FcBP consisting of 13 amino acids specifically binds to Immunoglobulin G Fc domain. Initially, we utilized this peptide for preparation of antibody chip as a PEG composite for enhanced solubility. After then, the peptide conjugate was immobilized on agarose resin, resulting in highly efficient affinity column for antibody purification. The efficiency was comparable to commercial Protein A column. Recently, this peptide was conjugated with cell penetratingpeptide (CPP) on a backbone of GFP, affording antibody transducer, which carries antibody into live cells by simple mixing of antibody and the transducer in cell culture media. Antibody transduction into cells was monitored by live cell imaging. More recently, the FcBP was fused to ferritin cage, which consists of 24 ferritin protein molecules. The FcBP-ferritin cage showed greatly increased binding affinity to human IgG. Its binding was analyzed by QCM and SPR analysis. Finally, it was selectively delivered by Herceptin to SKBR3, a breast cancer cell, over MCF10A, non-tumorigenic cells.
Baculovirus 발현 시스템은 박테리아 발현 시스템, 특히 복잡한 번역 후 변형을 필요로 하는 것과 비교하여 다량의 재조합 단백질을 생성하는 빠르고 비용 효율적인 방법을 포함하는 많은 알려진 장점을 갖는다. 특히 재조합 baculovirus는 광범위한 포유류 세포 유형에서 벡터를 전달하고 재조합 단백질을 발현 할 수 있다. 본 연구에서는 pcDNA3.1로부터 재구성된 baculovirus 벡터를 사용하였는데 이 벡터는 cytomegalovirus (CMV) 프로모터, uroplakin II promoter, polyhedron promoter, 수포 구내염 바이러스 G (VSVG), 녹색 형광 단백질 (EGFP), 단백질 전달 도메인 (PTD) 유전자 등 다양한 유전자들로 재조합 되어 개발되었다. 이러한 재구성 된 벡터를 다양한 세포 및 세포주에 감염시켰다. 이렇게 개발된 baculovirus 벡터는 재조합된 유전자들의 전이성 및 발현성을 기존의 일반적인 벡터와 비교하여 분석하였다. 본 연구결과로 이렇게 개발된 baculovirus 벡터는 기존의 대조군 벡터보다 전이성 및 발현성면에서 더 높은 효율을 갖는다는 것을 확인하였다.

We examined the ways in which fenobam could promote not only the transduction of PEP-1-FK506BP into cells and tissues but also the neuroprotective effect of PEP-1-FK506BP against ischemic damage. Fenobam strongly enhanced the protective effect of PEP-1-FK506BP against $H_2O_2$-induced toxicity and DNA fragmentation in C6 cells. In addition, combinational treatment of fenobam with PEP-1-FK506BP significantly inhibited the activation of Akt and MAPK induced by $H_2O_2$, compared to treatment with PEP-1-FK506BP alone. Interestingly, our results showed that fenobam significantly increased the transduction of PEP-1-FK506BP into both C6 cells and the hippocampus of gerbil brains. Subsequently, a transient ischemic gerbil model study demonstrated that fenobam pretreatment led to the increased neuroprotection of PEP-1-FK506BP in the CA1 region of the hippocampus. Therefore, these results suggest that fenobam can be a useful agent to enhance the transduction of therapeutic PEP-1-fusion proteins into cells and tissues, thereby promoting their neuroprotective effects.

암의 유전자치료에서 암세포로의 선택적 유전자전달 매체의 부족은 치료효과의 감소를 야기하는 문제이다. 본 연구에서는 난소암 유전자치료의 효율을 높이기 위한 목적으로 난소암세포로 선택적인 유전자전달을 하도록 개량된 아데노바이러스 벡터를 제조하고, 그 효율성을 난소암세포주를 이용하여 조사하였다. 난소암세포에 과다발현하는 분자인 ErbB receptor를 표적하도록 아데노바이러스 외피단백질 fiber에 ErbB receptor에 대한 ligand인 heregulin으로부터 유래한 펩티드를 부착하였다. 53개의 아미노산으로 구성된 외부 펩티드를 fiber에 부착하였을 때 바이러스 감염에 중요한 기능을 하는 fiber 삼량체 구조 형성에 영향을 미치지 않았다. Fiber를 조작한 개량 아데노바이러스는 야생형 fiber를 가진 1세대 아데노바이러스 벡터에 비해 선택적으로 난소암으로 유전자를 전달하는 비율이 증가하였다. 특히 항암제에 저항성을 가진 난소암세포주 OVCAR3에서 유전자전달 효율이 약 5배 증가되었다. 따라서 난소암의 유전자치료에서 개량된 아데노바이러스로 치료 유전자를 전달하면 치료의 효율성을 향상시킬 수 있을 것이다.
There has been interest in developing gene therapy based on naked plasmid DNA for treating serum protein deficiencies and human erythropoietin (hEPO) is one of the candidate for gene therapy being Investigated most enthusiastically. We constructed novel plasmid DNA vectors pVAC-hEPOI/II/III which contain one, two, three hEPO gene(s) respectively for producing high level expression and secretion of hEPO in vitro and in vivo. NIH3T3 and COS7 cells were transfected transiently with these vectors and increase in hEPO expression in medium reached 2-5 fold in comparison with pSecTagB-hEPO. Intra muscular administrations of pVAC-hEPOI/II/III vectors into mice resulted in high level secretion of hEPO in the serum and corresponding increases in hematocrit level. In conclusion the transduction efficiency of naked plasmid vectors is one of the critical factors of a gene delivery system and these novel plasmid vectors will contribute to various gene therapy based on naked plasmid DNA.
glandular kallikrein에 광범위한 상동성을 가지는 인간 전립성 산성 인산 가수분해 효소는, 전립선암의 대표적인 혈청 biomarkers이다. 수지상세포는 유력한 항원 제시 세포이며, 바이러스, 미생물 병원체 및 종양에 대하여 면역 계통에서 강력한 T 세포 응답을 유도할 수 있다. 따라서, 종양 특이적인 항원으로 감작된 수지상세포를 이용한 면역요법은 anti-tumor 면역 유도를 위한 강력한 방법중의 하나이다. 크레아젠(주)에서 개발된 CTP (세포막 투과성 펩티드) 기술은 세포 내로의 높은 침투 효율성을 가지며 핵산이나 단백질과 같은 생체 고분자 물질을 접합하여 세포내로 수송할 수 있는 기술이다(36). 하지만 활성형의 인간 전립성 산성 인산 가수분해 효소는 세포사멸을 매개할 수 있기 때문에, 본 연구진은 항암 치료용 백신으로의 수지상세포 감작을 위해 비활성형 형태의 다중체 (multimer) 항원을 개발하였다. 본 연구에서, 수지상 세포의 감작과 활성화에 안전하고 효율적인 다중체 형태 (multimeric form)의 세포막 투과성 펩티드가 융합된 인간 전립성 산성 인산 가수분해 효소를 얻기 위한 정제공정을 기법을 개발하였고 젤 여과 크로마토그래피, western blot과 Dynamic Light Scattering을 이용하여 확인하였다. 아울러, 정제된 다중체 형태 (multimeric form)의 세포막 투과성 펩티드가 융합된 인간 전립성 산성 인산 가수분해 효소는 수지상 세포의 감작시 세포질 내로 효과적으로 침투되었다. 결과적으로 세포의 사멸의 부작용이 없이 MHC class I 분자를 통해 수지상세포의 표면으로 효과적으로 제시되었다.
재조합 배큘로 바이러스는 배양 된 곤충 세포에서 이종 유전자를 발현하는데 널리 사용된다. 재조합 배큘로 바이러스는 광범위한 포유류 세포 유형에서 재조합 단백질의 발현을위한 유전자 전달 벡터로서 작용할 수있다. 바큘로 바이러스 시스템은 안전성, 대규모 및 높은 수준의 유전자 발현 관점에서 중요한 이점을 갖는다. 본 연구에서는 pOPINEneo-3C-GFP 벡터로부터 재구성 된 바큘로 바이러스 벡터를 사이토 메갈로 바이러스 (CMV) 프로모터, 강화 된 녹색 형광 단백질 (EGFP) 및 p53과 NcoI 및 XhoI로 재조합시켰다. 이러한 재조합 벡터를 다양한 세포 및 세포주에 감염시켰다. 이와 같이 개발 된 바큘로 바이러스 벡터는 재조합 유전자의 전이 및 발현을 통상적 인 벡터와 비교하여 분석 하였다. 이러한 결과는 바큘로 바이러스 벡터가 대조군 벡터보다 전이 및 전이에서 더 높은 효율을 갖는다는 것을 시사한다. 본 연구는 과학 기술부, 한국 정보 기술 진흥 기금 (MSIP)이 후원하는 한국 연구 재단(NRF)을 통해 중견 연구원 프로그램 (NRF-2016R1A2B4016552)을 통해 지원되었다.
목적: 줄기 세포에서 렌티바이러스와 아데노바이러스를 이용해 유전자를 전달하였을 때 유전자 발현 정도를 정량적으로 비교하는 연구는 보고되지 않았다. 저자들은 쥐의 중간엽줄기세포(이하 rMSC)에 사람 나트륨/옥소 공동수송체 (이하 hNIS) 유전자를 렌티바이러스와 아데노바이러스를 통하여 전달하고 발현 정도를 정량적으로 비교해 보았다. 대상 및 방법: hNIS를 발현하는 rMSE (lenti-hNIS-rMSC)의 생산은 hNIS유전자를 렌티바이러스 벡터인 pLenti/UbC/V5-DEST (Invitrogen)에 클로닝하여 pLenti-hNIS를 얻은 후 rMSC에 감염시켜서 3주간 blasticidin으로 선별하였다. hNIS를 발현하는 재조합 아데노바이러스(Rad-hNIS)는 homologous recombination 방법에 의하여 생산하였으며 Rad-hNIS의 rMSC에 대한 transduction efficiency는 Rad-GFP를 사용하여 MOI 1, 5, 20, 고리고 100에서 평가하였고 그 결과는 각각 $19.1{\pm}4.7%$, $54.0{\pm}6.4%$, $85.7{\pm}8.7%$, 그리고 $98.4{\pm}1.3%$이었다. 두 바이러스 전달 시스템에서 hNIS의 발현정도를 ummunocytochemistry, western blot 그리고 방사성옥소 섭취실험을 통해 평가하였다. 결과: Immunocytochemistry와 western blot으로 hNIS 단백질의 양을 비교하였을 때 lenti-hNIS-rMSC에서 발현하는 hNIS 단백질의 양은 Rad-hNIS를 rMSC에 MOI 20으로 감염시킨 경우보다는 많았으나 MOI 50 보다는 작았다. 그러나 방사성옥소 섭취실험에서 lenti-hNIS-rMSC ($29,704{\pm}6,659\;picomole/10^6\;cells$)는 MOI 100의 Rad-hNIS를 rMSE에 감염시킨 경우($6,168{\pm}2,134\;picomole/10^6\;cells)$ 보다도 높은 섭취율을 보였다. 결론: 렌티바이러스를 통하여 hNIS를 rMSC에서 발현하도록 했을 때 아데노바이러스를 전달 벡터로 사용한 경우에 비하여 단백질 발현 양은 상대적으로 적었으나 hNIS의 기능은 더 우수하였다. hNIS를 리포터 유전자로 사용한 줄기세포추적은 벡터에 따른 유전자 발현효율의 차이를 고려하고 시행되어야 할 것으로 생각한다.

All living organisms use numerous signal-transduction pathways to sense and respond to their environments and thereby survive and proliferate in a range of biological niches. Molecular dissection of these signalling networks has increased our understanding of these communication processes and provides a platform for therapeutic intervention when these pathways malfunction in disease states, including infection. Owing to the expanding availability of sequenced genomes, a wealth of genetic and molecular tools and the conservation of signalling networks, members of the fungal kingdom serve as excellent model systems for more complex, multicellular organisms. Here, we employed Cryptococcus neoformans as a model system to understand how fungal-signalling circuits operate at the molecular level to sense and respond to a plethora of environmental stresses, including osmoticshock, UV, high temperature, oxidative stress and toxic drugs/metabolites. The stress-activated p38/Hog1 MAPK pathway is structurally conserved in many organisms as diverse as yeast and mammals, but its regulation is uniquely specialized in a majority of clinical Cryptococcus neoformans serotype A and D strains to control differentiation and virulence factor regulation. C. neoformans Hog1 MAPK is controlled by Pbs2 MAPK kinase (MAPKK). The Pbs2-Hog1 MAPK cascade is controlled by the fungal "two-component" system that is composed of a response regulator, Ssk1, and multiple sensor kinases, including two-component.like (Tco) 1 and Tco2. Tco1 and Tco2 play shared and distinct roles in stress responses and drug sensitivity through the Hog1 MAPK system. Furthermore, each sensor kinase mediates unique cellular functions for virulence and morphological differentiation. We also identified and characterized the Ssk2 MAPKKK upstream of the MAPKK Pbs2 and the MAPK Hog1 in C. neoformans. The SSK2 gene was identified as a potential component responsible for differential Hog1 regulation between the serotype D sibling f1 strains B3501 and B3502 through comparative analysis of their meiotic map with the meiotic segregation of Hog1-dependent sensitivity to the fungicide fludioxonil. Ssk2 is the only polymorphic component in the Hog1 MAPK module, including two coding sequence changes between the SSK2 alleles in B3501 and B3502 strains. To further support this finding, the SSK2 allele exchange completely swapped Hog1-related phenotypes between B3501 and B3502 strains. In the serotype A strain H99, disruption of the SSK2 gene dramatically enhanced capsule biosynthesis and mating efficiency, similar to pbs2 and hog1 mutations. Furthermore, ssk2, pbs2, and hog1 mutants are all hypersensitive to a variety of stresses and completely resistant to fludioxonil. Taken together, these findings indicate that Ssk2 is the critical interface protein connecting the two-component system and the Pbs2-Hog1 pathway in C. neoformans.
