The present work aims to estimate the effect of heat treatment on the in vitro protein digestibility and formation of trypsin inhibitor or trypsin indigestible substrate(TIS) of raw and defatted flounder. It was also carried out to assess the formation of lipid-protein complexes under the conditions of different ratio of lipid addition. The in vitro protein digestibility increased when steamed for 5 min showing $88.09\%$ in raw and $90.56\%$ in defatted samples, respectively. After 40 min steaming, the digestibility decreased by $2{\sim}4\%$. As for microwaving, heating for 1 min resulted in slight increase of digestibility, however, heating for 7 min did decrease of digestibility by $3{\sim}4\%$ for both raw and defatted materials. There was no difference in fatty acid composition found with heat treatment. The major fatty acids of flounder meat were $C_{16:0},\;C_{16:1},\;C_{18:1},\;C_{20:5},\;C_{22:6}$ and the ratio of the unsaturated to saturated was 67.3:32.6. Fat oxidation and nonenzymatic browning were enhanced by heat treatment and protein solubility decreased necessarily as the brown pigment formation increased. On the other hand, the effects on the digestibility and TIS of the complexes formed from interaction of lipid and myofibrillar or meat protein of flounder were examined. The interaction of protein with lipid was considered to mostly contribute to the drop of digestibility of fish products. The digestibility of myofibrillar protein was $93.72\%$ for flounder, and it generally decreased as the amount of lipid added to protein and reaction time increased. Also mixed and heated samples were more active in digestibility decline than those mixed after heating. The result probably indicated that lipid-protein interaction was involved in the drop of digestibility which coincided with protein denaturation.
The study was conducted to evaluate the nutritional value of seeds and various seed fractions of different varieties of cotton (MNH 147, CIM 240, NIAB 78, FH 87, CIM 109, MNH 93, FH 682, GOHAR 87, SLS I and B 557). Linter, hull, kernel and meal were obtained from cottonseed by physical and chemical methods. Free gossypol and available lysine contents of seed and it's fractions were determined. In vitro protein digestibility of cottonseed meal was also determined. Free gossypol and available lysine contents ranged between 0.22-2.26% and 0.64-1.32% in seed, and 0.03-0.29% and 1.38-2.36% in meal, respectively. FH 87 was highest both in free gossypol and available lysine content, and NIAB 78 was lowest in free gossypol content and FH 862 was lowest in available lysine content. In vitro protein digestibility of cottonseed meal ranged between 66,02-79,96%. Statistical analysis revealed significant (p<0.05) varietal differences in free gossypol, available lyslne and in vitro protein digestibility of cottonseed and derived products.
To determine the nutritional quality and physical properties of ginseng-chicken meat porridge, 10 kinds of ginsengchicken meat porridge samples containing waxy and/or non-waxy rice were analyzed for in vitro protein digestibility and their degree of starch hydrolysis. Viscosity and spreadness were determined for the gelatinized pastes of the porridge samples. Microphotographs of the starch granules and pastes were studied to confirm structural changes in the rice starch during cooking. The starch paste from non-waxy rice porridge had higher viscosity than the starch paste from the waxy rice porridge; however, in the case of the ginseng-chicken meat porridge, the difference in viscosity was negligible. Microphotograph comparisions between the waxy rice porridge and non-waxy rice porridge indicated apparent differences in the shapes of their starch granules and gels. The granule surface of the non-waxy rice was very rough while that of the waxy rice was very smooth; this difference would lead to organoleptical discrepancy. The added ginseng increased the protein digestibility of the chicken meat; however, the protein digestibility of the ginseng-chicken meat porridge was lower than that of the chicken meat or rice porridge due to inhibited protein digestion by the gelatinized starch. Finally, the rice porridge had increased starch hydrolysis with additions of chicken meat and vegetables.

This study was conducted to obtain accurate data on intake, apparent digestibility and nitrogen balance of dietary protein which the korean take in habitually. Subjects were 8 male college students, aged from 20 to 26, and maintained their menu and life pattern as usual during a 4-week study. The same amount of diet that the subjects had consumped, and feces and urine were collected and measured to extract their nitrogen content data by Kjeldahl method. From above data, the apparent digestibility and the body nitrogen balance were estimated by determining daily protein intake and excretion. The daily protein intake measured by Jjeldahil method was 88.3$\pm$ 0.9g(1.45/kg of body weight /day) which marked 8.3% higher than that estimatd by food analysis table. The proportional of animal protein against total protein intake was 50.4$\pm$ 2.3%. Daily fecal protein loss was 14.3$\pm$0.6g and the apparent digestibility was 83.8$\pm$ 0.7%. The urinary nitrogen excretion was 10.30$\pm$0.19g. The nitrogen balance considering nitrogen excretion from feces indicated positive balance of 1.06$\pm$0.20g.

To predict the apparent total tract digestibility (ATTD) of crude protein (CP) in dogs we developed an in vitro system using an in vitro digestion method and a statistical analysis. The experimental diets used chicken meat powder as the protein source, with CP levels of 20% (22.01%, analyzed CP value as dry-based), 30% (31.35%, analyzed CP value as dry-based), and 40% (41.34%, analyzed CP value as dry-based). To simulate in vivo digestive processes a static in vitro digestion was performed in two steps; stomach and small intestine. To analyze ATTD the total fecal samples were collected in eight neutered beagle dogs during the experimental period. CP digestibility was calculated by measuring CP levels in dog food, in vitro undigested fraction, and dog feces. In result, CP digestibility at both in vivo and in vitro was increased with increasing dietary CP levels. To estimate in vivo digestibility the co-relation of in vivo ATTD and in vitro digestibility was investigated statistically and a regression equation was developed to predict the CP ATTD (% = 2.5405 × in vitro CP digestibility (%) + + 151.8). The regression equation was evaluated its feasibility by using a commercial diet. The predicted CP digestibility which was calculated by the regression equation showed high index of similarity (100.16%) with that of in vivo in dogs. With that, it would be a feasible non-animal method to predict in vivo CP digestibility by using in vitro digestion method and the proposed linear regression equation in adult dogs.
A regression equation was proposed for predicting protein digestibility using pH drop and free amino acid content. Results were compared with those determined by the pH drop method of Satterle et al. and with apparent in vivo digestibility in rats. Measurd free amino acid content prior to four enzyme digestion had an influence on calculating digestbiilty . Results from new equation correlated more highly (r=0.8434, difference average=2.304) with in vivo digestibility than the results of pH drop method (r=0.7603, difference average=10.099).
Previous studies conducted with swine have reported that the mobile nylon bag technique (MNBT) does not always accurately predict in vivo nutrient digestibilities. Therefore, in this study, the MNBT was modified so that nutrient digestibilities would more closely resemble those from conventional (Con) digestibility studies obtained using the indicator method. A total of 19 feeds were tested including five cereal grains, five legumes, three high protein sources and six mixed diets. The principle changes to the MNBT included the use of a fecal collection harness which minimized the number of bags lost. In addition, previous protocols involved pooling of bags within pig while in the present experiment all bags were analyzed separately to increase the precision of the test. Finally, chemical analyses were done using the entire nylon bag plus residue rather than opening.the bags and scraping out the contents. With the exception of the barley sample (p=0.01), dry matter digestibility (DMD) coefficients obtained with the MNBT were not significantly different from those obtained with the indicator method. The linear regression equation relating the MNBT to the indicator method was Con DMD=-O.77+1.02 MNBT DMD ($r^2=0.93$: p<0.0001). There was no significant (p>0.05) difference in gross energy digestibility (GED) coefficients determined using the MNBT or the indicator method for any of the 19 feeds. The regression line equation relating the MNBT to the indicator method was Con GED=-5.68+1.06 MNBT GED ($r^2=0.94$: p<0.0001). The MNBT was less effective in predicting in vivo crude protein digestibility (CPD) than it was in predicting dry matter and energy digestibility. Differences greater than five percentage units were observed for two of the legumes, Kabuli chickpeas (p=0.02) and the extruded pea-canola seed mixture (p=0.01) as well as for three of the mixed diets including the unheated hulled barley-based diet (p=0.01), the unheated hulless-barley based diet (p=0.08) and the barley-soybean meal based diet (p=0.008). The regression equation relating the MNBT to the indicator method was Con CPD=5.75 + 0.90 MNBT CPO ($r^2=0.76$; p<0.0001). This study indicates that the modified MNBT can be used for the rapid determination of dry matter and energy digestibility in a wide variety of ingredients. For the measurement of crude protein digestibility, the technique produces results similar to conventional digestibility studies for cereal grains and high protein feeds but tends to overestimate protein digestibility for legumes and mixed diets.

The daily protein intake was 56.9$\pm$1.4g and daily fecal protein loss was 6.3$\pm$0.2g. The apparent digestibility of protein was 89.6$\pm$0.7$\%$. The daily nitrogen intake measured by Kjeldahl method was 9.43$\pm$0.2g. The urinary nitrogen excretion was 7.64$\pm$0.23g and fecal nitrogen excretion was 1.02$\pm$0.03g. The interaction of myofibrillar protein with lipid or oxidized lipid was considered to be mostly contributing to the drop of digestibility of fish meat products. The digestibility of myofibrillar protein was $92.11\%$ for flounder and $88.04\%$ for hairtail fish, repectively, and as a rule it decreased as both the amount of lipid and reaction time increased. It also decreased with increase in the amount of added linoleate and oxidized linoleate. However, when the reaction continued for 6 hours or more the digestibility rather increased, which was provably due to the unfolding of protein structure. The hot air dried hairtail fish showed the lowest C-PER values among all dried fish products. The protein quality of flounder, hairtail fish and their dried ones except hot air dried ones measured by C-PER procedure were superior to that of ANRC casein. DC-PER values of all samples were greater than those of C-PER values and the greater discrepancies were noted in hairtail fish (fatty fish) products which possessed the lower in vitro protein digestibilities. Predicted diegstibilities, which were calculated using amino acid profiles, of all samples except raw ones were overestimated in comparison with in vitro protein digestibilities. From the observations so far, formation of complex of lipids and protein was thought to be the most important factor in lowering protein digestibility of the dried fish meat products.

The objective of this research was to determine the effects of synchronizing the rate of dietary energy and nitrogen release on: ruminal fermentation, microbial protein synthesis, blood urea nitrogen, and nutrient digestibility in beef cattle. Four, two-and-a-half year old Brahman-Thai native crossbred steers were selected for the project. Each steer was fitted with a rumen cannula and proximal duodenal cannula. The steers were then randomly assigned in a $4{\times}4$ Latin square design to receive four dietary treatments. Prior to formulation of the dietary treatments, feed ingredients were analyzed for chemical composition and a nylon bag technique was used to analyze the treatments various ingredients for degradability. The treatments were organized in four levels of a synchrony index (0.39, 0.50, 0.62 and 0.74). The results showed that dry matter digestibility trend to be increased (p<0.06), organic matter and acid detergent fiber digestibility increased linearly (p<0.05), while crude protein and neutral detergent fiber digestibility were not significantly different (p>0.05). Higher concentration and fluctuation of ruminal ammonia and blood urea were observed in the animal that received the lower synchrony index diets. As the levels of the synchrony index increased, the concentrations of ruminal ammonia nitrogen and blood urea nitrogen, at the 4 h post feeding, decreased linearly (p<0.05). Total volatile fatty acid and bacteria populations at the 4 h post feeding increased linearly (p<0.05). Microbial protein synthesis trend to be increase (p<0.08). The results of this research indicate that synchronizing the rate of degradation of dietary energy and nitrogen release improves ruminal fermentation, microbial protein synthesis and feed utilization.
