Research on the Quality Characteristics of Domestic Colostrum according to the Processing Methods Employed (국내산 초유의 가공방법에 따른 품질특성 연구)

  • The characteristics of Holstein colostrum according to the methods that were employed in processing it were analyzed in this study to improve its industrial utilization. Colostrum samples were collected from the dairy farm of the National Institute of Animal Science (NIAS). The milk fat, protein, lactose, and SNF contents of colostrum were 4.34, 6.99, 3.37, and 11.10%, respectively. The effects of spray drying, freeze drying, freezing, acidification, and inoculation of lactic-acid bacteria on the characteristics of colostrum were then compared. The freezing of colostrum was found to be proper for long-term storage in a farm. Freeze-dried colostrum powder could not meet the processing requirements and the component standards for animal products in terms of the total bacterial and coliform bacteria counts, but spray-dried colostrum powder could meet the microbiological requirements because of its bactericidal effect during the spray-dry treatment. The inoculation of lactic-acid bacteria showed a better inhibitory effect on coliform than the acidification treatment, but protein precipitation appeared because of the low pH and the high acidity. To estimate the effects of the processing methods employed on the IgG of colostrum, the IgG contents of the milk treated by long temperature long time (LTLT) ($65^{\circ}C$, 30 min), by inoculating the lactic acid bacteria starter, by spray drying, and by freeze drying were measured. The IgG contents of the colostrum were changed significantly by the processing treatment employed, from 53.98 mg/mLto 33.28, 34.82, 21.98, and 36.89 mg/mL, respectively.

Effect of Different Growing Stages of Winter Cereal Crops on the Quality of Silage Materials and Silages (맥류의 수확시기가 사일리지의 재료적 특성 및 품질에 미치는 영향)

  • This study was carried out to determine the effect of different growing stages of winter cereal crops on the quality of silage materials and silages. Silages were made from the silage materials harvested at four growing stages(boot, heading, flowering, and yellow ripe) of barley, rye, oat, and wheat. Approximately 1 kg of silage materials harvested from each growing stage stored in vinyl bags with vacuum packing method and fermented at room temperature for 40 days. As the growing stages progressed, the moisture and crude protein contents of the silage materials decreased, and fiber contents(NDF, ADF and hemicellulose) increased. All the silage materials showed significantly higher contents of water soluble carbohydrate in the boot stages than in the flowering and yellow ripe stages. There was no tendency in acetic acid contents of silage materials cut at different growing stages. The overall pH of silage materials were in the range of 5.91-6.01, and there was no significant difference among growing stages. Buffering capacity of silage materials were in the range of 26.23-29.47meq/100g DM, and showed a tendency to decline as the growing stages proceeded. The moisture and crude protein contents of silages decreased significantly in all species as the growing stages proceeded, and the fiber contents vice versa. As the growing stages proceeded, the pH of the silages tended to increase, and the acetic, butyric, and lactic acid contents tended to decrease. The buffering capacity of silages had a tendency to decrease as the growing stages of winter cereal crops proceeded. Therefore, these features described above should be taken into consideration in order to make silages from winter crops economically.

Component Analysis and Antioxidant Activity of Oenanthe javanica Extracts (돌미나리 추출물의 함유성분 분석과 항산화 활성)

  • The purpose of this study was to determine the possibility of using Oenanthe javanica as a natural health food source. To accomplish this, its general and biological activities were measured. Its carbohydrate, crude protein, crude lipid, and ash contents were 44.7, 9.8, 8.9, and 27.8%, respectively. The K content was largest for minerals followed by Ca, P, and Mg, which means that Oenanthe javanica is an alkali material. The concentrations of total phenol and flavonoids of OJE were $88.9{\pm}0.46$ mg GAE/g, and $28.6{\pm}0.64$ mg QE/g, respectively. Gallic acid, catechin, chlorogenic acid, and caffeic acid in OJE as measured by using HPLC were $0.9{\pm}0.23$, $1.2{\pm}0.19$, $227.1{\pm}0.62$, and $4.0{\pm}0.35$ mg/g. The DPPH and ABTS radical scavenging activities of OJE were 72.2%, and 66.1%, respectively, at $1,000{\mu}g/mL$. The FRAP and reducing power of OJE were 0.79, and 0.41 absorbance units value respectively, at $1,000{\mu}g/mL$. OJE possessed significant antioxidant properties, which suggests its great potential as a functional ingredient for food applications.

Outcome and Prognosis in Critically III Children Receiving Continuous Renal Replacement Therapy (소아 중환자에서 지속적 신대체요법의 치료 결과와 예후)

  • Purpose : Continuous renal replacement therapy(CRRT) has been the first choice for the treatment of acute renal failure in critically ill children not only in western countries but also in Korea. However, there are very few studies that have analyzed the outcome and prognosis of this modality in Korean children. We performed this study to evaluate the factors associated with the outcome and prognosis of patients treated with CRRT. Methods : We retrospectively reviewed the medical records of 32 children who had received CRRT at Severance hospital from 2003 to 2006. The mean age was 7.5 years(range 4 days-16 years) and the mean body weight was 25.8 kg (range 3.2-63 kg). Results : Eleven(34.4%) of the 32 patients survived. Bone marrow transplantation and malignancy were the most common causes of death and underlying disease leading to the need for CRRT Mean patient weight, age, duration of CRRT, number of organ failures, urine output, estimated glomerular filtration rate(eGFR), C-reactive protein, and blood urea level did not differ significantly between survivors and nonsurvivors. (1) Pediatric risk of mortality(PRISM) III score at CRRT initiation($9.8{\pm}5.3$ vs. $26.7{\pm}7.6$, P<0.0001), (2) maximum pressor number ($2.1{\pm}1.2$ vs. $3.0{\pm}1.0$, P=0.038), and (3) the degree of fluid overload($5.2{\pm}6.0$ vs. $15.0{\pm}8.9$, P=0.002) were significantly lower in survivers than in nonsurvivors. Multivariate analysis revealed that fluid overload was the only independent factor reducing survival rate. Conclusion : CRRT was successfully applied to the treatment of acute renal failure in a wide range of critically ill children. To improve survival, we suggest the early initiation of CRRT to prevent the systemic worsening and progression of fluid overload in critically ill children with acute renal failure.   • Background/Aims: It is well recognized that all aerobic cells have the protective mechanisms in order to minimize the tissue damage induced by various reactive oxygen species(ROS). Thioredoxin peroxidase(TPX) which has been recently identified and characterized functions to convert peroxide to water. The protein is also found in various subtypes(TPX-A & B, MER5, HS22 and HORF-06) and is known to be ubiquitous in most human cells. Especially, ischemic brain injuries, partial hepatectomy and radiation induced DNA damages. In treating lung cancer, radiation therapy has a major place in the local control and the relief of symptoms, but radiation induced free radical injury and resulting pulmonary fibrosis has been the major drawback of the therapy. However, little is known about the protective mechanisms and biologic modulations against radiation-induced tissue damages. Methods: Eighteen mice were divided into six groups, 3 in each group, and fifteen had received 900cGy of radiation. The mice were sacrificed according to the pre determined time schedule; immediate, 1, 2, 3 and 6 weeks after irradiation. Extracts were made from the lungs of each mice, Western blot analysis of various subtypes of TPX were done after SDS-P AGE. Examination of H & E stained slides from the same irradiated specimens and the control specimens were also performed. Results: No difference in the intensity of the immunoreactive bands in the irradiated lung samples of the mice compared to the unirradiated control was observed regardless of the time intervals, although H & E examination of the sample specimens demonstrated progressive fibrotic changes of the irradiated lung samples. Conclusion: In conclusion, according to our data, it is suggested that various thioredoxin peroxidase subtypes and catalase which are known to be increased in many repair processes may not be involved in the repair of the radiation injury to the lung and subsequent fibrosis.

Effect of Nitrogen Top Dressing Levels on Productivity, Feed Value, and Anthocyanin Content of Colored Barley (질소 추비량이 유색 보리의 생산성, 사료가치 및 안토시아닌 함량에 미치는 영향)

  • his experiment was carried out to evaluate the effects of nitrogen top dressing levels on the growth, feed value, and anthocyanin content for developing functional feed of colored barley. A colored barley cultivar, Boanchalbori, was tested in this experiment. Nitrogen top dressing levels was six (0, 20, 40, 60, 80, 100%) and top dressing time was a regeneration time. In case of productivity, heading data was get behind and dry matter rate was significant decreased with higher nitrogen top dressing levels (p<0.05). Fresh yield, dry matter yield and TDN yield was increased with higher nitrogen top dressing levels, but not significant. In case of feed value, crude protein content was significant increased with higher nitrogen top dressing levels (p<0.05), and higher in the order of spike, whole and leaf culme of the plant. Percent NDF and ADF was decreased with higher nitrogen top dressing levels in leaf culme, but no difference in spike and whole (p<0.05), and higher in the order of leaf culme, whole and spike of the plant. TDN was increased with higher nitrogen top dressing levels in leaf culme, but no difference in spike and whole (p<0.05), and higher in the order of spike, whole and leaf culme of the plant. Total anthocyanin content was significant decreased with higher nitrogen top dressing levels in leaf culme and whole (p<0.05), and higher in the order of leaf culme, whole and spike of the plant. Specially, cyanidin-3-glucoside (C3G), delphinidin (Del), malvidin-3-glucoside (M3G) and malvidin (Mal) show a significant decrease. So there are an accumulation of anthocyanin in the culm, and standard nitrogen top dressing levels dressing on the regeneration time for produces high anthocyanin content of the colored barley.

Quality Changes of a Fully Ripe Korean Native Pumpkin, Yangsan, during Long-term Storage, and High Temperature and Pressure Treatment (장기저장 및 고온고압 처리에 따른 한국재래종 호박 '양산'의 품질변화)

  • We have studied quality changes of fully ripe fruit of Korean native pumpkin 'Yangsan' regarding the following parameters: pH, sugar content, weight, water content, contents of crude protein and amino acids during 60 days storage at room temperature. As the results, there was no changes in sugar contents according to the storage period, but pH was changing to a little acidic direction with slight decrease in weight and water content. Contents of total crude proteins and comprising amino acids were increased during the storage period. The main contents of amino acids of the Korean native pumpkin, Yangsan, were glutamic acid (15.5%), aspartic acid (10.1%), lysine (8.7%), valine (7.5%), leucine (7.1%) and alanine (6.6%), which were not highly influenced during storage period. Additionally we have investigated the content of free amino acids and color changes during processing of Yangsan under high temperature at $121^{\circ}C$ and high pressure at $1\;kg/cm^2$. In fully ripe fruits, a total of 29 kinds of free amino acids were detected including 8 kinds of essential amino acids (histidine, isoleucine, leucine, lysine, phenylalanine, methionine, threonine and valine). More than 35% of total free amino acids were aspartic acid (20.3%) and asparagine (15.4%); ornithine, citrullin, and arginine, which are related to Ornithine cycle, were also detected in fully ripe fruits. But when treated with high temperature and high pressure, glutamic acid and arginine were decreased rapidly whereas ammnonium chloride was relatively increased. Moreover "b" value as yellow color indicator was decreased from 17.45 to 9.14 while treated for 60 minutes with high temperature and pressure, caused by the degradation of ${\beta}-carotene$ and other yellowish pigments in Yangsan.

  • This study was carried out to investigate the physicochemical compositions and antioxidative effects of Eriobotrya japonoca Lindl (Loquat). The proximate compositions of the loquat leaf on a dry matter basis were 8.78% moisture content, 6.74% crude protein, 7.87% crude fat, 6.99% crude ash, 43.61% dietary fiber and 26.01% carbohydrate. In analysis of free amino acids, 16 kinds total amino acid components, 17 kinds of components were isolated from loquat. The essential amino acids contained in loquat leaf accounted for 50.15% of total amino acids, while the non-essential amino acids accounted for 49.85%. In analysis of total fatty acids, only 5 kinds of acid were detected: lauric acid, myristic acid, pentadecanoic acid, stearic acid and oleic acid. The contents of vitamin A, vitamin E and vitamin C were 0.039 mg%, 0.096 mg% and 0.575 mg%, respectively. The mineral contents of loquat leaf were greater in order of Zn

  • 'Dakyeong' (Avena sativa L.), a winter oats for forage use, was developed by the breeding team at National Institute of Crop Science, RDA in 2016. It was derived from a cross between 'CI7505'(IT133304) and 'Swan'(IT197920). Subsequent generations followed by the cross were handled in bulk and pedigree selection programs at Iksan and Jeonju, respectively. After preliminary and advanced yield test for 2 years, 'SO2004009-B-B-10-8-3-9', designated as a line name of 'Gwiri91', were subsequently evaluated for earliness and forage yield during 3 years in four parts such as Jeju (upland), Yesan (upland), Iksan (upland), and Jeonju (paddy), from 2014 to 2016, and finally named as 'Dakyeong'. Cultivar 'Dakyong' has leaves of dark green color, thick diameter culm and long grain of brown color. Over 3 years, the heading date of 'Dakyeong' was about 5 days earlier than that of check cultivar 'Samhan' (April 30 and May 5, respectively), and their average forage dry matter yield harvested at milk-ripe stage was higher 12% ($15.7tone\;ha^{-1}$) than $14.0tone\;ha^{-1}$ of check cultivar. Cultivar 'Dakyeong' was lower than the check cultivar 'Samhan' in terms of the protein content (6.1% and 7.0%, respectively) and total digestible nutrients (62.1%, and 62.5%, respectively), while the TDN yield was more than the check ($7.79tone\;ha^{-1}$ and $7.64tone\;ha^{-1}$, respectively). Fall sowing cropping of 'Dakyeong' is recommended only in areas where average daily minimum mean temperatures in January are higher than $-6^{\circ}C$, and it should not be cultivated in mountain areas, where frost damage is likely to occur.

Superoxide Dismutase Gene Expression Induced by Lipopolysaccharide in Alveolar Macrophage of Rat (폐포대식세포에서 내독소 자극에 의한 Superoxide Dismutase 유전자발현의 조절 기전)

  • Background: In the pathogenesis of acute lung injury induced by lipopolysaccharide(LPS), oxygen radiclls are known to be involved in one part. Superoxide dismutase(SOD) protects oxygen radical-induced tissue damage by dismutating superoxide to hydrogen peroxide. In eukaryotic cells, two forms of SOD exist intracellularly as a cytosolic, dimeric copper/zinc-containing SOD(CuZnSOD) and a mitochondrial, tetrameric manganese-containing SOD(MnSOD). But there has been little information about SOD gene expression and its regulation in pulmonary alveolar macrophages(PAMs). The objective of this study is to evaluate the SOD gene expression induced by LPS and its regulation in PAMs of rat. Method: In Sprague-Dawley rats, PAMs obtained by broncholaveolar lavage were purified by adherence to plastic plate. To study the effect of LPS on the SOD gene expression of PAMs, they were stimulated with different doses of LPS($0.01{\mu}g/ml{\sim}10{\mu}g/ml$) and for different intervals(0, 2, 4, 8, 24hrs). Also for evaluating the level of SOD gene regulation actinomycin D(AD) or cycloheximide(CHX) were added respectively. To assess whether LPS altered SOD mRNA stability, the rate of mRNA decay was determined in control group and LPS-treated group. Total cellular RNA extraction by guanidinium thiocyanate/phenolfchlorofonn method and Northern blot analysis by using a $^{32}P$-labelled rat MnSOD and CuZnSOD cDNAs were performed. Results: The expression of mRNA in MnSOD increased dose-dependently, but not in CuZnSOD. MnSOD mRNA expression peaked at 8 hours after LPS treatment. Upregulation of MnSOD mRNA expression induced by LPS was suppressed by adding AD or CHX respectively. MnSOD mRNA stability was not altered by LPS. Conclusion: These findings show that PAMs of rat could be an important source of SOD in response to LPS, and suggest that their MnSOD mRNA expression may be regulated transcriptionally and require de novo protein synthesis without affecting mRNA stability.
