This study was carried to identify a correct species and asses genetic diversity within the same species of Trametes spp. preserved in Division of applied Microbiology The morphological and cultural characteristics of preserved strains were observed through microscope and investigated on PDA, respectively. Contaminated isolates showed different growth rates, morphology and color of hyphae. We have reconstructed the phylogenetic tree of a select group of Trametes spp. using nucleotide sequences of the internal transcribed spacer region(ITS) region. The phylogenetic tree was constructed by using the neighbor-joining method. PELF primers of 20-mer were used to assess genetic diversity of preserved isolates. Sequence analysis showed that five strains were different species and six strains were identified completely different nomenclature. According to the analysis of ITS sequences, the genus Trametes clustered into four distinct group, most of which correlated with species-groups identified by RAPD method. Seven isolates included TM 01 strain showed high similarity with Trametes versicolr, TM 07 and TM 10 high similarity with Trametes gibbosa, and TM 05 high similarity with Trametes elegans. But isolates collected in the United States was identified as T. junipericola. T. gibbosa and T. versicolor by RAPD analysis of genetic polymorphisms showed a very different band patterns and these strains showed different band patterns on areas. As the result of RAPD and ITS region sequences analysis for preserved isolates, it seems likely that 11 isolates of Trametes spp. may be need to reclassify or eliminate from preserved catalogue.

Bangpungtongseong-san(BPTSS, Fangfengtongsheng-san in Chineses) is a traditional herbal formula comprising 18 medicinal herbs. In the present study, we performed the simultaneous analysis for four compounds of BPTSS and examined anti-allergic effects in human keratinocytes and mouse splenocytes. The column for separation of four compounds was used Luna C18 column and maintained at 40℃. The mobile phase for gradient elution consisted of two solvent systems. The analysis was carried out at a flow rate of 1.0 mL/min with PDA detection at 254 and 280 nm. To evaluate production and expression of Th2 chemokines, ELISA and RT-PCR were conducted in tumor necrosis factor (TNF)-α and interferon (IFN)-γ-stimulated HaCaT cells with or without BPTSS or silymarin, a positive control for skin inflammation. To measure Th2 cytokines, primary mouse splenocytes were treated with BPTSS and performed ELISA for interleukin (IL)-4, 5, 13. Calibration curves were acquired with r2>0.9999. The contents of geniposide, liquiritin, baicalin, and glycyrrhizin in BPTSS were 5.06 ㎎/g, 7.33 ㎎/g, 27.56 ㎎/g, and 7.81 ㎎/g, respectively. BPTSS reduced TARC and RANTES production and mRNA expression in TNF-α and IFN-γ-treated HaCaT cells. BPTSS inhibited IL-4, 5, and 13 production in mouse splenocytes. Our data will be a helpful information to upgrade quality control and anti-allergic effects of BPTSS.
During the year 2014, black locust (Robinia pseudoacacia L.) had been observed with dark brown spots on the leaves at Andong, Cheongsong, Mungyeong in Korea. Symptoms initially appeared as small, black lesions on the leaves, and sometimes, the leaves become yellow and ultimately leads to fall off the leaves. The pathogenic fungus grown in potato dextrose agar was white or sometime gray with mycelia in tufts and from which numerous conidia were produced. The conidia were straight and fusiform in shape and measured $8.3-17.2{\times}2.5-4.1{\mu}m$. Internal transcribed spacer (ITS) rRNA sequence analysis for sequence similarity of the ITS region revealed 100% identity with nucleotide sequences for Colletotrichum acutatum. The morphological characteristics, pathogenicity and molecular data have been confirmed that the symptomatic pathogen was C. acutatum. This is the first report of anthracnose caused by C. acutatum on black locust in Korea.

Endophytes provide multifarious benefits such as promotion of plant growth and yield, suppression of phyto-pathogens, phosphate solubilising and fixation nitrogen. A study has been carried out to explore growth promotion and antifungal activities of endophytes of mulberry (Morus spp.). Endophytic bacteria were isolated from mulberry plants and studied their cultural, morphological characters, growth promotion as well as their antifungal activity against Rhizoctonia bataticola and Fusarium oxysporum , two mulberry root rot associated pathogens. Except two isolates, all bacteria were colourless and the colony size of eight isolates was small. The margin of five isolates was irregular and the consistency of three isolates was creamy, six isolates was slimy and one was mucoid. Texture of seven isolates was convex and others were flat. Eight isolates were gram positive and the rest Gram negative, five were cocci and others were bacilli (rod shaped). Four isolates were motile and all were catalase positive and only three isolates were oxidase positive. Spore staining was positive only for two isolates. The growth promotion study showed that there was significant difference in root length and seedling length. The antagonistic effect of the bacterial isolates was tested against R. bataticola showed significant (p <0.05) influence of the bacteria, days after inoculation and their interaction on the inhibition of fungal growth. The isolate En-7 completely inhibited the fungus followed by En-5 (66.67%). The bacterial isolates significantly (p <0.05) inhibited growth of F. oxysporum in PDA. The mean inhibition was higher (70.45%) in case of En-7 followed by En-8 (68.65%) and En-10 (66.44%). The study reveals that some endophytic bacteria associated with mulberry have growth promotion and antifungal activity and could be explored for promotion of mulberry growth and managing root rot disease.
With an explosive growth of internet and a change of consumer's demand, Post PC is emerging as a substitute for the existing desktop computer and laptop computer. Post PC is a new computer which is portable with wireless networking, cheaper than the existing PCs and specialized in the specific function that consumers want. Furthermore, the shape and the ability of Post PC are dependent on its specific function. The rudimentary Post PCs such as PDA, web-pad and smart-phone are already usable. And in the future, owing to the progress of technologies, the heterogeneity of relate companies and the low entry barrier, various Post PCs will be developed. In this paper, we analyze consumers' willingness-to-pay for Post PC with the attributes such as the portability, the speed of CPU, the data input method (using key board, or notepad etc), the size of monitor and price. To estimating consumers' willingness-to-pay for the attributes of Post PC, we apply a contingent ranking method, which makes the respondents rank hypothetical Post the portability, the speed of CPU, the data input method (using key board, or notepad etc),the size of monitor and price. We had screened the Monascus strain capable of producing monacolin K dominantly among 29 Monascus strains. Red yeast rice was prepared by culturing each Monascus sp. with 200 g of steamed rice (12$0^{\circ}C$, 20 min) at 3$0^{\circ}C$ for 10 days and drying at 8$0^{\circ}C$ for 20 min. As a result, red yeast rice cultured by M. purpureus ATCC16457, M. purpureu IFO 32316, M. purpureus IFO 32228, M. kaoliang ATCC 46595 and M. kaoliang ATCC 46596 produced lots of red pigment and monacolin K. An unidentified Monascus sp. showed the highest productivityof red pigment and monacolin K among 29 Monascus strains. Its production of red pigment and monacolin K was 1.3∼39 times and 2.4∼8 times higher than other strains, respectively. Although the morphological characteristics of unidentified Monascus strain were a little different from the typical M. purpureus, it was identified as M. purpureus CBS 281.34 from the result of sequencing of ITS (Internal transcribed spacer) and 28S ribosomal RNA (partial).

Antibacterial activities of 33 antibiotics against Pseudomonas tolaasii causing the brown blotch disease on the edible mushroom Pleurotus ostreatus, were tested in vitro for the control of the disease. Tetracyclin, kanamycin, kasugamycin, and streptomycin showed strong antibacterial activity against P. tolaasii, having the minimal inhibitory concentration of 10, 10, 100 and 200ppm, respectively. These antibiotics showed similar control value of 72.9, 71.2, 68.1 and 62.7%, respectively when applied on the artificially infected mushroom beds. Mushroom yields in the tetracycline treated boxes were increased about 31.8% comparing to the control ones. Mycelial growth of P. ostreatus on the PDA supplemented with streptomycin and kanamycin were not affected, but were inhibited 10~20% and 40% with tetracyclin and kasugamycin treatment, respectively.
A stem and root rot disease of kalanchoe (Kalanchoe sp.) which is a succulent plant that provides consumers with a durable flowering pot was found in Koyang, Kyounggi province, Korea in May 1998. We found that stems and roots of potted kalanchoe had dark brown spots at the soil level. The causal organism was identified as Pjytophtora nicotianae on the basis of mycological characteristics. The fungus produced markedly papillate, ovoid to spherical sporangia, and abundant chlamydospores. Sporangia were 20∼48$\times$24∼64㎛(avg. 35.0$\times$47.3㎛) in size, and optimum temperature for the mycelial growth of the isolate was 30$\^{C}$. The fungus showed relatively different pathogenicity to 14 kalanchoe cultivars including K. blossfelana cv. Florus. cultivars florsu, calypso, Maya, and Redsing were susceptible to thedisease in root dip inoculation. This is the first report demonstrating the stem rot on kalanchoe caused by P. nicotianae in Korea.

Food products selected based on their annual sales and international acrylamide research data were analyzed for quantitation of acrylamide. Samples including raw food, substitute meal, snack, drink, and sauce products were analyzed by LC/MS/MS methods adopted by PDA. Upon comparison, concentrations of acrylamide in these products were similar to those analyzed in other countries.
A Rhizopus soft rot caused by Rhizopus stolonifer occurred on Momordica charantia at Daesan-myon, Chang-won city, Gyeongnam province from 2003 to 2005. The disease usually started from wound on fruit with water-soaking lesions. The lesion rapidly expanded and softened. The fungal mycelia grew vigorously and sporangia, sporangiophores, and stolons were formed on surface of the fruit. Spoyangiophores were $15\~30{\mu}m$ in width. Sporangia were globose or hemispheric and $90\~180{\mu}m$ in size. The color of sporangia was white and cottony at first and turned to brownish black with many spores at maturity. Columella were hemispheric and $80\~150{\mu}m$ in size. Sporangiospores were irregular round or oval, brownish-black streaked and $7\~18{\times}6\~12{\mu}m$ in size. The optimum temperature for mycelial growth of the fungus on PDA was $25^{\circ}C$. On the basis of mycological characteristics and pathogenicity to host plants, the causal fungus was identified as Rhizopus stolonifer (Ehrenberg ex. Fr,) Lind. This is the first report of Rhizopus soft rot on M. charantia caused by R. stolonifer in Korea.
