Influence of Tyrosol on Cell Growth Inhibition of KB Human Oral Cancer Cells

  • Tyrosol, a phenylethanoid and a derivative of phenethyl alcohol, possesses various biological properties, such as anti-oxidative and cardioprotective activity. Olive oil is the principal source of tyrosol in the human diet. However, so far the anti-cancer activity of tyrosol has not yet been well defined. This study therefore undertakes to examine the cytotoxic activity and the mechanism of cell death exhibited by tyrosol in KB human oral cancer cells. Treatment of KB cells with tyrosol induced the cell growth inhibition in a concentration- and a time-dependent manner. Furthermore, the treatment of tyrosol induced nuclear condensation and fragmentation of KB cells. Tyrosol also promoted proteolytic cleavage of procaspase-3, -7, -8 and -9, increasing the amounts of cleaved caspase-3, -7, -8 and -9. In addition, tyrosol increased the levels of cleaved PARP in KB cells. These results suggest that tyrosol induces the suppression of cell growth and cell apoptosis in KB human oral cancer cells, and is therefore a potential candidate for anti-cancer drug discovery.

Purification and Properties of HPS (Halitosis Prevention Substance) Isolated from Cumin (Cuminum cyminum L.) Seed

  • Halitosis is mainly caused by the presence of volatile sulfur-containing compounds (VSC's) produced by proteolytic periodontopathic bacteria in the oral cavity. Various mouth-rinses have been offered on the market as solutions to reduce halitosis. The aim of this study was to find a potent substance for the prevention of halitosis. The halitosis prevention substance (HPS) from cumin seed powder was purified by solvent extraction, silica gel column chromatography and preparative TLC to yield an oil phase (0.98%). Instrumental analysis such as FT-IR, $^1H$-NMR and $^{13}C$-NMR showed that HPS contained an -OH group, -HC=CH-, -COO-, and long chain acyl group. HPS was therefore determined to be 2-hydroxyethyl-${\beta}$-undecenate. HPS inhibited the growth of Fusobacterium nucleatum and Porphyromonas gingivalis, by 72.44% and 64.37% at $1{\times}10^{-2}\;M$, and by 99.85% and 91.62% at $5\;{\times}\;10^{-2}\;M$, respectively. It also inhibited the activity of L-methionine-${\alpha}$-deamino-${\gamma}$-mercaptomethane-lyase (METase), which was produced by oral microbes. Furthermore, the VSC production by oral microbes in the human mouth air decreased with increasing HPS concentration. These results suggested that HPS from cumin seed is an efficient halitosis prevention agent.

  • The alkaline protease produced by Sarcodon aspratus(Berk) S. Ito. was purified from its fruit bodies. The enzyme was purified by using ammonium sulfate fractionation, tris-acryl CM-cellulose column chromtography and chromatofocusing. The protease migrated as one major band with a molecular weight of about 29,000 dalton on sodium dodecylsulfate-polyacrylamide gel electrophoresis. The amino acid sequence of the N-terminal residues(21) of the enzyme was determined by automated sequence analysis. The sequence was Val-Thr-Thr-Lys-Gln-Thr-Asn-Ala-Pro-Trp-Gly-Leu-Gly-Asn-Ile-Ser-Thr-Thr-Asn-Lys-Leu. Comparison of this sequence with the N-terminal sequence of the p-roteinase K from Tritirachium album showed high similarity, i. e. 57.8% identical residues. The protease displayed a relatively high stability in sodium dodecyl sulfate.

고감성 Easycare Wool의 제조 및 특성 연구 (A Study on the Manufacturing and Properties of Hightech Easycare Wool)

  • To manufacture hightech easycare wool, there are several methods which use strong oxidising agent or the resin treatment, however, neither are environmentally friendly methods. Moreover it may deteriorate the handle. The aim of this study is to manufacture the hightech easycare wool using the modified Fenton method which can be formed by hydrogen peroxide and ferric sulfate and enzyme treatment. The method was pretreated by ferric sulfate on the wool surface and then the surface of wool scale was selectively removed by ferric ion catalyst. Subsequently the Enchiron which is one of the proteolytic enzymes was treated on the wool surface. The treated wool had the result of having optimum weight loss and excellent whiteness and good handle. Therefore implications of these results suggest that this method using the modified Fenton method and enzyme treatment may be one way of manufacturing the hightech easycare wool.

Partial Purification and Characterization of a Cysteine Protease Inhibitor from the Plerocercoid of Spirometra erinacei

  • Helminthic cysteine proteases are well known to play critical roles in tissue invasion, nutrient uptake, and immune evasion of the parasites. In the same manner, the sparganum, the plerocercoid of Spirometra mansoni, is also known to secrete a large amount of cysteine proteases. However, cysteine protease inhibitors regulating the proteolytic activities of the cysteine protease are poorly illustrated. In this regard, we partially purified an endogenous cysteine protease inhibitor from spargana and characterized its biochemical properties. The cysteine protease inhibitor was purified by sequential chromatographies using Resource Q anion exchanger and Superdex 200 HR gel filtration from crude extracts of spargana. The molecular weight of the purified protein was estimated to be about 11 kD on SDS-PAGE. It was able to inhibit papain and 27 kDa cysteine protease of spargana with the ratio of 25.7% and 49.1%, respectively, while did not inhibit chymotrypsin. This finding suggests that the cysteine protease inhibitor of spargana may be involved in regulation of endogenous cysteine proteases of the parasite, rather than interact with cysteine proteases from their hosts.

Use of Glucose Oxidase Immobilized on Magnetic Chitosan Nanoparticles in Probiotic Drinking Yogurt

  • The aim of this study was to investigate the effect of glucose oxidase (GOX) immobilized on magnetic chitosan nanoparticles (MCNP) on the viability of probiotic bacteria and the physico-chemical properties of drinking yogurt. Different concentrations (0, 250, and 500 mg/kg) of free and immobilized GOX were used in probiotic drinking yogurt samples. The samples were stored at $4^{\circ}C$ for 21 d. During storage, reduction of the number of probiotic bacteria in the samples with enzyme was lower than the control sample (without enzyme). The sample containing 500 mg/kg immobilized enzyme had the highest number of Bifidobacterium lactis and Lactobacillus acidophilus. The samples containing immobilized enzyme had lower acidity than other samples. Moreover, moderate proteolytic activity and enough contents of flavor compounds were observed in these samples. It can be concluded that use of immobilized GOX is economically more feasible because of improving the viability of probiotic bacteria and the physico-chemical characteristics of drinking yogurt.

Biological Potential of Novel Specific Casein-Derived Peptides

  • This study aimed to evaluate the biological potential of functional food, i.e., specific peptides obtained from the hydrolysis of milk protein, by assessing their antioxidant and antibacterial properties. For the preparation of casein hydrolysates, commercial enzymes were added to 10% casein solution in a 1:200 (w/v) ratio, and samples were collected each hour. Based on the assessment of the degree of hydrolysis (DH) of casein hydrolysates, it was observed that the concentration of all enzymatic hydrolysates increased rapidly from 30 to 40 minutes. However, no change was observed in their concentrations after 150 minutes. Protamex® and Neutrase® exhibited the highest DH when compared to other enzymes. Furthermore, SDS-PAGE was performed for analyzing the proteolytic pattern of each enzyme, except for Flavourzyme®, and peptides in the size range of 20-25 kDa were identified. Subsequently, peptides produced by two enzymes were isolated using a preparative liquid chromatography system. Overall, NF3, NF4, PF5, and PF6 showed higher antioxidant potential than other peptide fractions. Moreover, NF7 and PF3 exhibited the highest antibacterial activity. In this study, we evaluated the biological potential of novel casein-derived peptides that may find application in the food and healthcare industry.

Immunostimulatory Effect of Ovomucin Hydrolysates by Pancreatin in RAW 264.7 Macrophages via Mitogen-Activated Protein Kinase (MAPK) Signaling Pathway

  • Ovomucin (OM), which has insoluble fractions is a viscous glycoprotein, found in egg albumin. Enzymatic hydrolysates of OM have water solubility and bioactive properties. This study investigated that the immunostimulatory effects of OM hydrolysates (OMHs) obtained by using various proteolytic enzymes (Alcalase®, bromelain, α-chymotrypsin, Neutrase®, pancreatin, papain, Protamax®, and trypsin) in RAW 264.7 cells. The results showed that OMH prepared with pancreatin (OMPA) produced the highest levels of nitrite oxide in RAW 264.7 cells, through upregulation of inducible nitric oxide synthase mRNA expression. The production of pro-inflammatory cytokines such as tumor necrosis factor-α and interleukin-6 were increased with the cytokines mRNA expression. The effect of OMPA on mitogen-activated protein kinase signaling pathway was increased the phosphorylation of p38, c-Jun NH2-terminal kinase, and extracellular signal-regulated kinase in a concentration-dependent manner. Therefore, OMPA could be used as a potential immune-stimulating agent in the functional food industry.

Comparison of changes in functional characteristics of fermented soybean with different microbial strains

  • The purpose of this study was to compare the effect of solid-state fermentation on soybean using three microbial strains under four different fermentation times. Soybean was fermented for 12, 24, 36 or 48 hours with highly proteolytic microbes, either Bacillus amyloliquefaciens (BA), B. subtilis (BS), or B. subtilis var. natto (BN), and levels of total protein concentration, protein distribution, and antioxidant activity were analyzed. Total protein was highest in the BS 12 h group (9.21 ㎍·µL-1) and lowest in BN 48 h (6.80 ㎍·µL-1), respectively (p < 0.001). Furthermore, three microbes decomposed large molecular weight proteins as well as major allergens of soybean such as β-conglycinin, Gly m Bd 30K, and glycinin. Each treatment group showed the highest degradation rate at 48 h fermentation and among the three microbes, BS showed a relatively higher degradation rate. The radical scavenging ability, known as an indicator of antioxidant activity, showed a significant increase in all treatment groups except BA 24 h. The results from this study suggest that protein concentration, and degradation and antioxidant activity were affected by different types of microbial trains and fermentation period and that B. subtilis fermentation might be the most effective way to increase nutritional and functional properties of soybean.

Bacillus subtilis MJP1이 생산하는 Bacteriocin-Like Substances (Characterization of Bacteriocin-Like Substances Produced by Bacillus subtilis MJP1)

  • 메주로부터 항곰팡이 활성을 보이는 균주 1종과 이에 감수성을 나타내는 곰팡이 3종을 분리하였다. 분리된 균주는 형태학적, 생화학적 특성 조사와 16S rRNA 염기서열 결정을 통한 균주 동정결과 Bacillus subtilis MJP1으로 명명하였고, 3종의 곰팡이는 ITS-5.85 rRNA염기서열 분석을 통하여 Aspergillus petrakii PF-1, A. ochraceus PF-2, 그리고 A. nidulans PF-3로 명명하였다 B. subtilis MJP1은 곰팡이 에 대한 강한 저해활성 뿐만 아니라 Candida 속 효모들과 그람 양성균에 대한 넓은 범위의 저해활성을 나타내었다. B. subtilis MJP1의 생육에 따른 항균 활성을 측정한 결과 항진균 활성은 배양 16시간부터 최대 활성(3,200 AU/ml)을 나타내어 균이 사멸기에 접어든 후에도 활성을 그대로 유지한 반면, 항세균 활성은 대수기 중반인 12시간부터 25시간까지 가장 높은 활성 (1,600 AU/ml)을 보이다가 72시간 이후에는 활성을 상실하였다. pH 안정성 실험에서 항진균 활성과 항세균 활성 모두 pH $6{\sim}10$ 구간에서 비교적 안정한 결과를 보였으나, 열처리 실험에서 항진균 활성은 영향을 받지 않은 반면, 항세균 활성은 $30^{\circ}C$ 이상의 온도에서는 불안정한 결과를 보였다. 각종 효소에 대한 안정성 실험에서 항진균 활성은 어떠한 효소에도 영향을 받지 않았으나, 항세균 활성은 단백분해효소 처리 후에 활성이 실활 됨으로써 항균 물질이 단백질성 물질임을 추정하였다. $C_{18}$ Sep-Pak column으로 부분 정제한 항균 물질이 항진균 활성과 항세균 활성을 나타내므로 소수성을 가지는 물질임을 알 수 있으며, Tricine-SDS-PAGE및 direct detection을 통하여 항진균 물질의 분자량은 약 2.4 kDa 정도이며, 항세균 물질의 분자량은 약 4.5 kDa으로_ 확인되었다. 따라서 B. subtilis MJP1은 항진균 활성과 항세균 활성을 가진 bacteriocin-like substances를 생산함을 알 수 있고 이와 같은 새로운 항미생물 물질은 천연 식품보존제 및 사료보존제 뿐만 아니라 항생제 대체 의약품으로도 활용이 기대되며, 이를 위하여 향후 이 물질들의 보다 정확한 구조 및 특성 규명 등의 연구가 필요하다.
