Microbial Distribution in Refrigerated Beef (냉장 우육내의 미생물의 분포)

  • Isolation and identification of mesophilic and psychrotrophic bacteria distributed in Korean refrigerated beef were attempted. Total isolated colonies were 192, and identified as 5 genera and 10 species. Among them, mesophilic bacteria were Enterobacter aerogenes, E. agglomerans, Serratia liquefaciens, Proteus mirabilis, and "psychrotrophic" bacteria were Pseudomons fluorescens, P. putida, P. pickettii, P. mendocina, P. stutzeri, Alcaligenes faecalis. Dominant species was Serratia liquefaciens as mesophiles, and Pseudomonas putida as psychrotroph.chrotroph.

Evaluation of Antimicrobial Activities of Sequential Spray Applications of Decontamination Treatments on Chicken Carcasses

  • The objective of this study was to evaluate the effects of sequential applications of ${\varepsilon}$-polylysine (EPL) or lauramide arginine ethyl ester (LAE) sprays followed by an acidic calcium sulfate (ACS) spray on inoculated chicken carcasses to reduce Salmonella (Salmonella enterica serovars including Salmonella typhimurium and Salmonella enteritidis) contamination during 6 days of storage ($4.4^{\circ}C$). Secondly, reductions of the resident microflora were studied on uninoculated chicken carcasses following the sequential application of the treatments, chilling and 10 days of storage at $4.4^{\circ}C$. The treatment of Salmonella inoculated carcasses with 300 mg/L EPL followed by 30% ACS (EPL300-ACS30) sprays reduced Salmonella counts initially by 1.5 log cfu/mL and then by 1.2 log cfu/mL (p<0.05) following 6 days of storage at $4.4^{\circ}C$. Likewise, 200 mg/L LAE followed by 30% ACS (LAE200-ACS30) treatment reduced initial Salmonella counts on poultry carcasses by 1.8, 1.4 and 1.8 log cfu/mL (p<0.05), respectively, after 0, 3, and 6 days storage. Immediately after the treatments, EPL300-ACS30 and LAE200-ACS30 both reduced Escherichia coli counts significantly by 2.6 and 2.9 log cfu/mL, respectively. EPL300-ACS30 and LAE200-ASC30 were effective in lowering psychrotroph counts by 1 log cfu/mL on day 10 when compared to the control and distilled water treatments. This study demonstrated that EPL300-ACS30 and LAE200-ACS30 were effective in reducing Salmonella on inoculated chicken carcasses both after treatment and during the storage at $4.4^{\circ}C$ for up to 6 days. In addition, reductions in psychrotroph counts indicated that these treatments might have the potential to increase the shelf-life of poultry carcasses.

On the Psychrophilic Yeasts Isolated from the Han River (한강에서 분리한 pwychrophilic yeasts에 관하여)

  • Psychrophilic yeast strains, HJ3011, HJ3023, and HJ3021, which were isolated from the Han River during winter season of December 1980 to February 1981 were identified as Candida sp., Candida nivalis, and Rhodotorula rubra respectively. The effect of temperature on growth was carried out in this study. Growth rate constants of strains HJ3011 and HJ3023 were the highest value at $15^{\circ}C\;and\;16{\sim}20^{\circ}C$ respectively, and both growth rates diminished gradually above $20^{\circ}C$ and were decreased to zero at $25^{\circ}C$. According to the results obtained in this experiment, above two strains of yeast were confirmed as psychrophile. On the other hand since the strain of HJ3021 exhibited the highest growth rate constant at $15^{\circ}C$ and could slightly grow above $25^{\circ}C$, this strain was defined as psychrotroph.

Inhibition Effect of Germ-resistant Sponge on Microbial Growth in Kitchen Hygiene (주방위생에서 항균수세미의 효과)

  • It was intended to investigate the effect of the microbiological kitchen hygiene such as dishclothes and scrubbers. The 8indicator organisms (standard plate counts, coliform, heterotroph, enterococcus, staphylococcus, heat-stable bacteria, psychrotroph, Pseudomonas aeruginosa) were detected highly in dishwaters, dishcloth and scrubber. Coliform and Staphylococcus aureus were appeared on dishcloth dominantly than the scrubber, and the scrubbers were intruded by hetrotrophs and psychrotrophs numerously than dishclothes. The germ-resistant sponge inhibited the growth of the most of test strain, and appeared the about 100% reduction rate after 24 hr, but did not affect Pseudomonas aeruginosa and P. fragi so typically after 24 hr. The anti-microorganism durability of germ-resistant sponge, treated with food soil, was maintained by 10 days, the early stage strain density was founded in 20 days, and the strains grew after 30 days.

  • The effects of garlic concentrations on the properties of squid sik-hae such as the chemical and microbial changes were investigated. pH, the amounts of lactic acid showed significant differences at early stages of fermentation and no significant differences at later stages with increasing garlic concentrations. The amount of TMAO decreased with increasing storage periods and garlic concentrations, whereas that of TMA increased significantly at early stages of fermentation and showed no significant differences with increasing garlic concentrations. At later stages of fermentation, the amount of TMA at $2\%$ garlic concentration was higher than those at 3 and $4\%$. The number of lactic acid bacteria increased significantly garlic concentrations at early stages of storage and had no significant differences at later stages. The numbers of proteolytic bacteria, psychrotroph, and fungi increased in proportion to length of storage and garlic concentrations at early stages of storage whereas, the number of bacteria decreased significantly at later stages of storage with increasing garlic concentrations.

Screening and Characterization of Psychrotrophic, Lipolytic Bacteria from Deep-Sea Sediments

  • Of 23 psychrotrophic bacteria isolated from the west Pacific deep-sea sediments, 19 were assigned to the $\gamma$-Proteobacteria, 3 to the <$\beta$-Proteobacteria, and 1 to the Gram-positive bacteria, as determined by their 16S rDNA sequences. Ten psychrotrophs, affiliated to the Psychrobacter, Pseudoalteromonas, and Pseudomonas genera in the $\gamma$-Proteobacteria group, were screened for lipolytic bacteria. The majority of the lipolytic isolates had growth temperatures between 4-$30^\circ{C}$, and all of them were neutrophilic, aerobic, or facultatively anaerobic, and some were able to produce multiple kinds of ectohydrolytic enzymes. The deep-sea strains Psychrobacter sp. wp37 and Pseudoalteromonas sp. wp27 were chosen for further lipase production analysis. Both strains had the highest lipase production when grown at 10 to $20^\circ{C}$; their highest lipase production occurred at the late-exponential growth stage; and the majority of the enzymes were excreted to the outside of the cells. Lipases from both strains had the same optimal reaction temperature and pH (20-$30^\circ{C}$, pH 7-8) and could retain about 60% of their highest activity at $4^\circ{C}$. Furthermore, SDS-PAGE and an in-gel activity test showed that they had the same high molecular mass of about 85 kDa.

Isolation and Characterization of Psychrotrophic and Halotolerant Rhodococcus sp. YHLT-2

  • A psychrotrophic bacterium was isolated from oil-contaminated groundwater and identified as Rhodococcus sp. YHLT-2. Growth was observed at the temperature of 4 to $30^{\circ}C$. This strain degraded various petroleum hydrocarbons such as crude oil, diesel oil, and gasoline over the whole range of temperatures tested. The Rhodococcus sp. YHLT-2 was capable of growing even at $4^{\circ}C$, exhibiting 90% of oil biodegradation after 20 days. Degradation of crude oil occurred at low temperature in nature. This strain was also able to grow at 7% NaCl, and utilized not only short chain alkenes $(C_9\;to\;C_{12})$, but also a broad range of long chain alkenes $(C_{19}\;to\;C_{32})$ present in crude oil at $4^{\circ}C$. The Rhodococcus sp. YHLT-2 is expected to be of potential use in the in situ bioremediation of hazardous hydrocarbons under low-temperature and high-salt conditions.

Cold-Adapted and Rhizosphere-Competent Strain of Rahnella sp. with Broad-Spectrum Plant Growth-Promotion Potential

  • A phosphate-solubilizing bacterial strain isolated from Hippophae rhamnoides rhizosphere was identified as Rahnella sp. based on its phenotypic features and 16S rRNA gene sequence. The bacterial strain showed the growth characteristics of a cold-adapted psychrotroph, with the multiple plant growth-promoting traits of inorganic and organic phosphate solubilization, 1-aminocyclopropane-1-carboxylate-deaminase activity, ammonia generation, and siderophore production. The strain also produced indole-3-acetic acid, indole-3-acetaldehyde, indole-3-acetamide, indole-3-acetonitrile, indole-3-lactic acid, and indole-3-pyruvic acid in tryptophan-supplemented nutrient broth. Gluconic, citric and isocitric acids were the major organic acids detected during tricalcium phosphate solubilization. A rifampicin-resistant mutant of the strain exhibited high rhizosphere competence without disturbance to the resident microbial populations in pea rhizosphere. Seed bacterization with a charcoal-based inoculum significantly increased growth in barley, chickpea, pea, and maize under the controlled environment. Microplot testing of the inoculum at two different locations in pea also showed significant increase in growth and yield. The attributes of cold-tolerance, high rhizosphere competence, and broad-spectrum plant growth-promoting activity exhibited the potential of Rahnella sp. BIHB 783 for increasing agriculture productivity.

Strain Selection of Psychrotrophic Leuconostoc mesentroides Producing a Highly Active Dextransucrase from Kimchi (김치로부터 고활성 dextransucrase를 생성하는 저온성 Leuconostoc mesenteroides 균주선발)

  • Leuconostoc mesenteroides, the major bacterium in the initial phase of lactate-fermentation in kimchi, produces lactic acid, acetic acid, mannitol, and $CO_2$. It also secrets dextransucrase, which catalyzes the transfer reaction of glucose from sucrose to maltose, synthesizing mainly panose ($6^_2-{\alpha}-D-glucopyranosylmaltose)$, a probiotic oligosaccharide. To use the strain as a starer culture to produce high amount of panose during kimchi fermentation, we screened psychrotrophic strains showing fast growth rate at low temperature among the isolates of Leuconostoc sp. and selected two strains showing high dextransucrase activity. The strains were identified as Leuconostoc mesenteroides, which can be used as function added-starters for lactate-fermented foods.

Contamination Level of Hygiene Indicator and Prevalence of Foodborne Pathogens in Retail Beef in Parallel with Market Factor

  • In this study, the contamination levels of hygienic indicators and foodborne pathogens in retail meat products were investigated in relation to the various market factors including processing temperature, processing area, and market type. Ground beef samples (n=80) were purchased from 40 meat markets and investigated for microbiological quality. Beefs processed below $20^{\circ}C$ had significantly lower numbers of total coliforms (TC) than these processed over $20^{\circ}C$ (2.01 vs. 2.79 log CFU/g; p<0.05). Interestingly, separation of processing area did not affect the contamination levels. Remarkably, the contamination levels of hygienic indicator differ among market types, indicating that not only processing condition but distribution structure that is directly related with storage period could affect the final microbiological loads of the meat products. In addition, the prevalences of Listeria monocytogenes (a psychrotroph), Enterococcus faecium, and Enterococcus faecalis were 7.5% (6/80), 10.0% (8/80), and 20.0% (16/80), respectively, which is irrelevant to market factors except meat products from wholesale markets where no L. monocytogenes were found among 30 samples. The results of this study indicate that the contamination level of hygiene indicator and foodborne pathogens in retail beef is more related with processing temperature and storage period than other environmental factors.
