Assessing the Activity Concentration of Agricultural Products and the Public Ingestion Dose as Result of a Nuclear Accident

  • Background: A model to assess the activity concentration of agricultural products and the public ingestion dose as result of a nuclear accident is necessarily required to manage the contaminated agricultural systems by the accident, or to estimate the effects of chronic exposure due to food ingestion at a Level 3 PSA. Materials and Methods: A dynamic compartment model, which is composed of three sub-modules, namely, an agricultural plant contamination assessment model, an animal product contamination assessment model, and an ingestion dose assessment model has been developed based on Korean farming characteristics such as the growth characteristics of rice and stockbreeding. Results and Discussion: The application study showed that the present model can predict well the characteristics of the activity concentration for agricultural products and ingestion dose depending on the deposition date. Conclusion: The present model is very useful to predict the radioactivity concentration of agricultural foodstuffs and public ingestion dose as consequence of a nuclear accident. Consequently, it is expected to be used effectively as a module for the ingestion dose calculation of the Korean agricultural contamination management system as well as the Level 3 PSA code, which is currently being developed.

Preparation and Characterization of Poly(vinyl alcohol)/Poly(N-vinylpyrrolidone)/Glycerin/Chitosan Hydrogels by Radiation (방사선 가교에 의해 제조된 Poly(vinyl alcohol)/Poly(N-vinylpyrrolidone)/글리세린/키토산 하이드로겔의 제조 및 특성)

  • In this study, hydrogels from mixtures of poly (vinyl alcohol) (PVA)/poly(N-vinylpyrrolidone) (PVP)/glycerin/chitosan were prepared by γ-ray irradiation and the mechanical properties such as gelation, water absorptivity, and gel strength were examined to evaluate the applicability of these for wound dressing. Then PVA:PVP was weight ratio of 6 : 4, the concentration of chitosan was 0.3 wt%, the concentration of glycerin was in the range of 0∼5 wt%t. The solid concentration of PVA/PVP/glycerin/chitosan solution was 15 wt%. Gamma irradiation doses of 25, 35, 50, and 60 kGy were exposed to a mixture of PVA/PVP/glycerin/chitosan to evaluate the effect of irradiation dose. Gel content and gel strength increased as glycerin concentration in PVA/PVP/glycerin/chitosan decreased, and as irradiation dose increased. Swelling degree increased as glycerin concentration in PVA/PVP/glycerin/chitosan increased, and as irradiation dose decreased. The glycerin in PVA/PVP/glycerini/chitosan hydrogel prevented the transformation of shape. These hydrogel dressings had better curing effect than vaseline gauge.

Effects of Selenium and Metallothionein on the Toxicity of Cadmium in Rats (카드뮴 투여후 셀레늄과 Metallothionein의 독성효과)

  • To find out an effective way of treating the toxicity of cadmium, concentrations of $K^+,\;Ca^{2+}$ and c-AMP, and activities of GOT and GPT in blood of rats were determined at definite time intervals after administration of cadmium to rats. Cadmium caused more decrease of $Ca^{2+}$ concentration than $K^+$ concentration but increases of GOT and GPT activities and of c-AMP concentrations. The decreased concentration of $Ca^{2+}$ was not recovered by selenium or metallothionein. However, the increased activities of GOT and GPT, and the c-AMP concentration were down regulated remarkably by selenium but not by metallothionein. These results suggested that selenium might have the compensate effects concerning toxic index against cadmium such as increases of GOT and GPT activities, and of c-AMP concentration.

  • A deuterium oxide leakage accident occurred on October 4, 1999, at nuclear power plant in Korea. The concentration of tritium in air increased and 22 workers were exposed by tritium at that time. It is well known that tritium causes internal exposure. Therefore, we examined complete blood cell count, physical and biological dosimetry fur 13 workers among whole 22 workers to check the health effect and to evaluate the dose estimation of tritium exposure. The leukocyte count test, one of general blood test, was normal. The estimated doses were 0 - 4.44 mSv by physical dosimetry and 0-37 mGy by biological dosimetry. This dose does not exceed radiation dose limit, and the clinical symptoms of the exposed workers were not shown. The consistency between clinical sign and estimated dose means that physical and biological dosimetry were very useful especially in accident evaluation.

Radioprotective effects of delphinidin on normal human lung cells against proton beam exposure

  • BACKGROUND/OBJECTIVES: Exposure of the normal lung tissue around the cancerous tumor during radiotherapy causes serious side effects such as pneumonitis and pulmonary fibrosis. Radioprotectors used during cancer radiotherapy could protect the patient from side effects induced by radiation injury of the normal tissue. Delphinidin has strong antioxidant properties, and it works as the driving force of a radioprotective effect by scavenging radiation-induced reactive oxygen species (ROS). However, no studies have been conducted on the radioprotective effect of delphinidin against high linear energy transfer radiation. Therefore, this study was undertaken to evaluate the radioprotective effects of delphinidin on human lung cells against a proton beam. MATERIALS/METHODS: Normal human lung cells (HEL 299 cells) were used for in vitro experiments. The 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay assessed the cytotoxicity of delphinidin and cell viability. The expression of radiation induced cellular ROS was measured by the 2'-7'-dicholordihydrofluorescein diacetate assay. Superoxide dismutase activity assay and catalase activity assay were used for evaluating the activity of corresponding enzymes. In addition, radioprotective effects on DNA damage-induced cellular apoptosis were evaluated by Western blot assay. RESULTS: Experimental analysis, including cell survival assay, MTT assay, and Western blot assay, revealed the radioprotective effects of delphinidin. These include restoring the activities of antioxidant enzymes of damaged cells, increase in the levels of pro-survival protein, and decrease of pro-apoptosis proteins. The results from different experiments were compatible with each to provide a substantial conclusion. CONCLUSION: Low concentration ($2.5{\mu}M/mL$) of delphinidin administration prior to radiation exposure was radioprotective against a low dose of proton beam exposure. Hence, delphinidin is a promising shielding agent against radiation, protecting the normal tissues around a cancerous tumor, which are unintentionally exposed to low doses of radiation during proton therapy.

  • Electrospun nanofibers prepared with synthetic biodegradable polymer have some limitations in regulating adhesion, proliferation, and spreading of cells because of their surface hydrophobicity and absence of cell-interaction. In this study, we functionalized the electrospun poly(L-lactide-co-${\varepsilon}$-caprolactone) (PLCL) nanofibers with acrylic acid (AAc) to modulate their surface hydrophilicity using electron-beam irradiation method and then measured grafting ratio of AAc, water contact angle, and ATR-FTIR of AAc-grafted nanofibers. A grafting ratio of AAc on the nanofibers was increased as irradiation dose and AAc concentration were increased. AAc-grafted nanofibers also have higher wettability than non-modified nanofibers. In conclusion, those surface-modified nanofibers may be an essential candidate to regulate cell attachment in tissue engineering applications.

Sensory Quality Improvement of Gamma-irradiated Kimchi after Addition of Paprika Oleoresin and Artificial Kimchi Flavor (파프리카색소와 김치향 첨가 후 감마선 조사된 김치의 관능적 품질 개선)

  • Paprika oleoresin (PO) and artificial Kimchi flavor (AKF) were added to Kimchi to improve the sensory qualities deteriorated by gamma irradiation in terms of color and flavor. Optimum concentration of both PO and AKF resulting from the sensory evaluation was 0.2%. The redness and capsanthin contents of Kimchi decreased by gamma irradiation at 25 kGy during storage at $35^{\circ}C$. However, the redness and capsanthin contents of gamma-irradiated Kimchi was increased by the addition of 0.2% PO, and maintained during the storage at $35^{\circ}C$ for 30 days. The result from the sensory evaluation indicated that sensory qualities of gamma-irradiated Kimchi were effectively improved by the addition of PO and AKF during storage at $35^{\circ}C$. Therefore, the combined treatment of additives (PO and AKF) and gamma irradiation can be considered as an effective method to improve the redness and sensory qualities of Kimchi sterilized by high-dose gamma irradiation.

  • Monodispersed colloidaly yttria(Y2O3) can be used in a variety of applications such as phosphors. IR transparent materials and fine ceramics. For preparing monodispersed yttria homogeneous precipitation has been regarded as a fovorable method that is monodispersed yttria can be obtained through calcining monodispersed colloidal compound of yttrium (eg:Y(OH)CO3.nH2O)which can be prepared by homogeneous precipitation with urea. It is however still required to find out the quantitative effects of important variables of precipitation such as concentration of yttrium and urea reaction temperature and initial pH of reactant even though homogeneous precipitation of Y3+ with urea has been studied extensively. Among the effects of these variables we investiga-ted 1) the effect of yttrium concentration on the shape and size of precipitate and the reation rate 2) range of yttrium concentration required to make monodispersed colloidal particles 3) the reason for limited concentra-tion range of yttrium and 4) the effect of ultrasonic radiation on the limited concentration.

Lack of Association between Fingernail Selenium and Thyroid Cancer Risk: A Case-Control Study in French Polynesia

  • Background: Numerous studies have suggested that selenium deficiency may be associated with an increased risk for several types of cancer, but few have focused on thyroid cancer. Materials and Methods: We examined the association between post-diagnostic fingernail selenium levels and differentiated thyroid cancer risk in a French Polynesian matched case-control study. Conditional logistic regression models were used to estimate odds ratios and 95% confidence intervals. Results: The median selenium concentration among controls was $0.76{\mu}g/g$. Significantly, we found no association between fingernail selenium levels and thyroid cancer risk after conditioning on year of birth and sex and additionally adjusting for date of birth (highest versus lowest quartile: odds-ratio=1.12, 95% confidence interval: 0.66-1.90; p-trend=0.30). After additional adjustment for other covariates, this association remained non-significant (p-trend=0.60). When restricting the analysis to thyroid cancer of 10 mm or more, selenium in nails was non-significantly positively linked to thyroid cancer risk (p-trend=0.09). Although no significant interaction was evidenced between iodine in nails and selenium in nails effect (p=0.70), a non-significant (p-trend =0.10) positive association between selenium and thyroid cancer risk was seen in patients with less than 3 ppm of iodine in nails. The highest fingernail selenium concentration in French Polynesia was in the Marquises Islands ($M=0.87{\mu}g/g$) and in the Tuamotu-Gambier Archipelago ($M=0.86{\mu}g/g$). Conclusions: Our results do not support, among individuals with sufficient levels of selenium, that greater long-term exposure to selenium may reduce thyroid cancer risk. Because these findings are based on post-diagnostic measures, studies with prediagnostic selenium are needed for corroboration.

Comparative Study on Human Risk by Ionizing Radiation and Pesticide as Biological Information about Environmental Disaster (환경재해에 관한 생물정보로서의 이온화 방사선과 살충제의 인체 위해성 비교 연구)

  • Environmental risk factors such as ionizing radiations, heavy metals, and pesticides can cause environmental disasters when they exist in excess. The increases in use of ionizing radiation and agricultural pesticide are somewhat related to the possibility of the disaster. The risk of radiation and pesticide was evaluated by means of the single cell gel electrophoresis (SCGE) assay on the human blood lymphocytes. The lymphocytes were irradiated with $0{\sim}2.0Gy$ of $^{60}Co$ gamma ray. Another groups of lymphocytes were exposed to various concentrations of parathion. Significantly increased tail moment, which was a marker of DNA strand breaks in SCGE assay, showed a clear dose- or concentration-response relationship. Parathion of a recommended concentration for agricultural use ($1mg {\ell}^{-1}$ ) has a strong cytotoxic effect on lymphocytes, which is equivalent to damage induced by 0.1 Gy of ${\gamma}$-ray. Furthermore, $2mg{\ell}^{-1}$ of parathion can give rise to DNA damage equivalent to that induced by 0.25 Gy at which the radiation-induced damage can start to develop into clinical symptoms. The comparative results of this study can provide an experimental basis and biological information for the prevention of environmental disaster.
