Previous experiment was performed under abiotic condition. Steady-state abiotic experiments in the sand-tank reactor with air flowing through the reactor headspace demonstrated that oxygen supply through the water table interface into the saturated zone was enhanced when an LNAPL (dodecane) pool was present at the water table. Biotic condition was considered in this study. Biotic experiments performed after inoculating the reactor with Pseudomonas putida mt-2, which does not grow on dodecane, indicated that the enhanced oxygen supply in the presence of the LNAPL pool also enhanced biodegradation of a solute (glucose) plume passing beneath the LNAPL pool at steady-state.
Six strains of an obligate predatory bdellovibrio isolate that preys on Burkholderia glumae in rice paddy field water and rhizosphere soil, were identified and characterized. The numbers of Bdellovibrio cells varied from $3.2{\times}10^3$ to $9.2{\times}10^3$ plaque-forming unit/g after enrichment in cells of B. glumae. Prey range tests with six Bdellovibrio strains and 17 prey strains of rice-pathogenic, antibiosis-related, or nitrogen-fixing bacteria resulted in unique predation patterns in related prey cells. Strain BG282 had the widest prey range on 7 plant pathogenic bacteria among the 17 prey strains tested. However, no predation occurred with strains of Azospirillum brasilense, Paenibacillus polymyxa, Pseudomonas fluorescens, P. putida, and Serratia marcescens that are associated with antibiosis or nitrogen fixation in the rice ecosystem. Identification was confirmed by the presence of typical bdelloplast in the prey cells of B. glumae and by a PCR assay using B. bacteriovorus-specific primers. Furthermore, 16S rDNA sequencing of the six bdellovibrio strains showed a homology range of 97.2% to 99.2% to the type strain of B. bacteriovorus.
The study was carried out to evaluate the characteristics of biodegradation by Pseudomonas putida G7 in soil column. The reactor system was used to investigate mass transfer of VOCs as Toluene in a column of unsaturated soil. Determination of the fate of VOCs in unsaturated soil is necessary to evaluate the feasibility of natural attenuation as a VOCs remediation strategy. The objective of this study was to develop a mechanistically based mathematical model that would consider the interdependence of VOC transport, microbial activity, and sorptive interactions in a moist, unsaturated soil. Because the focus of the model was on description of natural attenuation, the advective VOCs transport that is induced in engineered remediation processes such as vapor extraction was not considered. It can be concluded that the coefficient for gas liquid mass-transfer was found to be a key parameter controlling the ability of bacteria to VOCs. Finally, it appeared that bioremediation technology of VOCs which are difficult to be decomposed by chemical methods.
A strain able to grow up m-toluate minimal medium has been isolated after selective enrichment and given the name T81X, which was later identified as pseudomonase putida according to its morphological and biochemical characteristics. After treatment with plasmied specific curing agent, mitomycin C, followed by replica plating on m-toluate and xylene minimal agar plate, T81Xstrain has been shown to harbour a curable plasmid relating to the m-toluate and xylene metabolism. Spontaneous curing frquency of this plasmid was also greatly enhanced by growing on benzoate minimal medium. After then, it was also xylene metabogrowing on benzoate minimal medium. After then, it was found to be conjugally nontransmissible. From the comparative investigation of catechol 1,2-oxygenase and catechol 2,3-oxygenase activities in wild type and cured strain on various growth substrate, it appeared that T81X strain has both of these two enzymes while cured strain has catechol 1,2-oxygenase only. Growing on m-toluate minimal medium T81X strain should carry the genetic information necessary for coding the catechol 1,2-oxygense induced by m-toluate or benzoate, on that curable plasmid.
Purpose: The purpose of this study is to investigate whether wetland has antimicrobial activity on pathogenic bacteria in the toilet bowl. Research design, data and methodology: Air-dried mud obtained from "Jilmoe Bog" wetland was packed and dissolved in the autoclaved saline. Antimicrobial susceptibility was assessed against three Gram-negative bacteria using disk diffusion method and broth dilution method. Identification of specific bacterium presented in wetland supernatant was performed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Results: Incubation of three Gram-negative bacteria with wetland supernatant inhibited bacterial growth of the bacteria, otherwise increased prevalence of specific bacterium. It was confirmed that Pseudomonas putida was presented in wetland supernatant. Conclusions: The results presented in this study might provide the possibility to utilize wetland supernatant as a bioremediation of toilet bowl bacteria.
Bioluminescence방법에 의하여 세균내에 존재하고 있는 ATP를 추출하여 그양을 정량분석하므로서 제육표면에 생존하고 있는 세균수를 신속하게 측정하는데 응용될 수 있는 가능성을 조사하기 위하여 본 실험을 실시하였으며, 그 결과는 1. Luciferin-luciferase에 작용하는 ATP농도가 $10^{-10}M$부터 $10^{-6}M$까지 증가함에 따라 발생된 빛의 양도 비례적으로 증가했다. 2 . 계육에서 분리동정된 부패균인 E. coli, H, alvei, P. putida, A. hydrophila에서 추출된 ATP 농도와 세균수와의 관계는 거의 일직선으로 나타난다. 3. E. coli에 있는 ATP양은 생존중 거의 변화하지 않는 것으로 나타났다. 4 계육의 저장기간동안 증식된 세균수에 따라 ATP 농도도 비례 적으로 증가되며 이때 상판계수(r)는 0.95로 나타났다. 5. 계육에 오염된 세균수와 ATP 농도와의 관계에서 볼 때 ATP농도가 $10^{-7}M$부근에서부터 부패가 시작된다고 사료된다. 6. 개발된 방법에 의하여 계육표면에 존재하는 세균의 ATP 농도를 측정하여 세균수를 간접적으로 신속히 ( 1시간내) 조사할 수 있다.
녹조대발생의 원인종인 남조 Microcystis aeruginosa와 규조 Stephanodiscus hantzschii 에 대한 살조 및 섭식능을 갖는 박테리아 및 섬모충을 각각 단독 및 혼합처리하고 살조능을 비교하였다. 먼저 남조 M. aeruginosa에 대하여 살조세균 Streptomyces neyagawensis와 섬모충 Stentor roeselii를단독 및 혼합처리한 결과, 각각의 단독처리가 혼합처리보다 더 높은 살조능을 나타내는 두 생물제재의 길항적 살조효과를 나타냈다. 또한 규조 S. hantzschii 에 대해 살조능을 갖는 박테리아 P seudomonas putida와 S. roeselii를 전과 동일한 방법으로 단독 및 혼합처리하여 결과를 비교하였다. 전 실험과는 다르게 두 생물제재의 혼합처리가 단독처리보다 더 높은 살조능을 보이는 두 생물제재의 상승적 살조효과를 나타냈다. 이러한 결과들은 녹조 제어를 위한 생물제재의 적용시 유용하게 이용될 것이며, 다양한 생물제재를 이용함으로서 수중생태계의 혼란을 초l소화시킬 수 있을 것으로 사료되었다.

This study evaluates the utility of an antibacterial microneedle composed of green tea extract (GT) and hyaluronic acid (HA), for the efficient delivery of GT. These microneedles have the potential to be a patient-friendly method for the conventional sustained release of drugs. In this study, a fabrication method using a mold-based technique to produce GT/HA microneedles with a maximum area of ${\sim}60mm^2$ with antibacterial properties was used to manufacture transdermal drug delivery systems. Fourier transform infrared (FTIR) spectrometry was carried out to observe the potential modifications in the microneedles, when incorporated with GT. The degradation rate of GT in GT/HA microneedles was controlled simply by adjusting the HA composition. The effects of different ratios of GT in the HA microneedles were determined by measuring the release properties. In HA microneedles loaded with 70% GT (GT70), a continuous higher release rate were sustained for 72 h. The in vitro cytotoxicity assays demonstrated that GT/HA microneedles are not generally cytotoxic to chinese hamster ovary cells (CHO-K1), human embryonic kidney cells (293T), and mouse muscle cells (C2C12), which were treated for 12 and 24 h. Antimicrobial activity of the GT/HA microneedles was demonstrated by ~95% growth reduction of gram negative [Escherichia coli (E. coli), Pseudomonas putida (P. putida) and Salmonella typhimurium (S. typhimurium)] and gram positive bacteria [Staphylococcus aureus (S. Aureus) and Bacillus subtilis (B. subtilis)], with GT70. Furthermore, GT/HA microneedles reduced bacterial growth in the infected skin wound sites and improved skin wound healing process in rat model.
The transcriptional capacities of target genes are strongly influenced by promoters, whereas few studies have focused on the development of robust, high-performance and cross-species promoters for wide application in different bacteria. In this work, four novel promoters (Pk.rtufB, Pk.r1, Pk.r2, and Pk.r3) were predicted from Ketogulonicigenium robustum and their inconsistency in the -10 and -35 region nucleotide sequences indicated they were different promoters. Their activities were evaluated by using green fluorescent protein (gfp) as a reporter in different species of bacteria, including K. vulgare SPU B805, Pseudomonas putida KT2440, Paracoccus denitrificans PD1222, Bacillus licheniformis and Raoultella ornithinolytica, due to their importance in metabolic engineering. Our results showed that the four promoters had different activities, with Pk.r1 showing the strongest activity in almost all of the experimental bacteria. By comparison with the commonly used promoters of E. coli (tufB, lac, lacUV5), K. vulgare (Psdh, Psndh) and P. putida KT2440 (JE111411), the four promoters showed significant differences due to only 12.62% nucleotide similarities, and relatively higher ability in regulating target gene expression. Further validation experiments confirmed their ability in initiating the target minCD cassette because of the shape changes under the promoter regulation. The overexpression of sorbose dehydrogenase and cytochrome c551 by Pk.r1 and Pk.r2 resulted in a 22.75% enhancement of 2-KGA yield, indicating their potential for practical application in metabolic engineering. This study demonstrates an example of applying bioinformatics to find new biological components for gene operation and provides four novel promoters with broad suitability, which enriches the usable range of promoters to realize accurate regulation in different genetic backgrounds.
토양의 특성과 재배 환경이 상이한 13개 토양을 대상으로 미생물의 다양성을 분석하여 토양 환경별 미생물상 지표의 기준을 설정하기 위해서 실험한 결과는 다음과 같다. 희석 평판법에 의한 토양별 미생물의 군집은 호기성 세균은 $2{\sim}27{\times}10^6$, 형광성 Pseudomonas는 $1{\sim}1364{\times}10^5$, 그람 음성 세균은 $1{\sim}126{\times}10^4$, 중온성 Bacillus는 $1{\sim}110{\times}10^5$ 범위로 토양간에 현저한 차이를 보였다. 특히 그람 음성 세균은 충북 음성 고추 재배지 (4번 토양)과 충북 괴산 고추 재배지 (6번) 토양이 현저히 많았고, 형광성 Pseudomonas는 경남 진주의 시설재배지 (7번 토양)에서 현저히 높은 군락을 형성하였고, 이 들 토양은 작물의 생육이 양호한 토양이었다. 따라서 시설재배인 토양에서는 그람 음성 형광성 Pseudomonas 및 mesophillic bacillus의 군집이 토양 환경 및 작물 생육과 미생물의 지표로서 가능성을 제시하였다. 13개 토양에서 분리한 579 균주 중 호기성 세균은 Agrobacterium 102 isolates, Bacillus 112 isolates, Pseudomonas 32 isolates, Kocuria 44 isolates, Paenibacillus 34 isolates 등의 다양한 군집을 이루고 있었고, 또한 624균주의 그람 음성 세균은 Pseudomonas 속이 51%로 제일 많은 군집을 이루고 있었는데 이 들 속내에는 Pseudomonas putica, Pseudomonas fluorescens 등이 많았고 그 다음으로는 Senotrophomonas maltophilia, Bukhodenis cepacia 순으로 다양한 군집을 이루고 있었다. RAPD 분석에 의한 Pseudomonas의 종 다양성은 토양에 따라서 4-31개의 균주 types을 보였으며 특히 4, 6번 토양에서 균주의 다양성이 높게 나타났다. 특히 형광성 Pseudomonas의 군집을 많이 이루었던 7번 토양에서는 26균주에서 5개 균주 types을 보여 종 다양성이 단수하였다. 토양 환경별 미생물 군집은 논 토양에서 신규 세균으로 추정되는 다수의 새로운 분리균이 출현되어 새로운 지표 미생물의 가능성을 제시 하였으며, 또한 MIDI 분석에서 no match 되는 분리균에 대한 미생물 자원의 이용성이 높이 평가되었다.
