무즙의 돌연변이 억제 효과 및 그 특성 (Inhibitory Effect of Radish Juice on the Mutagenicity and Its Characteristics)

  • Salmonella typhimurium TA 98 균주를 사용한 Ames법을 실시하여 무즙의 돌연변이 억제효과 및 특성에 대해 조사하였다. 무즙은 간접 돌연변이원인 것으로 조사된 담배연기응축물(CSC), 태운 삼치의 메탄올 추출물(MECS) 2종에 대해서는 각각 100% 정도의 억제효과를 보였고, 발암물질인 2-aminofluorene(2-AF)에 대해서는 87% 정도의 억제효과를 나타내었다. 그리고 무즙은 양배추, 부추 등과 더불어 CSC에 대해서 조사된 다른 과 채류에 비해 높은 억제효과를 나타냈다. 무즙에서 CSC에 대해 억제효과를 나타내는 물질은 분자량이 50,000 이상으로 $100^{\circ}C$의 열처리시 5분 이내에 효과가 50% 이하로 감소하였으나 중 저온에서는 약 2주간 효과가 유지되는 것으로 조사되었다. 또한 ammonium sulfate $30{\sim}80%$로 포화시켜 얻은 무즙 침전물이 돌연변이 억제 효과가 큰 것으로 나타났으며 이를 native-PAGE를 실시하였던바 3종류의 band 추출물이 CSC에 대해 돌연변이 억제효과가 있는 것으로 나타났다.

결명자(決明子)(Cassia tora L.) 엽(葉)과 종자(種子)의 추출물(抽出物)과 휘발성분(揮發成分)의 타감성(他感性) 생육저해작용(生育沮害作用) (Allelopathic Inhibition by Extracts and Volatiles from Leaf and Seed of Sicklepod (Cassia tora L.))

  • 본 실험은 결명자(決明子)(Cassia tora L.)의 재배지에서 타작물이나 잡초의 생육이 저해되는 현상이 단순한 물리적 조건의 경쟁뿐 아니라 타감작용(allelopathy)에 기인함을 탐색함과 동시에 결명자의 자가독성을 확인하는 연구로서, 결명자 식물의 종자와 잎의 수용성 추출액 및 휘발성분이 무우(십자화과), 벼(화본과), 녹두(두파)등의 발아와 유묘성장에 미치는 영향을 살펴 본 결과는 다음과 같다. 1 결명자 종자의 수용성 타감물질에 의한 저해효과에 대해, 벼는 발아보다 유묘성장이 더욱 민감하게 저해되었고, 반면 무우는 발아가 배축의 길이 성장보나 더 민감하게 저해되었다. 2. 결명자 종사의 수용성 추출액 처리에 의해 추출액의 처리농도가 커질수록 벼 유묘의 엽록소 함량은 무처리구의 50%에서 65%까지 감소하였다. 3. 결명자 잎의 수용성 추출액 처리에 의해 무는 배추길이와 생체량이, 벼는 발아가 저해되었으나, 녹두는 단지 생체량에서 약간의 저해를 보였다. 4. 결명자 미숙종자의 휘발성분은 무우의 발아와 생육에 저해작용이 있으며, 특히 뿌리의 생장이 발아나 배축성장보다 더 민감하게 저해되었다. 5. 결명자 잎의 휘발성 성분은 무우, 벼, 녹두의 발아나 생육에 저해적인 타감작용 효과를 나타냈으며, 특히 발아보다는 유묘의 성장이 더 많이 저해되었다. 6. 결명자 잎의 휘발성분에 의해 결명자 스스로의 발아와 유근 성장이 저해되는 자가중독 현상이 확인되었다. 7. 결과적으로, 결명자의 종자와 잎에는 일부 타식물에 대하여 타감작용과 아울러 자가중독을 유발하는 수용성 및 휘발성 물질이 존재하는 것으로 인정된다.

국내 다소비 십자화과 새싹채소 추출물의 항산화 활성 (Antioxidant activities of commonly used Brassica spp. sprout vegetables in Korea)

  • 본 연구에서는 국내에서 다소비 되는 십자화과 새싹채소 7종(콜라비, 적무, 브로콜리, 배추, 유채, 무순 및 다채)의 항산화 및 지방세포 내 ROS 생성 억제 활성을 측정하였다. 총 페놀 및 총 플라보노이드 함량을 측정한 결과, 총 페놀 함량은 콜라비 새싹과 무순 추출물이 $23.97{\pm}0.46mg\;TAE/g$$24.40{\pm}1.24mg\;TAE/g$으로 유의적으로 가장 높은 값을 나타내었고, 총 플라보노이드 함량은 무순 추출물이 $15.30{\pm}1.35mg\;CE/g$으로 다른 새싹채소 추출물에 비해 높은 값을 나타내었다. DPPH radical 소거능과 ORAC value를 측정한 결과, 콜라비가 DPPH radical 소거능($IC_{50}=1.95mg/mL$)과 ORAC value($79,032.5{\mu}M\;TE/g$)에서 모두 항산화능이 뛰어난 것으로 나타났다. 한편, 다채를 제외한 새싹채소 추출물 6종 모두에서 세포 내 ROS 생성량을 감소시킴을 확인하였다. 이상의 결과로부터, 십자화과 새싹채소 중 콜라비 새싹과 무순 추출물의 경우 항산화활성 및 지방세포 내 ROS 생성 억제 활성을 가지며, 천연물 유래 항산화 소재로써 활용 가능성이 높은 것으로 기대된다.

수국차 잎 추출액을 첨가한 깍두기의 이화학적 및 관능적 특성 (The Physico-chemical and Sensory Characteristics of Kakdoogi Containing Hydrangea serrata Seringe Extract)

  • To evaluate the effect of Hydrangea serrata Seringe extract on kakdoogi(Korean radish kimchi), the kakdoogi was prepared, after supplementation, with Hydrangea serrata Seringe extract(0, 1.25, 2.5 and 5%(v/w)). The quality characteristics of the kakdoogi were investigated during 7 days of fermentation at $20^{\circ}C$. The proximate components of the Hydrangea serrata Seringe leaves were 3.48% crude protein, 6.45% crude lipid, 7.98% crude fiber and 9.32% crude ash. The pH values in the samples with the extract were higher than that of both the control group and the sugar group. The total acidity levels in the sugar group was higher than that of other groups. The sugar group showed higher L value(lightness) than that of kakdoogi fermented with the extract. A value(redness) and B value(yellowness) of the supplemented kakdoogi was lower than that of the control and the sugar group on day 7. The hardness of samples with the extract was higher than that of the sugar group. And the hardness of the groups with the extract was not reduced during the fermentation period. The control group showed higher salinity than the other groups by day 4. The samples with the extract were sweeter than the control group on day 0 then the difference disappeared after day 2. Sourness of all the groups increased during the fermentation period. However the extract did not affect the hardness or overall acceptability of the kakdoogi during fermentation.

무즙을 이용한 Candida utilis ATCC 42416균체 생산 (Chinese Radish Juice as a Growth Substrate for the Production of Candida utilis ATCC 42416 Biomass)

  • 단세포 단백질 자원으로 이용 가능한 균주로 알려진 Candida utilis ATCC 42416 균체를 생산하기 위하여 무를 파쇄하여 얻은 무즙을 배지로 균체배양 실험을 하였다. 무즙의 수용성 고형물과 총 당량은 각각 5.0~8.8 Brix와 3.5~6.5%였다. C. utilis ATCC 42416 균주를 무즙에 접종하여 $30^{\circ}C$에서 200rpm으로 진탕 배양하였을 때 24시간 이내에 증식이 끝났으며, 단위 소모당에 대한 최대 균체 생산량은 당농도 1% 회석 무즙에서 L당($5\;\times\;DCRI$) 21.5g의 건조균체를 생산하였다. 5배 회석무즙에 약간의 영양소 glucose(2.0%), yeast extract(0.2%), peptone(0.2%), ammonium sulfate(0.2%) 및 $KH_2PO_4(0.2%)$ 등을 각각 보강하여도 증식 속도, 균체생산 및 세포 단백질 함량 등에 차이가 거의 없어서 무즙자체가 C. utilis ATCC 42416 균체 생산에 좋은 기질이었음을 알 수 있다.

Comparative Investigation of Glutathione S-Transferases, Glyoxalase-I and Alliinase Activities in Different Vegetable Crops

  • Glutathione S-transferases(GSTs, EC 2.5.1.18), glyoxalase-I(EC 4.4.1.5) and alliin lyase(alliinase, EC 4.4.1.4) are important enzyme systems in plant bodies. The first two are mainly detoxifying enzymes that utilize glutathione(GSH) in the defense mechanism, and the last one is mainly involved in secondary metabolism and relevant to sulfur compounds derived from GSH. The activities of the three enzymes have been investigated in soluble extracts of vegetable crops, including pumpkin, cabbage, broccoli, radish, carrot, potato, sweet potato, mungbean, and onion. GST activities were detected in all of the vegetables, and the extract of onion bulb exhibited the highest specific activity(648 nmol/min/mgP). The putative GSTs of most of the vegetables were found to be induced by ethanol. The activities of GSTs in onion bulb were found to be markedly inhibited by S-hexyl glutathione and were also inhibited by S-butyl glutathione and S-propyl glutathione. The anti-CmGSTF1 antiserum recognized a thick band for putative onion GST. The estimated glyoxalase-I activity level was also high in onion bulb(4540 nmol/min/mgP), indicating that the thick band detected by Western blot analysis might result from partial recognition of glyoxalase-I by the antiserum. The specific activities for glyoxalase-I were moderate in radish and carrot, and the extracts of other vegetables had rather low levels of activities. The extract of onion also showed the highest specific activity level for alliinase(2069nmol pyruvate/mgP). The extracts of other vegetables also had alliinase activities, although the estimated values were much lower than that of onion.

멸치액젓잔사 추출물을 이용한 조미소재 개발 (Development of a Seasoning Sauce Using Hot Water Extracts from Anchovy Engraulis japonica Fish Sauce Processing By-products)

  • We developed a seasoning sauce using hot water extracts from anchovy Engraulis japonica fish sauce processing by-products. A temperature of 121℃ was maintained for 120 min and the resulting amino acid content, salinity, and pH were 183.6 mg/100 g, 6.86, and 17.4 g/100 g, respectively. Radish juice, sea tangle Saccharina japonica extract, and mushroom Lentinula edodes were added to improve the flavor. The glutamic acid content of the extract mixed with 10% sea tangle extract was 88.87 mg/100 g and the 5'-GMP (guanine 5'-monophosphate) content of the extract mixed with 10% mushroom extract was 9.67 mg/ g. This study was conducted to determine optimal processing conditions for seasoned products using response surface methodology (RSM). The optimal conditions for X1 (sea tangle extract concentration) and X2 (mushroom extract concentration) were 15.0% and 5.0%, respectively, and the predicted values of the multiple response optimal conditions were Y1 (5'-GMP: 17.36 mg/100 g) and Y2 (glutamic acid: 157.35 mg/100 g). Under the optimal conditions, the experimental values of Y1 and Y2 were 17.32 mg/g and 155.36 mg/100 g, respectively, which are similar to the predicted values. We confirmed the feasibility of developing a seasoning sauce using hot water extract from anchovy fish sauce processing by-products and additives.

  • This study is to examine the origin of Andong sikhe and to introduce the making process of it. The following facts are found after consulting literature and doing field-work to find out the origin of Andong sikhe. Andong sikhe is a kind of so-sikhe develolped as a sweet beverage, the making process of which is to ferment the mixture of boiled rice, radish slices, and red pepper extract with malt liquid. So-sikhe is made by fermenting with malt the mixture of boiled rice, radish slices, red pepper powder and flavorings such as a shallot, a garlic and salt, and o-sikhe is made by adding fish to the ingredients used to make so-sikhe. So-sikhe is distributed on the coastal areas of the East Sea and the inland areas adjacent to them as o-sikhe is. It is presumed that so-sikhe was introduced to Korea by the same way as o-sikhe was introduced eastward from the regions around Thailand by sea. It is also presumed that Andong sikhe has been developed into its present type after the second half of the 18th century when red pepper was introduced to Korea and then widely used.

New Stigmastane Steroids Constituents from Rice Hulls of Oryza sativa and Inhibitory Activity on Radish seed

  • Two new compounds 1'-(stigmast-11,20(21),25-trien 3$\alpha$,9$\alpha$-diolyl)-3'-(pimara-11,15-dien-3$\alpha$-olyl) glycerol (1) and stigmast-5-en-3$\alpha$,26-diol (2) along with known fatty acids n-hexacosanoic acid (3) and hexadecanoic acid (4), have been isolated from the methanol extract of rice hulls of Oryza sativa. The structures of the compounds were elucidated using 1D and 2D NMR spectral methods, viz; $^1H, ^{13}C, ^{13}C$-DEPT, $^1H-^1H$ COSY, $^1H-^{13}C$ HETCOR, HSQC and HMBC aided by IR, EIMS, FABMS and HRFABMS. Compound (1) showed inhibition to radish germination, growth of shoot and root length.

Induction of Hepatic Glutathione S-transferase Activity in Mice Administered with Various Vegetable Extracts

  • The effect of various vegetables commonly consumed by Koreans on the induction of glutathione S-trasferase(GST) activity in mice was assessd. The extract of vegetable dissolved in propylene glycol (5ml/kg body wt.) was administered to ICR female mice 6 to 8 weeks old via gavage during 5 days. The changes of body weight and liver weight of all treated groups were not significantly different compared with control group. Hepatic protein contents of trated groups compared with control group were not significantly different except BHT treated group. The induction of GST activity in liver cytosol of mice was the greatest with broccoli, followed by radish, wild green onion, turnip, and green onion. The induction of GST activity in liver cytosol increased up to 1.5 to 1.8-folds at a dose of 24 g fresh vegetable/mouse. The induction of combination between vegetables was the highest with the combination of broccoli and radish (1.83-fold), followed by that of broccoli and green onion (1.72-fold), and that of broccoli and turnip (1.50-fold).
