Purification of Si using Catalytic CVD

  • Silicon is commercially prepared by the reaction of high-purity silica with wood, charcoal, and coal, in an electric arc furnace using carbon electrodes, so called the metallurgical refining process, which produces ~98% pure Si (MG-Si). This can be further purified to solar grade silicon (SoG-Si) by various techniques. The most problematic impurity elements are B and P because of their high segregation coefficients. In this study, we explored the possibility of the using Cat-CVD for Si purification. The existing hot-wire CVD was modified to accommodate the catalyzer and the heating source. Mo boat (1.5 cm ${\times}$ 1 cm ${\times}$ 0.2 cm) was used as a heating source. Commercially available Si was purchased from Nilaco corporation (~99% pure). This powder was kept in the Mo-boat and heated to the purification temperature. In addition to the purification by cat-CVD technique, other methods such as thermal CVD, plasma enhanced CVD, vacuum annealing was also tried. It is found that the impurities are reduced to a great extent when treated with cat-CVD method.

산소용해수와 미생물제재를 이용한 호소 및 폐쇄수역의 정화기술 (Purification Technology in Closed Water like a Reservoir and Pond using Oxygen Solubilized Device and Standardized Microorganism Culture System)

  • The oxygen solubilized device(O.S.D) and standardized microorganism culture system is more efficient than physical and chemical purification techniques in closed water. This study was to determine how the O.S.D and standardized culture system is efficient in purification capacity in closed water based on the lab scale and pilot plant. In the batch test, inducing the quantitative results from pilot plant operation condition, removal efficiency of COD and TN were about 48.3% and 35% respectively, while SS and chlorophyll-a were 94.9% and 68.7%. The pilot plant results showed that suspended solid(SS) and chlorophyll-a removal efficiency were 60% and 59% respectively, due to coagulation characteristics by standardized culture. Total nitrogen(TN) and total phosphorus(TP)showed good effect for the purification of target pond water quality from field data. Additionally, released velocity was determined in control condition of $5.31mgPO{_4}^{3-}{\cdot}m^{-2}{\cdot}day^{-1}$ and $2486.8mgCOD{\cdot}m^{-2}{\cdot}day^{-1}$. Otherwise, phosphate and COD reflux in the aeration and microorganism condition was showed $-9.95mgPO{_4}^{3-}{\cdot}m^{-2}{\cdot}day^{-1}$ and $-397.88mgCOD{\cdot}m^{-2}{\cdot}day^{-1}$. This technology is the most effective not only removal of SS and chlorophyll-a but also control of phosphate and COD release which is very important phenomena in evaluating water quality in closed water like a reservoir and pond.

  • Streptomyces somaliensis가 생산하는 phospholipase D (PLD)를 정제하기 위하여 펩티드 항체 결합 면역 친화 크로마토그래피용 칼럼을 개발하였다. 단백질 구조 예측 프로그램과 Streptomyces PLD X-선 결정구조를 참조하여, S. somaliensis PLD의 1차 구조로부터 항원특성이 높고. 표면에 위치하는 것으로 예상된 5종류의 펩티드들을 epitope로 선정한 뒤, 이에 대한 항체로 면역친화 크로마토그래피용 칼럼을 제작하였다. 배양 농축액을 칼럼에 통과시켜 정제한 활성 분획을 SDS-PAGE 및 Western blot 결과, 칼럼 종류에 따라 순수한 PLD또는 35 kDa의 단백질 불순물만을 포함하는 PLD 정제 분획을 보여 면역친화 칼럼의 높은 항원결합 특이성을 보여주었다. 그러나 수용액상에서 PLD 자체의 구조적 불안정성 때문에 정제 후 PLD의 특이적 활성 및 정제 수율은 낮았다.

Microbial Activity of Gravel Intertidal Zone for Purification of Polluted Near Shore Water

  • Microbial activity of biofilm formed on the surface of gravels from intertidal zone was estimated using an aerobic respirometer system, and compared with that of suspended marine microorganisms contained in a near shore water. The maximum oxygen uptake rate of the suspended marine microorganisms was 0.15mg O$_2$/L/hr, indicating the potential of purification of polluted near shore water. For the gravels from the intertidal zone, the maximum uptake rate of oxygen was affected by the vertical positions, but their gross value was 0.77mg O$_2$/L/hr, which was around 5.1 times higher than the purification potential of polluted near shore water by the microorganisms contained in the near shore water. The nitrogen removed by the gravels from the intertidal zone and the marine microorganisms was about 1/20-1/39 times of the total consumption of oxygen, which was similar to that of the phosphate. The gravel intertidal zone contained lots of particulate organics, over than that in the near shore water, and this was confirmed from the large difference between total oxygen consumption and the removed soluble COD in the microbial activity test. This indicates that the gravel intertidal zone plays an important role in controlling the non-point source pollutants from land, as well as self-purification of polluted near shore water by trapping and degrading the particulate organics.

  • 나타마이신 배양액을 4$^{\circ}C$와 실온에서 14일 동안 보관하였을 때, 4$^{\circ}C$에서는 나타마이신의 활성이 80% 이상 유지되었으나 실온에서는 27%로 급격하게 감소하였다. 이러한 사실은 나타마이신의 손실을 최소화하기 위해서는 최단 시간 내에 배양액으로부터의 나타마이신을 회수할 필요성과 장기간 보관 시에는 4$^{\circ}C$이하에서 보관할 필요성을 제시하고 있다. 효율적인 나타마이신 정제과정을 개발하기 위해 배양액으로부터의 나타마이신의 추출 용매 및 적정 사용량에 대해 조사한 결과 2g의 나타마이신을 추출하는데 1 l의 methanol을 사용하는 것이 가장 효율적이었다. 확립된 나타마이신 추출에 필요한 methanol의 양과 Diaion HP-20을 이용한 column chromatography를 적용하여 4.2 g의 나타마이신이 함유된 1,800 ml의 배양액으로부터 2.9 g의 나타마이신을 획득하였다. 본 연구에서 개발된 정제과정을 통해 순도가 96.6%이고 회수율이 69.1%인 나타마이신을 얻을 수 있었다.

유채박 단백질의 추출 및 정제에 관한 연구 (Extraction and Purification of Rapeseed Protein)

  • 유채박 단백질의 효과적인 추출 및 정제공정을 확립하기 위하여 추출용매를 비교했으며, 1% SHMP 용매로 추출한 후 등전침전, 산세척, UF농축 처리에 의하여 정제한 단백질의 수율 및 glucosinolate와 phytate 잔존량을 조사하였다. 추출조건은 1% SHMP(pH 8.0) 용매가 가장 적당하였으며, glucosinolate 및 phytate 제거를 위한 정제효과 면에서는 2회 산세척시와 1회 산세척 및 UF(100K) 농축 처리한 공정이 가장 효과적 이었다. 수율과 단백질 함량은 2회 산세척의 경우는 37.1% 및 75.3%이었으며, 1회 산세척과 UF(100K) 농축의 경우는 42.1% 및 72.4%였다. 따라서 glucosinolate와 phytate 제거효과와 단백질의 수율면을 고려한 결과 가장 효과적인 정제공정은 등전침전${\rightarrow}$1회 산세척(pH 3.5)${\rightarrow}$중화(pH 7.5)${\rightarrow}$ UF(100K) 농축 처리하는 방법이다.

  • Poly(ethylene glycol)/salt가 형성하는 액상 2상계에서 균체내 효소인 $\beta$-galactosidase 분리를 최적화하는 실험을 수행하였다. L. sporogenes에 의해 생산된 $\beta$-galactosidase의 분획은 2상계를 형성하는 PEG 분자량이 작을수록 상층부로 이동하였으며, 상층부 PEG 농도가 증가할 때 salting-out 현상이 나타나서 단백질 뿐아니라 $\beta$-galactosidase는 상층부로 크게 이동하였다. 또한 균체내 효소 정제에서 문제가 되는 세포벽잔사(cell debris)들은 조성이 binodial line에 접근할수록 효소 fraction과 반대방향으로 이동되었다. PEG-salt를 이용한 $\beta$-galactosidase 분리는 조성을 binodial line레 접근시키면서 동시에 상층부의 부피를 줄이는 것이 효과적이었다. PEG1000, 300을 각각 단계적으로 이용한 2단계 추출(two-step extraction)로써 세포벽 잔사, bulk protein, 색소물질 및 핵산 등을 제거할 수 있었으며, 이때 효소의 회수율은 74%, 단백질 회수율 35%로서 정제도는 2 배 이상의 효과를 보였다.

산초유 정제공정에 따른 물리화학적 변화 (Changes in the Physicochemical Characteristics of Sancho Oil according to the Purification Process)

  • Background: Sancho oil extracted from Zanthoxylum schinifolium (Siebold & Zucc) is a useful edible oil that has been in use for a long time, but it is known to be susceptible to rancidity. Sancho oil purification can remove impurities to prevent rancidity. This study was performed in order to improve the quality of sancho oil and enhance its availability throughout the purification process. Methods and Results: Sancho oil extracted in Hadong, Korea was refined via the degumming and deoxygenation processes, following which we examined the changes in the polyphenol content, fatty acid content and antioxidant activity of the oil. Acetic acid was effective for deoxygenation of sancho oil and 2 N NaOH was effective for its deoxidation. The polyphenol content and antioxidant activity were reduced by the purification process. Saturated fatty acids contents did not vary with the degumming and deoxygenation processes, however the content of unsaturated fatty acids were slightly reduced. Conclusions: This study suggests that the process of sancho oil purification used in this study will contribute to the increased use and storage of sancho oil.

물리적 정제방법에 의한 규석의 고순도화 연구 (High Purification Characteristics of Quartz with Physical Separation Method)

  • 본 연구에서는 $0.1{\sim}0.3mm$ 입도의 규석을 대상으로 물리적 선별공정을 적용하였을 때의 정제효과를 조사하였다. $SiO_2$ 품위가 99.41 wt.%, 99.54 wt.%이고 불순물 함량이 각각 5,864 mg/kg과 4,568 mg/kg인 시료에 대하여 자력선별, 비중선별 및 scrubbing 공정을 단계별로 적용시킨 결과, 주요 불순물인 철 성분은 자력선별에 의하여, 알루미늄 성분은 비중선별에 의해 효과적으로 제거되었다. 최종 정제산물의 $SiO_2$ 품위는 99.98 wt.%였으며, 산출율은 각각 79.05 wt.%와 75.43 wt.%였다. 이때 불순물의 함유량은 각각 126 mg/kg, 174 mg/kg을 나타내어, 불순물의 제거율은 97.85wt.%, 96.19 wt.%였다. 따라서 물리적 분리 선별에 의하여 저급 품위(Low grade)의 규석을 불순물 함유량이 200 mg/kg 이하인 중 고급 품위(Medium-high grade) 이상으로의 품위 향상이 가능하였다.
