High-level Expression and Purification of Recombinant 4-Aminobutyrate Aminotransferases in Escherichia coli

  • The protein coding sequence of the 4-aminobutyrate aminotransferase was amplified by polymerase chain reaction (PCR) from a previously cloned cDNA of pig brain using a pair of primers based on the published sequence. The amplified DNA was introduced into a T7 expression vector. Recombinant 4-aminobutyrate aminotransferases were overexpressed in Escherichia coli. The inclusion bodies were formed when enzyme was overexpressed. The unfolded, overproduced proteins were purified by chromatography with hydroxyapatite and refolded by a sequential dialysis method. The renatured 4-aminobutyrate aminotransferase regained the catalytic activity. However, the purified mutant protein did not show the catalytic function of 4-aminobutyrate aminotransferase.

A Study on Formation and Concentration of Trihalomethanes in Water Treatment Process (정수처리공정의 THMs 생성과 농도변화에 관한 연구)

  • This study was carried out to investigate the effects of prechlorination and algae growth on THMs generation. The sample water obtained from Paldang Dam which is a main source of raw water for the Seoul metropolitan area. THMs concentration in the sample water was investigated in water treatment process prechlorifiation, chemical coagulation, and sand filtration. And also, THMs concentration were analyzed in the water which cultured algae in laboratory. The results were as follows 1. The THMs concentration produced by prechlorination unit process were increased in control (not purified) but decreased in process of purification. 2. The THMs concertration can reduce by increasing the number of cleaning filters. 3. The main precursor in raw water for the THMs generation was supplied by algae growth. So as to reduce the THMs concentration in water supplying system, it is the best method to manage algae growth in water body of Paldang reservoir.

  • An efficient method for the preparation of 1,4-disubstituted 1,2,3-triazole compounds is described using polymeric quaternary ammonium salts having azide or alkyne functionality to remove unreacted excess starting molecules (azide/alkyne). Copper metal could easily be removed by simple filtration with a short $Na_2SO_4$/silica cartridge, affording highly regioselective products in high yield and excellent purity without the need for work-up, extraction and chromatographic purification.

  • Within fractionating devices existing in separation and purification industries, sieve trays are widely used as tower internals and their choice is due to economical attractiveness. While operating a trayed distillation tower weeping phenomenon has a critical effect on the efficiency, in this case study a weeping phenomenon was undertaken by means of numerical model in a rectangular sieve tray. Eulerian-Eulerian Computational Fluid Dynamics (CFD) method was used and the obtained CFD results are in a good agreement with the experimental data in terms of weeping rate and pressure drop.

  • For in vitro myelination system, Schwann cells and neuronal cells of rat were cocultured. Schwann cells and neuronal cells, respectively, were obtained from dorsal root ganglion of rat embryos (E15). This method includes four steps: first step of suspension of the embryonic dorsal root ganglion cells, second step of addition of anti-mitotic cocktail, third step of purification of dorsal root cells, and fourth step of addition of Schwann cells to dorsal root ganglion cells. We made a highly purified population of myelination in a short period through this procedure and identified myelination basic protein using antibody of myelination basic protein.

  • Availability of X-ray photoelectron spectroscopy (XPS) for the identification of ionic liquids (ILs) was tested. Commercially available ionic liquids (1-butyl-3-methyl imidazolium tetrafluoroborate ([BMIM] $BF_4$), (1-butyl-3-methyl imidazolium trifluoromethanesulfonate ([BMIM] OTf), (1-butyl-3-methyl imidazolium hexafluorophosphate ([BMIM] $PF_6$), 1-hexyl-3-imidazolium hexafluorophosphate ([HMIM] $PF_6$), and 1-ethyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide ([EMIM] $Tf_2N$) were qualitatively and semi-quantitatively analyzed with XPS. In order to confirm whether the results of XPS were correct, conventional method such as a nuclear magnetic resonance (NMR) was performed. After the XPS results were convinced by NMR, we synthesized ILs (1-(4-sulfonic acid) butyl-3-butylimidazolium trifluoromethanesulfonate ([SBBIM] OTf), 1-(4-sulfonic acid) propyl-3-methylimidazolium trifluoromethanesulfonate ([SPMIM] OTf), and 1-(4-sulfonic acid) propyl-3-butylimidazolium trifluoromethanesulfonate ([SPBIM] OTf) and analyzed it with XPS and NMR as well. It was successful the usage of XPS to analyze ILs without any purification processes.

  • Carotenoids are the major pigment of pepper (Capsicum annuum) and tomato (Lycopersicon esulentum) which are very important foods in Korea. However the analysis of carotenoids is quite complicated because of their diversity and the presence of cis-trans isomeric forms of these compounds. The objective of this review is to collect the achievements on the field of the chromatographic separation of carotenoids in food and some vegetables, to describe and critically evaluate the techniques, And to compare the benefits and shortcomings of the various chromatographic methods such as adsorption and reversed-phase HPLC and thin-layer chromatography. HPLC equipped with ultra-violet or photodiode array detection is most often employed in routine use for the analysis of carotenoids. Here, the method to analyze carotenoids by HPLC separation after solvent extration and purification from pepper powder samples done in our laboratory is also mentioned.

  • Chitinase, a potential pathogensis- related portein, was induced in the leaves of 30 day - old bean( phaseolus vulgaris ) plant with the treatment of 10 $\mu $ /mℓ ethylen for 30hrs. Chitinase was purified from the mature tissue of bean leaves( phaseolus vulgaris ) by ammonium sulfate precipitation followed by affinity chromatography on a regenerated chitin and sephadex G-75 chromatography. The purified chitinase gave a single band SDS- PAGE to be 32,000 Dalton. In order to elucidate the three- dimensional structure of chitinase and to shed light on the functional mechanism of this class of enzymes, the enzyme was tried to crystallize( Sitting Drop Method). Crystals grew at room temperature to their final size within two weeks(0.2mm $\times $0.2mm $\times $ 0.1 mm ), This enzyme are still continuing to crystallize for the study of X- ray.

  • The conventional control techniques based a mathematical model are not well suited for dealing with ill-defined and uncertain system like a linear quadratic control. Recently, fuzzy control has been successfully applied to a wide variety of practical problems such as robot, water purification, automatic train operation system etc. In this paper, a design technique of robust Fuzzy-LQ controller for each subsystem is designed. Secondly , all the subsystem controllers are combined by fuzzy weighted averaging method. Finally the effectiveness of the proposed controller is verified through a series of computer simulations for an inverted pole system.

  • The study of whole patterns of changes in protein expression and their modifications, or proteomics, presents both technological advances as well as formidable challenges to biological researchers. Nutrition research and the food sciences in general will be strongly influenced by the new knowledge generated by the proteomics approach. This review examines the different aspects of proteomics technologies, while emphasizing the value of consideration of "traditional" aspects of protein separation. These include the choice of the cell, the subcellular fraction, and the isolation and purification of the relevant protein fraction (if known) by protein chromatographic procedures. Qualitative and quantitative analyses of proteins and their peptides formed by proteolytic hydrolysis have been substantially enhanced by the development of mass spectrometry technologies in combination with nanoscale fluidics analysis. These are described, as are the pros and cons of each method in current use.
