Isolation and Characterization of Bacillus sp. WRD-2 Extracellular Protease from Soil (토양에서 분리한 Bacillus sp. WRD-2가 생산하는 Extracellular Protease의 특성)

  • In order to produce alkaline protease, psychrotrophic Bacterium which have high enzyme activity, was isolated by using enrichment culture from soil samples and identified as genus Bacillus sp. The optimal pH and temperature for the enzyme activity were pH 6 and $40^{\circ}C$. The temperature range of high enzyme activity was $20{\sim}40^{\circ}C$. The optimal initial pH of culture condition for enzyme was pH 6. The most favorable carbon and nitrogen sources for the production of protease by Bacillus sp. WRD-2 were 3% maltose and 4% yeast extract, respectively.

Antimicrobial Effects of Pinus densiflora Sieb. et Zucc. Extract on Pathogenic Microorganisms (병원성 미생물에 대한 소나무(Pinus densifiora Sieb. et Zucc.) 추출물의 항균효과)

  • To develop natural food preservative, antimicrobial activity of Pinus densiflora Sieb. et Zucc. (PD) ethanol extract against pathogens were investigated. The growth of pathogens such as Staphylococcus aureus ATCC 29737, Bacillus subtils KCTC 1021, Bacillus megaterium KCTC 3007 and Vibrio parahaemolyticus ATCC 17802 was inhibited as much as 4 to 6 log cycle in tryptic soy broth containing 40mg/㎖ of PD extract but Salmonalla typhimurium ATCC 14028 and Pseudomonas fluorescens ATCC 21541 were not inhibited. The total, gram negative and psychrotrophic bacterial count were inhibited in minced pork containing 40mg/㎖ of PD extract for 20 days at 4$\^{C}$. The sensory quality of hamburger patty such as taste, flavor and overall acceptability were not decreased by the addition of 40mg/㎖ PD(p<0.05).

Distributions of Microorganisms and Identification of Pathogenic Bacteria Isolated in Raw Beef of Jangzorim (장조림 원료육의 미생물 분포 및 분리 병원성세균의 동정)

  • Raw beefs used fer Jangzorim production were evaluated fur contamination of pathogenic bacteria and microorganisms related to spoilage and food safety. Eleven groups of mesophilic, psychrotrophic, and anaerobic microorganisms, and total coliforms were selected to evaluate degree of food contamination. Nine strains including Bacillus cereus, Clostridium botulinum, C. perfringens, and Listeria monocytogenes were selected to evaluate incidences of pathogenic bacteria. Raw beefs harbored large populations of microorganisms, which decreased greatly after heat treatment. Psychrotrophic microorganisms were found to be more abundant than other microorganisms. B. cereus, C. perfringens, and L. monocytogenes were isolated from raw beefs, whereas C. botulinum, Escherichia coli O157:H7, Salmonella spp., Shigella spp., Staphylococcus aureus, and Yersinia enterocolitica were not isolated. Isoiates from Cereus selective agar, clostridium Perfringens agar, and Oxford agar were in 99.8, 99.9 and 98.6% agreements with B. cereus, C. perfringens, and L. monocytogenes at species level, respectively. B. cereus produced enterotoxin with CRET-RPLA method, whereas C. perfringens did not produce enterotoxin with PET-RPLA method.

Studies on the Bacteriological Properties of Loin Ham during Curing Period (염지경과에 따른 Loin Ham의 세균학적 특성에 관한 연구)

  • In order to examine related bacteria on the maturation of cured loin ham, bacteria isolated during curing periods from ham, which counted coliform group, psychrotrophic and halo-tolerant bacteria. The results are as follows; The isolated bacteria from ham during the curing period were Staphylococcus spp. 24 strains, Bacillus spp. 21 strains, Lactobacillus spp. 10 strains, Coryneform 2 strains. Microbacterium spp. 2 strains and Gram negative rods 8 strains. Micrococcus spp. were identified M. varians 12 strains and M. luteus 3 strains, and Streptococcus spp. identified S. faecium 14 strains, S. lactis 2 strains. Lactobacillus spp. were isolated L. Plantarum 4 strains, L. brevis and L. casei 1 strains. In the case of cured ham, the number of coliform group and psychrotrophic bacteria were decreased but halo-tolerant bacteria were increased for 10 days of curing period. On the brine solution. the number of coliform group, psychrotrophic and halo-tolerant bacteria were increased for 10 days. 4 days and 20 days, respectively.
