Comparison of Infrequent Restriction Site-Polymerase Chain Reaction and Pulsed-Field Gel Electrophoresis for Molecular Typing of Staphylococcus aureus and Escherichia coli (황색포도구균과 대장균의 기준형별 결정에 있어서 Infrequent Restriction Site Polymerase Chain Reaction과 Pulsed-Field Gel Electrophoresis의 변별력 비교)

  • Background: Staphylococcus aureus (s. aureus) and Escherichia coli (E. coli) are major pathogens in community and hospital. And they sometimes cause the outbreak in hospital in the immunocompromised patients. Pulsed-field gel electrophoresis (PFGE) has been regarded as a standard method for genotyping in epidemiologic studies, but it is laborious and time-consuming. Infrequent restriction site-polymerase chain reaction (IRS-PCR), a new genotyping methods, was performed to compare the applicability with PFGE. Methods: We performed PFGE and IRS-PCR on S. aurues (n=120) and E. coli (n=117) which were collected clinically in 4 different hospitals. We assessed each method in terms of discriminatory power, quality, and efficiency. Results: In E. coli, the discriminatory power of IRS-PCR was $46.7{\sim}86.7%$, and that of PFGE was $88.9{\sim}96.7%$ according to hospital. But in S. aurues, the discriminatory power of IRS-PCR was $20{\sim}56.7%$, and that of PFGE was $40{\sim}90%$ according to hospital. The typablity and reproducibility of IRS-PCR were 100% of each. PFGE needed four days to complete the procedure, but IRS-PCR could be performed within one day, IRS-PCR showed better resolution than PFGE. Conclusion: In case of gram negative bacteria (like E. coli), IRS-PCR could be a reliable alternative for epidemiologic typing due to better efficiency and comparable discriminatory power. But in the case of gram positive bacteria (like S. aureus), IRS-PCR does not seem to be suitable for the strain-to-strain differentiation. More trials and changes of restriction enzymes or primers could reveal the efficacy of IRS-PCR in the field of molecular typing.

A clinical study of the power control of Nd : YAG laser for painless irradiation on intraoral soft tissues (구강내 연조직에 대한 무통적조사를 위한 Nd:YAG laser의 출력조절에 관한 임상적 연구)

  • Most dentists are very interested in laser therapy on the intraoral soft tissue lesions because they want to accomplish the analgesic and aseptic surgery with little or no bleeding. In order to determine the difference of pain threshold according to different gingival tissues with or without inflammation, 25 patients with inflammatory periodontal disease and 10 volunteers with good general and oral health were selected as the inflamed group and the normal group, respectively. Interdental papilla, marginal gingiva, attached gingiva, and alveolar mucosa were irradiated by the contact delivery($300{\mu]m$ fiber optic, for 5 seconds) of a pulsed Nd:YAG laser(EN.EL.EN06O, Italy). And the laser power was gradually increased from 0.5W by the increment of 0.1W. The highest laser power was recorded as the first painful power when the painful gesture was recognized at first. The difference of the first painful power of laser according to different gingival tissues with or without inflammation was statistically analyzed by paired t-test in MICROSTAT program. Following results were obtained: 1. In the comparison related with the inflammation, the first painful power was significantly lower in the inflamed group than in the normal group, regardless of interdental papilla and marginal gingiva(p<0.05). 2. In the comparison related with the tissue structure, the first painful. power was significantly lower in alveolar mucosa than in attached gingiva(p<0.05). The results suggest that, for the painless therapy by a pulsed-Nd:YAG laser irradiation, the laser surgery over 2.0W of power should be necessarily accomplished under the local anethesia, and the local anesthesia should be considered according to the degree of inflammation, the tissue structure, and the purpose of laser therapy.

  • Industrial-scale pulse corona process to remove $SO_2$ and $NO_x$ simultaneously from combustion flue gas has been studied. The pilot plant built in the present study treats 2,000 $Nm^3$/hr of flue gas from a boiler. The geometry of the pulse corona reactor is similar to that of an electrostatic precipitator commonly used in industry, A thyratron switch and magnetic pulse compressors, which can generate up to 130 kV of peak pulse voltage and up to 30 kW of average pulse power, have been used to produce pulsed corona. The removal efficiencies of $S0_2$ and $NO_x$ with the present process are maximum of 95 % and 85 %, respectively. Electrical power consumption to produce the pulsed corona, which has been one of the major difficulties to apply this process to industry, has been evaluated in the present study. The results showed that the power consumption can be reduced significantly by simultaneous addition of hydrocarbon injection and heterogeneous phase reactions to the process.

  • In pulsed-power techniques. Marx generator is generally used for the high-power device. but this generator has insulation and spatial problems. So we will suggest a pulse transformer that has a small size to generate the high voltage pulse instead of Marx generator. In this paper, Pulse duration is 4 [${\mu}s$] and the ratio of input and output voltage is 40[kV]/200[kV](step-up ratio=5). The output voltage and the process of pulse compression for pulse circuit are simulated by EMTP (Electro-Magnetic Transient Program). The secondary voltage of pulse transformer is about 200[kV] and pulse width is 4[t/s]. When the secondary winding of the pulse transformer is saturated. the pulse width is 1.25[${\mu}s$]. We selected dummy load 50[$\Omega$] for impedance matching. The pulse voltage of dummy load is 100[kV] and pulse width is 500[ns].

  • High power microwave sources operating in relativistic regions, high current reletivistic electron beams are obtained by using cold cathodes with the help of explosive emission. For these relativistic devices, the pulsed power and magnetic field systems are very large and heavy. The phase velocity of electromagnetic mode should be showed down close to the beam velocity, ensuring enough beam coupling with electromagnetic modes. By using the annular electron beam, a weakly relativistic oversized bwo consisting of rectangularly corrugated cylindrical waveguide is demonstrated.

  • We have studied the backward wave oscillator, a power-pulsed generator oscillator at 20 GHz has higher frequency then current one. An absolute instability linear analysis was used for the purpose of designing the slow wave structure. A large diameter of the slow wave structure was adopted to prevent the breakdown brought about by the increase of power density.

A STUDY OF ANALGESIC EFFECT ON PULP INDUCED BY PULSED Nd : YAG LASER (Pulsed Nd ; YGA 레이저의 치수마취효과에 관한 연구)

  • The purpose of this study was to evaluate the analgesic effect on pulp induced by pulsed Nd : YAG laser according to the power density, irradiated time and the method of application (Non-contact vs contact) in vivo. Experiments were carried out on 90 volunteers who have sound 4 canines. Each canine was grouped by the intensity and the time of irradiation of pulsed Nd : YAG laser was measured. Mean EPT response(units) was measured at 5 min, 30 min, and 60 min after irradiation with various intensity(1.5-3W, 15-30Hz), irradiated time (1.5min, 3min), and application method (Non-contact and contact). The results were as follows: 1. 5 minutes after irradiation, the mean EPT response(units) was increased by 32 % in the non-contact method and by 35% in the contact method of application. Which showed significant difference in mean EPT response(units) compared to not-lased canine in each groups(P<0.05). 2. The mean EPT response(units) with irradiating condition according to the various laser intensity, and the irradiation time stastically did not show significant difference in each groups as time elapsed. 3. There was no significant difference between the non-contact and contact method in the mean EPT response(units). 4. The mean EPT response(units) returned to the baseline value within 30 miuntes. In regard to the above results, the analgesic effect on pulp induced by pulsed Nd : YAG laser resulted a slight increase of pulpal pain threshold, but it was not as high as to replace the role of drug analgesia as whole. This technique can be used for inducing pulpal analgesia as adjunct to other pain control methods and is needed to develop better analgesic effects.

  • A forward-looking automotive radar typically utilizes the frequency modulated continuous wave(FMCW) or pulsed-Doppler waveform for the Information acquisition of the target range and velocity. In order to obtain the high resolution target information, however, a narrow pulse width and wide bandwidth are inherently required, thus resulting in high peak power and high speed digital converter processing. In this paper, a stepped-frequency pulsed-Doppler(SFPD) waveform algorithm is proposed for high resolution forward looking automotive radar application. The performance of the proposed SFPD waveform technique is analyzed and compared with the conventional FMCW and PD method. Since this technique can be used for the high resolution target imaging with arbitrary range and Doppler resolution, it is expected to be useful In automotive radar target classification for the precision collision avoidance applications in the future.

Diamond-Like Carbon Films Deposited by Pulsed Magnetron Sputtering System with Rotating Cathode

  • Extended cylindrical magnetron sputtering system with rotating 600-mm long and 90-mm diameter graphite cathode and pulsed power supply voltage generator were developed and fabricated. Time-dependent Langmuir probe characteristics as well as carbon films thickness were measured. It was shown that ratio of ions flux to carbon atoms flux for pulsed magnetron discharge mode was equal to $\Phi_{i}$ $\Phi$sub C/ = 0.2. It did not depend on the discharge current in the range of $I_{d}$ / = 10∼60 A since both the plasma density and the film deposition rate were found approximately proportional to the discharge current. In spite of this fact carbon film structure was found to be strongly dependent on the discharge current. Grain size increased from 100 nm at $I_{d}$ = 10∼20 A to 500 nm at $I_{d}$ = 40∼60 A. To deposit fine-grained hard nanocrystalline or amorphous carbon coating current regime with $I_{d}$ = 20 A was chosen. Pulsed negative bias voltage ($\tau$= 40 ${\mu}\textrm{s}$, $U_{b}$ = 0∼10 ㎸) synchronized with magnetron discharge pulses was applied to a substrate and voltage of $U_{b}$ = 3.4 ㎸ was shown to be optimum for a hard carbon film deposition. Lower voltages were not sufficient for amorphization of a growing graphite film, while higher voltages led to excessive ion bombardment and effects of recrystalization and graphitization.

  • This paper describes the successful development and the performance of X-band 50 W pulsed power amplifier using a 50 W GaN-on-SiC high electron mobility transistor. The GaN HEMT with a gate length of $0.25{\mu}m$ and a total gate width of 12 mm were fabricated. The X-band pulsed power amplifier exhibited an output power of 50 W with a power gain of 6 dB in a frequency range of 9.2~9.5 GHz. It also shows a maximum output power density of 4.16 W/mm. This 50 W GaN HEMT and X-band 50 W pulsed power amplifier are suitable for the radar systems and related applications in X-band.
