This study was conducted to evaluate the estrogen activity of silkworm (Bombyx mori) pupa extracts and their fractions. Powdered samples of freeze-dried silkworm pupa were extracted at room temperature (RT), $40^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$, and $100^{\circ}C$ in water (D.W), chloroform, ethyl acetate, and methanol for 6h and then filtered (0.45 um). The extracts were then freeze-dried. The estrogenic activity of these extracts was then investigated by competition binding assays using estrogen receptor ${\alpha}\;(ER{\alpha})$ and $ER{\beta}$, and by evaluating their effects on the proliferation of the human breast cancer cell line, MCF-7. Among the extracts evaluated, water extracts prepared at RT showed the highest binding affinity to $ER{\alpha}$ ($IC_{50}$, 1.76 ug/ml) and $ER{\beta}$ ($IC_{50}$, 0.07 ug/ml). In addition, MCF-7 cells that were treated with 62.5 ug/ml of the RT extract showed the greatest increase in proliferation (2-fold; 1291.79%) when compared to control cells (659.82%). Next, the water extract that was prepared at RT (sample 1) was dissolved in D.W. and further fractionated using a Dowex 50W - 8X ($H^+$) column. The flow-through and wash were then pooled together and freeze-dried (sample 2). The bound materials were then eluted with 20 mM NaCl, after which they were applied to a Dowex 1X2 - 200 ($Cl^-$) column and washed with D.W. to remove the sodium ions. The eluants were then freeze-dried (sample 3). Of these fractions, sample 2 showed the highest binding affinity to ER{\alpha} ($IC_{50}$, 1.44 ug/ml) and $ER{\beta}$ ($IC_{50}$, 1.18 ug/ml). In addition, MCF-7 cells that were treated with sample 2 (15.6 ug/ml) showed the largest increase in growth (1159.39%) when compared to control cells (525.26%). Taken together, these results suggest that the fraction of the RT water extract of silkworm pupa referred to as sample 2 may be useful as a phytoestrogen.
The aim of this study was to develop a seasoning paste fermented by Aspergillus oryzae and Bacillus subtilis using three edible insects: Tenebrio molitor larvae, Gryllus bimaculatus, and Bombyx mori pupa. No significant changes in pH and titratable acidity were observed between the insect pastes and soybean control during a 5-week fermentation process. The amino nitrogen was 0.35-0.50% (w/w) in the early stage of the fermentation and increased to 0.72-0.78%. The total nitrogen was 2.36-3.62% (w/w) in the early stage and was preserved during the fermentation period. Free amino acids of the insect pastes were similar to those in soybean paste. In general, the fermented insect pastes showed a lower flavor and color than soybean paste but B. mori pupa-fermented paste did not show any significant difference in preference from soybean. Glutamate, aspartate, lysine, phenylalanine, arginine, and serine were found to affect taste preference. It was possible to ferment edible insect protein to produce a fermented seasoning paste like Korean doenjang.
The external anatomy of the immature stages (last instar larva and pupa) of the crane fly, Tipula (Nippotipula) sinica Alexander, 1935 (Diptera: Tipulidae) is described and illustrated from specimens first collected in Korean peninsula (South Korea). Comments concerning natural history and microhabitats of larvae are provided. This is the first detailed description with illustrations for the last instar larva and pupa of Tipula (Nippotipula) sinica. Pupal characteristics resemble those of most species from advanced lineages within the subfamily Tipulinae. And, also it described with habitats and biological notes of Tipula (Nippotipula) sinica. Crane fly larvae, categorization as found in this survey indicate a taxonomic stream and are expected to help.
The serotonin-immunoreactive (5-HTil neurons have been investigated in the brains of lanra, pupa and adult from Pieris ropae. There are ca. 54 5-HTi neurons in 5-instar larva, ca. 20 in 2-dav-old pupa and ca. 118 in 1-day-old adult, respectively. Most of these 5-HTi neurons are interneurons, but efferent and afferent 5-HTi neurons were also observed. Most of the 5-HTi neurons project into the central neuropils of postembrvonic brains. The larval brain contains abundant 5-HTi processes in the central neuropils, including those in three cerebral commissures. But in the pupal brain the 5-HTi processes are restricted in small numbers to the given regions of central neuropil. The adult brain contains a large number of 5-HTi processes in mushroom body, central body complex, lateral protocerebrum, protocerebral bridge, antennal lobe, and tritocerebral and suboesophageal neuropils. However, the 5-HTi processes are not found in the optic lobe of the brains. One prominent feature of the 5-HTi fibers in the postembrvonic brains is the fact that they are greatly arborized.
1. The differentiation process of the cluster cells of compound eye in wild typed Drosophila melanogaster, born in Heuk San Do, Korea, was observed. 2. The eye disc of Drosophila melanogaster is originated from the frontal sac before hatching. 3. Clusteris bgin to form at the posterior end of the eye disc at the 2 nd instar larval stage, and the completed to differentiate in the eye disc at the 96 hours after hatching . 4. At the pupa stage, the eye cups are formed by clusters. 5. The eye cup begins to elongate and is changed into an ommatidium. 6. All organs of an ommatidium are completed in an omatidium at the old pupa stage(156-160 hours after hatching. )
Larvae and pupae of Simulium (Eusimulium) satsumense were collected and recorded from Korea for the first time. Detailed descriptions and photographs of both immatures are provided. Currently, 42 species are known from the subgenus Eusimulium, which is found globally, mainly in the Holarctic Region, but unknown from Korea. Members of the subgenus Eusimulium are morphologically homogeneous in male, female, and pupa, making them extremely difficult to distinguish from one another. S. (E.) satsumense can be distinguished from other Korean black flies by the following combination of the characteristics: Pupa: Gill of four slender filaments, dorsalmost filament strongly divergent from other filaments; cocoon slipper shaped without anterodorsal projection; Larva: Head spots distinctly positive; postgenal cleft short, apically rounded or squared; antenna with proximal segment brown; abdominal segments IX with prominent ventral tubercles; and rectal papillae of three simple lobes. In addition to descriptions for larvae and pupae of the species, a key to genera and subgenera of Simulium of Korean black flies are also provided.
In order to eradicate harmful insect by use of sterile male techniques, it is necessary to disperse about 10 times as many male insect as in wild condition. However, it is so difficult to discriminate male insect in pupal stage that usually the population are likely to be contaminated by female insect when that are released in field. The present study was conducted to establish a useful method capable of differenciating the male pupae of Stomawys calcitsans by means of measuring the pupal weight and length. The results were summarized as follows; 1. Male pupa was lighter and shorter than female and high correlation existed between length and width of pupa. 2. There was significant relationship between the length of pupa and width of adult 3. When the pupal sex of stable ay was identified by median of mean length and mean weight of the pupae, the Ratio of female to male was 1 : 1. 4 and 1 : 2. 2, respectively. Therefore, median of pupal weight seemed to be applicable for obtaining more number of male pupae.
Changes in the protein concentration in the hemolymph and the oxidation of respiratory substrates at various developmental stages in cabbage worm, Pieris rapae Linne were measured using Biuret method and Warburg manometric method, respectively. The results are summarized as follows: 1. The hemolymph protein contentration decreased gradually until the six-day pupa and increased thereafter. In the fifth larval instar the concentration reached a maximum and was about two times the value for the six-day pupa. 2. Endogenous respiration was very high at the prepupal stage, decreasing at two-day pupa, followed by an increase at the late pupal stage. 3. Glucose showed the marked activity throughout the whole stages, and had the striking influence on both the prepupal and adult stages. 4. The changes in protein concentration and the rate of oxygen consumption exhibited a general pattern of U-shaped curve through the process of life cycle according to histogenesis of the adult organ and histolysis of the larval organ.
The forming process of scale and socket of Pieris rapae L. during in 30 hr. pupa to in adult was morphologically investigated with scanning electron microscopy and transmission electron microscopy. 1. The scale forming cells which were distinguished from other epidermal cells were first observed in 30 hr. pupa. In the aspect that scale forming cell beared some morphological relations to socket forming cells and in the distribution of its organelles, scale forming cell was divided into three regions-basal region in which nucleus located, neck region which was surrounded by socket forming cells and scale region that was the cytoplasmic projection region over the wing surface. In process of the development of scale forming cell neck region and scale region were extended into the molting space and at this time, the changes of surface structure of scale region have occurred initially. 2. There was a more distinct process that scale region changed into the scale. Scale region which was first originated as clublike projection of the cell body was subsequently elongated and flattened out by broadening of the cytoplasm. After that, in the surface of scale were formed longitudinal ridges and microribs. In the late pupa, the cytoplsam of scale region have autolyzed by lysosome-like bodies and at length, scale which had air spaces, trabecula, pigment granules, longitudinal ridges and transverse ridges. 3. The major protion of socket forming cell located beside neck region of scale forming cell under the wing surface but the processing portion of the cell lay over the wing surface, suggesting that socket forming cells have actively processing. In extending to the molting space of neck and scale region, socket forming cells developed to the molting space and constructed socket.
The chitin was isolated from various kinds of insects such as exuvia of Psacothea hilaris Pascoe, silkworm pupa, Agrius convolvuli or from cuticle of cockroach by treatment with dilute HCI and NaOH. The chemical and crystalline structure was characterized by FT-IR and X-ray diffractometer. All of the chitins extracted from insects showed characteristic ${\alpha}$-chitin peaks at the Bragg angle 2$\theta$=9.3$^{\circ}$, 19.4$^{\circ}$, and 23.5$^{\circ}$by X-ray diffraction analysis. The transition from chitin to chitosan was confirmed by IR spectra and the degree of deacetylation of the crab shell, silkworm pupa, cockroach, and Psacothea hilaris Pascoe was 70.9, 76.4, 75.5, and 74.1%, respectively. The double diffraction peaks of insect chitosan were observed at 2$\theta$=10$^{\circ}$and 20$^{\circ}$, indication the characteristic of hydrated crystalline structure of chitosan.
