Correlation of Lignan Contents with Protein and Oil Contents in the Seeds of Sesamum indicum L.

  • The contents of lignan, protein, and oil of nine Sesamum indicum cultivars were analyzed. To determine the lignan contents, sesamin 1 and sesamolin 2 were isolated from the ethylacetate extracts of the Suwon cultivar, and the structures were fully characterized by spectral and physical methods. The seeds of nine cultivars were screened for two lignans, which were determined by HPLC using a $C_{18}$ reversed phase column coupled with a photodiode array detector. Suwon cultivar showed the highest concentration (1: $6.24{\pm}0.04$ and 2: $3.58{\pm}0.01\;mg/g$), whereas Soonheuk displayed the lowest (1: $0.91{\pm}0.01$ and 2: $0.73{\pm}0.01\;mg/g$). The average content ratio of sesamin 1 (3.64 mg/g) was significantly higher than that of sesamolin 2 (2.57 mg/g). The protein content ranged from $21.52{\pm}0.35$ to $31.22{\pm}0.25%$, Suwon containing the highest level and Soonheuk had the lowest. Kwangbaek showed the highest oil level ($49.84{\pm}0.40%$), while Soonheuk exhibited the lowest ($42.52{\pm}0.05%$). Sesamin 1 exhibited a stronger radical-scavenging activity in the ABTS ($IC_{50}:\;63.2{\pm}2.4\;{\mu}M$) than its DPPH radical-scavenging activity ($IC_{50}>200\;{\mu}M$). These results lead to the conclusion that lignan content is affected by protein and oil contents. Concentration of the chemical components in the cultivars could be a key factor in the selection process of a high quality species.

조피볼락(Sebastes schlegeli)의 성숙에 따른 간세포 활성과 간 및 근육의 단백질$\cdot$총 RNA 함량변화 (Activities of Hepatocytes and Changes of Protein and Total RNA Contents in Liver and Muscle of Sebastes schlegeli with the Gonadal Maturation)

  • 난태생 어류인 조피볼락(Sebastes schlegeli)의 성숙과 안정적인 출산을 위한 기초 정보를 얻고자, 간중량지수의 월별 변화와 간세포의 활성이 생식소의 발달과 어떤 관계에 있는지 조사하고, 아울러 생식소 발달에 따른 간 및 근육조직내의 단백질과 총 RNA 함량변화도 조사하였다. 암컷의 HSI는 9월부터 높아지기 시작하여 2월에 최대 값(2.58)에 달한 뒤, 3월(성숙기) 부터 5월(출산기) 사이에는 낮아졌다 수컷에서는 10월(성숙기)부터 높아지기 시작하여 4월(회복기)에 최대 값(3.20)에 달했다. 암컷의 난황형성기에 간세포내 글리코겐과 지방적의 양이 점차 감소되나, 총 RNA의 함량은 증가하였다. 그러나 수컷에서는 다량의 지방적들이 소모되지 않고 성숙기에도 세포질내에 잔존하고 있었다. 생식소의 발달과 함께 간 세포질에는 잘 발달된 조면소포체 가 증가하였는데, 이 들은 단백질 합성과 난황 전구물질의 형성에 중요한 역할을 하는 것으로 간주된다. 암컷의 간과 근육조직의 단백질 및 총 RNA 함량은 1월(성장기)부터 점차 증가되기 시작하여 4월 초순(성숙기)에 최대 값을 나타낸 다음, 4월 하순(임신기)에 감소되는 경향을 보였다. 수컷에서는 초기 성숙기인 10월부터 점차 증가하여 성숙 및 교미기인 12월에 최대 값을 보인 다음, 퇴화기인 1월에 다시 감소하였다.

Foraging behaviour and preference of pollen sources by honey bee (Apis mellifera) relative to protein contents

  • Background: Pollen is an important source of protein and lipids for many animals including honey bees. In order to understand the foraging behaviour of honey bee colonies and preference among the available floral resources, pollen collections from three experimental healthy colonies of honey bees were analysed in the month of June. Results: The amount of pollen collections were related to the colony's need which was indicated by the number of larval and adult bees present in the hive. Interesting was the sequence of pollen collection from different floral sources. All honey bee colonies collected pollens from Trifolium repens first, then Erigeron annus and the third choice was Coreopsis drummondii and Oenothera biennis flowers. Total protein content of Trifolium pollen was the highest (20.0 g/100 g DM), and the others were in the range of 8.9-11.4 g/100 g DM. Conclusion: The results indicated that the first criteria for honey bee foraging preference of pollens would be the nutritional contents of protein and the resource availability of the lesser nutritious floral sources. This information can help pollinator protection programmes of habitat manipulation using flowering plants for nectar and pollen sources.

Hydrocolloids Decrease the Digestibility of Corn Starch, Soy Protein, and Skim Milk and the Antioxidant Capacity of Grape Juice

  • Hydrocolloids have many applications in foods including their use in dysphagia diets. We aimed to evaluate whether hydrocolloids in foods affect the digestibility of starch and protein, and their effects on antioxidant capacity. The thickening hydrocolloids: locust bean gum and carboxymethyl cellulose, and the gel-forming agents: agar agar, konjacglucomannan, and Hot & Soft Plus were blended with corn starch and soy protein, skim milk, or grape juice and were examined for their in vitro-digestability by comparing the reducing sugar and trichloroacetic acid (TCA)-soluble peptide, for antioxidant capacity by total polyphenol contents and the 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity. The hydrocolloids resulted in a decrease in starch digestibility with the gel-forming agents. Hydrocolloids diminished TCA-soluble peptides in skim milk compared to soy protein with the exception of locust bean gum and decreased free radical scavenging capacities and total phenolic contents in grape juice. Our findings may provide evidence for the use of hydrocolloids for people at risk of nutritional deficiencies such as dysphagia patients.

삼지구엽초의 멜라닌합성 촉진과 작용기전 (Melanogenic Effect and the Mechanism of Epimedium koreanum Nakai)

  • In this study, ethanol extract of Epimedium koreanum Nakai(EEKN) enhanced melanogenesis by inducing expression of tyrosinase and tyrosinase-related protein-1 (TRP-1). But EEKN did not increase the protein expression of tyrosinase-related protein 2 (TRP-2). Moreover, EEKN enhanced tyrosinase activity and melanin contents of B16F10 cells. EEKN raised the expression of CREB phosphorylation and microphthalmia-associated transcription factor (MITF) as a key transcription factor for tyrosinase expression regulating melanogenesis. And PKC inhibitor H89 supressed that EEKN induced tyrosinase activity, melanin contents, and expression of tyrosinase, TRP-1. These results suggest that melanogenesis-promoting effect of EEKN was correlated with regulation of tyrosinase and TRP-1 protein through cAMP/PKC pathway.

단백질(蛋白質) 급원(給源) 식품(食品)이 김치의 발효와 Ascorbic Acid의 안정도(安定度)에 미치는 영향(影響) (Effects of Protein Sources on Kimchi Fermentation and on the Stability of Ascorbic Acid)

  • Effects of protein-sources on Kimchi fermentation and on the stability of ascorbic acid were investigated at $7{\pm}1^{\circ}C.$ Kimchi samples with various protein sources showed the higher pH and total acidity through the fermentation period than kimchi without the proteins. The results revealed that the proteins have acted as good buffer and lactobacilli-growth enhancer in the fermentation, and the milk proteins showed the most significant effect among the samples. The lactobacilli were at the highest growth at 15th day in all the samples. The amounts and changes in ascorbic acid content during the fermentation did not differ significantly between the control and protein added samples. The fresh, unfermented kimchi contained 14.5-15.7mg of ascorbic acid per 100g of sample and decreased continuously by the 12th day of fermentation. After then the vitamin increased in all the samples and then again decreased slowly after 18th day. The contents of ascorbic acid at the end of the 3 weeks-fermentation(16.3-17.3mg/100g) were still higher than the contents of fresh, unfermented kimchi.

수유기간에 따른 Preterm Milk의 질소 함량 변화에 관한 연구 (Changes in Contents of Nitrogen of Preterm Milk during Lactation)

  • Nitrogen(N) concentration of preterm(PT) and term (T) milk in various fractions, such as total, protein, nonprotein, whey protein and casein were determined at 2-5 days, 1, 2, 4 and 6 weeks of postpartum. The purpose of this study was to investigate the nitrogen concentration of human milk from mothers delivering at preterm and term, and the propriety of preterm milk for premature infants. The concentration of total N, nonprotein N, protein N, whey protein N and casein N in preterm milk was decreased significantly with time postpartum. Total N was 374mg/이 at colostrum, 232mg/dl at mature milk. Whey protein N was decreased from 42mg/dl at 2-5 days to 32mg/dl at 4-6 weeks. Protein N was 332mg/dl at colostrum, 202mg/dl at mature milk. The proportion of whey protein N and casein N were 39:61 at colostrum, 28:72 at mature milk. No difference were found between T and PT milk for total nitrogen excepted 2 weeks. In this report we show that nitrogen concentration except casetpt casein N is smaller in milk from mothers giving birth prematurely than in milk from mothers giving birth at term, over the first two weeks of lactationl But protein N was higher in preterm milk than term milk, whey protein nitrogen was lower. By comparing predicted nitrogen intakes to estimated requirements of preterm infants fed 150 to 200ml/kg/day of their own mother's milk, we predict that the quantities of protein provided would be adequate to meet the requirements of the prematured infants during the early weeks of life.

EFFECTS OF DIETARY PROTEINS ON THE ACTIVITIES OF LIPOGENIC ENZYMES IN THE LIVER OF GROWING CHICKS

  • In Experiment 1, when fasted chicks were fed diets containing various sources of protein for 3 days, the activities of lipogenic enzymes (acetyl-CoA carboxylase, fatty acid synthetase, citrate cleavage enzyme and malic enzyme) in the liver of growing chicks were significantly lower in the soybean protein or gluten diet than in the casein or fish protein diet. Triglycride contents of the liver and plasma of chicks fed the casein or fish protein diet were significantly lower than that of those fed soybean protein or gluten diet. In Experiment 2, the effects of dietary amino acid mixture simulating casein or protein on the activities of hepatic lipogenic enzymes were examined. The activities of acetyl-CoA carboxylase and fatty acid synthetase in the liver of chicks fed the casein diet were significantly higher than that of those fed the soybean protein diet or two diets of amino acid mixtures. Furthermore, there were no significant differences between the two diets of amino acid mixture based on casein or soybean protein. However, the activities of malic enzyme and citrate cleavage enzyme tended to be lower in the soybean-type amino acid diet than in the casein-type amino acid diet. Thus, some effects can be ascribed to the protein itself and some to the amino acid composition of the protein sources.

피혁폐기물(皮革廢棄物)인 Shaving scraps으로 부터 가수분해(加水分解) 단백질(蛋白質)의 제조(製造) 및 특성(特性) (Preparation and Characterization of the Hydrolyzed Protein from Shaving Scraps of Leather Waste Containing Chromium)

  • 피혁 제조시 발생되는 크롬을 함유한 피혁 고형폐기물인 shaving scraps의 단백질 자원화 가능성을 검토하기 위하여 알카리제로 MgO를 농도별로 처리하여 가수분해시켜 회수한 hydrolyzed protein의 특성은 다음과 같다. MgO 처리에 의한 shaving scrap의 알카리 가수분해방법에서 크롬이 불용화 되는 pH 8.0 이상을 유지하기 위해서는 MgO를 5% 이상 처리해야 하는 것으로 나타났으며, MgO 처리에 따른 alkaline condition하에서 shaving scrap의 용해도는 약 60% 수준으로서 낮았다. MgO 처리에 의해 shaving scraps으로 부터 회수한 hydrolyzed protein의 평균 분자량은 약 80~100 KD이었고, collagen protein을 구성하고 있는 glycine, alanine 및 proline이 대부분을 차지하고 있었으며, 산성 아미노산인 aspartic acid와 glutamic acid도 상당량 함유되어 있는 것으로 나타났다. 무기성분중 Mg, Ca 및 Na는 액체비료의 무기양분으로서는 다소 많은 양이 함유되어 있었으며, Cr은 약 30~40ppm으로서 원시료 (40,000~42,000ppm)에 비해 크게 감소되었다.

위암 조직과 세포주에서 mDNA와 OXPHOS 단백질 분석 (Alterations in Mitochondrial DNA Copy Numbers and Mitochondrial Oxidative Phosphorylation (OXPHOS) Protein Levels in Gastric Cancer Tissues and Cell Lines)

  • 위암 환자에서 미토콘드리아 DNA (mtDNA)의 양적 변화가 보고 되고 있으며 이러한 변화가 위암의 발암이나 진행에 관여되는 것으로 추정되고 있다. 그리나 위암에서 미토콘드리아 단백질이나 mtDNA에 의해 암호화된 산화적 인산화(OXPHOS) 단백질의 양적 변화에 관한 연구는 아직까지 미비한 실정이다. 본 연구에서는 위암환자 조직 및 세포주를 이용하여 mtDNA 양 그리고 미토콘드리아 단백질 및 OXPHOS 단백질의 양을 분석하였다. 또한, mtDNA 양적 변화와 위암 환자의 임상병리학적 특징을 연관 분석하였다. MtDNA 양을 분석하기 위하여 qPCR 기법을 그리고 단백질 분석에는 Western blot 기법을 각각 활용하였다. 총 27개의 위암 환자 샘플에서 약 80%에 해당하는 22개의 환자 위암조직에서 정상조직에 비해 mtDNA 양이 감소하였으며, 나머지 환자에서는 mtDNA 양이 증가하였다. 이러한 mtDNA 양이 감소한 위암 조직 샘플에서는 미토콘드리아 단백질 및 OXPHOS 단백질의 양도 같이 감소하였다. 한편, 본 연구에 사용된 총 5개의 위암 세포주 모두에서 mtDNA 양이 감소하였다 그러나 위암 세포주에서는 mtDNA 양적 감소와 미토콘드리아 단백질 및 OXPHOS 단백질의 양적 감소가 항상 일치하지는 않았다. 이러한 연구결과는 위암 조직 및 세포주에서 mtDNA 양의 감소가 흔하며 이는 mtDNA 양적 변화가 위암의 생성에 관여함을 제시한다.
