Contents of Nitrogen Fractions and the Degree of Whey Protein Denaturation in Market Milks (시유의 질소분획물 함량과 유청단백질 변성정도)

  • To find out the effective indicators for identification and classification of different heat treatment, the contents of nitrogen fractions and the degrees of whey protein denaturation in market milks were investigated by Kjeldahl method. The contents of nitrogen fractions per 100ml raw milk were total nitrogen (431.3mg), casein nitrogen (341.0mg) and non-casein nitrogen(90.3mg), in which non-protein nitrogen (31.6mg) and denatured whey protein nitrogen (58.8mg), while those of LTLT, HTST, UHT pasteurized and UHT sterilized showed different values. The degrees of whey protein denaturation were 26.7%(LTLT), 32.9%(HTST), 60.7%(UHT pasteurized) and 38.4%(UHT sterilized), respectively. As the higher temperature was applied for the treatment of milk, the degree of the whey protein denaturation was higher. Remarkable differences in the degree of whey protein denaturation according to the heating methods were observed.

A Study on the Change of Serum Components in Rats by Feeding the Diet with Soft Drink (시판(市販) 음료수(飮料水)가 백서(白鼠) 혈청내(血淸內) 성분(成分)에 미치는 영향(影響))

  • This experiment was designed to observe some influence on the serum components in rats by feeding basal diet (Protein contents: 20.0%; lipid contents: 3.5%) with soft water (Water, Carbonated cider, Cola and Fanta). Fourty male Albino rats were used as the experimental animals and the subjects were devided into four feeding group and each group was fed on the corresponding diet for six weeks. The contents of protein, lipid and cholesterol were determined in their serum. The obtained results are summarized as follows. 1) The protein contents in the serum was decreased by the feeding of the Carbonated cider, Cola and Fanta compared with Water experimental group. 2) The lipid contents in the serum was similar to each feeding experimental group. 3) The total cholesterol and ester form cholesterol in the serum were decreased by the feeding of the Carbonated cider compared with Water, but were similar to the feeding of the Cola and Fanta.

Effect of Proteases on the Extraction of Crude Protein and Reducing Sugar in Pollen (화분에서의 조단백질 및 환원당 추출시 단백질 분해효소가 미치는 영향)

  • This study was conducted to increase crude protein and reducing sugar contents in pollen extracts by proteases. Four commercial neutral proteases (Alcalase 2.4L, Protamex, Flavozyme and Protease A) and two alkaline proteases (Protease S and Protease P) were used to prepare acorn and Darae pollen extracts. Contents of moisture, ash, crude protein and crude fat of acorn pollen were 5.2%, 2.7%, 6.2% and 22.3%, respectively, while those of Darae pollen were 5.4%, 2.8%, 1.8% and 27.8%, respectively. Contents of crude protein and reducing sugar in pollen extracts were increased by proteases. Alcalase 2.4L was the most effective in increasing protein contents while Protease A in increasing reducing sugar contents. It is suggested the use of proteases is one of the potential methods for increasing the contents of crude protein and reducing sugar in preparation of pollen extracts.

Chemical Composition of Seed in Medicinal Soybean Collected in Korea

  • Production of medicinal soybean [Glycine max (L.) Merrill], characterized with black seed, white stripe at hilum border, yellow cotyledon and small seed, is increasing with increasing consumption. The objective of this study was to investigate the chemical composition of medicinal soybean seed and to provide basic information, for the characterization of these soybeans among genetic resources. Forty-four lines of medicinal soybeans collected from Korea and two control cultivars, 'Hwangkeumkong' (Yellow seed coat) and 'Geomjeongkong l' (Black seed coat) were planted at the Research Farm of the College of Natural Resources, Korea University, located at Namyangju City on May 25, 1996. Seeds of these lines were harvested at full maturity and analyzed for protein, oil, sugar, starch and mineral contents. Mean protein and oil content of the medicinal line seeds were 42.6 and 16.1%, respectively, and those of the control cultivars were in the middle range for protein and oil content. However, sugar and starch content of the medicinal line seeds appeared to be in the lower range of the distributions compared to the control cultivars and were 10.0 and 1.68%, respectively. Mean P, K, Ca, and Mg contents of the seeds of medicinal soybean lines were 15.9, 21.5, 3.11, and 2.81 mg/g, respectively, indicating that these lines had higher P, K, and Mg and lower Ca contents when compared to the control cultivars. Mean Na and Fe contents were 671 and 224 mg/kg, respectively, showing lower Na and similar Fe contents. The observed results provided that chemical compositions of medicinal soybean were, on average, different from those of the general soybean cultivars.

Quality Improvement in Perilla III. Varietal Differences of Protein Content and Amino Acid Composition in Perilla (들깨의 성분개량 연구 III. 들깨 품종의 단백질 함량과 아미노산 조성)

  • Korean local collected strains of perilla were analyzed on their protein and amino-acid contents in the seed. The strains analyzed were 80 of which grown in the experimental field of Crop Experiment Station at Suwon in 1991. This experiment was performed to obtain the basic informations on the varietal differences of protein and amino-acid contents and further applied in the breeding project for quality improvement of perilla. The seed protein contents of 80 perilla varieties ranged from 17.9% to 24.6% and the average content was 24.6%. There were no differences in protein contents according to the maturity and 1,000 seed weight, but differed by the seed coat color showing 1.4% higher content in light gray than that of drak brown, which may suggest that the seed coat color could be used as a marker gene for breeding high protein varieties. While the total essential amino-acid contents of collected perilla ranged from 36.41% to 44.85%, the mean of 40.55%, the non-essential amino-acid contents was 47.23%~60.01% with the mean of 55.29%. Moreover, there was high positive correlation between the essential amino-acid contents and seed coat color. The essential amino-acids of perilla has an exceptionally high content of arginine, leucine, but methionine indeed being almost marginal. Of the essential amino-acids, lysine, methionine and valine were contained higher compared with the FAO recommendation level.

  • Functional prediction of unannotated proteins is one of the most important tasks in yeast genomics. Analysis of a protein-protein interaction network leads to a better understanding of the functions of unannotated proteins. A number of researches have been performed for the functional prediction of unannotated proteins from a protein-protein interaction network. A chi-square method is one of the existing methods for the functional prediction of unannotated proteins from a protein-protein interaction network. But, the method does not consider the topology of network. In this paper, we propose a novel method that is able to predict specific molecular functions for unannotated proteins from a protein-protein interaction network. To do this, we investigated all protein interaction DBs of yeast in the public sites such as MIPS, DIP, and SGD. For the prediction of unannotated proteins, we employed a modified chi-square measure based on neighborhood counting and we assess the prediction accuracy of protein function from a protein-protein interaction network.

The Effects of Age and Dietary Protein Level on Ca Metabolism in Rats (나이가 다른 단계에서 식이단백질 수준이 흰쥐의 Ca 대사에 미치는 영향)

  • To study the effects of the age and the dietary protein content on Ca metabolism male rats of 1 month 6 month 12 month of age were fed experimental diets containing 5%, 15% or 50% casein for 4 weeks. Food and ca intake were higher in old rats and in high protein groups. The weight ash and Ca contents of femur and tibia were higher in old rats. The higher dietary protein level resulted in higher skeletal weigh ash and Ca contents. But high protein diet(50% casein) lead to reduced bone mineral density(ash/dry bone weight) and Ca density(Ca/dry bone weight) in 1 month old rats. Low protein diet(5% casein) on the other hand reduced the bone growth even though the bone density was higher in this group. The ill effect of low protein diet was not evident in 12 month old rats. Glomerular filteration rate(GFR) and urinary Ca excretionincreased with age and with dietary protein level especially in 12 month old rats. Serum immunoreactive parathyroid hormone(iPTH) level tended to be higher in aged rats but was not affected by dietary protein level except 1 month old rats where 50% protein group showed significantly higher value. This study showed that the dietary protein level seemed to have different effect on Ca metabo-lism in rats of different age., The low bone density in the high protein group of growing rats may be due to the higher iPTH level and increased urinary Ca. The dietary protein level however had no effects on the bone composition in aged rats even though the higher urinary Ca excretion. In conclusion this study suggests that high protein intake from young may lead to less peak bone mass and to increase the bone loss in later years, which would increase the risk for osteporosis.

Effects of Different Body Color Strain and Improvement on Serium Cholesterol and Protein Contents in Chickens (닭의 체모색 계통별 및 개량상태가 혈중 Cholesterol과 Protein 함양에 미치는 영향)

  • The present study was undertaken to determine the effect of different body hair color (Native type) and commercial white laying hen (Improvement, Nick Chick strain) on cholesterol, triglyceride and protein concentration in serium. Experimental hens were selected out of yellow-brown body hair color strain (10 heads), green-black body hair color strain (13 heads), white body hair color strain (7 heads), black-bone strain (10 heads) as native type and commercial white laying hen (30 head,3) as improved variety. Experimental diet was fed identically for 4 weeks in total groups. The results of the experiment obtained were summerized as follows:1. Serium cholesterol contents were the highest in commercial white laying hen ($172.50\pm$13.45 mg/100 $m\ell$) (p<0.01;, and higher in white color strains than body hair color strain groups. Body hair color strain groups were similar in serium cholesterol contents. 2, Serium triglyceride concentration also showed no response among native type groups, however commercial white laying hen group shelved significatly difference. (p<0.01) 3. Serium protein contents were the highest in commercial white laying hen group ($6.24\pm$0.23 mg/100 $m\ell$). (p<0,01) Increasing the serium cholesterol, triglyceride and protein contents in commercial white laying hen group than color strain group of native type was considered to improved grade.

Analysis of Paddy Soil Chemical Properties and Rice Quality in Central Area (Sejong) in Korea

  • This study was carried out to review and compare crop cultivations upon chemical properties of paddy soil and qualitative characteristics of rice in Sejong-city from a point of view of farming extension to rice farmers and to utilize the result of the study as a basic guideline for precise agricultural practice. The pH in soils of Sejong was about 6.1-6.6 and had no difference with an average pH of paddy soils in Chung-Nam with pH 6.1. However, the average of organic matter, calcium (Ca) and available silicate in Sejong was lower than the average of them in Chung-Nam. The yields of rice were higher in 2010 than in 2011 and 2012, and the protein contents of rice were the highest in 2011 while the lowest water contents of rice in 2011. The protein contents upon regions were the highest in 2011 with 6.1%, and the amylose contents were the highest in Yeondong-myun, Jeoneu-myun, and Yeonseo-myun in 2010 while Kumnam-myun and Jeondong-myun were the highest in 2012. With the increase of precipitation, the protein content level in rice was increased while the amylose content level tended to decrease. Correlations between the chemical properties of paddy soil and the quality of rice and between level of organic matter in soil and amylose contents were negative (r = -0.507), and the correlation between the moisture contents and amylose contents (r = 0.419) and between the water contents and whiteness (r = 0.485) were positive. Because the quantity and quality of rice yield is determined by the soil characteristics, the consultation to farmers for the proactive soil analysis and for the maintenance of stable level of pH, organic matter and available silicate based on historical results of analysis is highly recommended. Also, the analysis on the effect of the weather and the soil characteristics affecting the quality and quantity of rice would be another good way.

Effects of Soy Protein and Calcium on Lipid Metabolism in Rats Fed Unsaturated Fat Diet (불포화 고지방식을 섭취한 흰쥐의 체내지질대사에 대한 대두 단백질과 칼슘의 섭취 효과)

  • This study was performed to observe how soy protein and calcium in rats fed unsaturated fat-enriched diet affect the total lipid and cholesterol contents of blood and tissues male Sprague-Dawley rats weighing approximately 220g were fed four purified diets which contained 18% (w/w) corn oil, 1% (w/w) cholesterol, two sources of protein : casein or isolated soy protein (ISP) and two levels of dietary calcium : 0.1% or 1.0%, first for four weeks (Expt. 1) and second for eight weeks (Expt. 2). The contents of total lipid, cholesterol and triglyceride in blood, liver, heart and feces were determined. After four weeks feeding, the serum lipid and cholesterol concentration was not significantly different among the groups. After eight weeks feeding, these concentrations were significantly lower in ISP-High calcium group. The serum lipid concentration was influenced by dietary protein sources and calcium levels. These results indicated that the hypolipidemic effects of soy protein and high calcium intake were partly due to decrease in lipid absorption and these effects were not detected at 4 weeks feeding but 8 weeks feeding.
