The present study was conducted to examine the effects of dietary protein (20, 22, 24%) with a constant protein-to-energy ratio on clenbuterol-induced performance in broilers. The protein-to-energy ratio was based on adequate level (22% protein, 3,100 kcal of energy). Female broiler chickens were used for a $3{\times}2$ factorial arrangement and fed diets with or without 1 ppm clenbuterol from 14- to 32-days of age. Feed efficiency improved with increasing dietary protein level, regardless of clenbuterol treatment. The dietary clenbuterol increased weights of breast and leg muscles (gastrocnemius and peroneus longus), and clenbuterol markedly reduced protein content of leg muscles in chickens fed the 20% protein diet, but did not in chickens fed the 22 and 24% protein diets. Feeding the 24% protein diet with clenbuterol improved the protien accretion (peroneus longus) by 8.4%. Clenbuterol decreased DNA content and increased the protein/DNA ratio in breast muscle regardless of dietary protein intake. Clenbuterol had no effect on RNA content in both breast and leg muscles. The present results demonstrated that various protein levels which retain the same protein-to-energy ratio in the diet markedly alter the protein accretion induced by ${\beta}$-agonist in broilers.
In this paper, we propose a probabilistic framework to predict the interaction probability of proteins. The notion of domain combination and domain combination pair is newly introduced and the prediction model in the framework takes domain combination pair as a basic unit of protein interactions to overcome the limitations of the conventional domain pair based prediction systems. The framework largely consists of prediction preparation and service stages. In the prediction preparation stage, two appearance pro-bability matrices, which hold information on appearance frequencies of domain combination pairs in the interacting and non-interacting sets of protein pairs, are constructed. Based on the appearance probability matrix, a probability equation is devised. The equation maps a protein pair to a real number in the range of 0 to 1. Two distributions of interacting and non-interacting set of protein pairs are obtained using the equation. In the prediction service stage, the interaction probability of a protein pair is predicted using the distributions and the equation. The validity of the prediction model is evaluated fur the interacting set of protein pairs in Yeast organism and artificially generated non-interacting set of protein pairs. When 80% of the set of interacting protein pairs in DIP database are used as foaming set of interacting protein pairs, very high sensitivity(86%) and specificity(56%) are achieved within our framework.
A feeding trial of four dietary protein levels (20, 30, 40, and 50%) and two lipid levels (7 and 14%) with a factorial design was conducted to determine the optimal dietary protein and lipid levels for juvenile Israeli carp Cyprinus carpio. Triplicate groups of fish (average body weight, $1.3{\pm}0.02g$) were fed the experimental diets for 9 weeks. Survival of fish was not affected by either dietary protein or dietary lipid level. Weight gain and feed efficiency increased as dietary protein levels increased up to 40 and 50%, respectively. Weight gain was higher in fish fed the high-lipid diets with 20 and 40% protein content. Feeding efficiency increased as the dietary lipid level increased for the 30, 40, and 50% protein diets. Daily feed intake decreased with increasing protein level and the minimum feed consumption was observed in fish fed the 50% protein diet with 14% lipid content. Moisture and lipid contents of the whole body were affected by both dietary protein and lipid levels. The crude lipid content of fish fed the 14% lipid diet was higher than that of fish fed the 7% lipid diet at each protein level. The results of this study indicate that a diet containing 40% protein with 14% lipid content is optimal for the growth and effective protein utilization of juvenile Israeli carp.

Here we report the molecular cloning of a LIM protein cDNA of the CRP (cysteine-rich protein) family from the mulberry longicorn beetle, Apriona, geramri. The A. germari LIM protein cDNA contains an open reading frame of 276 bp encoding 92 amino acid residues with a calculated molecular weight of approximately 10 kDa. The A. germari LIM protein contains the cysteine-rich consensus sequence of LIM domain and the glycine-rich consensus sequence observed in cysteine-rich protein family 1 (CRP1). The potential nuclear targeting signal is retained. The deduced amino acid sequence of the A. germari LIM protein cDNA showed 81 % identity to both Bombyx mori muscle LIM protein (Mlp) and Drosophila melanogaster Mlp60A and 77% to Epiblema scudderiana Mlp. Northern blot analysis showed that A. germari LIM protein is highly expressed in epidermis and muscle, and less strongly in midgut, but not in the fat body.
이 연구에서는 법랑모세포 분화과정에서 APin의 기능을 알아보고자 APin-protein microarray를 시행한 후 치아발생과 관련이 있는 MEF2, Aurora kinase A, BMPR-IB와 EF-hand calcium binding protein을 분석하여 다음과 같은 결과를 얻었다. 1 CMV-APin construct를 transfection하여 APin의 과발현을 유도한 경우에는 MEF2와 Aurora kinase A 둘 모두에서 발현이 현저히 감소한 반면에, APin의 발현억제를 유도한 경우에는 둘 모두 변화가 없었다. 2. APin의 과발현을 유도한 경우에는 BMPR-IB와 EF-hand calcium binding protein 모두에서 발현이 크게 증가한 반면, APin을 발현억제 시킨 경우에는 BMPR-IB는 변화가 없었고, EF-hand calcium binding protein은 현저히 감소하였다. 위의 결과들로 보아 APin 단백질은 MEF2, Aurora kinase A, BMPR-IB, EF-hand calcium binding protein과 상호작용하여 법랑모세포의 분화와 석회화 과정 중에 중요한 역할을 하는 것으로 사료된다.
Random sequencing of expressed sequence tags in roots of Chinese cabbage led to isolation of a partial cDNA clone, BR77, which encoded a putative protein kinase. Using the BR77 cDNA as a probe, we isolated a full-length cDNA encoding the Brassica campestris protein kinase 1 (Bcpk1). The Bcpt1 cDNA contained one open reading frame encoding a polypeptide of 439 amino acids. The putative polypeptide consisted of a short N-terminal region and a protein kinase catalytic domain. The catalytic domain of Bcpkl showed a high homology to cAMP- and calcium- phospholipid-dependent subfamilies of serine/threonine protein kineses. Eleven major catalytic domains in protein kineses were well conserved in Bcpk1. However, Bcpk1 contained a unique nonhomologous intervening sequence between subdomains VII and VIII, which was not found in protein kineses of animals and lower eukaryotes. Genomic DNA gel blot analysis showed that Bcpt1 genes might be present as three copies in the Chinese cabbage genome. These imply that Bcpk1 belongs to a plant-specific serine/threonine protein kinase subfamily.
목적: 본 연구에서는 콘택트렌즈 단백질제거제 개발 시 요구되는 단백질 제거효능의 적절한 평가를 위한 시험법을 확립하고 이를 실제 콘택트렌즈에 침착된 단백질의 제거효율을 측정한 결과와 비교, 분석하여 단백질제거제의 효능 평가법으로 제시하고자 하였다. 방법: 대한약전에 제시된 단백소화력 시험법을 이용하여 파파인, 판크레아틴, 섭틸리신 A 및 프로테아제와 각각의 효소가 포함된 단백질제거 정 또는 용액의 효능평가에 적절한 시험조건을 찾고자 하였다. 또한, balafilcon A 재질 렌즈에 인위적으로 침착시킨 단백질을 시판되고 있는 단백질제거 정 또는 용액으로 제거하여 세척효율을 확인하고 이들 방법의 상관관계를 분석하였다. 결과: 판크레아틴과 판크레아틴 함유 제품의 경우 단백소화력 시험법으로 평가하였을 때 둘 다 약전에서 제시하는 판크레아틴 단백소화력 기준값인 28 IU/mg를 충족하였다. 프로테아제와 삼염화아세트산 B 용액으로 실험한 섭틸리신 A의 경우는 단백소화력 시험법으로 평가하였을 때 제조사에서 제시한 효소활성 값을 충족하였으나, 파파인과 삼염화아세트산 A 용액으로 실험한 섭틸리신 A는 제조사에서 제시된 효소활성 값에 해당하는 단백소화력이 측정되지 않았다. 시판되는 단백질제거제의 경우 판크레아틴을 함유한 제품을 제외한 나머지 세 가지 제품은 단백소화력 시험법으로는 제조사에서 명시한 효소들의 단백질 효소활성 값을 확인할 수 없었다. 그러나 실제 렌즈에 침착된 단백질의 제거정도를 측정하였을 때에는 파파인을 함유한 제품을 제외한 3종의 단백질제거제는 모두 90%가 넘는 단백질 제거효율을 보였다. 파파인 함유 단백질제거제의 경우 단백소화력 시험법으로는 효능 측정이 불가능하였으나 실제 렌즈에 침착된 누액단백질 제거효율은 73.72%에 이르렀다. 결론: 본 연구결과로 콘택트렌즈 단백질제거제의 효능은 함유되어 있는 단백분해효소의 종류에 따라 시험법을 달리하여 평가되어야 함을 확인할 수 있었다. 즉, 판크레아틴, 프로테아제, 섭틸리신 A를 함유하는 단백질제거제는 단백소화력 시험법과 단백질제거효율 측정법으로 효능평가가 가능하고, 파파인을 함유하는 단백질제거제의 평가는 콘택트렌즈를 이용한 단백질제거효율 측정법만이 효율적임을 제안할 수 있다.
In E. coli, chromosomal DNA associated with proteins is condensed into an organized structure known as nucleoid. Using a nitrocellulose filter binding assay to identify proteins forming nucleoid, a 21 kDa protein was purified from E. coli. The molecular weight of the purified protein was 21 kDa on SDS-polyactylamide gel electrophoresis and 24 kDa on gel permeation chromatography. A molecular weight of 21 kDa on SDS-polyacrylamide gel electrophoresis is unique among known proteins which are believed to be involved in the formation of nucleoid in E. coli. The 21 kDa protein nonspecifically binds to both double-stranded and single-stranded DNA. Sedimentation in a sucrose gradient revealed that the protein induced significant condensation of both supercoiled plasmid DNA and linear bacteriophage $\lambda$ DNA On the basis of quantitative Western-blot analysis, approximately 40,000 molecules of the protein were estimated to exist in an E. coli. The biochemical properties and cellular abundance of the 21 kDa protein suggest that this protein participates in the formation of nucleoid in E. coli.
Muscle protein synthesis in vitro was measured in chicks fed low-protein(10% CP) and control(20% CP) diets. Right leg muscles (M. gastrocnemius) were mounted on a support made of stainless steel to stretch in constant tension, whereas left leg muscles were unmounted. Both leg muscles were incubated in Dulbecco's modified Eagle's medium including L-[$4-^3H$] phenylalanine for 60 min to measure in vitro protein synthesis. There was no significant difference in fractional synthesis rate(FSR) of muscle protein between both dietary protein levels, whereas FSR with stretch in constant tension was significantly higher than that without constant tension due to an increase in the absolute synthesis rate(ASR) per unit RNA(the efficiency of RNA to synthesize protein). The ASR of muscle protein in chicks fed the control diet was significantly higher than that in the low-protein diet group.
Adaptation of infections and hosts has resulted in several metabolic mechanisms adopted by intracellular pathogens to combat the defense responses and the lack of fuel during infection. Human tuberculosis caused by Mycobacterium tuberculosis (MTB) is the world's first cause of mortality tied to a single disease. This study aims to characterize and anticipate potential antigen characteristics for promising vaccine candidates for the hypothetical protein of MTB through computational strategies. The protein is associated with the catalyzation of dithiol oxidation and/or disulfide reduction because of the protein's anticipated disulfide oxidoreductase properties. This investigation analyzed the protein's physicochemical characteristics, protein-protein interactions, subcellular locations, anticipated active sites, secondary and tertiary structures, allergenicity, antigenicity, and toxicity properties. The protein has significant active amino acid residues with no allergenicity, elevated antigenicity, and no toxicity.
