The Effect of Vegetable Extracts on the Activity of Alcohol Dehydrogenase from Saccharomyces cerevisiae

  • We investigated the effects of bean sprouts (Glycine max), dropwort (Oenanthe javanica), and radish (Raphanus sativus var. hortensis for. acanthiformis) extracts on alcohol dehydrogenase (ADH). The extracts from three kinds of vegetables were prepared by extracting with boiling water, distilling water, and ethyl alcohol. Among extracts, boiling water extract showed the highest activating effect on ADH, respectively and distilled water extract had a greater effect on ADH activation than that of alcohol extract. The ADH facilitating effect of bean sprout extract by distilled water was significantly higher than dropwort or radish, hut the effect of the bean sprout extract by ethyl alcohol was lower than others. The facilitating effect on ADH of mixture extracts of bean sprout and dropwort were mixed at 1 : 1 mixture of boiled-water extract showed the highest effectiveness. And bean sprout extract separated below 3000 molecular weight (MW) range of extract fraction had greater ADH activity than large MW parts.

Physiological Activity of Extracts from Radish (Raphanus sativus L.) Leaves (무청 추출물의 생리활성)

  • This study was conducted to investigate yield of extract, total phenolic compounds content, electron donating activity, nitrite scavenging activity, superoxide dismutase like activity of the radish leaves (Raphanus sativus L.) extracts. Radish leaves extracts were prepared using water extraction, 50% ethanol extraction and 50% methanol extraction. The yield of Kwandong radish leaves extracts without blanching treatment (DRLK) was $26.56{\sim}32.56%$ and the extract yield of Minongdanbaek radish leaves without blanching treatment (DRLM) was $34.34{\sim}36.90%$. On the other hand, the yield of samples with blanching treatment was a lower range value of $18.70{\sim}23.56%$ than samples without blanching treatment. In the total contents of phenolic compound, samples with and without blanching treatment were $49.1{\sim}59.4mg/g\;and\;53.41{\sim}62.08mg/g$, respectively. Minongdanbaek radish leaves extracts (DRLM, BDRLM) showed higher contents value than Kwandong radish leaves extracts (DRLK, BDRLK) in the total phenolic compounds. Samples without blanching treatment showed electron donating activity above 80%, nitrite scavenging activity of $36.63{\sim}51.17%$ and superoxide dismutase like activity of $38.53{\sim}45.38%$. Generally, Kwandong samples showed a little higher activity more than Minongdanbaek samples in the electron donating activity, nitrite scavenging activity and superoxide dismutase like activity. Also, radish leaves extracts without blanching treatment showed high physiological activities.

Preparation of Active Fraction from Radish Water Extracts for Improving the Intestinal Functions and Constipation Activities (무(Raphanuse sativa var. nigra L.) 물 추출물로 부터 장기능 및 변비질환 개선을 위한 활성 분획의 제조)

  • The aim of this study was to develop an activator, 3-10 kDa fraction from radish water extracts, that will improve the intestinal function and bowel movement in the colons. Radish water extracts were investigated for their intestinal function effects according to the charcoal meal transit method, employing Balb/c mice: also, their anti-constipation activities were compared utilizing the loperamide-induced constipation method, employing SD rats. The result suggested that the effects of the charcoal meal transit increased remarkably in radish water extract administrated rats in comparison to loperamide administrated rats. Futhermore, the effects of various solvent extracts of radish on charcoal meal transit in Balb/c mice increased remarkably in radish water fraction administrated rats than in different solvent fraction administrated rats. Radish extraction was tested and isolated into 4 groups: below 3 kDa, 3-10 kDa, 10-300 kDa, and over 300 kDa. 3-10 kDa was the most effective on the intestinal function and bowel movement in the colons; also, 3-10 kDa fraction of radish water extraction was found to be the most effective charcoal meal transit. The dry weight and moisture content of feces remarkedly increased in the 3-10 kDa administrated rats group than in the loperamide only group. Experimental results revealed that 3-10 kDa fraction of radish water extract was the most effective on the intestinal function and bowel movement was the crypt epithelial cells that contained more MUC2 in the 3-10 kDa administrated group than the loperamide only group: in addition, the thickness of mucus layer stained with alcian blue was significantly thicker in 3-10 kDa administrated rats than in loperamide administrated rats. Crypt epithelial cells secreted more MUC2 in the 3-10 kDa administrated group than the loperamide only group and the stained cells clearly showed the MUC2 with antibody Biogenex AM358.

The Chemopreventive Effect of Heat-treated Radish Complex Extracts on Rat Colonic Aberrant Crypt Foci Induced by 1,2-Dimethylhydrazine (1,2-Dimethylhydrazine에 의해 유발된 Colonic Aberrant Foci에 대한 초임계 열처리된 무복합추출물의 대장암 효과)

  • The present study was designed to investigate the chemopreventive effects of supercritical heat radish extracts with anti-constipation acitivity in loperamide-treated rats, on 1,2-dimethylhydrazine(DMH)-induced aberrant crypt foci(ACF) in rat colonic mucosa. Supercritical heat radish extracts was administered in the drinking water(3.2 mg/ml) during the initiation (for 12 weeks) or postinitiation phase(for 9 weeks) of DMH-induced carcicogenesis. Supercritical heat radish extracts coupled with DMH for 12 weeks decreased significantly total number of aberrant crypts(pre-neoplastic lesions) and the number of ACF to 35% and 30%, respectively. The number of ACF with more than four aberrant crypts (crypt multiplicity) was significantly reduced in the colon of rats receiving Supercritical heat radish extracts for 12 weeks. In the group receiving Supercritical heat radish extracts after post-initiation for 9 weeks, only the number of ACF with ${\geq}4$ abberant crypts significantly decreased. Supercritical heat radish extracts, given alone for 12 weeks, did not induced the development of ACF. These results indicate that Supercritical heat radish extracts possess chemopreventive effects on chemically induced rat colon carcinogenesis.

  • Fibrionolytic activity(FA) of kimchi in rat and FAs of water and methanol extracts of kimchi ingredient were determined using fibrin plate method to see where the active principles for FA are present in kimchi. Nine Sprague-Dawley per each group were fed diet containing 3, 5 or 10% of freeze dried kimchi for 6 weeks. The FA of plasma obtained from 10% kimchi group showed stronger FA than that of control. FAs of 3 and 5% kimchi group were not noticeable. FA from methanol extract of kimchi was approximately 6 times higher than that from water extract assuming that the active material may be present in a fat soluble fraction. From water extract of kimchi ingredients, dropwort, green onion, radish and baechu showed FA in descending order. And from methanol extracts, red pepper powder, radish, green onion and baechu showed FA in decending order. The Fa of methanol extract of red pepper powder was the most noticeable among samples.

Comparison of Physiological Activities of Radish Bud (Raphanus sativus L.) according to Extraction Solvent and Sprouting Period (추출용매 및 발아시기에 따른 무순 추출물의 생리활성 비교)

  • This study extracted radish bud (Raphanus sativus L.) and investigated its nitrite scavenging activity, superoxide dismutases (SOD)-like activity, tyrosinase inhibition activity, xanthine oxidase inhibition activity, and angiotensin-converting-enzyme (ACE) inhibition activity according to extraction solvent and sprouting period. For nitrite scavenging activity, each extract recorded its highest level of 81.44~89.71% at pH 1.2. Radish bud extracts on sprouting days 4 and 8 showed greater scavenging activities than those on sprouting day 12 at pH 1.2 and pH 4.0. There were differences in scavenging activity according to extraction solvent based on water extract exhibiting improved scavenging activity. Ethanol extract recorded scavenging activity of 16.12% at pH 6.0, which was similar to those of ethanol and methanol radish bud extracts on sprouting day 12. SOD-like activity of radish bud extracts was in the range of 4.57~27.05%. For comparison purposes, SOD-like activity of L-ascorbic acid was 52.15%, which was higher than that of radish bud extracts. Acetone and methanol extracts showed high SOD-like activities on sprouting day 8. SOD-like activity of radish bud extracts on sprouting day 12 significantly decreased to 4.57~15.59%. Radish bud extracts recorded good tyrosinase inhibitory activities on sprouting 8 and 12, whereas methanol extracts recorded the greatest tyrosinase inhibitory activity at 62.65~84.89%. Radish bud extracts recorded xanthine oxidase inhibition activity of 21.26~29.52% on sprouting day 4, and acetone extracts showed the highest level of xanthine oxidase inhibition activity. Xanthine oxidase inhibitory activity tended to decrease with sprouting period compared early on. ACE inhibitory activity was in the range of 12.48~51.78% according to sprouting period and extraction solvent. Ethanol extracts on sprouting day 8 showed the highest ACE inhibitory activity of 51.78%. These results will hopefully contribute to research into the identification of materials and development of products for natural functional foods.

미생물 생육에 대한 무 추출물의 억제 및 촉진 효과

  • Extracts active on the growth of food-born microorganisms were prepared from radish by using acetone, methanol and water as the solvents. Strains of bacteria and yeast, such as L. brevis, L. fermentum, L. faecalis, L. mesenteroides, B. subtilis, S. flexneri, S. aureus and C. albicans. Addition of 1 $m\ell$ of acetone extract exhibited the strongest growth effect on L. brevis. The water extracts turned out to stimulate growth of all Lactic acid bacteria. Especially, it was observed that the same material produced the highest stimulatory effect(by about 40-50 times) on L. faecalis and L. fermentum and L. mesenteroides. Also, it was proved to have a weak stimulate effect on C. albicans, S. flexneri and S. aureus. But, on the contrary to this, when applied to B. subtilis it showed growth stimulation by about 10 times.

Cytotoxicity of Ethanol Extract of Raphanuse Sativus on a Human Lung Cancer Cell Line (인체 폐암 세포주에 대한 무의 에탄올 추출물의 세포독성)

  • This study was desinged to investigate the cytotoxic effect of ethanonl extract of radish, Raphanuse sativus on human lung cancer cell lines (A-549) using MTT(3-〔4,5-dimethylthiazol-2-yl〕-2-yl)-2,5-diphenyltetrazolium bromide) assay. Radish trunk and radish root were extracted by a mixed solvent of water and ethanol (5 : 5, v/v) after drying, The extracts were used for anticancer activity assay. $IC_{50}$/ (50% inhibitory concentration) of radish trunk and radish root were 0.015% and 0.03%, respectively. Radish trunk extract showed higher anticancer activity than radish root extract at the same concentration of 0.01%.

Allelopathic Inhibition by Extracts and Volatiles from Leaf and Seed of Sicklepod (Cassia tora L.) (결명자(決明子)(Cassia tora L.) 엽(葉)과 종자(種子)의 추출물(抽出物)과 휘발성분(揮發成分)의 타감성(他感性) 생육저해작용(生育沮害作用))

  • The growth of weeds and some other plants has been considered to be inhibited by sicklepod(Cassia tora L.) sharing the habitat. The study was conducted, for the first time, to propose that this phenomenon is not only due to competition for physical and nutritional conditions but also due to allelopathy. In addition, autotoxicity of sicklepod was examined. The results are summarized as follows. 1. Rice sheath length reduced progressively from 18 % to 36 % with increasing the concentration of treating aqueous extracts of sickle pod seeds, but rice germination was not affected. In contrast, radish hypocotyl length was not reduced by the aqueous extract treatment but radish germination was significantly reduced by 66 % at 1 : 10 and 1 : 5 treatment. 2. Total chlorophyll contents in rice seedling decreased from 50 % to 65 % by treatment of seed aqueous extracts diluted from 1 : 50 to 1 : 5 ratio. 3. Aqueous extracts of sicklepod leaves significantly reduced hypocotyl length and fresh weight in radish and germination in rice, but mung bean was slightly affected by aqueous extracts only in fresh weight. 4. Volatiles from fresh, immature seeds with husk reduced the radish germination and seedling growth and radish root growth appeared to be more sensitive to the exposure to volatiles from fresh immature seeds than both germination and hypocotyl growth. 5. Volatiles from sickepod leaves inhibited germination and growth of radish, rice and mung bean, and seedling growth was more sensitive to volatiles from leaves than germination. 6. Volatiles from sicklepod leaves reduced germination and radicle length of sicklepod itself. 7. Collectively, it is concluded that there are water-soluble and volatile substances responsible for allelopathy in sicklepod.

  • Allelopathic inhibition by substance(s) originated from sicklepod(Cassia tora L.) seed on germination of other plant seeds, was confirmed and used to elucidate general mechanism of allelopathy which was occurred widly in natural and/or agricultural ecosystems. The mechanism was deduced from changes in water contents, ${\alpha}$-amylase activities, protease activities, concentrations of abscisic acid and total phenolic compounds during germination of rice and radish seeds treated with water-soluble extracts of sicklepod seeds. The results obtained were summarized as follows. 1. The % germinations of radish, rice, lettuce, barley seeds were decreased by substances originated from crushed sicklepod seeds. 2. By the treatment of water-soluble extracts of sicklepod seeds, the inhibition of germination of radish seeds was occured with the sequential phenomena of increase of protease activity(synthesis), decrease of water content and increase of total phenolic compounds content. 3. In rice, the inhibition of seed germination by the water-slouble extracts of sicklepod seeds was related to increase of abscisic acid concentration and then decrease of ${\alpha}$-amylase activity(synthesis).
