, Pb and Zn) in the sediment and Cipangopaludina chinensis malleata from three sites of the wetland and to assess the potential of the gastropod as a bioindicator for heavy metal levels. The gastropods were dissected into shell and soft tissue without the digestive and excretive organs. The levels of Cd, Cu and Pb were below the guideline of Soil Environment Conservation Act and the heavy metals except Cr were slightly different among the sites. Cd was higher in Upo site ($0.32{\mu}g/g$) than Sajipo site ($0.28{\mu}g/g$). Cu and Zn showed the highest value in Sajipo as $43.5{\mu}g/g\;and\;39.8{\mu}g/g$, respectively while the concentrations of Pb and Zn were the highest in Upstream as $58.8{\mu}g/g\;and\;138{\mu}g/g$, respectively. In the soft tissues and shells of the gastropod, the overall common trend in the concentrations of the heavy metals was revealed with the following order: Zn > Cu > Cr > Ni > Pb > Cd and Ni > Zn > Cr > Cu > Pb > Cd, respectively. Although the soft tissues exhibited higher concentrations of the heavy metals except Ni than the shell in the gastropod, the levels of Cd and Pb in the gastropod were generally below the restrictive values set up by Korea Food & Drug Administration. From Duncan's Multiple Range Test (DMRT) results, the concentrations of Pb and Zn in the sediments among the sites were reflected on the soft tissue (Pb) and the shell (Pb and Zn) of the gastropod in the same order. The lower value of coefficient of variation (CV) in Pb concentration of the shell than in that of the soft tissue supports the usefulness of the shell as a bioindicator for Pb pollution. Although the CV value in the shell was a little higher than in the soft tissue, DMRT results and the stability of incorporated Zn into the shell support the use of the shell of the gastropod as a potential bioindicator for long-term contamination of Zn.

A study on the concentration and health risk of trihalomethanes in drinking water (음용수중 Trihalomethane의 오염과 그 위해성에 관한 조사연구 - 주요 14개도시를 대상으로 (1988. 9-1989. 2) -)

  • This study was conducted as a nationwide survey In Korea to determine the THM (trihalomethane) concentration levels in the drinking water of 14 selected cities. The survey was underdtaken in one city (Kwangju) during September and October of 1988 and 14 cities in January through February of 1989. The results were as follows : 1. The $KMnO_4$ consumption was 10.6mg/l-11.6mg/l in Pusan and Ulsan, exceeding the drinking water standard of 10mg/l. Pusan, Inchon, Kwangju, Ulsan, Mokpo and Junju areas also exceeded the ammonia nitrate standard of 0.5mg/l. Other tap waters surveyed were detected below the drinking water standards. 2. The THM concentrations of tap water measured in January and February of 1989 were detected in the range $1.20{\mu}g/l-150.8{\mu}g/l$. 3. In the comparative study of the THM concentration of tap water measured in the Kwangju area during September and October of 1998, the average THM concentation of $145.63{\pm}70.72{\mu}g/l$ showed a sixfold increase compared to that of $23.8{\pm}8.31{\mu}g/l$ surveyed in January and February 1989. 4. The proportion of the four THM compounds found in tap water was bromoform, 47% ; chloroform, 30% ; chlorodibromomethane, 13% ; and dichlorobromomethane, 10%. 5. Since the results indicate that the concentration of bromoform was 2-10 times higher than that of chloroform measured in the seaside district of Pusan, Ulsan and Cheju Island, it is reasonable to assume that the raw water was somehow Influenced by seawater. 6. The average lifetime cancer rate of the population exposed to chloroform measured in the surveyed areas was 17 cancer incidences per 1 million population. From the above results, the existence of THM in the distribution systems seems to be inevitable, since chlorine disinfection is performed in water treatment plants In our country. There seems to be a trend of increasing. THM cncentrations due to the contamination of raw water. In order to establish my form of regulations, health risk assessment is an imminent subject.

Contamination Characteristics of Hazardous Air Pollutants in Particulate Matter in the Atmosphere of Ulsan, Korea (울산시 미세먼지의 유해대기오염물질 오염 특성)

  • Recently, long-range atmospheric transport (LRAT) from China is regarded as a major reason for elevated levels of particulate matter (PM) in Korea. However, local emissions also play an important role in PM pollution, especially in large-scale industrial cities. In this study, PM samples were collected at suburban, residential, and industrial sites in Ulsan, Korea. Polycyclic aromatic hydrocarbons (PAHs) and heavy metals were analyzed, and a potential human health risk assessment was conducted. The concentrations of PAHs and heavy metals in total suspended particles (TSP) increased during high $PM_{10}$ episodes, and backward trajectory analysis verified the influence of LRAT from China during the high episodes. Furthermore, the concentrations of PAHs and heavy metals in $PM_{2.5}$ and $PM_{10}$ at the industrial site were higher than those at the residential site. The risk assessment of PAHs and heavy metals in $PM_{2.5}$ suggested no significant health effects. The highest levels of PAHs were measured in the particle size of $0.32{\sim}0.56{\mu}m$ at the residential site, and those of heavy metals were detected in the particle size of 1.8~5.6 and $>18{\mu}m$, reflecting different major emissions sources for both groups. On the basis of this preliminary study, we are planning long-term monitoring and modeling studies to quantitatively evaluate the influence of industrial activities on the PM pollution in Ulsan.
