Inhibition of Quorum Sensing Regulated Virulence Factors and Biofilm Formation by Eucalyptus globulus against Multidrug-Resistant Pseudomonas aeruginosa

  • Objectives: The quorum-sensing-inhibitory and anti-biofilm activities of the methanol extract of E. globulus leaves were determined against clinically isolated multidrug-resistant Pseudomonas aeruginosa. Methods: The preliminary anti-quorum-sensing (AQS) activity of eucalyptus was investigated against a biosensor strain Chromobacterium violaceum ATCC 12472 (CV12472) by using the agar well diffusion method. The effect of sub-minimum inhibitory concentrations (sub-MICs) of the methanol extract of eucalyptus on different quorum-sensing-regulated virulence factors, such as swarming motility, pyocyanin pigment, exopolysaccharide (EPS), and biofilm formation, against clinical isolates (CIs 2, 3, and 4) and reference PA01 of Pseudomonas aeruginosa were determined using the swarm diameter (mm)-measurement method, chloroform extraction method, phenol (5%)-sulphuric acid (concentrated) method, and the microtiter plate assay respectively, and the inhibition (%) in formation were calculated. Results: The preliminary AQS activity (violacein pigment inhibition) of eucalyptus was confirmed against Chromobacterium violaceum ATCC 12472 (CV12472). The eucalyptus extract also showed concentration-dependent inhibition (%) of swarming motility, pyocyanin pigment, EPS, and biofilm formation in different CIs and PA01 of P. aeruginosa. Conclusion: Our results revealed the effectiveness of the E. globulus extract for the regulation of quorum-sensing-dependent virulence factors and biofilm formation at a reduced dose (sub-MICs) and suggest that E. globulus may be a therapeutic agent for curing and controlling bacterial infection and thereby reducing the possibility of resistance development in pathogenic strains.

인돌의 항균 효과에 의한 Pantoea agglomerans의 바이오필름 생성 억제 조절에 관한 연구 (Study on the Control of Biofilm Formation Inhibition on Pantoea agglomerans by Anti-bacterial Effect of Indole)

  • In this study, we investigated the effects of indole on biofilm formation inhibition in Pantoea agglomerans (P. agglomerans). In the biofilm growth assay, indole inhibited biofilm formation across all the growth time. Depending on biofilm growth stage, indole exhibited biofilm inhibition and anti-bacterial effects on planktonic cells. Through the analysis of the proportion rate between biofilm and Colony Forming Units (CFU) and inhibition rate of indole, we confirmed that depending on the biofilm stage of P. agglomerans, indole treatment timing was more important than the treatment duration. By comparing gene expression rates through rt-qPCR P.agglomerans affected by indole was found to significantly change quorum sensing (pagI/R) and indole transportation (bssS) gene expressions. Throughout all, indole exhibited both antimicrobial and anti-biofilm effects on P. agglomerans. In addition, we confirmed the anti-biofilm effects of indole on mature biofilm. In conclusion, indole as a signal molecule, can exhibit anti-biofilm effects through bacterial quorum sensing inhibition and indole affects. Therefore, indole can regulate biofilm bacteria especially gram-negative opportunistic pathogens.

  • A new series comprising 7 analogs of N-(sulfanyl ethanoyl)-L-HSL derivatives, 2 analogs of N-(fluoroalkanoyl)-$_L$-HSL derivatives, N-(fluorosulfonyl)-L-HSL, and 2,2-dimethyl butanoyl HSL were synthesized using a solid-phase organic synthesis method. Each of the 11 synthesized compounds was analyzed using NMR and mass spectroscopies, and molecular modeling studies of the 11 ligands were performed using SYBYL packages. Thereafter, a bacterial test was designed to identify their quorum-sensing inhibition activity and antifouling efficacy. Most of the synthesized compounds were found to be effective as quorum-sensing antagonists, where antagonist screening revealed that 10 among the 11 synthesized ligands were able to antagonize the quorum sensing of A. tumefaciens.

박테리아의 Quorum Sensing 및 생물막 형성 억제를 위한 Quorum Quenching 연구 동향 (Bacterial Quorum Sensing and Quorum Quenching for the Inhibition of Biofilm Formation)

  • 본 총설은 N-acyl-homoserine lactone (AHL)에 기반한 quorum sensing(QS)을 비롯한 다양한 QS 시스템 및 생물막 형성과의 관련성에 대한 연구 동향을 정리하였다. 또한 anti-QS으로서 quorum quenching 전략을 이용한 생물막 억제 연구 동향에 대해 중점적으로 서술하였다. 세균의 독특한 신호전달 체계인 QS는 AHL과 같은 특정한 신호분자의 농도에 의해 세균의 집단적 행동 양식이 결정되는 세포밀도-의존성 유전자 발현 조절 메커니즘이다. QS 시스템은 미생물의 부착 및 생물막 형성에 있어 중요한 역할을 한다. AI-1이나 AI-2에 의한 QS는 생물막 형성 과정에 필요한 세포외 다당류, 단백질, 세포 외 DNA 등 주요한 구성 성분 등의 생산뿐만 아니라, 세균의 운동성 조절, 부착, 생물막 해체 과정까지도 조절하는 기능을 한다. 일부 세균의 경우 QS시스템 이외에도 second messenger로 알려진 c-di-GMP에 의한 signaling이 QS와 서로 연결되어 생물막 형성이나 병독성과 같은 타깃들을 함께 조절한다. 생물막은 병원성 세균에 의한 감염 시 여러 가지 병독성 가운데 가장 중요한 요소 중 하나이기 때문에, 생물막 형성을 조절하는 QS를 차단하기 위한 다양한 anti-quorum sensing 전략이 연구되고 있다. Anti-QS 접근 방식은 의학적 이용뿐만 아니라 물에 노출되어있는 MBR을 비롯한 많은 산업적 장치 등에서 생물막 형성으로 인한 손상 및 오염을 방지하기 위해 쓰일 수 있다. Anti-QS 전략 중 신호분자인 AHL을 무력화 시키는 quorum quenching 효소(AHL-lactonase, AHL-acylase, oxidoreductas)를 이용하여 생물막 형성을 억제할 수 있으며, 막을 이용한 수처리 공정에서 막에 발생하는 biofouling을 완화시킬 수 있는 새로운 anti-fouling 처리 기술로서 이러한 QQ 효소의적용 가능성을 보여 주고 있다.

  • Recent studies have revealed that bacterial biofilm production by the gram-negative bacteria is regulated by the quorum sensing signaling molecules, AHLs (N-acyl homoserine lactones). This suggests that inhibiting the AHLs could enhance the effects of antibacterial agents. Halogenated furanones purified from the red algae Delisea pulchra have been known to decrease quorum sensing responses by competitive inhibition of the AHLs. (omitted)

  • 재조합 Agrobacterium tumefaciens균주를 사용한 Liquid Culture Assay는 quorum sensing autoinducer를 검색하는 생물학적 분석 방법으로 개발되었다. 그러나 이 시스템에 사용되는 해양천연물 시료들이 일반적으로 낮은 수용액에 대한 용해도를 갖기 때문에 활성검색에 걸림돌이 되고 있다. 시료의 용해도는 유기용매 혹은 계면활성제의 첨가로 증가될 수 있으나, 유기용매나 계면활성제 자체가 검색결과의 정확한 해석을 방해할 수 있으므로 적절한 조건의 확립이 매우 중요하다. Methanol, ethanol, 1-propanol, DMSO와 DMF를 0~10% 농도 범위에서 재조합 A. tumefaciens균주에 대한 세포 성장에 미치는 영향과 ${\beta}$-galactosidase activity에 미치는 영향을 살펴본 결과 methanol 2%이하의 농도가 가장 적합하였으며, 계면활성제의 경우, Tween 20, Tween 80보다는 Triton X-100이 약 0.05% 농도에서 세포내로의 활성물질의 전달효율을 높이는데 효과적이었다.

Inhibitory Effect of Pentose on Biofilm Formation by Oral Bacteria

  • A number of bacterial species coexist in oral cavities as a biofilm rather than a planktonic arrangement. By forming an oral biofilm with quorum sensing properties, microorganisms can develop a higher pathogenic potential and stronger resistance to the host immune system and antibiotics. Hence, the inhibition of biofilm formation has become a major research issue for the future prevention and treatment of oral diseases. In this study, we investigated the effects of pentose on biofilm formation and phenotypic changes using wild type oral bacteria obtained from healthy human saliva. D-ribose and D-arabinose were found to inhibit biofilm formation, but have no effects on the growth of each oral bacterium tested. Pentoses may thus be good candidate biofilm inhibitors without growth-inhibition activity and be employed for the future prevention or treatment of oral diseases.

천연물로부터 Quorum Sensing 저해제의 탐색 (Detection of a Quorum-Sensing Inhibitor from the Natural Products)

  • 인간이 서로 간의 의사소통을 위해 언어를 사용하듯이, 세균의 경우도 외부 환경 변화를 신속히 감지하여 서로 효과적으로 대응하기 위해서 주변 세포들과 소통할 수 있는 세균만의 독특한 화학적 언어를 사용하는 것으로 알려져 있다. 특히, 일정 세포 농도에 도달했을 때 자체적으로 생산된 화학적 신호를 통해 개체 수를 인지하고 그에 따라 특정 유전자의 발현을 동시에 조절하는 quorum sensing (QS) 기작은 다양한 세균 종들에서 광범위하게 존재한다. 본 연구는 다양한 천연물 추출물들을 대상으로 QS 저해 활성을 확인하였는데 QS 지시균주인 Agrobacterium tumefaciens NT1과 화학적으로 합성한 QS autoinducers을 사용한 bioassay를 수행하였다. 그 결과 양배추, 파, 양파의 추출물들에서 QS 저해 활성을 확인하였고, recycling preparative HPLC (prep-HPLC)를 통한 정제 과정을 통해, 83분 지점의 peak에 해당하는 성분들이 공통으로 QS 저해 활성을 가지고 있음을 확인하였다. 따라서 그 QS 저해 성분을 QSI-83으로 지정하고 thin layer chromatography (TLC)를 통해 P. syringae pv. tabaci의 autoinducers 합성을 저해하는 활성을 가지고 있음을 확인하였다. 또한 열에 대한 안정성과 세균 생장에서의 영향을 조사하였는데, 그 결과 QSI-83은 열에 안정하며 세균의 생장에는 영향을 끼치지 않는 물질임을 확인하였다. 따라서 우리는 천연물로부터 분리된 새로운 성분이 QS 저해제로서 이용될 수 있음을 제안한다.

Thymol Rich Thymbra capitata Essential Oil Inhibits Quorum Sensing, Virulence and Biofilm Formation of Beta Lactamase Producing Pseudomonas aeruginosa

  • Infections with Pseudomonas aeruginosa are difficult to treat not only because it is often associated with multidrug-resistant infections but also it is able to form biofilm. The aim of this study was to evaluate the antibiofilm and anti-Quorum Sensing (QS) activities of Thymbra capitata essential oils (EOs) against Beta Lactamase (BL) producing P. aeruginosa and the reference strain P. aeruginosa 10145. GC/MS analysis showed that thymol (23.25%) is the most dominant compound in T. capitata EOs. The MICs of T. capitata EOs against P. aeruginosa (BL) and P. aeruginosa 10145 were 1.11%. At sub MIC (0.041, 0.014 and 0.0046%), the EOs of T. capitata remarkably inhibited the biofilm formation of both strains tested and complete inhibition of the biofilm formation was reported at 0.041%. The EOs of T. capitata were found to inhibit the swarming motility, aggregation ability and hydrophobic ability of P. aeruginosa (BL) and P. aeruginosa 10145. Interestingly, the EOs of T. capitata reduce the production of three secreted virulence factors that regulated by QS system including pyocyanin, rhamnolipids and LasA protease. The potent antibiofilm and anti-QS activities of T. capitata EOs can propose it as a new antibacterial agent to control pseudomonas infections.

Quorum Quenching Bacteria Isolated from the Sludge of a Wastewater Treatment Plant and Their Application for Controlling Biofilm Formation

  • Bacteria recognize changes in their population density by sensing the concentration of signal molecules, N-acyl-homoserine lactones (AHLs). AHL-mediated quorum sensing (QS) plays a key role in biofilm formation, so the interference of QS, referred to as quorum quenching (QQ), has received a great deal of attention. A QQ strategy can be applied to membrane bioreactors (MBRs) for advanced wastewater treatment to control biofouling. To isolate QQ bacteria that can inhibit biofilm formation, we isolated diverse AHL-degrading bacteria from a laboratory-scale MBR and sludge from real wastewater treatment plants. A total of 225 AHL-degrading bacteria were isolated from the sludge sample by enrichment culture. Afipia sp., Acinetobacter sp. and Streptococcus sp. strains produced the intracellular QQ enzyme, whereas Pseudomonas sp., Micrococcus sp. and Staphylococcus sp. produced the extracellular QQ enzyme. In case of Microbacterium sp. and Rhodococcus sp., AHL-degrading activities were detected in the whole-cell assay and Rhodococcus sp. showed AHL-degrading activity in cell-free lysate as well. There has been no report for AHL-degrading capability in the case of Streptococcus sp. and Afipia sp. strains. Finally, inhibition of biofilm formation by isolated QQ bacteria or enzymes was observed on glass slides and 96-well microtiter plates using crystal violet staining. QQ strains or enzymes not only inhibited initial biofilm development but also reduced established biofilms.
