Raffinose와 stachyose 함량이 낮은 콩과 두부의 품질특성 (The Quality Characteristics of Low Raffinose and Stachyose (LRS) Soybean Cultivars and their Tofu)

  • LOX-1,2,3 결핍과 비소화성 당류인 raffinose와 stachyose 함량이 낮은 품종(LRS)인 진양콩, 하영콩 및 대복콩을 원료로 하여 제조한 두부의 품질특성을 태광콩 두부와 비교하였다. 태광콩과 하영콩의 백립중은 여타 시료에 비해 유의적으로 높았다. 조단백질 함량은 태광콩에 비해 진양콩 및 하영콩에서 유의적으로 많았다. 두부의 수율은 태광콩, 하영콩 및 대복콩으로 만든 두부가 비슷한 수준이었다. 두부의 경도는 태광콩 및 대복콩으로 만든 두부가 가장 낮았으며, 하영콩으로 만든 두부에서 유의적으로 높았다. 콩 및 두부의 이소플라본 함량은 대조구에 비해 LRS 콩은 약 1.4~1.6배, 두부는 약 1.8~3.4배 높은 함량이었다. 콩의 총 페놀 함량은 대조구에 비해 하영콩 및 대복콩에서 유의적으로 높았다. 두부의 총 페놀 및 플라보노이드 함량은 대조구에 비해 하영콩 및 대복콩으로 만든 두부에서 유의적으로 높았다. 콩의 항산화 활성은 태광콩이 LRS 콩에 비해 높은 활성이었으나, 두부에서는 하영콩 및 대복콩으모 만든 두부가 대조구에 비해 유의적으로 높았다. 따라서 LRS 콩은 두부 제조에 적합하나 특히 하영콩이 콩의 단백질, 이소플라본 함량 및 두부의 항산화 활성 측면에서 가장 우수한 것으로 판단된다.

Glucansucrases를 이용한 새로운 올리고당의 합성 (Enzymatic Synthesis of New Oligosaccharides Using Glucansucrases.)

  • 본 연구에서는 dextransucrase hyper-producing Leuconostoc mesenteroides B-512FMCM과 dextransucrase constitutive mutants인 B-742CB 및 B-1355C에서 얻은 dextransucrase 및 alternansucrase를 이용하여 maltose, lactose, gentiobios 및 raffinose의 수용체반응을 수행하였다. B-512FMCM의 경우 수용체로서 maltose와 gentiobiose는 설탕에 대한 농도비가 1/1이하일 때 수용체에 ${alpha}$l-6으로 D-glucose가 결합한 일련의 올리고당을 생성하였고, 9/1이상에서는 단 두 가지의 수용체산물로서 maltose의 경우에는 panose와 isomaltosyl maltose를, gentiobiose의 경우에는 6$^2$-${alpha}$-D-glucopyranosyl gentiobiose와 isomaltosyl gentiobiose만을 생성하였다. 그러나 lactose는 유일한 산물인 2$^1$-${alpha}$-D-glycopyranosyl lactose만을 생성하였다. 효소양의 변화에 대해서 maltose와 gentiobiose의 수용체반응에서 생성된 수용체산물(올리고당)의 수와 양은 효소양이 증가함에 따라 증가하였으나 lactose의 경우에는 효소양의 증가에 대해 수용체산물 양의 변화가 크지 않았다. B-742CB dextransucrase와 B-1355C alternansucrase를 사용하면 B-512FMCM dextransucrase에서는 단 한가지만의 수용체산물을 생산하였던 lactose와 raffinose를 수용체로 하여 여러 가지의 수용체산물(올리고당)을 생산하였다.

완두종자분필물과 Pythium에 의한 묘립고병에 대한 감수성과의 관계 (Relationship Between Susceptibility to Pythium Seedling Blight and Pea Seed Exudates)

  • Pythium ultimum에 의한 묘립고병에 감수성인 완두품종의 종자는 저항성품종의 종자보다 다량의 환원당이 종자분필물내에 포함되었으며 분필속도도 감수성인 완두품종의 종자에서 더 빨랐다. 묘립고병에 저항성인 완두품종의 종자와 감수성인 품종의 종자분필물에서 모두 Raffinose, Meilbiose, Sucrose, Glucose 및 Fructose가 확인되었으나 Glucose와 Sucrose는 감수성 품종의 종자분필물에서 더 많이 검출되었다. Pythium ultimum균은 감수성인 종자 및 종자분필물에서 더 잘 자랐다.

A Co-expression Network of Drought Stress-related Genes in Chinese Cabbage

  • Plants have evolved to adapt to abiotic stresses, such as salt, cold, and drought stress. In this study, we conducted an in-depth analysis of drought resistance mechanisms by constructing a gene co-expression network in Chinese cabbage (Brassica rapa ssp. pekinensis L.). This drought stress co-expression network has 1,560 nodes, 4,731 edges, and 79 connected components. Based on genes that showed significant co-expression in the network, drought tolerance was associated with the induction of reactive oxygen species removal by raffinose family oligosaccharides and inositol metabolism. This network could be a useful tool for predicting the functions of genes involved in drought stress resistance in Chinese cabbage.

The Effects of Carbon Sources on the Biosynthesis of the Phospholipid and the Fatty Acid Composition of Mitochondria in Chlorella ellipsoidea

  • The biosynthesis of phospholipid and the composition of fatty acid were analyzed in mitochondria isolated from Chlorella ellipsoidea treated with carbon sources (glucose, sucrose, raffinose) during the culture. The growth of Chlorella and total lipid contents in mitochondria treated with various carbon sources was increased to compare with the control. When Chlorella mitochondria was treated with various carbon sources, four kinds of phospholipid were increased predominantly. The major fatty acids utilized for the biosynthesis of the phospholipid were analyzed linoleic acid (average 25.18%) and stearic acid (average 10.52%) in the control. But, it was shown that the major fatty acids in Chlorella mitochondria treated with glucose were stearic acid (average 30.93%), palmitic acid (average 17.47%) and stearic acid (average 20.31%), linoleic acid (average 16.68%) in sucrose treatment and oleic acid (average 17.17%), palmitic acid (average 15.64%) in raffinose treatment.

Purification and Characterization of Guar Galactomannan Degrading $\alpha$-Galactosidase from Aspergillus oryzae DR-5

  • $\alpha$-Galactosidase from A. oryzae DR-5 was induced in the presence of melibiose, raffinose, galactose, and locust bean galactomannan. The enzyme was purified to homogeneity by precipitation with acetone followed by ion-exchange chromatography using DEAE-Sephacel. The purified enzyme showed a single band in both nondenaturing-PAGE and SDS-PAGE. The enzyme was a glycoprotein in nature by activity staining. The molecular weight of the purified enzyme was 93-95 kDa by SDS-PAGE. The enzyme exhibited the optimum pH and temperature at 4.7 and $60^\circ{C}$, respectively. $\alpha$-Galactosidase activity was strongly inhibited by $Ag^{2+}, Hg^{2+}, Cu^{2+}$, and galactose. EDTA, 1,10-phenanthraline, and PMSF did not inhibit the enzyme activity, whereas N-bromosuccinimide completely inhibited enzyme activity. Investigation by TLC showed complete hydrolysis of stachyose and raffinose in soymilk in 3 h at pH 5.0 and $50^\circ{C}$.

고려인삼(Panax ginseng) Invertase의 정제와 그 특성 (Purification and Characterization of Invertase from Korean Ginseng Panax ginseng)

  • In An invertase (EC 3.2.1.26) was extracted from Korean giseng (Panax ginseng C.A. Meyer) with distilled tvater The ginseng invertase was purified about 62.6 folds purified by procedures including ammonium sulfate fractionation , DEAE-cellulofine chromatography and gelfiltrations through Sephadex G-75 and the recovery of enzyme activity was 11.1%. The homogeneity of the purified enzyme was probed by polyacrylamide gel disc electrophoresis. The purifled enzyme was divided into two different subunits by treating with a mixture of SDS and 2-mercautoethanol, and the molecular weight of the large subunit was estimatedtobe 116,000 and that of the small one to be 14,000. The optimal VH and temperature of the enzyme were pH 6 and 45$^{\circ}C$, respectively. The enzyme hydrolyzed specifically the hydrolyzation of the -fructofuranosides such as sucrose, raffinose and inulin. The Km values of the enzyme for sucrose and raffinose were determined to be 0.85 and 0.6 mM, respectively.

유청의 갈락토올리고당을 이용한 Bifidobacteria 의 생육촉진 (Use of Galactooligosaccharides from Cheese Whey for Growth of Bifidobacteria)

  • Effect of galactooligosaccharides produced by the $\beta$-galactosidase from Aapwefillua niger CAD 1 on the growth of Bifidobacterium infantis KCTC 3127 Bifidobacterium longum KCTC 3128 and Bifidobacterium bif-idum ATCC 11863 were investigated. Bifidbacterium infantis Bifidobacterium longum and Bifidobacterium bif-idum were in the logarithmic growth phase after 6hr incubation at 37$^{\circ}C$. Bifidobacterium infantis was in the stationary phase after 24hr incubation at 37$^{\circ}C$. The growth rate of B. bifidum containing galactooligo-saccharides and raffinose in MRS broth increased up to 18%, 8% and 7% compared to glucose galac-tose and lactose during 48hr incubation. The growth rate of B. infantis and B. longum contatining galacto-oligosaccharides and raffinose in MRS broth increased up to both 6% and 8% and both 13% and 10% compared to glucose and galactose during 48hr incubation.

국내 포도밭 토양에서 분리된 야생 효모 미기록종들의 균학적 특성과 탄소원 활성 (Mycological Characteristics and Carbon Source Activities of Unrecorded Wild Yeast Species from Vineyard Soil in Korea)

  • This study aimed to isolate and characterize wild yeast strains from the vineyard soil in Korea. Twenty yeast strains were isolated from vineyard soil in Gimpo-si, Gyeonggi-do, Korea, where Vitis labrusca cv. Campbell Early is grown. Eighteen strains were previously recorded in Korea. The remaining two, Cyberlindnera mrakii VG-21-10C and Starmerella bacillaris GR9 were not previously recorded in Korea. The mycological characteristics of VG-21-10C and GR9 were investigated. Both were oval-shaped, convex, and smooth. However, differences were evident in colony color and carbon assimilation activities. Strain VG-21-10C is white-colored and assimilates glucose, glycerol, D-xylose, D-cellobiose, D-saccharose, and D-raffinose as carbon sources. Strain GR9 is cream-colored and assimilates glucose, D-saccharose, and D-raffinose as carbon sources.

Characterization of the $\alpha$-Galactosidase Gene from Leuconostoc mesenteroides SY1

  • Leuconostoc mesenteroides SY1, an isolate from kimchi, was able to ferment $\alpha$-galactosides, such as melibiose and raffinose. $\alpha$-Galactosidase ($\alpha$-Gal) activity was higher in cells grown on melibiose and raffinose than cells grown on galactose, sucrose, and fructose. $\alpha$-Gal activity was not detected in cells grown on glucose, indicating the operation of carbon catabolite repression (CCR). A 6 kb DNA fragment was PCR amplified using a primer set based on the nucleotide sequence of a putative $\alpha$-galactosidase gene (aga) from L. mesenteroides ATCC 8293. Nucleotide sequencing of the 6 kb fragment confirmed the presence of aga and other genes involved in the galactosides utilization, and the gene order was galR (transcriptional regulator)-aga-gaIK (galactokinase)-gaIT (galactose-1-phosphate uridylyltransferase). Northern blotting experiment showed that aga, gaIK, and gaIT constituted the same operon, that the transcription was induced by galactosides, such as melibiose and raffinose, whereas gaIR was independently transcribed as a monocistronic gene, and that the level of transcription was fairly constant. The aga was overexpressed in E. coli BL21 (DE3) using pET26b(+) vector, and $\alpha$-Gal was accumulated in E. coli as an inclusion body.
