눈꽃동충하초 배지별 인공재배법과 성분분석에 관한연구 (Study on analysis of components and artificial cultural practice on several culture media of Paecilomyces japonica)

  • 1. 누에를 이용한 눈꽃동충하초 재배에서 종균접종은 누에 5령1일째 실시하였으며, 감염율은 72% 되었으며 2. 눈꽃동충하초 대량생산을 위한 인공재배 최적배지는 "현미80g+번데기가루20g" 처리구에서 자실체 길이 11.4cm,무게 51.6g, 짙노랑색으로 아주 양호했으며, 현미 처리구, 번데기+PDA처리구에서도 양호했음. 3.누에동충하초에서 발생된 눈꽃동충하초는 번데기 1마리당 1.2g(번데기포함), 길이 3.0~3.5cm정도 되었으며, 일부는 누에상태에서도 동충하초가 발생하였음. 4. $\beta$-glucan함량은 누에 5령 1일 접종 처리구가 40.5%로 가장 높고 저비용 배지인 현미 16.4%, 번데기 20.7%, 현미+번데기가루 23.1%, 소나무 톱밥+밀기울 28.7%로 $\beta$-glucan 함량이 높았다. 5. 지네, 굼벵이, 누에가루처리구에서는 균사생육이 부진하였고 분생포자만 발생하였다.

초등예비교사들의 곤충의 탈피와 변태에 관한 개념 (Elementary Preservice Teachers' Conceptions on Molt and Metamorphosis of Insect)

  • Elementary preservice teachers' understanding about the insect molt and metamorphosis was examined. Data were collected through the paper-pencil test of 448 junior students at a National University of Education. The instrument used was developed by Son(2003) and modified for the test. The results are as follows: First, Only 3.1$\%$ of students had a scientific concept on the molt periods in the insect life cycle. Students had an alternative conception that molt occurs either only in the larval period (29.9$\%$) or only in the period from pupa to adult (22.3$\%$). The percentage of the students thinking that molt occurs in the period from larva to pupa was low. Second, 98$\%$ of students stated that molt has to do with the larval growth, but 39.1$\%$ of students had an alternative conception that molt occurs after the larval growth at each instar. 25.1$\%$ had a conception that molt is related with the larval growth, but did not show any further understanding. Third, most students understood the correct meaning of metamorphosis. 34..2$\%$ of students had an alternative conception that metamorphosis occurs only in the period changing from pupa to adult, and only a few thought that it occurs in the period from larva to pupa. 24.8$\%$ of students had a scientific concept on the periods in which metamorphosis occurs. Fourth, some students understood the hatching process as molt in the sense that the egg shell is taken off (21.0$\%$), and as metamorphosis in terms of the appearance change from egg to larva (25.0$\%$). Fifth, 35.5$\%$ of students selected bees as an insect of incomplete metamorphosis, and 35.3$\%$ responded 'I have no idea', showing that they had poor understanding about the insect life cycle.

An efficient production management of the silkworm variety "Daewhangjam" through low temperature in a pupa

  • The Daehwangjam has excellent marketability and quality. However, precise silkworm breeding data on the recent high-temperature environment is not enough collected, and it is necessary to check whether stable Daewhangjam silkworm egg production was possible in poor environments. In this study, the timing of metamorphosis of the Daewhangjam parents Jam323 and Jam324 silkworms, in an optimal condition and a general condition similar to breeding grounds, was investigated to confirm that appropriate mating was achieved. Surprisingly, a number of female moths were discarded due to the metamorphosis of Jam324 female moths much earlier than Jam323 male moths in either an optimal or a general condition. To improve this, during the Jam324 female pupation period, the mild low-temperature of 15℃ was treated for the pupation period to delay the metamorphosis into female moths. Depending on the treatment period, the metamorphosis of the female pupa was delayed by more than 4 days, allowing normal mating with the Jam323 male moth. The eye pigments in a pupa also slowly accumulated in these treatments. In order to discover the side effects of low-temperature treatments, the voltinism of the Daewhangjam eggs was investigated, but most of them remained univoltine. Taken together, the low-temperature treatment during the female pupation period made it possible to use Jam324 female moths that were should be discarded, which means that stable and efficient management of Daewhangjam egg production is possible.

동충하초균주로 발효한 감초의 주요성분 함량 변화 및 NO 생성 억제 효과 (Changes in Cordycepin and Liquiritigenin Content and Inhibitory Effect on NO Production in Fermented Licorice and Dongchunghacho)

  • 약용식물의 발효는 새로운 식품의 소재 개발이 가능하나, 발효 균주는 대부분 이스트, 유산균, 박테리아 등이 이용되고 있다. 본 연구에서는 감초와 번데기 단독 또는 감초에 발효원인 번데기를 20%와 50%로 첨가한 혼합물의 배지에 눈꽃 동충하초(Paecilomyces tenuipes)와 밀리타리스 동충하초(Cordyceps militaris)를 이용하여 고체배양방법을 확립하였다. 동충하초 발효물을 식품소재로 개발하기 위하여 식용 가능한 용매인 에탄올 95%, 70%, 50%, 25% 및 물로서 추출한 다음 동충하초로부터 생성된 cordycepin과 감초의 지표성분인 liquiritin, liquiritigenin과 glycirrhizin의 함량 및 NO생성 억제효과를 조사하였다. Cordycepin함량은 감초에 번데기를 50%로 혼합한 배지에 밀리타리스 동충하초 균주을 접종하여 발효한 발효물을 70% EtOH추출하였을 경우에 가장 많았으며, 번데기를 첨가하지 않은 밀리타리스 동충하초 발효물 추출물보다 함량이 33배 정도 증가하였다. 또한 추출용매의 극성이 70% EtOH보다 높거나 낮아지면 감소하는 경향이었으며, 특히 발효원으로서 번데기의 첨가는 cordycepin의 함량을 현저하게 증가시켰다. Liquiritin의 함량은 발효하지 않은 감초보다 눈꽃 동충하초와 밀리타리스 동충하초로 발효한 모든 추출물에서 감소하였다. Liquiritigenin의 함량은 눈꽃 동충하초로 발효한 추출물이 밀리타리스 동충하초 발효 추출물보다 현저히 증가하였으나, 밀리타리스 동충하초 균쥬의 발효 추출물은 발효하지 않은 감초 추출물과 거의 차이가 없었으며, 두 균주 모두 번데기의 첨가량이 증가할수록 liquiritigenin의 함량이 감소하는 경향이었다. 감초에 번데기의 첨가량 또는 추출 용매의 극성이 증가하면 liquiritin과 glycyrrhizin의 함량은 현저히 감소하였다. 이상의 결과로부터, cordycepin 함량은 C. militaris 균주로 liquiritigenin은 P. tenuipes로 발효시에 현저하게 증가하였으나, liquiritin과 glycyrrhizin은 감소하였다. 감초를 동충하초로 발효시에 번데기의 첨가는 주요 성분의 변화를 현저하게 유도하였다. 동충하초 발효 추출물은 NO생성 억제효과가 증가하였으며, 고극성 용매 추출물에서 그 효과가 현저하였다. 감초의 발효시에 생성된 cordycepin과 liquiritin, liquiritigenin 및 glycyrrhizin의 함량은 발효원으로서 첨가되는 번데기, 추출용매의 극성, 발효 균주의 종류 등에 따라서 현저한 차이가 있었다. 이러한 결과는 동충하초 균주를 이용한 기능성 식품 소재를 개발하기 위한 기초 자료로서 활용이 가능할 것으로 기대된다.

Microsporidian Multiplication and Spore Production in Various Tissues of Pupa and Adult, in Relation to Age and Development of Silkworm, Bombyx mori L.(Lepidoptera: Bombycidae)

  • Multiplication and spore production of three microsporidia(Nosema bombycis, Nosema sp. 1 and Nosema sp. 2) in selected tissues of pupa and adult of silkworm, Bombyx mori L. were studied in two seasons (SI, SII) with distinct temperature (SI: $20.1{\pm}0.8^{\circ}C$ and SII: $25.1{\pm}0.7^{\circ}C$) regimes. Multiplication of the microsporidia followed a logistic pattern with a lag phase, an exponential phase and a stationary phase. In SII, spore production was significantly (P<0.01) higher in various tissues. Highest spore production was observed 30 days post inoculation (p.i.) in SI and in SII, it was $21{\sim}23$ days p.i. Spore production was significantly (P<0.01) higher in the gut tissues than other tissues. Nosema sp. 2 registered significantly (P<0.01) higher spore production in both the seasons compared to Nosema bombycis and Nosema sp. 1. Results indicate that the multiplication and spore production of microsporidia are tissue specific and extremely sensitive to the temperature at which the host is reared. Through this study, the precise day that the spore numbers of the microsporidia are maximized can be predicted in both pupa and adult in case the infection is initiated in the first instar.

Rate of Oxygen Uptake in Antheraea mylitta in Various Stages of Development and during Diapause

  • Respiration in Antheraea mylitta was studied using constant pressure respirometer. Oxygen uptake in different stages of life showed that moth has highest rate of oxygen uptake as compared to larva and pupa. At each stage significant sexual differentiation was observed. Depression in the rate of oxygen uptake in diapausing pupa was found to be higher in male ($65.75\%$) than female ($60.65\%$) as compared to non-diapausing counterparts. During diapause lowest oxygen uptake was recorded in the month of February, and were in the order of February < January < December < March < November < April < May < June in male, and, February < January < December < March < April < November < May < June in female. Significant sexual differentiation in oxygen uptake was recorded throughout the period of diapause (November to June) where female pupae registered lower rate of oxygen uptake than that of male. Oxygen uptake in female pupae declined upto $28.89\%$ in non-diapausing and $18.29\%$ in diapausing broods over male. Highest respiratory quotient value was recorded in the moth of November (0.68 in male and 0.70 in female) and, the lowest in the month of March in male (0.54) and May in female (0.55). Percentage loss in pupa weight always remained at a significantly higher level in male (except in February and March). The lowest oxygen uptake rate and weight loss in the month of February shows that the pupae were at peak of their dormancy during this month.

누에번데기 유래 불포화지방산을 이용한 기능성 효과 시험 (Study on the Unsaturated Fatty Acids in the Pupae of Silkworm, Bombyx mori)

  • 누에유래 불포화지방산의 분리법을 확립하여 특허출원 하였다(출원번호 :2004-76364호). 분리 정제된 누에유래 불포화지방산의 비율은 70% 이상이었다. 누에유래 불포화지방산의 기능성을 시험해본 결과, 고지혈증 개선의 효과를 보였고, 피부단백질인 콜라겐 생성을 유의적으로 증가시키며, 콜라겐 분해효소 활성을 유의적으로 억제하였다. 또한, 피부건조증 개선에 빠르고 탁월한 효과가 있었다.

Anti-Oxidation of Cultured Cordyceps militaris Growing on Silkworm Pupa

  • Cordyceps, one of the well-known traditional Chinese medicines, consists of the dried fungus C. sinensis growing on the larva of caterpillar. It is commonly used as a tonic of restoring vital body functions. In order to evaluate the pharmacological effects of C. militaris, the anti-oxidation and antiaging effect of cultured C. militaris growing on pupa of silkworm Bombyx mori, were investigated. The results showed! that the contents of lipofusin in myocardium were reduced and the SOD activity in the liver were elevated in a dose-dependent manner while no effect was observed for blood GSH-Px activity and liver MDA content in mice perfused with the cultured C. militaris compared to those untreated mice. Furthermore, the average life span, maximum life span and 50% death time of the fruit flies treated with cultured C. militaris were markedly prolonged. The results suggest that the cultured C. militaris growing on pupa of silkworm possesses anti-oxidation and delay senescence activity.

The Expression of Corazonin Neurons in Pupa and Adult Stage of Scuttle Fly

  • The scuttle fly is a fly species in the Phoridae family. Scuttle fly which moves abruptly after standing for a while and stop suddenly to rush off again. These characteristic behaviors of the scuttle fly seem to be related to muscular and nervous system or neurotransmitters. Thus, we focused at the neurotransmitter, corazonin (Crz) that is known to be related to resistance to stress and investigated the developmental process of the neurons in the scuttle fly. In a previous studies, we found that there are three groups of corazoninergic neurons in the larval CNS of the scuttle. Larva has 3 pairs of Crz neurons at the dorsolateral area of the brain, 1 pair at the dorsomedial brain and 8 pairs at the ventral nerve cord. In this studies, among these neurons, 1 pair of dorsomedial brain and 8 pairs of ventral nerve cord disappear in early pupal stage after metamorphosis. Only the 3 pairs of dorsolateral brain persist expression of Crz gene through all the period of pupa stage. This group of neurons converge gradually to frontal center of the brain and situated at the medial region. These pairs of corazoninergic neurons keep their number and location in adult stage. In the future, we expect further studies on the histological characteristics of corazonin-expressing cells and the expression of corazonin gene.

Characterization, Antioxidant Capacity and Protective Effect of Peptides from Cordyceps militaris Cultivated with Tussah Pupa on Oxidative Injured HepG2 Cells

  • The antioxidant capacity and protective effect of peptides from protein hydrolysate of Cordyceps militaris cultivated with tussah pupa (ECPs) on H2O2-injured HepG2 cells were studied. Results indicated ECP1 (<3 kDa) presented the strongest antioxidant activity compared with other molecular weight peptides. Pretreated with ECPs observably enhanced survival rates and reduced apoptosis rates of HepG2 cells. ECPs treatment decreased the ROS level, MDA content and increased CAT and GSH-Px activities of HepG2 cells. Besides, the morphologies of natural peptides from C. militaris cultivated with tussah pupa (NCP1) and ECP1 were observed by scanning electron microscopy (SEM). Characterization results suggested the structure of NCP1 was changed by enzymatic hydrolysis treatment. Most of hydrophobic and acidic amino acids contents (ACC) in ECP1 were also observably improved by enzymatic hydrolysis. In conclusion, low molecular weight peptides had potential value in the development of cosmetics and health food.
