  • In order to observe the time of the first appearance of Bowie positive pepsinogen granules in the gland cells of the chicken proventriculus, histological examination was performed on the chicken embryo proventriculus at 7, 10, 14, 18, 19, 20 and 21 days postincubation. The first appearance of Bowie positive pepsinogen granules in the gland cells of the chicken proventriculus was at 21 days postincubation.

Effects of Dietary Supplementation with Astaxanthin on Histamine Induced Lesions in the Gizzard and Proventriculus of Broiler Chicks

  • Astaxanthin (ASX) is a xanthophyll pigment isolated from crustaceans and salmonids. Owing to its powerful antioxidant activity, ASX has been reported to have the potential to protect against gastric ulcers and a variety of other illnesses. Histamine (His) is a dietary factor that causes gastric erosion and ulceration in young chicks. In this study, we examined whether ASX had protective effects on dietary histamine-induced lesions in the gizzard and proventriculus of broiler chickens. Four experimental treatment groups were planned: basal diet (BD), BD+His, BD+ASX, and BD+ASX+His, with four chicks (5 days old) in each group and three replications (i.e., a total of 12 chicks per group). The BD was supplemented with either 0.4% His or 100 ppm ASX. The birds were fed ad libitum for 3 weeks, and diets contained no antimicrobial compounds. Supplementing the diet with His significantly decreased body weight gain, but increased the weights of the gizzard and proventriculus of the chicks as compared with those of chicks in the BD group (p<0.05). ASX did not affect His-dependent changes in chick body weight or weights of the gizzard and proventriculus. The loss of gastric glands in the proventriculus, which was observed in His-treated chicks, was not prevented by ASX administration. The frequency of proventricular ulceration, however, was lowered by treatment with ASX, without significant differences between the two supplementation levels. In conclusion, our data showed that ASX might be helpful for alleviating structural damage to the digestive system in poultry under certain stressful conditions.

Immunohistochemical study on the stomach of the duck, Anas platyrhynchos platyrhyncos Linne (청둥오리 위 내분비세포에 대한 면역조직화학적 연구)

  • The relative frequency and distribution of bovine chromogranin(BCG)-, serotonin-, somatostatin(SOM)-, and S-100 protein-immunoreactive cells in the proventriculus, gizzard and proventriculus-gizzard junction(P-G junction) of the duck(Anas platyrhynchos platyrhyncos, Linne) were studied using immunohistochemical methods. BCG-, and SOM-immunoreactive cells were detected in all ages of this study in the proventriculus and the gizzard, and BCG-immunoreactive cells were also detected in the P-G junction. SOM-immunoreactive cells were detected rarely on occasions only in 1 week, 2 weeks, 3 weeks and 9 weeks after hatching. But no S-100 protein-immunoreactive cells were detected in this study.

  • Clinically healthy 12 female Manina breed chicken (6 of 75 days old : group 46 of 145 days old: group B, female) were examined to establish physiological basic data on organ tissues total Creatine phosphokinase (CPK) activities and CPK isoenzymes fractions. The tissues examined were the Lung, Heart, Liver, Proventriculus, Gizzard, Duodenum, Colon and Muscle. The results obtained were summarized as follows : 1. Total CPK activities were high with decreasing order of the Muscle > Proventriculus >Giz-zard >Heart >Duodenum> Colon >Lung >Liver in group A and Muscle >Proventriculus >Giz-zard >Heart > Colon >Duodenum > Lung > Liver in group B. Significance of total CPK activities in group difference was only found Colon, group B showed higher values than that of group A (p< 0.01). 2. In the pattern of CPK isoenzymes fractions, Lung, Heart, Liver, Proventriculus, Gizzard, Duodenum and Muscle were high with decreasing order of CK2 >CK3, Colon showed the pattern with decreasing order of CK3 >CK2. Significance of CPK isoenzymes fractions in group difference was only found Liver, CK2 in group B(P<0.01) and CK3 in group A(P<0.01) were higher than that of the other group.

A Case Report of Proventriculotomy for Impaction of Proventriculus in Ostrich (타조에서 전위절개술을 통한 매복의 치유예)

  • A 120 kg, 3-year-old, male ostrich (Struthio camelus) was submitted to the veterinary teaching hospital, Chonbuk national university, for chronic weight loss, anorexia, small hard fecal pellets, and lethargy. Illness was first noticed approximately 2 weeks before submission. Although the bird was able to stand, he was extremely weak and cachectic. Blood examination indicated that the bird was dehydration (PCV: 55%). Fluoroscopic and lateral abdominal radio-graphic view indicated impaction of distended proventriculus with bigger rocks than normal. Anesthesia was induced with ketamine (10 mg/kg, IV)-xylazine (1 mg/kg, IV) combination and maintained with ketamine (10 mg/kg, IV) at intervals of 8 to 15 minutes during the entire procedure. After induction of general anesthesia, the bird was positioned in dorsal recumbency. The skin was incised approximately 15cm caudal to the caudal margin of the sternum and extended 20 to 25 cm caudally. After the proventriculotomy was peformed, the impacted material that weighed 3kg contained rocks in 4 to 6cm diameter was removed. The incision of proventriculus was closed in 2 layers. Cephalothin sodium (30 mg/kg, IV, q12hrs) and dexamethazone (2 mg/kg, IM, q24hrs) was administered before and after 5 days of the operation. The bird was eating well and defecating normal feces after 3 weeks of the surgery

Ontogeny, distributions and relative frequencies of the somatostatin-immunoreactive cells in the proventriculus of the chicken embryos with incubation periods (닭 태자 샘위의 somatostatin 면역반응세포의 출현시기, 분포 및 상대적 빈도에 관한 연구)

  • Ontogeny, distributions and relative frequencies of somatostatin-immunoreactive cells were investigated in the proventriculus of the chicken embryos with incubation periods. Samples were taken from 10 groups(10 days of incubation to hatching) and studied by immunohistochemical methods. The findings were as follows. Somatostatin-immunoreactive cells were observed from 12 days of incubation in the proventricular glands and after that increased with incubation periods. The first observation time of these cells in the epithelium were at 15 days of incubation in the basal portion but in 16 and 17 days of incubation, no immunoreactive cells were observed in the epithelium but after that a few immunoreactive cells were observed in the basal portion and gastric gland regions. The shapes of these cells were spherical to spindle in the proventricular glands and spherical to round in the epithelium and gastric gland.

Immunohistochemistry and RT-PCR for pathogenesis of Newcastle disease in chickens (닭 뉴캣슬병의 발병기전 규명을 위한 RT-PCR 및 면역조직화학적 연구)

  • The present experiment was carried out to study the pathogenesis of Newcastle disease(ND), ND virus (NDV) antigens and genes in various organs from NDV inoculated chickens were detected by immunohistochemistry and RT-PCR. Immunohistochemically, NDV antigens were detected in the spleen, thymus, cecal tonsil, proventriculus, trachea and lungs at 12 hour post-inoculation (hpi). Viral antigens were localized mainly in the cytoplasm of lymphocytes and macrophages. After 48 hpi, clinical findings of the affected chickens were open-mouth breathing, conjunctivitis, watery diarrhea and edema around the eye and neck. After 72 hpi, chickens showed muscular tremor, paralysis of the legs and wings, and coma. Histopathological results consist of multi-focal necrosis with hemorrhages in lymphoid aggregates of the intestinal tracts, necrosis of the lymphoid tissues, neuronal degeneration and necrosis, and perivascular cuffing. Using RT-PCR, virus genes were detected in the spleen and proventriculus at 48 hpi, and in the brain at 60 hpi.

An immunohistochemical study on the gastrointestinal endocrine cells in the bean goose, Anser fabalis Latham (기러기 위장관 내분비세포에 관한 면역조직화학적 연구)

  • The regional distributions and relative frequencies of the gastrointestinal endocrine cells in the bean goose (Anser fabalis Latham) were investigated by immunohistochemical methods using bovine Sp-1/chromogranin (CG), serotonin, gastrin, cholecystokinin (CCK)-8, somatostatin and glucagon antisera. BCG-immunoreactive cells were widespread throughout the gastrointestinal tract (GIT) with moderated frequencies except for the gizzard and proventriculus which were a few frequencies. Serotonin-immunoreactive cells were detected throughout the GIT except for the proventriculus and gizzard. These cells were observed in the pylorus with rare frequencies but numerous cells were detected in the intestinal tract. Gastrin-immunoreactive cells were restricted to the gizzard, pylorus and duodenum. These cells were most predominant in the pylorus and a few or rare in the gizzard and duodenum, respectively. CCK-8-immunoreactive cells were observed from the gizzard to ileum. The highest frequencies of endocrine cells were observed in the duodenum. These cells were increased from the gizzard to duodenum but thereafter decreased. Somatostatin-immunoreactive cells were detected in the GIT except for the large intestine. In the proventriculus and pylorus, numerous immunoreactive cells were demonstrated but a few cells were present in the other regions. Glucagon cells were observed in the gizzard, pylorus, ileum, colon and rectum with a few or moderated numbers.

Candidiasis in layer chickens in Korea (산란계에서의 칸디다증 국내 발생 사례)

  • Candidiasis is a mycosis caused by the mycelial yeast of the Candida genus which is opportunistic pathogen of humans, animals, and birds. Under some conditions such as prolonged antibiotic therapy, overcrowding, and immunosuppression, the opportunistic Candida can cause disease. Chicken candidiasis is sporadically occurred and characterized by unsatisfactory growth, listlessness, roughness of feathers, and death. A case of 23 weeks old layer with history of increased mortality and anemia was submitted to our Lab. At necropsy, the characteristic lesions were observed in the crop and proventriculus. The whitish pseudomembrane, that are peeled easily, was found in the crop. Proventriculus was swollen and the mucosa was covered with hemorrhagic exudate. The histological changes of the affected crop are epithelial hyperplasia, hydropic degeneration, and mycelia formation. Smears made from the necrotic mucosal surfaces of the crop revealed the presence of large number of yeast cells and mycelia. Pure cultures of yeast colonies were obtained from the potato dextrose agar. The yeast cells were identified as Candida albicans by gene sequencing. To our knowledge, this is the first report of candidiasis in chickens with anemia in Korea.

Morphological Study on the Mast Cell of Proventriculus in Pheasant (Phasianus colchicus) (꿩 전위의 비만세포에 관한 형태학적 연구)

  • Mast cells have been studied extensively in various animals including rats and mice, whereas little is known the morphological data about pheasant mast cells. Here, morphological features of Korean pheasant mast cells are described in this study using light and electron microscopes. For light microscopy, mast cells had many metachromatic granules stained with toluidine blue in the cytoplasm. The fixation with $10\%$ neutral buffered formalin blocked staining of most mast cells but a modified Karnovsky solution proved to be a good fixative. In Korean pheasants, toluidine blue stained more mast cells than did alcian blue. For electron microscopy, the mast cells of the Korean pheasant were round, oval, spindle-like and irregular form and occasionally had a few short cytoplasmic processes. These cells had membrane-bounded granules and poorly developed organells. Some granules in the cytoplasm of the mast cells had bilayer membrane. Most granules were round shape and the membrane of several granules was concave or convex. The granules were composed of three parts, homogenous, particulate and reticular pattern.
