Application of Differential Item Functioning to Test Adaptation (차별문항기능 기법의 응용 : 교육 및 심리검사의 번안과정에서)

  • This paper is concerned with evaluating the fidelity of a non-cognitive test adaptation for use in multiple languages and cultures using two differential item functioning(DIF) techniques: (a) PSIBTEST, and (b) Logistic Discriminant Function Analysis(LDFA). In particular, this study focused on how DIF research can best be extended to the problem of evaluating the equivalence of tests across cultures and languages. The Sixteen Personality Factor (16PF) questionnaire was administered in English to 844 American college students and in Korean to 538 Korean college students. This study attempted to identify the best matching criterion for the translated tests by using both a multivariate matching technique and iterative purification process. The results generally showed a small number of DIF items on each scale, except for scales A and N where about half of the items showed DIF. The choice of matching variables based on a combination of internal measures appeared to have little effect and the iterative purification method was unsuccessful. Finally, the results were discussed and methodological implications were also presented.

Separation of Human Breast Cancer and Epithelial Cells by Adhesion Difference in a Microfluidic Channel

  • A simple, label-free microfluidic cell purification method is presented for separation of cancer cells by exploiting difference in cell adhesion. To maximize the adhesion difference, three types of polymeric nanostructures (50nm pillars, 50nm perpendicular and 50nm parallel lines with respect to the direction of flow) were fabricated using UV-assisted capillary moulding and included inside a polydimethylsiloxane (PDMS) microfluidic channel bonded onto glass substrate. The adhesion force of human breast epithelial cells (MCF10A) and human breast carcinoma (MCF7) was measured independently by injecting each cell line into the microfluidic device followed by culture for a period of time (e.g., one, two, and three hours). Then, the cells bound to the floor of a microfluidic channel were detached by increasing the flow rate of medium in a stepwise fashion. It was found that the adhesion force of MCF10A was always higher than that of MCF cells regardless of culture time and surface nanotopography at all flow rates, resulting in a label-free detection and separation of cancer cells. For the cell types used in our study, the optimum separation was found for 2 hours culture on 50nm parallel line pattern followed by flow-induced detachment at a flow rate of $300{\mu}l/min$.

Purification and biological activity of recombinant human bone morphogenetic protein-2 produced by E. coli expression system (E. coli 발현 시스템에 의해 생산된 recombinant human bone morphogenetic protein-2의 정제와 생물학적 활성)

  • Purpose: Bone morphogenetic protein-2(BMP-2) has been shown to possess significant osteoinducitve potential. There have been attempts to overcome a limitation of mass production, and economical efficiency of BMP. The aim of this study was to produce recombinant human BMP-2(rhBMP-2) from E. coli in a large scale and evaluate its biological activity. Materials and Methods: The E.coli strain BL21(DE3) was used as a host for rhBMP-2 production. Dimerized rhBMP-2 was purified by affinity chromatography using Heparin column. To determine the physicochemical properties of the rhBMP-2 expressed in E. coli, we examined the HPLC profile and performed Western blot analysis. The effect of the purified rhBMP-2 dimer on osteoblast differentiation was examined by alkaline phosphatase (ALP) activity and representing morphological change using C2C12 cell. Results: E. coli was genetically engineered to produce rhBMP-2 in a non-active aggregated form. We have established a method which involves refolding and purifying a folded rhBMP-2 dimer from non-active aggregates. The purified rhBMP-2 homodimer was characterized by SDS-PAGE as molecular weight of about 28kDa and eluted at 34% acetonitrile, 13.27 min(retention time) in the HPLC profile and detected at Western blot. The purified rhBMP-2 dimer stimulated ALP activity and induced the transformation from myogenic differentiation to osteogenic differentiation. Conclusion: rhBMP-2 was produced in E. coli using genetic engineering. The purified rhBMP-2 dimer stimulated ALP activity and induced the osteogenic differentiation of C2C12 cells.

Characterization and Partial Purification of Storage Protein-i Receptor in the Fat Body of Hyphantria cunea (미국흰불나방 지방체에서 저장단백질-1의 수용체의 특성과 부분정제)

  • In vitro tissue culture of fat body of Hyphantria cunea in the medium containing [35S]-methionine reveaied that storage protein-i (SP-1) is taken up into fat body of prepupae and 1-day-old pupae. Using Western blotting and ligand binding method, we were able to identify the protein band of the SP-1 receptor protein. For the partial purification, the membrane proteins of fat body cells were solubilixed with 1% Triton X-1OO and applied to anion exchange chromatography. The results revealed the molecular weight of the receptor protein to be about 80 kl)a in SDSPAGE, and the P1 was estimated to be about 6.1. The mobility of the receptor protein in 8D8-PAGE was highly dependent on both temperature during electrophoresls and the condition of samples whether they were in reducing or nonreducing.

  • Decomposition of reactive organic compound dispersed in microemulsion media by hydroxide ions and proton ions generated during electrolysis was tried and the half-lifes for decomposition were compared. Absorbance of p-nitrophenoxide produced from the decomposition of p-nitrophenylacetate (PNPA) was followed to find the rate of decomposition. The applied voltage, temperature, and the amount of substrate were changed to see the effects on the decomposition rate. The advantages of electrolysis in microemulsion system were the high solubilizing capacity of substrate, easy control of decomposition rate, low operation cost, no need for any addition of chemicals, and no byproducts. The mechanism of decomposition and the application to water purification were discussed.

Recycling of Organic Materials Using Purification by Recrystallization for Solution-Processed OLEDs (재결정화법에 의한 유기물 재활용 및 이를 이용한 습식 OLED 제작)

  • We have investigated the possibility of recycling of an organic material that is wasted during thermal evaporation. To this end, we have collected a wasted organic material (N,N'-diphenly-N,N'-bis(1,1'-biphenyl)-4,4'-diamine(NPB)) from a vacuum chamber, purified it by recrystallization, and fabricated bilayer organic light-emitting diodes (OLEDs) with the recycled NPB. It is found that the surface roughness of thin films coated with the purified NPB is much enhanced. OLEDs fabricated by thermal evaporation of the purified NPB show lower device efficiency than OLEDs with the original NPB. However, the power efficiency of OLED fabricated by spin coating of the purified NPB is comparable with that of OLED with the original NPB. Therefore, such a recycling method by recrystallization would be more suitable for solution-processed OLEDs.

Preparation of High Purity ZnO Powder from zinc-bearing waste by the Hydrometallurgical Process (함아연 폐기물로부터 습식법에 의한 고순도 ZnO 분말의 제조)

  • A process development for direct synthesis of high pure ZnO powders from zinc-bearing waste was investigated. This waste contains a 55% of zinc and it was extracted by the sulfuric acid(leaching). After removal of iron ion by precipitation from the zinc solution, the purification through a solvent extraction by the use of D2EHPA as an extractant was carried out. Then, loaded zinc in organic solution was stripped and precipitated simultaneously using a precipitant such as oxalic acid. The synthesized $ZnC_2O_4$ powders by the precipitation stripping method was calcined to obtain more than 99.9% of ZnO powders. The effect of sulfuric acid concentration, leaching time, pulp density on the extraction of zinc was studied and the optimum conditions for the solvent extraction were obtained through the investigation of purification of zinc solution. The size, morphology and size distribution of synthesized $ZnC_2O_4$ powders were studied in terms of oxalic acid concentration, temperature, surfactant added, precipitation time, etc.

Purification of Total Ginsesides with Macroporous Resins and Their Biological Activities

  • Total ginsenosides were purified and their antioxidant, antibacterial and anticancer activities were measured. The crude extracts of ginseng, which were extracted with 75% ethanol by ultrasonification method, were firstly purified on AB-8 macroporous adsorption column to remove water soluble impurities, and decolored on Amberlite IRA 900 Cl anion-exchange column. Then, they were purified on Amberlite XAD16 adsorption column to delete the non-polar impurities. Total ginsenosides contents of the purified extracts were 79.4%, 71.7% and 72.5% in cultured wild ginseng, red ginseng and white ginseng, which were significantly increased than those of crude extracts. All of the three extracts showed concentration-dependant scavenging activities against DPPH radicals, among which white ginseng showed the most powerful activity. Cultured wild ginseng roots showed strongest effect against both B. subtilis PM 125(Gram-positive) and E. coli D31 (Gram-negative) bacteria, while red ginseng and white ginseng only showed the activity against B. subtilis. According to the result of the MTT assay, ail of the three extracts inhibited the growth of U-937 human hohistiocytic lympma cell, which were significantly different (p < 0.05) when compared to the control.

A study on Measurement and Improvement of Indoor Air Quality in Dental Clinic

  • The purpose of this study is to propose a management method to maintain the pleasant indoor air quality of the dental clinic by measuring and analyzing the indoor air quality of the dental clinic. The measurement was conducted in two rooms, a lobby where many residents stay in the reception room for waiting for medical treatment, and a VIP room where treatment activities are mainly performed. Measurement items are Temperature, Humidity, $CO_2$, CO, $NO_2$, $CH_2O$, VOC, $PM_{10}$ and measurement were taken on April 27, 2018. As a result of analyzing the temperature and humidity of the dental clinic, it was analyzed that the average indoor temperature was maintained at $25^{\circ}C$ and the humidity was kept at around 50%, maintaining proper indoor temperature and humidity environment. $CO_2$ was 855ppm in the VIP Room, which satisfied the maintenance standard. In the case of the lobby, it was analyzed to be 1,160ppm, which exceeded the maintenance standard and it is judged that the carbon dioxide generated by the respiration of the people staying in the lobby is the main reason. The mean concentration of formaldehyde in the VIP room was analyzed as $436{\mu}g/m^3$, exceeding the maintenance standard, and $2,100{\mu}g/m^3$ for the VOC exceeded the recommended standard. It was analyzed that the concentration was relatively higher due to the use of disinfectant and other drugs. The mean concentration of PM-10 in the lobby was analyzed as $65{\mu}g/m^3$ and it was analyzed that it satisfied the maintenance standard. To maintain a pleasant indoor air quality in a dental clinic it is necessary to minimize the effects of formaldehyde, VOC, $CO_2$ in the VIP rooms and lobby. For this purpose, the entire ventilation system and air purification system of the dental clinic should be installed. In case of the VIP room, local exhaust ventilation should be installed and workers should wear personal protective equipment.

Review on Antifouling Membranes with Surface-Patterning for Water Purification (물 정화를 위한 표면패턴화된 내오염성 분리막에 대한 총설)

  • As clean water continues to be a demand in this global water crisis, development of separation membrane technology for water purification becomes a necessity. The effectiveness of separation membranes is hindered in the water crisis due to fouling of membranes. To address this problem, the application of patterns on flat membranes via various methods have been recently studied and experimented. Patterned membranes have shown to not only reduce the fouling effects of membranes, but also increase the fluxes depending on the method and materials used. Each application has shown benefits that include, but not limited to, enhanced surface area, higher pure-water permeability, and increased number of filtration cycles. In this review, the effects of patterned membranes against antifouling is summarized and discussed.
