In this study the physiochemical characteristics and antioxidant activity of soybean curd were prepared with saltwort (Salicornia herbacea L.). The moisture and protein of the saltwort soybean curd showed no difference among the samples. Ash and lipid were found to be the highest in the soybean curd to which 20% saltwort was added. The contents of mineral(Fe, K, Mg, Zn) were highest in the soybean curd to which 20% saltwort was added, and Ca content was high in the soybean curd to which 16% and 20% saltwort was added. DPPH (1,1-dipheny1-2-picryl-hydrazyl) radical scavenging activity is a method of measuring the antioxidant power of food; this was high in the soybean curd to which 20% saltwort was added. Adding more saltwort led to higher scavenging activity. From the above results, it could be seen that the soybean curd, which is made by adding saltwort, contributed to enhancing the functionality of antioxidant activation and product quality.
Calmodulin (CaM) proteins, members of the EF-hand family of $Ca^{2+}$-binding proteins, represent important relays in plant calcium signals. Here, OsCam1-1 was isolated by PCR amplification from the rice genome. The gene contains an ORF of 450 base pairs with a single intron at the same position found in other plant Cam genes. A promoter region with a TATA box at position-26 was predicted and fused to a gus reporter gene, and this construct was used to produce transgenic rice by Agrobacterium-mediated transformation. GUS activity was observed in all organs examined and throughout tissues in cross-sections, but activity was strongest in the vascular bundles of leaves and the vascular cylinders of roots. To examine the properties of OsCaM1-1, the encoding cDNA was expressed in Escherichia coli. The electrophoretic mobility shift when incubated with $Ca^{2+}$ indicates that recombinant OsCaM1-1 is a functional $Ca^{2+}$-binding protein. In addition, OsCaM1-1 bound the CaMKII target peptide confirming its likely functionality as a calmodulin.
In this study, silkworms fed Cudrania tricuspidata leaves were powdered, and hydrolytic enzymes, including viscozyme, papain, and flavourzyme, were added to verify the functionality of the different mixtures. In the general component analysis, the C. tricuspidata silkworm (CS) group exhibited higher crude protein and ash content than did the other groups, and the enzyme-treated groups exhibited higher carbohydrate content than the CS group. The DPPH and ABTS radical scavenging activities were significantly higher in the CS treated with viscozyme (CSV) and the CS treated with viscozyme/flavourzyme (CSVF) than in the other groups, with the CSV group showing the highest reducing power. ACE inhibitory activity was significantly higher in the CS treated with visocozyme/papain (CSVP) than in the CS group. In conclusion, rather than using powdered silkworms fed C. tricuspidata leaves, it would be more effective to use hydrolysates from C. tricuspidata silkworms as raw materials for functional foods.
The objective of this study was to assess the quality characteristics of almond Dasik prepared with the addition of purple sweet potato powder (PSPP). Five different levels (0, 5, 10, 15, and 20%) of PSPP were added and the physicochemical properties of the Dasik were examined. Crude protein and crude fat contents decreased as the level of PSPP increased (p<0.05). The Hunter L-value decreased while the a-value increased as the PSPP level increased. Texture measurements showed that the addition of PSPP increased hardness, cohesiveness, and brittleness. Total polyphenol content and antioxidant activity were assessed in terms of estimating the functionality of the Dasik. The results showed that incorporating PSPP into the Dasik provided higher polyphenol content and antioxidant activity compared to the control Dasik (p<0.05). The results of a consumer acceptance test revealed that the Dasik sample made with 15% PSPP was most preferred and the 20% sample was the least preferred. Therefore, it is suggested that purple sweet potato powder can be incorporated into Dasik up to 15% without reducing its quality.

The rise in plant-based food consumption is propelled by concerns for sustainability, personal beliefs, and a focus on healthy dietary habits. This trend, particularly in alternative meat, has attracted attention from specialized brands and eco-friendly food companies, leading to increased interest in plant-based alternatives. The dominant plant-based proteins, derived mainly from legumes, include soy protein isolates, which significantly impact sensory factors. In the realm of plant-based fats, substitutes are categorized into fat substitutes based on fats and fat mimetics based on proteins and carbohydrates. The production of these fats, utilizing gums, emulsions, gels, and additives, explores characteristics influencing the appearance, texture, flavor, and storage stability of final plant-based products. Analysis of plant-based proteins and fats in hamburger patties provides insights into manufacturing methods and raw materials used by leading alternative meat companies. However, challenges persist, such as replicating meat's marbling characteristic and addressing safety considerations in terms of potential allergy induction and nutritional supplementation. To enhance functionality and develop customized plant-based foods, it is essential to explore optimal combinations of various raw materials and develop new plant-based proteins and fat separation.
선천성 당화장애(CDG)는 당화 과정의 결함으로 인해 발생하는 다양한 유전 대사 장애 질환을 포함한다. 당화는 단백질 접힘, 안정성 및 세포 간 신호전달에 필수적인 생화학적 과정이다. CDG는 1980년대에 처음 발견된 이후로 분자생물학과 유전학의 발전에 따라 현재까지 163개의 아형이 발견되었고 트랜스페린 등전점 전기영동이 선별검사로 사용되고 있으며 유전학적 진단기법의 발달로 CDG의 진단이 확연히 늘었으며, 다양한 선천성 당화장애의 결함에 대한 진단 기법이 연구되고 있다. CDG의 치료는 주로 대증요법에 의존하며, 일부 아형에서 단당류, 망간, 우라신, 피리독신 등의 경구 보충요법과 간 이식, 조혈모세포 이식이 사용되고 있으며 약리학적 샤페론, 유전자 치료, 그리고 약물 재배치 연구가 진행되고 있다. CDG 환자들의 진단과 치료에 대한 지속적인 연구와 협력이 필요하다.
가소제로서 솔비톨 혼합물의 첨가가 분리대두단백 필름의 수분흡습특성, 수분투과도, 색, 인장강도 및 신장률, 그리고 용해 특성에 미치는 영향을 조사하였다. 솔비톨 혼합물은 액상형과 결정형의 상태로 다양한 비율로 혼합하여 첨가하였고, 가소제로 글리세린을 첨가한 대조구 필름의 물리적 특성과 비교하였다. 솔비톨이 첨가된 필름은 대조구 필름에 비해 높은 인장강도를 갖는 반면 낮은 수분투과도 및 신장률을 보였다. 한편 솔비톨 첨가 형태에 따른 영향은 필름 신장률에는 유의적 차이는 보였으나 기타 수분투과도, 색, 인장강도 등의 필름특성에는 커다란 차이는 보이지 않았다. 솔비톨 첨가된 필름은 높은 수분차단 효과 및 필름강도를 갖는 대신 높은 용해성을 보여, 상업적 활용성을 증진시키기 위해서는 이를 극복하는 연구가 필요할 것으로 사료된다.
본 실험은 두유에 칼슘을 강화하기위한 사전실험으로 두유단백질을 단백분해효소로 처리한 뒤 효소처리 전, 후에 나타나는 단백질의 기능적 특성을 비교 분석하였다. 두유단백질과 단백분해효소의 비율이 100 : 1이 되도록 첨가하여 pH7.5, 5$0^{\circ}C$에서 10분간 처리하였다. pH에 따른 용해도는 두유단백(SMP)과 분리대두단백 (SPI)인경우 pH4.7에서 최소를 보이고 산성 및 알칼리성으로 갈수록 증가하였으나 전반적으로 매우 낮게 나타난 반면 효소처리된 두유단백질(PT-SMP)은 전 pH범위에서 SMP에 비해 높은 용해도를 보였고 특히 pH4.7에서는 20배 정도의 높은 용해도를 나타내었다. pH에 따른 유화력은 3가지 시료 모두 용해도와 같은 패턴으로 pH4.7에서 최소를 보이고 산성 및 알칼리성으로 갈수록 증가하는 경향을 나타내었다. SMP와 SPI는 높은 유화력을 나타냈으며 PT-SMP도 이와 유사한 정도의 유화력을 나타내었다. 반면 PT-SMP의 유화안정도는 다소 낮게 나타났다. SMP와 SPI는 pH4.7에서 최소의 거품형성력을 나타내면서 그 외의 pH에서는 약간 증가하다가 pH 12에서는 높은 거품형성력을 보여주었다. PT-SMP는 pH2와 4.7에서 높은 거품형성력을 나타내었고 pH8까지 차츰 감소하다가 pH12까지 다시 증가하였다. 거품안정성은 3가지 시료 모두 30분까지 급격한 감소를 보이다가 차츰 감소정도가 완만하게 나타났다. SMP와 SPI는 pH4.7에서 비교적 높은 안정도를 보였고 pH 12에서는 가장 낮은 안정도를 보였다. PT-SMP는 모든 pH에서 다른 두시료에 비해 낮은 안정도를 나타내었으나 pH4.7에서는 다소 높은 안정도를 나타내었다. PT-SMP는 SMP에 비해서 pH4.7을 제외한 pH에서 높은 열응고성을 나타내었고 가열후 감소된 가용성 단백질의 양을 보면 모든 pH에서 PT-SMP가 높게 나타났다. 이상의 결과로 볼 때 두유단백질에 효소처리를 하게 되면 높은 용해도와 거품형성력, 또 비슷한 정도의 유화력을 나타냄을 알 수 있다. 특히 산성 pH에서 월등한 용해도와 거품형성력을 나타내어 효소처리하여 부분가 수분해된 두유에 과일을 첨가하는 것도 가능하리라 생각되며, 또 효소처리된 두유단백질이 청량음료나 마요네즈, 과일첨가후식의 첨가물로 사용될 때 높은 기능적 특성을 발휘하리라 사료된다.
대두단백 가수분해 산물의 맛과 향을 개선하기 위해 효소에 의한 가수분해 system을 확립하기 위하여 단백질 가수분해 패턴이 서로 다른 효소로 생산된 단백 분해산물의 가수분해도와 표면 소수도 등을 측정하였다. 이들 분해물의 pattern을 SDS 전기영동으로 조사하였고, 효소반응에 의한 단백질분해물의 관능검사를 실시하였다. 각 균주가 생산한 단백질 분해효소의 pH 변화에 따른 효소의 활성은 No. 16 효소(Bacillu megarerium Bl6)와 No. 4효소(Aspergillus oryzae M4)는 pH 7.0에서 No. 95효소(Bacillu subtilis YG 95)와 No. 5효소(Mucor circinelloides M5)는 8.0에서 가장 높은 효소반응 활성을 보였다. 또한 반응온도에 따른 효소활성의 크기는 4가지 효소 모두 45$^{\circ}C$에서 가장 높은 활성을 나타내었다. 대두단백질 분해물의 SDS 전기영동 pattern변화에서, Bacillus megaterium Bl6과 Mucor circinelloides M5의 효소 (No. 16, No. 5)는 반응 후 분자량이 비교적 큰 peptide가 많이 생성되었으며, 효소반응 3시간 경과 후에도 분자량 66KD의 peptide를 확인할 수 있었다. 반면에 Bacillus subtilis YG 95와 Aspergillus oryzae M4의 효소(No. 95, No. 4)는 분자량 15KD∼45KD 미만의 작은 분자량의 peptide 물질이 주로 생성되었으며, 반응 2시간 경과후에는 30KD 미만의 저분자 Peptide가 주로 생성되었다. 이와 같은 결과는 HPLC 분석 결과와 일치하였다. 가수분해가 진행됨에 따라서 SDS 표면소수도가 크게 저하되었으며, Aspergillus oryzae M4 효소의 분해물의 가수분해도가 가장 높았다. 관능검사 결과, No. 4(Aspergillus oryzae M4)와 No. 95(Bacillus subtilis YG 95) 가 강한 쓴맛을 나타내었다. 각각의 효소들을 조합해서 분해한 단백분해물을 관능검사한 결과, Aspergillus oryzae M4 효소와 조합된 것의 대두단백질 분해물이 비교적 강한 쓴맛을 나타내었다. 분해물의 단맛은 시료별로 큰 차이가 나타나지 않았으나 Bacillu megarerium Bl6과 Aspergillus oryzae M4를 조합하였을 때 상대적으로 단맛의 정도가 높게 나타났다. 따라서 이와 같은 효소특성을 이용하여 대두단백질을 가수분해를 하였을 때 다양한 단백질 분해물의 제조에 이용이 가능할 것으로 사료된다.
한국 전통장류의 기능성 탐색 연구의 일환으로 청국장 발효과정 중 고혈압을 유발하는 angiotensin converting enzyme(ACE)의 저해 peptide를 분리하고 저해효과를 검토하였다. 청국장은 Bacillus subtilis CH-1023을 이용하여 제조하였고, $20^{\circ}C,\;30^{\circ}C,\;40^{\circ}C,\;50^{\circ}C,\;60^{\circ}C$에서 $0{\sim}72$시간동안 배양하면서 protein양, protease activity, ACE 저해율을 측정하고 저해활성을 가지는 peptide를 정제 후 아미노산조성을 분석하였다. 청국장의 발효시간이 경과함에 따라 protein 함량이 증가하여 $40^{\circ}C$, 60시간에서 최대를 나타낸 후 감소하였으며, 발효시간에 따른 protease activity와 ACE 저해율은 $40^{\circ}C$, 60시간에서 최대로 나타난 후 점차 감소하였다. 따라서 Bacillus subtilis CH-1023을 이용한 청국장 제조는 $40^{\circ}C$에서 60시간 배양이 최적의 배양조건이었다. 최적 발효조건에 따라 청국장을 제조한 후 20 mM sodium phosphate buffer(pH 7.0)를 가해 추출한 추출물을 Amicon YM-3 membrane filtration과 Sephadex G-10, G-25를 이용한 gel filtration으로 부분정제 하였다. 또한 정제한 peptide는 첨가함량이 높아질수록 저해활성은 높게 나타났으며, 0.5 mg 정도의 peptide 함량으로 94.3%의 저해율을 나타내었다. 정제한 peptide의 아미노산 조성은 histidine, alanine, phenylalanine 함량이 높은 것으로 나타났다.
