Degradation of Sulfonamide Antibiotic Substances by Ozonation: An Experimental and Computational Approach (설폰아미드계 항생물질의 오존산화분해에 대한 계산화학적 해석 및 실험적 검증)

  • Concern has grown over a presence of micropollutants in natural water since sulfonamide antibiotic substances such as sulfamethazine, sulfamethoxazole, sulfathiazole have been frequently detected in Nakdong River, Korea. The current work investigates the degradation of the three sulfonamide substances by using quantum chemistry calculations of density functional theory (DFT) and experimental measurement techniques of Fourier transform infrared spectroscopy (FT-IR) and ultraviolet-visible spectrophotometer (UV-VIS). DFT calculations demonstrate that the lowest energy gap between the highest occupied molecular orbital (HOMO) and the lowest unoccupied molecular orbitals (LUMO) lies in sulfanilamide functional group of sulfonamide, implying that the sulfanilamide functional group would be the most active site for ozone oxidation. Also, UV-VIS spectra and FT-IR analysis reveal that 260 nm band originated from sulfanilamide group was absent after ozone oxidation, indicating that a functional group of amine (N-H) was removed from sulfanilamide. Both theoretical and experimental observations agree well with each other, demonstrating the DFT calculation tool can be an alternative tool for the prediction of chemical reactions in purification treatment processes.

$^{13}C$ NMR Analysis for the Characterization of Heme Electronic/Molecular Structure in Horse Myoglobin Cyanide (Myoglobin 시안 단백질에 포함된 Heme에 대한 전자 및 분자구조 규명을 위한 $^{13}C$ NMR분석)

  • The reverse detection heteronuclear multiple quantum coherence, HMQC study of metcyano complex of horse myoglobin(MbCN) has provided the complete assignment of hyperfine shifted resonances of heme carbons attached with proton(s). The application of HMQC experiment to the paramagnetic low-spin MbCN gives clear $^1H$ and $^{13}C$ coherences for the paramagnetic amino acid residues as well as heme side chains, and can be extended to the low-spin paramagnetic hemoprotein derivative for the assignment of natural abundance $^{13}C$ resonances. This assignment strategy can avoid possible ambiguities that may result from the sole utilization of $^1H$ nuclear Overhauser effect for the assignment of heme $^1H$ signals resonating in the diamagnetic region. The resulting 2,4-vinyl ${\alpha}$-carbons and 7-propionate ${\beta}$-carbon follow anomalous anti-Curie behavior, and are indicative of incoplanarity with heme plane. Magnetic/electronic asymmetry of heme induced by proximal histidine(His) makes spread that the hyperfine shifted heme carbon resonances over the range of 250 ppm at $25^{\circ}C$. These heme carbon resonances would be the much more sensitive probe than those of proton resonances in analyzing the nature of heme electronic structure of myoglobin.

Development of Wide-Band Compensation Film to Improve Viewing Angle of Vertical Alignment Liquid Crystal Display (수직배향 액정디스플레이의 시야각 향상을 위한 광대역 보상필름 개발)

  • To expand the viewing angle of vertical alignment liquid crystal display (VA-LCD), a wide-band compensation film has been newly developed. VA-LCD needs a retardation film with improved oblique viewing property. The retardation film for VA-LCD has been fabricated by solvent casting and subsequent drawing triacetylcellulose (TAC) containing optical additives. Here, optical birefringence and wavelength dispersion of the retardation film strongly depend on the chemical structures of optical additives. Utilizing quantum mechanical computation, the polarizing anisotropy of retardation film was calculated with respect to additives. It is also realized that the wavelength dispersion of polarizing anisotropy depends on the type and location of substituent to the selected main isomer. When two propionates are substituted to the meta position of the selected main isomer, the wavelength dispersion of polarizing anisotropy shows the most gentle slop, which is well matched with the experimental results. The most gentle slop of wavelength dispersion means that the viewing angle characteristics of LCD are less influenced according to the wavelength of light source of LCD, and it's possible to make better image quality than the present level.

A topological metal at the surface of an ultrathin BiSb alloy film

  • Recently there has been growing interest in topological insulators or the quantum spin Hall (QSH) phase, which are insulating materials with bulk band gaps but have metallic edge states that are formed topologically and robust against any non-magnetic impurity [1]. In a three-dimensional material, the two-dimensional surface states correspond to the edge states (topological metal) and their intriguing nature in terms of electronic and spin structures have been experimentally observed in bulk Bi1-xSbx single crystals [2,3,4]. However, if we want to know the transport properties of these topological metals, high purity samples as well as very low temperature will be needed because of the contribution from bulk states or impurity effects. In a recent report, it was also shown that an intriguing coupling between the surface and bulk states will occur [5]. A simple solution to this bothersome problem is to prepare a topological metal on an ultrathin film, in which the surface-to-bulk ratio is drastically increased. Therefore in the present study, we have investigated if there is a method to make an ultrathin Bi1-xSbx film on a semiconductor substrate. From reflection high-energy electron diffraction observation, it was found that single crystal Bi1-xSbx films (0${\sim}30\;{\AA}A$ can be prepared on Si(111)-$7{\times}7$. The transport properties of such films were characterized by in situ monolithic micro four-point probes [6]. The temperature dependence of the resistivity for the x=0.1 samples was insulating when the film thickness was $240\;{\AA}A$. However, it became metallic as the thickness was reduced down to $30\;{\AA}A$, indicating surface-state dominant electrical conduction. Figure 1 shows the Fermi surface of $40\;{\AA}A$ thick Bi0.92Sb0.08 (a) and Bi0.84Sb0.16 (b) films mapped by angle-resolved photoemission spectroscopy. The basic features of the electronic structure of these surface states were shown to be the same as those found on bulk surfaces, meaning that topological metals can be prepared at the surface of an ultrathin film.   • The mechanisms of $CH_4$ uptake into and release from the ocean are not well understood due mainly to complexity of the biogeochemical cycle and to lack of regional-scale and/or process-scale observations in the marine boundary layers. Without complete understanding of oceanic mechanisms to control the carbon balance and cycles on a various spatial and temporal scales, however, it is difficult to predict future perturbation of oceanic carbon levels and its influence on the global and regional climates. High frequency, high precision continuous measurements for carbon isotopic compositions from dissolved $CH_4$ in the surface ocean and marine atmosphere can provide additional information about the flux pathways and production/consumption processes occurring in the boundary of two large reservoirs. This paper introduces recent advances on optical instruments for real time $CH_4$ isotope analysis to diagnose potential applications for in situ, continuous measurements of carbon isotopic composition of dissolved $CH_4$. Commercially available, three laser absorption spectrometers - quantum cascade laser spectroscopy (QCLAS), off-axis integrated cavity output spectrometer (OA-ICOS), and cavity ring-down spectrometer (CRDS) are discussed in comparison with the conventional isotope ratio mass spectrometry (IRMS). Details of functioning and performance of a CRDS isotope instrument for atmospheric ${\delta}^{13}C-CH_4$ are also given, showing its capability to detect localized methane emission sources.

Photosynthetic Characteristics of Benthic Microalgae Measured by HPLC and Diving Pulse Amplitude Modulated (PAM) Fluorometry on the Nakdong River Estuary of the Korean Peninsula (HPLC 및 Diving-PAM을 이용한 낙동강 하구 저서미세조류의 광합성 특성)

  • Daemadeung, located in the estuary of the Nakdong River, is formed by sand dunes and possesses well-developed intertidal flats. This study aimed to investigate the habitat of benthic microalgae, photosynthetic pigments, and photosynthetic efficiency in the intertidal flats of Daemadeung from January to December 2011. The inorganic nitrogen content in the sediment pore water was primarily composed of ammonium, while nitrate + nitrite was dominant in the upper layer water. The concentration of chlorophyll a and fucoxanthin in the sediment surface was significantly higher than the mean of all the sediment layer. The average Fv/Fm of benthic microalgae during the entire survey period was 0.52±0.03, with the highest value (0.61±0.08) observed in February. The rETRmax showed a seasonal trend, being high from spring to early autumn (April to October) and low from winter to early spring (January to March, November, December), with the highest value (153.05±2.30 µmol electrons m-2 s-1) in July and the lowest (38.49±5.17 µmol electrons m-2 s-1) in January. The average Fv/Fm of diurnal microalgae was 0.48±0.03, with the highest value (0.61±0.08) observed at noon. The rETRmax showed a highest peak at noon (54.24±11.35 µmol electrons m-2 s-1) and reached its lowest point at 16:00 (26.17±4.75 µmol electrons m-2 s-1). These findings suggest that the productivity of benthic microalgae varies significantly depending on the survey time and sediment depth. Therefore, to quantify the productivity of benthic microalgae using Diving-PAM, surveys should be conducted based on tidal conditions, and simultaneous pigment analysis of sediment layers should also be performed.

Shading Effects on the Growth and Physiological Characteristics of Osmanthus insularis Seedlings, a Rare Species (희귀 식물 박달목서 유묘의 생장 및 생리적 특성에 대한 차광 효과)

  • This study was conducted to determine the optimal light conditions for the in situ and ex situ conservation and restoration of Osmanthus insularis, a rare plant species in South Korea. Evaluations included the growth performance, leaf morphological features, photosynthetic characteristics, and photosynthetic pigment contents of seedlings grown from April to November under different light conditions (100%, 55%, 20%, and 10% relative light intensity). The shoot lengths and root collar diameters did not differ significantly with relative light intensity. The dry weights of leaves, stems, and roots and the leaf number were highest at 55% relative light intensity. The leaf shape showed morphological acclimation to light intensity, with leaf area decreasing and thickness increasing as the relative light intensity increased. Several leaf parameters, including photosynthetic rate and stomatal conductance at light saturation point, net apparent quantum yield, and dark respiration, as well as chlorophyll a, chlorophyll b, and carotenoid contents, were all highest at 55% relative light intensity. Under full light conditions, the leaves were the smallest and thickest, but the chlorophyll content was lower than at 55% relative light intensity, resulting in lower photosynthetic ability. Plants grown at 10% and 20% relative light intensity showed lower chlorophyll a, chlorophyll b, and carotenoid contents, as well as decreased photosynthetic and dark respiration rates. In conclusion, O. insularis seedlings exhibited morphological adaptations in response to light intensity; however, no physiological responses indicating enhanced photosynthetic efficiency in shade were evident. The most favorable light condition for vigorous photosynthesis and maximum biomass production in O. insularis seedlings appeared to be 55% relative light intensity. Therefore, shading to approximately 55% of full light is suggested for the growth of O. insularis seedlings.

Photosynthesis and Formation of UV-absorbing Substances in Antarctic Macroalgae Under Different Levels of UV-B Radiation (중파자외선에 대한 남극산 해조류의 광합성 및 자외선 흡수물질 형성 반응)

  • Effects of artificial and solar W-B radiation on five rhodophytes (Curdiea racovitzae, Gigaytina skottsbergii, Mazzaella obovata, Myriogramme manginii, Palmaria decipiens) from Antarctica have been investigated using PAM fluorescence in laboratory and in the field. Laboratory studies showed that there were significant differences in the UV sensitivity between different species, and that the differences appeared to be correlated with the depth of collection of the specimens. It was apparent from the observations that the samples such as M. manginii and P. decipiens collected from 20-30 m depths were move sensitive to W-B radiation compared with those collected from shallower depths, The present study confirmed that an acclimation to the surrounding light regime could be an important factor to determine the UV-sensitivity of a species or individuals and that PAM measurements are rapid and non-destructive methods to evaluate UV influences. From field studies on M. manginii and P. decipiens it was observed that both plants exhibited changes in the effective quantum yield, with the minimum values nt noon followed by n recovery in the evening. Photoinhibition occurred in these species could therefore be accounted for by so- called dynamic photoinhibition. It seems likely that this protective mechanism may contribute to survival of the species in shallow water where they may encounter intense solar radiation. The presence or absence of the W- B component under solar radiation differently affected the photosynthetic recovery process, and the rate of recovery was much stoney in UV- present than in W- absent conditions. Functional role of W- B appears to delay the recovery of photosynthesis in the studied macroalgae. Differential sensitivity to UV-B recognised between M. manginii and P. decipiens seemed to correspond well with the amount of UV-absorbing substances (UVAS) contained in the respective species. Higher tolerance to solar radiation by the latter species may be due to the higher amount of UVAS. There were variations of UVAS concentrations in algal thalli depending on the season and depth of collection.

The characteristic of InGaN/GaN MQW LED by different diameter in selective area growth method (선택성장영역 크기에 따른 InGaN/GaN 다중양자우물 청색 MOCVD-발광다이오드 소자의 특성)

  • In general, the fabrications of the LEDs with mesa structure are performed grown by MOCVD method. In order to etch and separate each chips, the LEDs are passed the RIE and scribing processes. The RIE process using plasma dry etching occur some problems such as defects, dislocations and the formation of dangling bond in surface result in decline of device characteristic. The SAG method has attracted considerable interest for the growth of high quality GaN epi layer on the sapphire substrate. In this paper, the SAG method was introduced for simplification and fabrication of the high quality epi layer. And we report that the size of selective area do not affect the characteristics of original LED. The diameter of SAG circle patterns were choose as 2500, 1000, 350, and 200 ${\mu}m$. The SAG-LEDs were measured to obtain the device characteristics using by SEM, EL and I-V. The main emission peaks of 2500, 1000, 350, and 200 ${\mu}m$ were 485, 480, 450, and 445 nm respectively. The chips of 350, 200 ${\mu}m$ diameter were observed non-uniform surface and resistance was higher than original LED, however, the chips of 2500, 1000 ${\mu}m$ diameter had uniform surface and current-voltage characteristics were better than small sizes. Therefore, we suggest that the suitable diameter which do not affect the characteristic of original LED is more than 1000 ${\mu}m$.

Plant Regeneration and Protein Analysis from Cadmium Resistant Callus of Tobacco (Nicotiana tabacum cv. BY4) (담배 (Nicotiana tabacum cv. BY4)카드뮴 저항성 캘러스로부터 식물체 재생과 단백질 분석)

  • Calli were induced from diploid and haploid tobacco after 4 weeks and maintained on MS medium with combination of 2.0 mg/L 2,4-D,0.1 mg/L BAP and 2.0 mg/L kinetin. Suspension cells were screened through 65 $\mu$m-nylon mesh and 100 $\mu$m-mesh, then they were smeared on selection medium combined with cadmium and PFP by using the low melting agarose of 0.8%. After 30days smeared cultures of the medium the cell was treated with 500 $\mu$M and 1000 $\mu$M to select the resistant cell line were selected. Plant regeneration was induced from the selected cell lines on medium with 0.5, 1.5, 2.0 mg/L BAP and on media with combination of auxin and BAP under 500 $\mu$M and 1000 $\mu$M cadmium. At this time, plant regeneration was achived on cadmium free medium. In case of haploid, occurred from the cell line which is selected in medium with cadmium and PFP. In case of diploid regeneration occurred is in the medium with cadmium alone. The plantlet regenerated from cadmium resistant calli grew well in cadmium 500 $\mu$M. Protein pattern of leaf, root, stem of regenerated plants was analyzed. The quantum was 6.5188 ug/mg.fr.wt in the leaf of plant, 5.3611 ug/mg.fr.wt in the stem, 3.0213 ug/mg.fr.wt in the root. On the other hand, 5.9652 ug/mg.fr.wt. in the leaf of control, 3.5974 ug/mg.fr.wt in the stem of the control, 4.3766 ug/mg.fr.wt. in the root of the control. The one dimension bends regenerated from cadmium resistant calli resistant to cadmium in leaf were 49 involving 198.7KD etc. Disappeared were 4 involving 160.5KD etc, The protein bends were combinized were 3 involving 83.4KD etc. The bends resistant to cadmium stress in stem were 41 involving 4.3KD etc. Disappeared were 5 involving 114.8KD etc. The protein bends combinized were 6 involving 128.7KD etc. The bends which had the resistance to cadmium stress in root is 27 in volving 166,9KD etc. The bends which disappeared were 198.7KD etc. There were 5 involving 83.4KD etc.
